Comparison of the Digestion and Absorption Characteristics of Docosahexaenoic Acid-Acylated Astaxanthin Monoester and Diester in Mice

Docosahexaenoic acid-acylated astaxanthin(DHA-AST)esters exhibit distinct bioactivities in improving brain ***,the digestion and absorption characteristics of DHA-AST esters in vivo are unclear,thereby restricting the molecular mechanism analysis of their superior *** study compared the digestion and absorption characteristics of DHA-AST monoester and diester by determining the levels of AST and DHA in the serum,liver,small intestinal content and wall,and feces at different time points after a single-dose oral administration of the *** oral gavage with 2 mg AST equivalent of the DHA-AST monoester and diester for 18 h,the excretion rates were approximately 51%and 84%,*** result indicates that DHA-AST monoester was better than diester for the absorption of *** results in serum,liver,and small intestinal content and wall also agreed with this ***,the excretion rates of DHA in the feces at 24 h in the DHA-AST monoester and diester groups were approximately 40%and 36%after gavage with 5 mg DHA equivalent,*** result indicates that DHA-AST diester exhibited a better tendency than monoester for the absorption of ***,the results in the liver and small intestinal wall showed an apparent difference,indicating that DHA-AST diester was better than monoester for the absorption of *** findings provide a scientific basis for the molecular mechanism analysis and utilization of DHA-AST monoester and diester as functional ingredients.

Identification of Fucans from Four Species of Sea Cucumber by High Temperature ~1H NMR

Acidic polysaccharide, which has various biological activities, is one of the most important components of sea cucumber. In the present study, crude polysaccharide was extracted from four species of sea cucumber from three different geographical zones, Pearsonothuria graeffei(Pg) from Indo-Pacific, Holothuria vagabunda(Hv) from Norwegian Coast, Stichopus tremulu(St) from Western Indian Ocean, and Isostichopus badionotu(Ib) from Western Atlantic. The polysaccharide extract was separated and purified with a cellulose DEAE anion-exchange column to obtain corresponding sea cucumber fucans(SC-Fucs). The chemical property of these SC-Fucs, including molecular weight, monosaccharide composition and sulfate content, was determined. Their structure was compared simply with fourier infrared spectrum analyzer and identified with high temperature 1H nuclear magnetic resonance spectrum analyzer(NMR) and room temperature 13 C NMR. The results indicated that Fuc-Pg obtained from the torrid zone mainly contained 2,4-O-disulfated and non-sulfated fucose residue, whereas Fuc-Ib from the temperate zone contained non-, 2-O- and 2,4-O-disulfated fucose residue; Fuc-St from the frigid zone and Fuc-Hv from the torrid zone contained mainly non-sulfated fucose residue. The proton of SC-Fucs was better resolved via high temperature 1H NMR than via room temperature 1H NMR. The fingerprint of sea cucumber in different sea regions was established based on the index of anomer hydrogen signal in SC-Fucs. Further work will help to understand whether there exists a close relationship between the geographical area of sea cucumber and the sulfation pattern of SC-Fucs.

Dietary Supplementation with Sea Cucumber Saponins and Exercise Can Significantly Suppress Adipose Accumulation in Mice Fed with High-Fat Diet

Dietary supplementation with sea cucumber saponins(SCS)and exercise have been confirmed to be effective in pre-venting the development of obesity and its related ***,the combined effectiveness of these interventions has not been ***,we studied whether the beneficial influences of exercise could be further enhanced by dietary supplementation with SCS in high-fat diet-fed KM(Kunming)*** were randomly divided into four groups,including the high-fat diet group(HF),the SCS group(HF-S),the exercise group(HF-E),and the combination of dietary SCS and exercise group(HF-S+E).There were eight mice in every *** results demonstrated that the combination of dietary SCS and exercise could synergistically reduce fat *** particular,white adipose tissue decreased by 63%in the HF-S+E group compared with that in the HF *** supplementation with exercise also improved peripheral markers,such as serum parameters and hepatic TG *** mechani-cal testing indicated that the combined effects of dietary SCS and exercise on inhibiting fat accumulation might be attributed to the inhibition of lipid synthesis in the liver and the activation of lipolysis in white adipose tissue to increase energy consumption.

Identification of Five Sea Cucumber Species Through PCR-RFLP Analysis

Sea cucumbers are traditional marine food and Chinese medicine in Asia. The rapid expansion of sea cucumber market has resulted in various problems, such as commercial fraud and mislabeling. Conventionally, sea cucumber species could be distinguished by their morphological and anatomical characteristics; however, their identification becomes difficult when they are processed. The aim of this study was to develop a new convenient method of identifying and distinguishing sea cucumber species. Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) analysis of mitochondrial cytochrome oxidase I gene(COI) was used to identifing five sea cucumber species(Apostichopus japonicus, Cucumaria frondosa, Thelenota ananas, Parastichopus californicus and Actinopyga lecanora). A 692 bp fragment of COI was searched for BamHI, KpnI, PstI, XbaI and Eco31 I restriction sites with DNAMAN 6.0, which were then used to PCR-RFLP analysis. These five sea cucumber species can be discriminated from mixed sea cucumbers. The developed PCR-RFLP assay will facilitate the identification of sea cucumbers, making their source tracing and quality controlling feasible.

Changes in the Contents of ATP and Its Related Breakdown Compounds in Various Tissues of Oyster During Frozen Storage

The changes in the contents of adenosine 5’-triphosphate (ATP) and its related breakdown compounds were investigated in the adductor muscle, mantle, gill and body trunk of oyster (Crassostrea gigas) during frozen storage at -20℃ and -30℃ and compared with that of the fresh oyster. The investigation was performed using an HPLC system. Different extents of ATP decomposi- tion were found in various tissues frozen at the two temperatures. The K, K’ and A.E.C values were calculated as the chemical fresh- ness indices. Considering the results of sensory evaluation, the A.E.C. value in body trunk at -20℃ and -30℃ could be used as the best freshness index for frozen oyster.

Study on the effects of the different polar group of EPA-enriched phospholipids on the proliferation and apoptosis in 95D cells

EPA-enriched phosphatidylcholine(EPA-PC)and EPA-enriched phosphatidylethanolamine(EPA-PE)are newly identified marine *** polar group of phospholipids is known to influence EPA-phospholipid ***,the differences in anti-tumor effects between EPA-PC and EPA-PE have not been *** this study,we evaluated the effects of two forms of EPA on the proliferation and apoptosis in the lung-cancer cell line 95D as well as possible molecular *** results showed that EPA-PC effectively inhibited proliferative activity and promoted apoptosis of 95D cells in a dose-dependent manner,while EPA-PE had no effect on cell proliferation,although it slightly promoted *** blot results showed that EPA-PC and EPA-PE upregulated the expression of PPARy,RXRoc,and PTEN,and downregulated the PI3K/AKT signaling ***,EPA-PC and EPA-PE induced the expression of the pro-apoptotic gene,Bax,and reduced the expression of the anti-apoptotic gene,***,EPA-PC and EPA-PE promoted the release of cytochrome c and activated the apoptotic enzyme-cleaved *** data suggest that the anti-tumor effect of EPA-phospholipids may be exerted via a PPARy-related ***-PC was more efficacious as compared to EPA-PE,which might be due to the different polar groups of phospholipids.

Effects of Alginate Gel on Rheological Properties of Hair-tail (Trichiurus lepturus) Surimi

Effects of alginate gel at different concentrations on rheological properties of hair-tail (Trichiurus lepturus) surimi were investigated. Alginate gel (1% - 3%) exhibited enhanced effects, especially when alginate gel concentration increased. The rheological properties of mixture samples were studied by the time sweep, frequency sweep and temperature sweep. The critical strain values of the mixture samples for the onset of non-linear viscoelasticity were about 5%. The storage modulus G' of the mixture samples increased with time for 4 h. The frequency sweep showed that G' was greater than G" for all the mixture gels with different alginate gel concentrations, and values of both n' and n" for all samples were low (<0.2), these constants corresponding to G' and G", and indicating the elasticity of mixture gels. The values of storage modulus G' decreased during heating process and increased with decreasing temperature.

水产品加工流通是实现渔业高质量发展的关键环节，对保障水产品常年优质安全供应、助推国家“食物安全战略”和“健康中国战略”实施发挥了重要作用。新中国成立后，通过自主创新、技术引进与消化吸收，水产品加工流通科技持续进步，产业快速发展。形成了冷冻冷藏水产品、鱼糜制品、休闲食品、干制品、罐藏食品、海藻食品、水产饲料、生物制品加工等全世界最完善的水产加工体系和以批发市场为主体、电商等新型物流模式快速发展的水产品流通体系。本文分析了水产品在提供优质蛋白质和ω-3多不饱和脂肪酸等必需营养素、平衡居民膳食结构、降低陆生动物性食物生产压力等方面的重要作用；系统总结了我国水产品加工流通产业科技发展现状、存在问题及发展方向；提出了“构建以消费引导加工、加工引领养殖的现代渔业产业体系，保障食物安全”、“构建水产品梯次加工利用技术体系，保障水产品安全优质供应”、“构建水产品功效成分的精准加工技术体系，助推建康中国战略实施”等保障我国水产品加工与流通产业高质量发展的新举措。

植物模拟蛋液体系构建及凝胶性质研究

本文以绿豆蛋白、甲基纤维素、κ-卡拉胶(κ-carrageenan,KC)和高酰基结冷胶(high acyl gellan gum,HA)等成分为原料,构建了以绿豆蛋白为基体的模拟蛋液体系。通过测定质构、动态流变、持水性和微观结构等指标,考察了主要凝胶剂KC和HA配比对模拟蛋液凝胶质构性质、流变性质、持水性和微观结构等的影响。质构分析和持水性测定结果表明,KC是模拟蛋液凝胶强度和硬度的决定性因素,而HA可提高凝胶的弹性、内聚性、咀嚼性和持水性。温度扫描结果显示模拟蛋液和天然蛋液在加热和冷却过程中储能模量(G′)始终大于损耗模量(G″),说明两者体系均以弹性为主体。随着HA质量比的提高,复合溶胶的黏度、储能模量(G′)和损耗模量(G″)增加,表明HA可改善模拟蛋液溶胶的黏性性质。扫描电镜结果显示,KC和HA可形成独立的互穿聚合物网络,具有一定的相容性和交联性。感官评定结果可知,当KC/HA配比为1.2:0.4时植物模拟煎蛋综合感官评分最高,和天然煎蛋在口感、组织结构和风味等方面最相近。综上所述,通过研究模拟蛋液体系中不同比例KC/HA的力学性能及相互作用机理,为植物基蛋类模拟制品的开发利用提供理论依据。

Long-Term Effects of Docosahexaenoic Acid-Bound Phospholipids and the Combination of Docosahexaenoic Acid-Bound Triglyceride and Egg Yolk Phospholipid on Lipid Metabolism in Mice

The bioavailability of docosahexaenoic acid(DHA) and eicosapentaenoic acid(EPA) depends on their chemical forms. This study investigated the long-term effects of DHA-bound triglyceride(TG-DHA), DHA-bound phospholipid(PL-DHA), and the combination of TG-DHA and egg yolk phospholipid(Egg-PL) on lipid metabolism in mice fed with a high-fat diet(fat levels of 22.5%). Male C57BL/6J mice were fed with different formulations containing 0.5% DHA, including TG-DHA, PL-DHA, and the combination of TG-DHA and Egg-PL, for 6 weeks. Serum, hepatic, and cerebral lipid concentrations and the fatty acid compositions of the liver and brain were determined. The concentrations of serum total triglyceride(TG), total cholesterol(TC), high-density lipoprotein cholesterol(HDL-c), and hepatic TG in the PL-DHA group and the combination group were significantly lower than those in the high-fat(HF) group(P < 0.05). Atherogenic index(AI) of the PL-DHA group was significantly lower than that of the combination group(P < 0.05). Hepatic TC level in the combination group was significantly lower than that in the HF group(P < 0.05), but no significant difference was observed between the combination group and the PL-DHA group. Both the PL-DHA and the combination groups showed significantly increased DHA levels in the liver compared with the HF group(P < 0.05). However, there were no obvious increases in the cerebral DHA levels in all DHA diet groups. These results suggest that PL-DHA was superior to the combination of TG-DHA and Egg-PL in decreasing the AI. Long-term dietary supplementation with low amount of DHA(0.5%) may improve hepatic DHA levels, although cerebral DHA levels may not be enhanced.
