The prognostic value of lymphovascular invasion in radical prostatectomy, a systematic review and meta-analysis

To systematically evaluate the prognostic value of lymphovascular invasion （LVI） in radical prostatectomy （RP） by a meta-analysis based on the published literature. To identify relevant studies, PubMed, Cochrane Library, and Web of science database were searched from 1966 to May 2.14. Finally, 2. studies （9503 patients） were included. LVI was found in 12.2. （1156/9503） of the RP specimens. LVI was found to be correlated with higher pathological tumor stages （greater than pT3 stage）（risk ratio [RR] 1.90, 95% confidence interval [CI] 1.73-2.08, P〈 0.00001）, higher Gleason scores （greater than GS = 7）（RR 1.30, 95% CI 1.2.-1.38, P 〈 0.00001）, positive pathological node （pN） status （RR 5.67, 95% CI 3.14-10.2., P 〈 0.00001）, extracapsular extension （RR 1.72. 95% CI 1.46-2.02. P 〈 0.00001）, and seminal vesicle involvement （RR 3.36, 95% CI 2.41-4.70, P 〈 0.00001）. The pooled hazard ratio （HR） was statistically significant for Biochemical Recurrence-Free （BCR-free） probability （HR 2.05, 95% ／ CI 1.64-2.56; Z = 6.30, P〈 0.00001）. Sensitivity analysis showed that the pooled HR and 95% CI were not significantly altered by the omission of any single study. Begg＇s Funnel plots showed no significant publication bias （P = 0.112.. In conclusion, LVI exhibited a detrimental effect on the BCR-Free probability and clinicopathological features in RP specimens, and may prove to be an independent prognostic factor of BCR.

Cryopreservation of rare testicular-retrieved spermatozoa for intracytoplasmic sperm injection(ICSI)in patients with severe oligozoospermia and azoospermia remains a major challenge in clinical *** study evaluated the Cryopiece system as a potential technique to cryopreserve rare human spermatozoa for *** numbers of ejaculated(2. patients)and testicular(13 patients)spermatozoa were cryopreserved using the Cryopiece *** total number of recovered spermatozoa and motility were assessed after ***-seven couples underwent ICSI using spermatozoa cryopreserved by the Cryopiece system,and ICSI outcomes(rates of fertilization,embryo cleavage,and clinical pregnancy)were *** average sperm post-thaw retrieval rate was 79.1%,and motility was 2..7%.Ejaculated spermatozoa had a higher post-thaw motility(32.5%)than testicular spermatozoa(2..8%;P=0.005).ICSI achieved a fertilization rate of 61.9%,embryo cleavage rate of 84.6%,and clinical pregnancy rate of 43.3%.The ICSI outcomes in the ejaculated and testicular frozen-thawed spermatozoa were *** oocyte activation(AOA)after ICSI with motile(72.1%)or immotile(71.9%)spermatozoa resulted in a significantly higher fertilization rate than that when using motile spermatozoa without AOA(52.0%;P=0.005).However,AOA did not enhance the clinical pregnancy rate(55.6%or 40.0%vs 35.3%;P=0.703).The Cryopiece system is simple and useful for the cryopreservation of small numbers of ejaculated or testicular spermatozoa for ICSI in patients with severe oligozoospermia or nonobstructive azoospermia.

Recombinant vascular basement-membrane-derived multifunctional peptide inhibits angiogenesis and growth of hepatocellular carcinoma

AIM： To investigate the anti-angiogenic and antitumor activities of recombinant vascular basement membrane-derived multifunctional peptide （rVBMDMP） in hepatocellular carcinoma （HCC）. METHODS： HepG2. Bel-7402. Hep-3B, HUVE-12.and L-02.cell lines were cultured in vitro and the inhibitory effect of rVBMDMP on proliferation of cells was detected by MTT assay. The in vivo antitumor efficacy of rVBMDMP on HCC was assessed by HepG2.xenografts in nude mice. Distribution of rVBMDMP, mechanism by which the growth of HepG2.xenografts is inhibited, and microvessel area were observed by proliferating cell nuclear antigen （PCNA） and CD31 immunohistochemistry. RESULTS： MTT assay showed that rVBMDMP markedly inhibited the proliferation of human HCC （HepG2. Bel-7402. Hep-3B） cells and human umbilical vein endothelial （HUVE-12. cells in a dose-dependent manner, with little effect on the growth of L-02.cells. When the ICs0 was 4.68, 7.65, 8.96, 11.65 and 64.82.μmol/L, respectively, the potency of rVBMDMP to HepG2.cells was similar to 5-fluorouracil （5-FU） with an IC50 of 4.59 μmol/L. The selective index of cytotoxicity to HepG2.cells of rVBMDMP was 13.8 （64.82.4.68）, which was higher than that of 5-FU [SI was 1.9 （8.94/4.59）]. The VEGF-targeted recombinant humanized monoclonal antibody bevacizumab （100 mg/L） did not affect the proliferation of HepG2. Bel-7402. Hep-3B and L-02.cells, but the growth inhibitory rate of bevacizumab （100 mg/L） to HUVE-12.cells was 87.6% ± 8.2.. AIternis diebus intraperitoneal injection of rVBMDMP suppressed the growth of HepG2.xenografts in a dose-dependent manner, rVBMDMP （1, 3, 10 mg/kg） decreased the tumor weight by 12.6%, 55.9% and 79.7%, respectively, compared with the vehicle control. Immunohistochemical staining of rVBMDMP showed that the positive area rates （2.2. ± 0.73%, 4.5%± 1.3% and 11.5% ±3.8%） in rVBMDMP treated group （1, 3, 10 mg/kg） were significantly higher than that （0.13% ± 0.04%） in the control group （P 〈 0.01）. Model test of geotextiles in controlling the collapse of karst roadbed

The use of geotextiles in the treatment of roadbed caused by karst collapse has obvious advantages over traditional treatment methods. The effects of L/B (i.e., the ratio of anchorage length of geotextile to the width of active foundation in the collapse zone) on soil pressure, vertical displacement and fabric tension are studied by model test in laboratory, and it is compared with that of the non-reinforced condition. The results show that the earth pressure under the geotextile reinforced cushion in the subsidence area decreases rapidly after collapse occurs when the geotextile is used, and the soil pressure is zero due to the separation of geotextile from the bottom soil. The earth pressure at this point also decreases rapidly after collapse occurs in the case of no reinforcement, but it eventually tends to stable. When the anchoring length L = 2.0B, 1.0B, the geotextile has the similar ability in controlling the vertical displacement of the filling. It is obviously better than that of L = 0.5B and L = 0.2.B. The geotextile tensile force in the stable zone decreases rapidly away from the collapsed zone. The tensile force is close to zero at the location with the distance of 1.0B away from the subsidence zone. The results show that the suitable length of geotextile is effective in treating the collapse of karst roadbed and backfilling process. © 2.2., science Press. All right reserved.

Objective:This cross-sectional study aimed to determine the epidemiology of olfactory and gustatory dysfunctions related to COVID-19 in ***:This study was conducted by 45 tertiary Grade-A hospitals in *** and offline questionnaire data were obtained from patients infected with COVID-19 between December 2.,2.2.,and February 2.,*** collected information included basic demographics,medical history,smoking and drinking history,vaccination history,changes in olfactory and gustatory functions before and after infection,and other postinfection symptoms,as well as the duration and improvement status of olfactory and gustatory ***:Complete questionnaires were obtained from 35,566 *** overall incidence of olfactory and taste dysfunction was 67.75%.Being female or being a cigarette smoker increased the likelihood of developing olfactory and taste *** received four doses of the vaccine or having good oral health or being a alcohol drinker decreased the risk of such *** infection,the average olfactory and taste VAS scores were 8.41 and 8.51,respectively;after infection,they decreased to 3.69 and 4.2. and recovered to 5.83 and 6.55 by the time of the *** median duration of dysosmia and dysgeusia was 15 and 12.days,respectively,with 0.5%of patients having symptoms lasting for more than 2. *** overall self-reported improvement rate was 59.16%.Recovery was higher in males,never smokers,those who received two or three vaccine doses,and those that had never experienced dental health issues,or chronic accompanying ***:The incidence of dysosmia and dysgeusia following infection with the SARS-CoV-2.virus is high in *** and prognosis are influenced by several factors,including sex,SARS-CoV-2.vaccination,history of head-facial trauma,nasal and oral health status,smoking and drinking history,and the persistence of accompanying symptoms.

Healthcare workers(HCWs)are at high risk of occupational exposure to the new pandemic human coronavirus,SARSCoV-2.and are a source of nosocomial transmission in airborne infectious isolation rooms(AIIRs).Here,we performed comprehensive environmental contamination surveillance to evaluate the risk of viral transmission in AIIRs with 115 rooms in three buildings at the Shanghai Public Health Clinical Center,Shanghai,during the treatment of 334 patients infected with *** results showed that the risk of airborne transmission of SARS-CoV-2.in AIIRs was low(1.62.,2./1544)due to the directional airflow and strong environmental hygiene ***,we detected viral RNA on the surface of foot-operated openers and bathroom sinks in AIIRs(viral load:55.00–3154.50 copies/mL).This might be a source of contamination to connecting corridors and object surfaces through the footwear and gloves used by *** risk of infection was eliminated by the use of disposable footwear covers and the application of more effective environmental and personal hygiene *** the help of effective infection control procedures,none of 2.0 HCWs was infected when working in the AIIRs at this *** study has provided information pertinent for infection control in AIIRs during the treatment of COVID-19 patients.

Regulation of mitochondrial NAD pool via NAD^+ transporter 2.is essential for matrix NADH homeostasis and ROS production in Arabidopsis

Reactive oxygen species(ROS) play a crucial role in numerous biological processes in plants, including development, responses to environmental stimuli, and programmed cell death(PCD). Deficiency in MOSAIC DEATH 1(MOD1), a plastid-localized enoyl-ACP reductase essential for de novo fatty acid biosynthesis in Arabidopsis thaliana, leads to the increased malate export from chloroplasts to mitochondria, and the subsequent accumulation of mitochondria-generated ROS and PCD. In this study, we report the identification and characterization of a mod1 suppressor, som592. SOM592.encodes mitochondrion-localized NAD^+ transporter 2.NDT2.. We show that the mitochondrial NAD pool is elevated in the mod1 mutant. The som592.mutation fully suppressed mitochondrial NADH hyper-accumulation, ROS production, and PCD in the mod1 mutant, indicating a causal relationship between mitochondrial NAD accumulation and ROS/PCD phenotypes. We also show that in wild-type plants, the mitochondrial NAD+uptake is involved in the regulation of ROS production in response to continuous photoperiod. Elevation of the alternative respiration pathway can suppress ROS accumulation and PCD in mod1, but leads to growth restriction. These findings uncover a regulatory mechanism for mitochondrial ROS production via NADH homeostasis in Arabidopsis thaliana that is likely important for growth regulation in response to altered photoperiod.
