Genetically modified non-human primate models for research on neurodegenerative diseases

Neurodegenerative diseases(NDs)are a group of debilitating neurological disorders that primarily affect elderly populations and include Alzheimer's disease(AD),Parkinson's disease(PD),Huntington's disease(HD),and amyotrophic lateral sclerosis(ALS).Currently,there are no therapies available that can delay,stop,or reverse the pathological progression of NDs in clinical settings.As the population ages,NDs are imposing a huge burden on public health systems and affected families.Animal models are important tools for preclinical investigations to understand disease pathogenesis and test potential treatments.While numerous rodent models of NDs have been developed to enhance our understanding of disease mechanisms,the limited success of translating findings from animal models to clinical practice suggests that there is still a need to bridge this translation gap.Old World nonhuman primates(NHPs),such as rhesus,cynomolgus,and vervet monkeys,are phylogenetically,physiologically,biochemically,and behaviorally most relevant to humans.This is particularly evident in the similarity of the structure and function of their central nervous systems,rendering such species uniquely valuable for neuroscience research.Recently,the development of several genetically modified NHP models of NDs has successfully recapitulated key pathologies and revealed novel mechanisms.This review focuses on the efficacy of NHPs in modeling NDs and the novel pathological insights gained,as well as the challenges associated with the generation of such models and the complexities involved in their subsequent analysis.

Vortex identification based on the Liutex method and its effect on fish passage upstream

Fishway research is important for mitigating the fragmentation of river habitats caused by hydraulic projects.The vertical slit fishway is a broadly used fishway type because of its high efficiency and adaptability to water levels.However,the resulting vortex current disrupts the fish passage hence directly affecting fish migration.This study aims to accurately capture the vortex structure in the fishway and analyze the effect of vortex elements(vortex structure,vortex intensity,etc.)on fish.We conducted an analysis of the 3-D current flow field in the fishway through the utilization of an experimental model and the large eddy simulation(LES)method.Moreover,we captured the vortex information in the fishway at different flow rates using the Liutex vortex identification method and investigated the effect of the vortex on fish migration.The results revealed that the structures inside the fishway pool occupy most of the room,however,the areas with higher vortex strength were primarily located in the vortex near the vertical seam and the mainstream,the vortex strength inside the fishway gradually increases with increasing flow,suppressing fish migration.Fish experienced significantly increased resistance when encountering strong vortices.This suggests that the vortex may act as a physical barrier to fish migration.These findings highlight the potential negative effects of vortex on fish movement and reiterate the importance of understanding vortex dynamics for aquatic environmental management.As an effective tool for identifying vortices in fluid flow,the Liutex method demonstrates features of vortex within the fishway,thereby providing important insights into the interaction between fluid dynamics and aquatic organisms.

The impact of Chinese "family" culture is deeply rooted in Chinese family business.This kind of culture,which has its origination from the patriarchal relations of Confucian culture,affects the Chinese family business at both positive and negative sides.When a professional manager,an outsider of the family,joins into the business and tries to help its development with his or her professional skills,the " family" culture may show its negative effects,hindering the role the professional manager should play.It is based on such consideration that we try to take the expectancy theory as a starting point and put forward the following suggestions:during the process of introducing professional managers,the private business should take full account of the attitudes and influences of family members into considerations,develop reasonable performance objectives and remuneration package with the professional manager,and achieve the purpose of effectively motivate professional managers to serve the enterprise by safeguarding the psychological contract between them.

CdS/LaFeO_(3) heterojunctions with improved optical properties for visible-light catalytic applications

Construction of heterojunctions is a normal and effective strategy to improve the photocatalytic performances of semiconductors,through which both the lifetime and the redox ability of electrons/holes can be improved,as co mpared to the respective component.On this basis,we constructed Z-scheme CdS/LaFeO_(3) heterojunctions(CdS/LFO),by in-situ growing different amounts of CdS on the surface of LaFeO_(3),for photocatalytic degradation of tetracycline hydrochloride(TC)in aqueous solution at room temperature.The crystal structure,surface morphology and optoelectronic properties of the COS/LFO heterojunctions were systemically characterized to correlate the reaction activity.Photocatalytic tests indicate that the CdS/LFO heterojunctions exhibit promising activity for TC degradation under visible light irradiation(λ≥420 nm),with 85%TC conversion obtained at reaction time of 45 min,which is 3.4 and 1.9 times higher than that of CdS and LaFeO_(3).The CdS/LFO heterojunctions are also stable in the reaction and can be reused for four cycles with no appreciable activity loss.The applicability of CdS/LFO to photocatalytic degradation of organic dyes,as well as the reaction mechanism,was also explored.

Objective:This study explores the mechanism of action of Danhongqing formula(DHQ),a compoundbased Chinese medicine formula,in the treatment of cholestatic liver fibrosis.Methods:In vivo experiments were conducted using 8-week-old multidrug resistance protein 2 knockout(Mdr2-/-)mice as an animal model of cholestatic liver fibrosis.DHQ was administered orally for 8 weeks,and its impact on cholestatic liver fibrosis was evaluated by assessing liver function,liver histopathology,and the expression of liver fibrosis-related proteins.Real-time polymerase chain reaction,Western blot,immunohistochemistry and other methods were used to observe the effects of DHQ on long non-coding RNA H19(H19)and signal transducer and activator of transcription 3(STAT3)phosphorylation in the liver tissue of Mdr2-/-mice.In addition,cholangiocytes and hepatic stellate cells(HSCs)were cultured in vitro to measure the effects of bile acids on cholangiocyte injury and H19 expression.Cholangiocytes overexpressing H19 were constructed,and a conditioned medium containing H19 was collected to measure its effects on STAT3 protein expression and cell activation.The intervention effect of DHQ on these processes was also investigated.HSCs overexpressing H19 were constructed to measure the impact of H19 on cell activation and assess the intervention effect of DHQ.Results:DHQ alleviated liver injury,ductular reaction,and fibrosis in Mdr2-/-mice,and inhibited H19expression,STAT3 expression and STAT3 phosphorylation.This formula also reduced hydrophobic bile acid-induced cholangiocyte injury and the upregulation of H19,inhibited the activation of HSCs induced by cholangiocyte-derived conditioned medium,and decreased the expression of activation markers in HSCs.The overexpression of H19 in a human HSC line confirmed that H19 promoted STAT3 phosphorylation and HSC activation,and DHQ was able to successfully inhibit these effects.Conclusion:DHQ effectively alleviated spontaneous cholestatic liver fibrosis in Mdr2-/-mice by inhibiting H19 upregulation in chol

Distributed Source-Load-Storage Cooperative Low-Carbon Scheduling Strategy Considering Vehicle-to-Grid Aggregators

The vehicle-to-grid(V2G)technology enables the bidirectional power flow between electric vehicle(EV)batteries and the power grid,making EV-based mobile energy storage an appealing supplement to stationary energy storage systems.However,the stochastic and volatile charging behaviors pose a challenge for EV fleets to engage directly in multi-agent cooperation.To unlock the scheduling potential of EVs,this paper proposes a source-load-storage cooperative low-carbon scheduling strategy considering V2G aggregators.The uncertainty of EV charging patterns is managed through a rolling-horizon control framework,where the scheduling and control horizons are adaptively adjusted according to the availability periods of EVs.Moreover,a Minkowski-sum based aggregation method is employed to evaluate the scheduling potential of aggregated EV fleets within a given scheduling horizon.This method effectively reduces the variable dimension while preserving the charging and discharging constraints of individual EVs.Subsequently,a Nash bargaining based cooperative scheduling model involving a distribution system operator(DSO),an EV aggregator(EVA),and a load aggregator(LA)is established to maximize the social welfare and improve the low-carbon performance of the system.This model is solved by the alternating direction method of multipliers(ADMM)algorithm in a distributed manner,with privacy of participants fully preserved.The proposed strategy is proven to achieve the objective of low-carbon economic operation.

MdbHLH162 connects the gibberellin and jasmonic acid signals to regulate anthocyanin biosynthesis in apple

Anthocyanins are secondary metabolites induced by environmental stimuli and developmental signals.The positive regulators of anthocyanin biosynthesis have been reported,whereas the anthocyanin repressors have been neglected.Although the signal transduction pathways of gibberellin(GA)and jasmonic acid(JA)and their regulation of anthocyanin biosynthesis have been investigated,the cross-talk between GA and JA and the antagonistic mechanism of regulating anthocyanin biosynthesis remain to be investigated.In this study,we identified the anthocyanin repressor MdbHLH162 in apple and revealed its molecular mechanism of regulating anthocyanin biosynthesis by integrating the GA and JA signals.MdbHLH162 exerted passive repression by interacting with MdbHLH3 and MdbHLH33,which are two recognized positive regulators of anthocyanin biosynthesis.MdbHLH162 negatively regulated anthocyanin biosynthesis by disrupting the formation of the anthocyanin-activated MdMYB1-MdbHLH3/33complexes and weakening transcriptional activation of the anthocyanin biosynthetic genes MdDFR and MdUF3GT by MdbHLH3 and MdbHLH33.The GA repressor MdRGL2a antagonized MdbHLH162-mediated inhibition of anthocyanins by sequestering MdbHLH162 from the MdbHLH162-MdbHLH3/33 complex.The JA repressors MdJAZ1 and MdJAZ2 interfered with the antagonistic regulation of MdbHLH162 by MdRGL2a by titrating the formation of the MdRGL2a-MdbHLH162 complex.Our findings reveal that MdbHLH162 integrates the GA and JA signals to negatively regulate anthocyanin biosynthesis.This study provides new information for discovering more anthocyanin biosynthesis repressors and explores the cross-talk between hormone signals.

Crystalline-amorphous M@MNx（M=Co,Fe,Ni）encapsulated in nitrogen-doped carbon for enhanced efficient and durable hydrogen evolution reaction

Developing earth-abundant electrocatalysts for hydrogen evolution reaction（HER） is important for the sustainable energy economy.Herein,efficient and stable heterocatalysts consisting of crystalline-amorphous M@MNx（M=Co,Fe,Ni） encapsulated in N-doped carbon layers supported with N-doped graphene sheets（denoted as M@MNx@NC-NG） are synthesized by facile hydrothermal reaction and nitridation process.During the nitriding process,metal ions in M（tzbc）2（H2O）4(tzbc=4-（1H-1,2,4-triazol-1-yl） benzoic acid) complexes are reduced to crystalline M cores,accompanied by the formation of amorphous MNxshells;the tzbc ligands are insitu carbonized to form outermost N-doped carbon（NC）layers that connect with inner MNxvia M-N-C motifs inherited from the complex precursors and inhibit the transition of MNxfrom amorphous to crystalline phase.The Co@CoNx@NC-NG catalyst exhibits excellent HER activity with small overpotentials of 45 and 64 mV at a cathode current density of 10 mA·cm-2and low Tafel slopes of 40 and 85 mV·dec-1in 0.5 mol·L-1H2SO4and1.0 mol·L-1KOH electrolytes,respectively.The Co@CoNx@NC-NG retains 97% of the initial overpotential after 100,000 s in both acidic and alkaline media.Such outstanding HER performance originates from the crystalline-amorphous Co@CoNxthat redistributes electrons around the heterointerfaces,facilitating the conversion process of H+/H2O to hydrogen and thereby promoting HER kinetics.The outermost NC layers serve as the armor of Co@CoNx,and graphene nanosheets act as carriers of egg-like Co@CoNx@NC and conduction paths for electron shuttles,ensuring stable and continuous electrocatalytic hydrogen production.

Ginsenoside Rd Induces Differentiation of Myeloid Leukemia Cells via Regulating ERK/GSK-3β Signaling Pathway

Objective: To investigate the role of ginsenoside Rd（GRd） in acute myeloid leukemia（AML） cell differentiation. Methods: AML cells were treated with GRd（25, 50, 100 and 200 μg/mL）, retinoic acid（RA, 0.1g/L） and PD98059（20 mg/mL） for 72 h, cell survival was detected by methylthiazolyldiphenyl-tetrazolium bromide and colony formation assays, and cell cycle was detected by flow cytometry. Cell morphology and differentiation were observed by Wright-Giemsa staining, peroxidase chemical staining and cellular immunochemistry assay, respectively. The protein expression levels of GATA binding protein 1（GATA-1）, purine rich Box-1（PU.1）, phosphorylated-extracellular signal-related kinase（p-ERK）, ERK, phosphorylated-glycogen synthase kinase-3β（p-GSK3β）, GSK3β and signal transducer and activator of transcription 1（STAT1） were detected by Western blot. Thirty-six mice were randomly divided into 3 groups using a random number table: model control group（non-treated）, GRd group [treated with 200 mg/（kg·d） GRd] and homoharringtonine（HTT） group [treated with 1 mg/（kg·d） HTT]. A tumor-bearing nude mouse model was established, and tumor weight and volume were recorded. Changes of subcutaneous tumor tissue were observed after hematoxylin and eosin staining. WT1 and GATA-1 expressions were detected by immunohistochemical staining. Results: The cell survival was inhibited by GRd in a dose-dependent manner and GRd caused G0/G1cell arrest（P<0.05）. GRd treatment induced leukemia cell differentiation, showing increased expressions of peroxidase and specific proteins concerning erythrogenic or granulocytic differentiation（P<0.05）. GRd treatment elicited upregulation of p-ERK, p-GSK-3β and STAT1 expressions in cells, and reversed the effects of PD98059 on inhibiting the expressions of peroxidase, GATA-1 and PU.1（P<0.05）. After GRd treatment, tumor weight and volume of mice were decreased, and tumor cells underwent massive apoptosis and necrosis（P<0.05）. Investigating the elliptic anisotropy of identified particles in p-Pb collisions with a multi-phase transport model

The elliptic azimuthal anisotropy coefficient(v_(2))of the identified particles at midrapidity(|η|<0.8)was investigated in p-Pb collisions at√s_(NN)=5.02 TeV using a multi-phase transport model(AMPT).The calculations of differential v_(2)based on the advanced flow extraction method of light flavor hadrons(pions,kaons,protons,andΛ)in small collision systems were extended to a wider transverse momentum(p_(T))range of up to 8 GeV/c for the first time.The string-melting version of the AMPT model provides a good description of the measured p_(T)-differential v_(2)of the mesons but exhibits a slight deviation from the baryon v_(2).In addition,we observed the features of mass ordering at low p_(T)and the approximate number-of-constituentquark(NCQ)scaling at intermediate p_(T).Moreover,we demonstrate that hadronic rescattering does not have a significant impact on v_(2)in p-Pb collisions for different centrality selections,whereas partonic scattering dominates in generating the elliptic anisotropy of the final particles.This study provides further insight into the origin of collective-like behavior in small collision systems and has referential value for future measurements of azimuthal anisotropy.
