Species identification of poisonous medicinal plant using DNA barcoding

The aim is to select a universal DNA barcode for identifying all poisonous medicinal plants in Chinese pharmacopoeia and their poisonous related species or adulterants. We chose 4 commonly used regions as candidate DNA barcodes(ITS2, psb A-trn H,matK and rbc L) and compared their identification efficiency in 106 species from 27 families and 65 genera totally. Data analysis was performed including the information of sequence alignment, inter/intra-specific genetic distance and data distribution, identification efficiency and the situation of Neighbor-Joining(NJ) phylogenetic trees. We found ITS2 sequence region had high variation, stable genetic distance and identification efficiency relatively. The topological structure of NJ phylogenetic tree showed monophyletic. Our findings show that ITS2 can be applied as a universal barcode for identifying poisonous medicinal plants in Chinese pharmacopoeia and their poisonous related species or adulterants.

The soft X-ray polarimeter(SXP)is a detector with a wide energy range,large area,and large field of view.A SXP will be mounted on the Chinese Space Station and will mainly focus on detecting the polarization of transient soft X-ray(2–10 keV)sources,especially gamma-ray bursts(GRBs).In this work,a polarimeter detector unit is taken as an example,and Geant4 and Garfield++software are used to simulate the detection efficiency and track *** improved track reconstruction algorithm is proposed and used to reconstruct two-dimensional images of the *** this method,the initial emission angle of photoelectrons is reconstructed from the initial part of the track by shortening or extending the initial part of the track until the remaining track is straight,and the number of pixels is within an adjustable *** modulation factor of the photoelectronic tracks after reconstruction reaches approximately 57%in the photon energy range of 7–10 keV.

The paper is concerned with the micro-flow self-sensing actuators,the work of which is based on the secondary piezoelectric effect. The piezoelectric ceramic stack can yield micro-displacement due to its first inverse piezoelectric effect. Therefore,we apply this micro-displacement to cell micro-flow injection. Moreover, due to the charge of the secondary direct piezoelectric effect,the piezoelectric ceramic stack is able to detect the force and displacement in the injection by itself. The experiments of first inverse piezoelectric effect and secondary direct piezoelectric effect are conducted. The experiment results show that,subjected to 0- 60 V input,the piezoelectric ceramic stack can generate 13. 45 μm displacement,and control accuracy can achieve 2 nm. It can completely meet the needs of cell micro-flow injection. Also,the experiments demonstrate that the micro-displacement due to the first inverse piezoelectric effect can be well self-sensed by the electric charge due to the secondary direct piezoelectric effect.

The developments of tough hydrogels in recent years have greatly expanded the applications of hydrogels as structural materials. However, most of the tough hydrogels are made of synthetic polymers. To develop biopolymer-based tough hydrogels has both fundamental and practical significances. Here we report a series of polysaccharides-based tough hydrogel films prepared by polyion complexation and solvent evaporation of chondroitin sulfate（CS） and protonated chitosan（CHT） solutions with different weight ratios. The obtained CS/CHT gel films with thickness of 40-80 μm and water content of 66 wt%-81 wt% possess excellent mechanical properties, with tensile breaking stress and breaking strain being 0.4-3 MPa and 160%-320%, respectively. We found that in the mixture solutions there are large amounts of excess CHT in terms of charges; after swelling the films in water, the acetic acid, which is used to protonate the amino groups of CHT, diffuses out of the gel matrix, enhancing the intermolecular interactions between CHT molecules and thus improving the mechanical properties of gel films, besides the ionic bonds between CS and CHT. Antimicrobial tests also showed that the gel films with low weight ratio of CS to CHT, corresponding to the case with excess CHT, have evident antimicrobial effect. These CS/CHT gel films with good mechanical properties and antimicrobial effect should extend the applications of hydrogels in biomedical fields.

Objective:To investigate the expression and significance of MMP-3 in synovium of knee joint at different stage in osteoarthritis(OA)***:Knee synovial tissue were collected in90 OA patients(the OA group).Patients in the OA group was divided into 3 subgroups:gradeⅠsubgroup(n=30),gradeⅡsubgroup(n=30),gradeⅢsubgroup(n=30).Thirty patients served as control group,lmmunohistochemical assay was used to detect the expression of MMP-3 protein in the knee synovial ***:MMP-3 protein was detected in all knee synovial *** expression of MMP-3 protein in the OA group was significantly higher that in the normal synovium(P<0.05),and the MMP-3 protein was mainly located in the *** was significant difference in the expression of MMP-3 protein between the gradeⅢsubgroup and the gradeⅠ,gradeⅡsubgroups(all P<0.05).The expression of MMP-3 protein was positively related to the severity of OA(r=0.912,P<0.05).Conclusions:The expression of MMP-3 protein are closely related to pathogenic mechanism of *** may be an important indicator of early diagnosis and the activity of the disease of osteoarthritis.
