cDNA cloning,sequence analysis,and recombinant expression of akitonin beta,a C-type lectin-like protein from Agkistrodon acutus

AIM: To clone the cDNA of a new member of snake venom C-type lectin-like proteins, to study its structure- function relationships and to achieve its recombinant production. METHODS: PCR primers were designed based on the homology and cDNA was amplified by RT-PCR using total RNA from snake venom gland as the template. The PCR products were cloned into the plasmid pGEM-T and sequenced. The deduced protein sequence was analyzed with some bioinformatic programs. A recombinant expression plasmid was constructed using pBAD- TOPO as vector and transformed into *** TOP10 competent cells. RESULTS: A novel cDNA sequence encod- ing akitonin β was found and accepted by GenBank (accession number AF387100). Akitonin β consists of a typical carbohydrate recognition domain (CRD) of C-type lectins, and it is homologous with other snake venom C-type lectin-like proteins. It was predicted to be a platelet antagonist. Upon induction with arabinose rAkitonin β express- ing in E coli was achieved at a high level (superior to 150 mg/L). The recombinant fusion protein exhibited inhibitory activities on rat platelet aggregation in vitro. CONCLUSION: A new member of snake venom C-type lectin-like proteins was discovered and characterized, and an efficient recombinant expression system was estab- lished for its production.

Alternative relay regulates the adenosine triphosphatase activity of Locusta migratoria striated muscle myosin

Locust(Locusta migratoria)has a single striated muscle myosin heavy chain(Mhc)gene,which contains 5 clusters of alternative exclusive exons and 1 differently included penultimate *** alternative exons of Mhc gene encode 4 distinct regions in the myosin motor domain,that is,the N-terminal SH3-like domain,one lip of the nucleotide-binding pocket,the relay,and the ***,we investigated the role of the alternative regions on the motor function of locust muscle *** Sf9-baculovirus protein expression system,we expressed and purified 5 isoforms of the locust muscle myosin heavy meromyosin(HMM),including the major isoform in the thorax dorsal longitudinal flight muscle(FL1)and 4 isoforms expressed in the abdominal intersegmental muscle(AB1 to AB4).Among these 5 HMMs,FL1-HMM displayed the highest level of actin-activated adenosine triphosphatase(ATPase)activity(hereafter referred as ATPase activity).To identify the alternative region(s)responsible for the elevated ATPase activity of FL1-HMM,we produced a number of chimeras of FL1-HMM and *** with the relay of AB4-HMM(encoded by exon-14c)substantially decreased the ATPase activity of FL1-HMM,and conversely,the relay of FL1-HMM(encoded by exon-14a)enhanced the ATPase activity of *** showed that the exon-14a-encoded residues Gly474 and Asn509 are responsible for the elevated ATPase activity of *** results indicate that the alternative relay encoded by exon-14a/c play a key role in regulating the ATPase activity of FL1-HMM and AB4-HMM.

Development and performance evaluation of high temperature resistant strong adsorption rigid blocking agent

As drilling wells continue to move into deep ultra-deep layers, the requirements for temperature resistance of drilling fluid treatments are getting higher and higher. Among them, blocking agent, as one of the key treatment agents, has also become a hot spot of research. In this study, a high temperature resistant strong adsorption rigid blocking agent(QW-1) was prepared using KH570 modified silica,acrylamide(AM) and allyltrimethylammonium chloride(TMAAC). QW-1 has good thermal stability,average particle size of 1.46 μm, water contact angle of 10.5., has a strong hydrophilicity, can be well dispersed in water. The experimental results showed that when 2.wt% QW-1 was added to recipe A(4 wt% bentonite slurry+0.5 wt% DSP-1(filtration loss depressant)), the API filtration loss decreased from 7.8to 6.4 m L. After aging at 2.0.C, the API loss of filtration was reduced from 2. to 14 m L, which has certain performance of high temperature loss of filtration. At the same time, it is effective in sealing 80-100mesh and 100-12. mesh sand beds as well as 3 and 5 μm ceramic sand discs. Under the same conditions,the blocking performance was superior to silica(5 μm) and calcium carbonate(2.6 μm). In addition, the mechanism of action of QW-1 was further investigated. The results show that QW-1 with amide and quaternary ammonium groups on the molecular chain can be adsorbed onto the surface of clay particles through hydrogen bonding and electrostatic interaction to form a dense blocking layer, thus preventing further intrusion of drilling fluid into the formation.

CCaP1/CCaP2.CCaP3 interact with plasma membrane H^(+)-ATPases and promote thermoresponsive growth by regulating cell wall modification in Arabidopsis

Arabidopsis plants adapt to warm temperatures by promoting hypocotyl growth primarily through the basic helix-loop-helix transcription factor PIF4 and its downstream genes involved in auxin responses,which enhance cell *** the current study,we discovered that cell wall-related calcium-binding protein 2.CCaP2.and its paralogs CCaP1 and CCaP3 function as positive regulators of thermo-responsive hypocotyl growth by promoting cell elongation in ***,mutations in CCaP1/CCaP2.CCaP3 do not affect the expression of PIF4-regulated classic downstream ***,they do noticeably reduce the expression of xyloglucan endotransglucosylase/hydrolase genes,which are involved in cell wall *** also found that CCaP1/CCaP2.CCaP3 are predominantly localized to the plasma membrane,where they interact with the plasma membrane H^(+)-ATPases AHA1/***,we observed that vanadate-sensitive H^(+)-ATPase activity and cell wall pectin and hemicellulose contents are significantly increased in wild-type plants grown at warm temperatures compared with those grown at normal growth temperatures,but these changes are not evident in the ccap1-1 ccap2.1 ccap3-1 triple ***,our findings demonstrate that CCaP1/CCaP2.CCaP3 play an important role in controlling thermo-responsive hypocotyl growth and provide new insights into the alternative pathway regulating hypocotyl growth at warm temperatures through cell wall modification mediated by CCaP1/CCaP2.CCaP3.

Surviving winter on the Qinghai-Xizang Plateau:Extensive reversible protein phosphorylation plays a dominant role in regulating hypometabolism in hibernating Nanorana parkeri

Changes in protein abundance and reversible protein phosphorylation(RPP)play important roles in regulating hypometabolism but have never been documented in overwintering frogs at high *** test the hypothesis that protein abundance and phosphorylation change in response to winter hibernation,we conducted a comprehensive and quantitative proteomic and phosphoproteomic analysis of the liver of the Xizang plateau frog,Nanorana parkeri,living on the Qinghai-Xizang *** total,5170 proteins and 5695 phosphorylation sites in 1938 proteins were *** on proteomic analysis,674 differentially expressed proteins(438 up-regulated,2.6 down-regulated)were screened in hibernating *** versus summer *** enrichment analysis revealed that higher expressed proteins in winter were significantly enriched in immune-related signaling pathways,whereas lower expressed proteins were mainly involved in metabolic processes.A total of 42.1 modified sites(4147 up-regulated,104 down-regulated)belonging to 1638 phosphoproteins(1555 up-regulated,83 down-regulated)were significantly changed in the *** hibernation,RPP regulated a diverse array of proteins involved in multiple functions,including metabolic enzymatic activity,ion transport,protein turnover,signal transduction,and alternative *** changes contribute to enhancing protection,suppressing energy-consuming processes,and inducing metabolic ***,the activities of phosphofructokinase,glutamate dehydrogenase,and ATPase were all significantly lower in winter compared to *** conclusion,our results support the hypothesis and demonstrate the importance of RPP as a regulatory mechanism when animals transition into a hypometabolic state.

Precise point positioning and its application in mining deformation monitoring

Precise point positioning (PPP)-based deformation monitoring scheme is presented for the use in mining deformation monitoring. Within the solutions of daily observation, outliers are detected and removed to avoid any potential misinterpretation of the results and then the deformation can be extracted by the coordinate differences between the two consecutive solutions. Meanwhile, because of the special location of a rover station in mining areas, the satellite geometry may be insufficient for a reasonable PPP solution, and the multipath impact an also be significant. Therefore, it is necessary to predict the satellite geometry before any daily observation. To evaluate the ability of extracting the deformation using the PPP-based method, various quality measures were introduced. The results of three datasets of the same station show that the precision of deformation monitored by PPP can reach up to cm level and even mm level.

TLR3 Ligand-Induced Accumulation of Activated Splenic Natural Killer Cells into Liver

It has been revealed that poly I：C is a potent stimulator for NK cells, which can induce NK cell rapid activation and preferential accumulation into liver. However, the process mediating the influx of NK cells remains obscure. In this study, we found that poly I：C administration increased the portion and absolute number of NK cells in liver, but largely decreased those in spleen. There were no obvious changes of these lymphocytes in other immune organs. The results from splenic adoptive transfer and splenectomy showed that the recruited spleen NK cells contributed to the accumulation of NK cells in liver, and this process was regulated by the production of chemokines and the presence of T cells.
