Background:Anaplastic lymphoma kinase(ALK)test in advanced non-small cell lung cancer(NSCLC)can help physicians provide target therapies for patients harboring ALK gene *** study aimed to investigate the real-world test patterns and positive rates of ALK gene rearrangements in advanced ***:In this real-world study(ChiCTR2000030266),patientswith advanced NSCLC who underwent an ALK rearrangement test in 30 medical centers in china between October 1,2018 and December 31,2019 were retrospectively *** training was conducted before the study was *** controls were performed at participating centers using immunohistochemistry(IHC)-*** positive ALK gene rearrangement rate and consistency rate were *** associated clinicopathological characteristics of ALK gene rearrangement were investigated as ***:The overall ALK gene rearrangement rate was 6.7%in 23,689 patients with advanced NSCLC and 8.2%in 17,436 patients with advanced lung *** quality control analysis of IHC-VENTANA-D5F3 revealed an intrahospital consistency rate of 98.2%(879/895)and an inter-hospital consistency rate of 99.2%(646/651).IHC-VENTANA-D5F3 was used in 53.6%,real-time polymerase chain reaction(RT-PCR)in 25.4%,next-generation sequencing(NGS)in 18.3%,and fluorescence in-situ hybridization(FISH)in 15.9%in the adenocarcinoma *** specimens tested with multiple methods,the consistency rates confirmed by IHC-VENTANA-D5F3 were 98.0%(822/839)for FISH,98.7%(1,222/1,238)forNGS,and 91.3%(146/160)for *** overall ALK gene rearrangement rateswere higher in females,patients of≤35 years old,never smokers,tumor cellularity of>50,and metastatic specimens used for testing in the total NSCLC population and adenocarcinoma subgroup(all P<0.05).Conclusions:This study highlights the real-world variability and challenges of ALK test in advanced NSCLC,demonstrating a predominant use of IHCVENTANA-D5F3 with high co

Propofol protects against blood-spinal cord barrier disruption induced by ischemia/reperfusion injury

Propofol has been shown to exert neuroprotective effects on the injured spinal ***,the effect of propofol on the blood-spinal cord barrier（BSCB） after ischemia/reperfusion injury（IRI） is poorly ***,we investigated whether propofol could maintain the integrity of the *** cord IRI（SCIRI） was induced in rabbits by infrarenal aortic occlusion for 30 ***,30 mg/kg,was intravenously infused 10 minutes before aortic clamping as well as at the onset of ***,48 hours later,we performed histological and m RNA/protein analyses of the spinal *** decreased histological damage to the spinal cord,attenuated the reduction in BSCB permeability,downregulated the m RNA and protein expression levels of matrix metalloprotease-9（MMP-9） and nuclear factor-κB（NF-κB）,and upregulated the protein expression levels of occludin and *** findings suggest that propofol helps maintain BSCB integrity after SCIRI by reducing MMP-9 expression,by inhibiting the NF-κB signaling pathway,and by maintaining expression of tight junction proteins.

Cold trapped ions can be excellent sensors for ultra-precision detection of physical quantities,which strongly depends on the measurement situation at *** stylus ion trap,formed by two concentric cylinders over a ground plane,holds the promise of relatively simple structure and larger solid angle for optical access and fluorescence collection in comparison with the conventional ion *** we report our fabrication and characterization of the first stylus ion trap constructed in china,aiming for studying quantum optics and sensing weak electric fields in the *** have observed the stable confinement of the ion in the trapping potential for more than two hours and measured the heating rate of the trap to be dε/dt=7.10±0.13 meV/s by the Doppler recooling *** work starts a way to building practical quantum sensors with high efficiency of optical collection and with ultimate goal for contributing to future quantum information technology.

Endoscopic submucosal dissection(ESD)is the standard treatment for early-stage gastric cancer,but the large post-operative ulcers caused by ESD often lead to serious side ***-ESD mu-cosal repair materials provide a new option for the treatment of post-ESD *** this study,we developed a polyurethane/small intestinal submucosa(PU/SIS)hydrogel and investigated its effi-cacy for accelerating ESD-induced ulcer healing in a canine ***/SIS hydrogel possessed great biocompatibility and distinctive pH-sensitive swelling properties and protected GES-1 cells from acid attack through forming a dense film in acidic conditions in ***,PU/SIS gels present a strong bio-adhesion to gastric tissues under acidic conditions,thus ensuring the reten-tion time of PU/SIS gels in *** a canine model,PU/SIS hydrogel was easily delivered via endoscopy and adhered to the ulcer ***/SIS hydrogel accelerated gastric ulcer healing at an early stage with more epithelium regeneration and slight *** findings reveal PU/SIS hydrogel is a promising and attractive candidate for ESD-induced ulcer repair.

In order to improve the thermostability of β-1,3-1,4-glucanase,evolutionary molecular engineering was used to evolve the β-1,3-1,4-glucanase from Bacillus subtilis *** process involves random mutation by error-prone PCR and DNA shuffling followed by screening on the filter-based *** mutants,EGsl and EGs2,were found to have four and five amino acid substitutions,*** substitutions resulted in an increase in melting temperature from Tm=62.5℃ for the wild-type enzyme to Tm=65.5℃ for the mutant EGsl and 67.5℃ for the mutant ***,the two mutated enzymes had opposite approaches to produce reducing sugar from lichenin with either much higher(28%)for the former or much lower (21.6%)for the latter in comparison with their parental *** results demonstrate that directed evolution is an effective approach to improve the thermostability of a mesophilic enzyme.

目的阐明创伤患者睡眠质量及创伤后应激障碍(PTSD)临床流行病学特征,系统分析睡眠质量对创伤住院患者PTSD发生的作用,为PTSD患者的临床早期识别提供参考。方法收集256例于2021年10月~2022年11月在遵义市4所综合医院招募创伤住院患者,226例完成PTSD调查评估。采用PSqi量表结合智能手环睡眠监测评估患者创伤前后睡眠质量;采用PCL-C量表随访测评PTSD发生情况。结果创伤患者1个月后PTSD检出率为19.47%,3个月后PTSD检出率为17.61%。在睡眠质量方面,PTSD患者创伤前PSqi量表评分显著增高(P1),其中患者PTSD发生前睡眠异常检出率为72.73%。在创伤后(入院7 d内)睡眠质量监测方面,PTSD患者评分降低,夜间清醒次数增多(P1个月与3个月后睡眠质量方面,PTSD患者PSqi量表评分均高于非PTSD人群(P<0.05)。结论创伤患者中创伤前睡眠质量差者更容易发生PTSD。

Objective:To confirm the efficacy and safety of Ganyushu Granule(GYSG)in treating premenstrual syndrome(PMS)in patients with Gan(Liver)depression and qi stagnation syndrome(GDQSS)and determine its effective ***:From June 2018 to March 2021,a total of 240 PMS women with GDQSS were included and randomly divided into 3 groups in a 1:1:1 ratio using central block randomization:high-dose GYSG group(n=78,GYSG 2 packs/time),low-dose GYSG group(n=82,GYSG and its simulant 1 pack/time),and placebo group(n=80,GYSG simulant 2 packs/time).Treatment with GYSG or placebo was given thrice daily and for up to 3 menstrual *** outcomes were PMS diary(PMSD)score and premenstrual tension syndrome self-rating scale(PMTS).Secondary outcomes were Chinese medicine(CM)syndrome ***,PMTS,and efficacy of CM were evaluated with menstrual cycles during the treatment *** indicators were analyzed after each menstrual *** analyses were performed using an intention-to-treat method,and clinical safety was ***:Of the 216 patients included in the effectiveness analysis,70,75,and 71 patients were in the high-,low-dose GYSG,and placebo groups,*** the 2nd treatment cycle,the change in PMSD scores in the high-and low-dose groups was lower than that in the placebo group(P3rd treatment cycle,that in the low-dose group was lower than that in the placebo group(P2nd treatment cycle,the high-dose GYSG group had the best CM syndrome efficacy(P<0.05).No serious adverse reactions were ***:GYSG was safe and well-tolerated at both doses for treating PMS patients with ***-dose GYSG might be the optimal dose for a phaseⅢtrial.
