Mammalian target of rapamycin(m TOR) inhibitor as an attractive drug target with promising antitumor effects has been widely investigated. High quality clinical trial has been conducted in liver transplant(LT) recipients in Western countries. However, the pertinent studies in Eastern world are paucity. Therefore, we designed a clinical trial to test whether sirolimus can improve recurrence-free survival(RFS) in hepatocellular carcinoma(HCC) patients beyond the Milan criteria after LT. This is an open-labeled, single-arm, prospective, multicenter, and real-world study aiming to evaluate the clinical outcomes of early switch to sirolimus-based regimens in HCC patients after LT. Patients with a histologically proven HCC and beyond the Milan criteria will be enrolled. The initial immunosuppressant regimens are center-specifc for the frst 4-6 weeks. The following regimens integrated sirolimus into the regimens as a combination therapy with reduced calcineurin inhibitors based on the condition of patients and centers. The study is planned for 4 years in total with a 2-year enrollment period and a 2-year follow-up. We predict that sirolimus conversion regimen will provide survival benefts for patients particular in the key indicator RFS as well as better quality of life. If the trial is conducted successfully, we will have a continued monitoring over a longer follow-up time to estimate indicator of overall survival. We hope that the outcome will provide better evidence for clinical decision-making and revising treatment guidelines based on Chinese population data.

Coronavirus disease 2019(COVID-19)is a public health disaster that has not been encountered for a hundred *** January 12,2020,the World Health Organization(WHO)confirmed and named the coronavirus which caused unexplained pneumonia as 2019 novel coronavirus(2019-nCoV).Since then,the disease caused by this virus had been named as coronavirus disease 2019(COVID-19).On February 11,2020,the International Committee on Taxonomy of Viruses announced that the English name of the novel coronavirus was severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)[1].At present,SARS-CoV-2 infection is still rampant *** of September 10,2021,there were about 222 million confirmed cases of COVID-19 and more than 4.5 million deaths worldwide[2].According to the American Centers for Disease Control and Prevention,by July 29,2021,4.19 million pediatric COVID-19 cases had been reported in the United States。

We operated a p-type point contact high purity germanium(PPCGe)detector(CDEX-1B,1.008 kg)in the China jinping Underground Laboratory(CJPL)for 500.3 days to search for neutrinoless double beta(0νββ)decay of ^(76)Ge.A total of 504.3 kg⋅day effective exposure data was *** anti-coincidence and the multi/single-site event(MSE/SSE)discrimination methods were used to suppress the background in the energy region of interest(ROI,1989–2089 keV for this work)with a factor of 23.A background level of 0.33 counts/(keV⋅kg⋅yr)was *** lower limit on the half life of^(76)Ge 0νββdecay was constrained as T_(1/2)^(0ν)>1.0×10^(23)yr(90%C.L.),corresponding to the upper limits on the effective Majorana neutrino mass:<3.2–7.5 eV.

目的建立测定人血清中甲芬那酸浓度的高效液相色谱检测法,并研究甲芬那酸分散片在健康人体内的药动学特征。方法甲芬那酸血清样品在酸性条件下,用乙腈-乙醚(2:1)混合液萃取。在DiscoveryC_(18)不锈钢分析色谱柱(4.6mm×250mm,5μm)上,以乙腈-水(55:45)(磷酸调pH=3.0)为流动相,流速1.8ml·min^(-1),检测波长280nm。结果测定方法的线性范围0.13～8.28mg·L^(-1),回归方程为ρ=3.903×10^(-6)A-0.01179(r=0.9993),血清中甲芬那酸的最低检测限为20mg·L^(-1)(按信噪比≥3计)。平均方法回收率为(97.30±4.54)%,日内与日间RSD均小于15%。药动学研究表明,甲芬那酸在12名健康人体内的主要药动学参数t_(max),ρ_(max),t_(1/2(Ke))和AUC(0～14h)分别为(1.04±0.50)h,(5.94±2.51)mg·L^(-1)。(2.12±0.69)h和(18.19±4.05)mg·h·L^(-1)。结论本方法具有良好的准确性、精密性和较高的灵敏度、简便快速,适用于甲芬那酸的血药浓度测定和药动学研究的需要。甲芬那酸分散片的药动学特征表现为达峰时间快,生物利用度较高。

Characterization of the Rod-Pinch Diode at 1MV as a Radiography Source

The rod-pinch diode(RPD)is a kind of new X-ray source for flash radiographic apphcations,especially in the small-scale hydrodynamic *** to theoretical analysis and the generator parameters,the 1MV rod pinch diode has been designed and studied *** a result,1.21 to 1.45rads at 1m in the forward direction (0°)are achieved in 18.1 to 27.5ns FWHM pulses,the spot sizes are 0.8 to 1.1mm with 1-mm-diameter tungsten rods. This suggests that the rod-pinch diode is a perfect X-ray source used in the small-scale hydrodynamic experiments.

Psoriasis is a chronic,recurrent,inflammatory systemic disease that is not only characterized by skin manifestations but may also be accompanied by various comorbidities,imposing a heavy burden on patients’physical and mental health and affecting their quality of *** the continuous approval of various biological agents for clinical use has provided more effective treatment options to patients with psoriasis,no consensus on the evaluation of comprehensive treatment goals has yet been *** treat-to-target(T2T)strategy requires consideration of multiple dimensions of treatment outcomes,the development of long-term management goals,and regular assessments of treatment conditions,which are often used in the management of chronic ***,based on the latest consensuses and guidelines,research data,and clinical experience as well as the combination of survey results and expert group discussions,the present consensus focuses on 4 dimensions of short-and long-term integrated management goals for biological agents:alleviating skin lesions,improving quality of life,screening and managing psoriasis comorbidities,and ensuring drug *** implementation methods,evaluation time,treatment monitoring,and program adjustments are also herein described to achieve comprehensive management of psoriasis to the maximum *** consensus provides a reference for clinical practice.

人C型凝集素样受体(human C-type lectin-like receptor-2,hCLEC-2)是新克隆的Ⅱ型跨膜受体分子,研究表明,它在病毒感染,血小板活化、聚集,肿瘤转移和信号转导等方面发挥重要作用.通过制备针对其胞外段糖类识别结构域(CRD)的抗体来进一步研究该蛋白质的生物学功能及相关信号通路.构建了pET23b-CRD重组表达质粒,将其转化到大肠杆菌BL21中,用于hCLEC-2-CRD-His融合蛋白的表达,经聚丙烯酰胺凝胶电泳检测表明,融合重组蛋白成功获得了高效表达,分子质量在18ku左右.经鉴定,发现hCLEC-2-CRD-His表达于包涵体中.为获得高纯度的可溶性蛋白,将包涵体溶于6mol/L盐酸胍,并用镍柱进行纯化.纯化后的蛋白质经过透析、再折叠后作为抗原,免疫家兔制备抗血清,抗血清经蛋白G亲合层析纯化后用Western blot等方法进行鉴定,结果显示该抗体能特异性地检测到GFP-hCLEC-2和GFP-CRD.通过使用该抗体,进一步发现,在用PMA和IL-4诱导单核细胞THP-1分化的过程中,内源性hCLEC-2蛋白水平下调,初步揭示了hCLEC-2的表达和单核细胞的分化存在一定的联系.因此,该抗体的成功制备将为进一步研究CLEC-2的生物学功能提供有利的条件.
