Effect of Tanshinone IIA on High Glucose-induced Apoptosis and Oxidative Stress Damage in Vascular Endothelial Cells

Objective: The primary cause of microvascular disease in diabetic complications is long-term hyperglycemia, wherein the damage and apoptosis of vascular endothelial cells play a significant role. Sodium tanshinone IIA sulfonate(STS) has been found to have beneficial effects on cardiovascular health. This study aimed to investigate the impact of STS on high glucose-induced apoptosis and oxidative stress damage in vascular endothelial cells, as well as its potential protective mechanisms. Methods:Human umbilical vein endothelial cells(HUVECs) were divided into five groups:low-glucose group, high-glucose group, and three STS groups(STS-a, STS-b, and STS-c). The low-glucose group was incubated with DMEM low-sugar medium containing 5.5 mmol·L-1glucose, while the high-glucose group was treated with 33.3mmol·L-1glucose. The STS groups were exposed to 10, 30, and 50 μg·m L-1of STS,respectively. Each group was cultured for 72 h, and the MTT method was utilized to assess cell proliferation. Additionally, flow cytometry was employed to monitor changes in cell apoptosis and cellular oxidative stress indicators at 24, 48, and 72 h of cell culture in each group. Results: As time went on, the cell proliferation ability and apoptosis rate of each group gradually increased. The high-glucose group exhibited lower proliferation ability compared to the other groups. The STS-c group demonstrated the highest OD value for proliferation ability(24 h: 1.19±0.12; 48 h: 1.20±0.13; 72 h:1.25 ±0.12), but it was still lower than that of the low-sugar group. Notably, the high-glucose group had the highest cell apoptosis rate, while the low-glucose group had the lowest. The apoptosis rate of the STS-c group(24 h: 8.02±0.13; 48 h: 10.10±0.12;72 h: 13.18 ±0.11)% was between that of the low-glucose group and the high-glucose group, and lower than the STS-a and STS-b groups. Recent advances in electrocatalytic generation of indole-derived radical cations and their applications in organic synthesis

Indole-derived radical cations, open-shell reactive species, display distinctive dual reactivity due to the carbon-centered radical and more electrophilic carbocation, which frequently appear in a variety of single electron oxidation reactions for synthesizing structurally diverse functionalized indoles and indolines. Electrocatalysis is considered as a synthetically attractive and environmentally friendly alternative for driving the single electron oxidation of indoles. Remarkable achievements in electrocatalytic indolederived radical cation-mediated indole functionalization have been realized so far. This review comprehensively summarizes the recent progresses in the applications of electrocatalytic indole radical cations,including C(sp~2)–H functionalization, dearomative 2,3-difunctionalization, and ring-opening reaction, emphasizing the vital single electron oxidation steps of indoles, the substrates scope and limitations, and the reaction mechanisms.

A Study on Idea Adoption Prediction Model Based on SMOTE-AdaBoost: Taking Salesforce Platform as an Example

In the context of information overload, companies often struggle to effectively identify valuable ideas on their open innovation platforms. In this article, we propose an idea adoption strategy based on machine learning. We used data from a well-known open innovation platform, Salesforce, and extracted characteristic variables using the Information Adoption Model. Four classification models were then constructed based on AdaBoost, Random Forest, SVM and Logistic Regression models. Due to significant differences in the number of positive and negative samples in the OIP, we used the SMOTE method to address the problem of data imbalance. The results of the study showed that the ensemble learning models were more accurate in identifying valuable ideas than the individual machine learning models. When comparing the two ensemble learning models, AdaBoost outperformed Random Forest in predicting both positive and negative class samples. The SMOTE-AdaBoost model achieved a recall of 0.93, a precision of 0.92 and an impressive AUC of 0.98 in identifying adopted ideas, which could well identify valuable ideas and has implications for improving the efficiency and quality of idea adoption in OIP. The shortcoming of this work is that it only investigated a single platform. In the future, we will consider extending this method to different platforms and multiple classification problems.

Acute pancreatitis(AP)is a potentially fatal condition with no targeted treatment *** inhibiting xanthine oxidase(XO)in the treatment of AP has been studied in several experimental models and clinical trials,whether XO is a target of AP and what its the main mechanism of action is remains ***,we aimed to re-evaluate whether XO is a target aggravating AP other than merely generating reactive oxygen species that trigger *** first revealed that XO expression and enzyme activity were significantly elevated in the serum and pancreas of necrotizing AP *** also found that allopurinol and febuxostat,as purine-like and non-purine XO inhibitors,respectively,exhibited protective effects against pancreatic acinar cell death in vitro and pancreatic damage in vivo at different doses and treatment time ***,we observed that conditional Xdh overexpression aggravated pancreatic necrosis and *** mechanism analysis showed that XO inhibition restored the hypoxia-inducible factor 1-alpha(HIF-1α)-regulated lactate dehydrogenase A(LDHA)and NOD-like receptor family pyrin domain containing 3(NLRP3)signaling pathways and reduced the enrichment of^(13)C_(6)-glucose to^(13)C_(3)-***,we observed that clinical circulatory XO activity was significantly elevated in severe cases and correlated with C-reactive protein levels,while pancreatic XO and urate were also increased in severe AP *** results together indicated that proper inhibition of XO might be a promising therapeutic strategy for alleviating pancreatic necrosis and preventing progression of severe AP by downregulating HIF-1α-mediated LDHA and NLRP3 signaling pathways.

WeldNet:A voxel-based deep learning network for point cloud annular weld seam detection

Weld seam detection is an important part of automated *** present,few studies have been conducted on annular weld seams,and a lot of defects exist in the point cloud model of the tube sheet obtained by RGB-D cameras and photography *** at the above problems,this paper proposed an annular weld seam detection network named WeldNet where a voxel feature encoding layer was adaptively improved for annular weld seams,the sparse convolutional network and region proposal network(RPN)were used to detect annular weld seam position,and an annular weld seam detection loss function was ***,an annular weld seam dataset was established to train the *** with the random sampling consistency(RANSAC)method,WeldNet has a higher detection accuracy,as well as a higher detection success rate which has increased by 23%.Compared with U-Net,WeldNet has been proven to achieve a better detection result,and the intersection over the union of the weld seam detection is improved by 17.8%.
