TLR2 and TLR4 signaling pathways are required for recombinant Brucella abortus BCSP31-induced cytokine production, functional upregulation of mouse macrophages, and the Th1 immune response in vivo and in vitro

Brucella abortus is a zoonotic Gram-negative pathogen that causes brucelosis in ruminants and humans. Toll-like receptors （TLRs） recognize Brucella abortus and initiate antigen-presenting cell activities that affect both innate and adaptive immunity. In this study, we focused on recombinant Brucella cell-surface protein 31 （rBCSP31） to determine its effects on mouse macrophages. Our results demonstrated that rBCSP31 induced TNF-α, IL-6 and IL-12p40 production, which depended on the activation of mitogen-activated protein kinases （MAPKs） by stimulating the rapid phosphorylation of p38 and JNK and the activation of transcription factor NF-KB in macrophages. In addition, continuous exposure （〉24 h） of RAW264.7 cells to rBCSP31 significantly enhanced I FN-y-induced expression of MHC-II and the ability to present rBCSP31 peptide to CD4＋ T cells. Furthermore, we found that rBCSP31 could interact with both TLR2 and TLR4. The rBCSP31-induced cytokine production by macrophages from TLR2-/- and TLR4-/- mice was lower than that from C57BL/6 macrophages, and the activation of NF-KB and MAPKs was attenuated in macrophages from TLR2-/- and TLR4-/- mice. In addition, CD4＋ T cells from C57BL/6 mice immunized with rBCSP31 produced higher levels of IFN-y and IL-2 compared with CD4＋ T cells from TLR2-/- and TLR4-/- mice. Macrophages from immunized C57BL/6 mice produced higher levels of IL-12p40 than those from TLR2-/- and TLR4-/- mice. Furthermore, immunization with rBCSP31 provided better protection in C57BL/6 mice than in TLR2-/- and TLR4-/- mice after B. abortus 2308 challenge. These results indicate that rBCSP31 is a TLR2 and TLR4 agonist that induces cytokine production, upregulates macrophage function and induces the Thl immune response.

Security Control for Uncertain Networked Control Systems under DoS Attacks and Fading Channels

This paper characterizes the joint effects of plant uncertainty,Denial-of-Service(DoS)attacks,and fading channel on the stabilization problem of networked control systems(NCSs).It is assumed that the controller remotely controls the plant and the control input is transmitted over a fading channel.Meanwhile,considering the sustained attack cycle and frequency of DoS attacks are random,the packet-loss caused by DoS attacks is modelled by a Markov process.The sampled-data NCS is transformed into a stochastic form with Markov jump and uncertain parameter.Then,based on Lyapunov functional method,linear matrix inequality(LMI)-based sufficient conditions are presented to ensure the stability of uncertain NCSs.The main contribution of this article lies in the construction of NCSs based on DoS attacks into Markov jump system(MJS)and the joint consideration of fading channel and plant uncertainty.

Computational Study on the Boundary Layer Suction Effects in the Supersonic Compressor Flow with a Bowed Blade

Flow control methodologies have been widely used to eliminating flow separation and increasing the blade load in axial compressor.Aiming at revealing the flow mechanism of coupled bowed blading and boundary layer suction in a supersonic compressor cascade,a cascade with a diffusion coefficient of 0.62 is numerically presented.First of all,according to the available experimental data,the numerical method was validated;then,different bowed blading effects on flow field in detail were investigated;at last,based on the flow physics of purely bowed blading,the positively bowed blade was coupled with boundary layer suction on blade suction surface,whereas the negatively bowed blade was coupled with endwall suction.For coupled control method,influence mechanism on flow field,especially on the shock structure was revealed,and different aspect ratios of coupled control method were investigated as well.Results showed that the coupled positively bowed blading and suction surface suction can eliminate the flow separation effectively.Compared with that of baseline supersonic cascade,the total pressure loss coefficient of the coupled scheme was reduced by 37.4%at most.At mid-span,the shock moved downstream and the single shock was separated to a dual-shock structure since the positively bowed blading reduced the static pressure of mid-span.The coupled negatively bowed blading and endwall suction also effectively enhanced the performance of cascade by removing the corner separation,with the loss coefficient reduced by as much as 41.9%.However,the suction coefficient of optimal coupled negatively bowed blading scheme reached 10.5%,which is too high for practical use.After coupled control,the 3 D shock structure became“C”shaped distribution along spanwise because of the difference in influence mechanism of negatively bowed blading on different spanwise location.Due to the opposite influence effect of positively and negatively bowed blading,the shock structure in the two different schemes of cascades were different and showed oppo

Prediction of undeformed chip thickness distribution and surface roughness in ultrasonic vibration grinding of inner hole of bearings

Ultrasonic vibration grinding differs from traditional grinding in terms of its material removal mechanism.The randomness of grain-workpiece interaction in ultrasonic vibration grinding can produce variable chips and impact the surface roughness of workpiece.However,previous studies used iterative method to calculate the unformed chip thickness(UCT),which has low computational efficiency.In this study,a symbolic difference method is proposed to calculate the UCT.The UCT distributions are obtained to describe the stochastic interaction characteristics of ultrasonic grinding process.Meanwhile,the UCT distribution characteristics under different machining parameters are analyzed.Then,a surface roughness prediction model is established based on the UCT distribution.Finally,the correctness of the model is verified by experiments.This study provides a quick and accurate method for predicting surface roughness in longitudinal ultrasonic vibration grinding.

With the deepening of China’s health-care reform,an integrated delivery system has gradually emerged with the function of improving the efficiency of the health-care delivery system.For China’s integrated delivery system,a medical consortium plays an important role in integrating public hospitals and primary care facilities.The first medical consortium policy issued after the COVID-19 pandemic apparently placed hope on accelerating the implementation of a medical consortium and tiered health-care delivery system.This paper illustrates the possible future pathway of China’s medical consortium through retrospection of the 10-year process,changes of the series of policies,and characteristics of the policy issued in 2020.We considered that a fully integrated medical consortium would be a major phenomenon in China's medical industry,which would lead to the formation of a dualistic care pattern in China.

Event-based sliding mode control under denial-of-service attacks

This paper deals with the problem of estimator-based sliding mode control against denial-of-service(DoS) attacks and discrete events via a time-delay approach. A networked system is considered an uncertain dynamical system with matched and mismatched perturbations and exogenous disturbance in the network environment. A network-resource-aware event-triggering mechanism is designed with aperiodically releasing system measurements. Furthermore, to describe the DoS attack duration and inter-event time, a time-delay modeling approach considers the DOS attack duration and inter-event time as a “time delay” of the measurements between the sensor and controller over the network is proposed. Consequently, an intervaltime-delay system with uncertainties is formulated. A state-observer-based sliding mode controller, by which the ideal sliding mode can be achieved, is proposed against the DoS attacks. The resulting sliding motion is proved to be robust and stable with an Lgain performance. Finally, the effectiveness and applicability of the present sliding mode control are validated in a simulated pendulum system.

网络文学的“爽文学观”是在消费主义语境中生成,并发展为当代消费社会的快感模式在新媒介环境下的延伸和弥补。基于消费主义所蕴含的时代语境,爽文学观生成了主体膨胀、技术化上升和伦理淡漠这一核心语法。由于爽文书写的内在不足,网络文学从形成对于现实的表征效力、树立写作主体的情动模式以及重构基于主体膨胀的语法等视角出发,形成了广泛的“反爽文”书写样态。“反爽文”美学的兴起源自对消费主义意识形态的悖反,体现了网络文学商业性和审美性的分化与平衡。
