Identification of the multiple bioactive derivatives and their endogenous molecular targets that may mediate the laxative effect of rhubarb in rats

Objective:To determine the mechanism of the laxative effect of rhubarb(Rheum tanguticum *** Balf.)in rats using a combined bioinformatic and in vivo ***:Substances derived from rhubarb that are present in the colorectum were identified using high-performance liquid chromatography,coupled with linear ion-trap quadrupole Orbitrap highresolution mass *** targets with a potential laxative effect were identified from databases and the literature,then used with rhubarb-derived substances in molecular docking *** expression of candidate endogenous target molecules that bound specific components of rhubarb was then measured in constipated rats that had or had not been administered rhubarb by western ***,potentially bioactive compounds were traced back to their prototype ***:We identified 17 anthraquinones and 21 anthrones derived from rhubarb in the colorectum of rats.G-scoring identified three potential mediators of the laxative effect:c-kit,5-hydroxytryptamine receptor 4(5-HT4),and aquaporin-3(AQP3).In addition,10 rhubarb-derived components(aloeemodin,emodin,rhein,chrysophanol,physcion,sennoside A,sennoside C,physcionanthrone,aloeemodinanthrone,and rheinanthrone),which have strong binding affinities for more than one of the three potential targets,were selected as likely active *** extract increased the expression of c-kit and 5-HT4,and reduced the expression of AQP3 in the colon of constipated rats,which might mediate its laxative *** also found that a single prototype component may be metabolized into several active metabolites,and a single active ingredient can also be generated from various prototype ***:The present study demonstrates that various anthraquinones and anthrones present in rhubarb may be metabolized to form bioactive compounds that have additive or synergistic effects to promote defecation via c-kit,5-HT4 and/or AQP3.

A model to calculate the thermodynamic functions of the pure metal nanocrystals has been developed, with the consideration of the effects of both the interfaces and the crystal in the nano-grain interior. As an example, the enthalpy, entropy and Gibbs free energy, as functions of the excess free volume at interfaces, temperature and grain size, are calculated for the Co nanocrystals. Furthermore, the characteristics of β-Co→α-Co phase transformation are studied, and the transformation temperatures at different levels of grain size, as well as the critical grain sizes at different temperatures, are predicted. The calculation results show that, the nano-grained β-Co （fcc） is thermodynamically stable at temperatures much lower than that for the conventional coarse-grained materials, and may also stably exist at room temperature when the grain size is reduced to be small enough. The present model is verified by comparisons between the experimental findings and the theoretical predictions.

Because of the limitations of sampling and seasonal study in polar regions, knowledge of dinoflagellate diversity, distribution and ecology are limited. Dinoflagellates have been incidentally reported from polar regions during some seasons and some populations have been reported as components of microalgae. Surveys of molecular diversity link the genotype of dinoflagellates from polar regions with environmental adaptation. In this study, 37 positive clones of dinoflagellates collected from different sites were used for genotype analysis, providing new insights into the biodiversity and distribution of these species based on 18S rRNA sequencing. Diverse genotypes were recorded for the summer season in Kongsfjorden （high Arctic） whilst a single novel genotype of dinoflagellate was recorded from winter samples from the Antarctic Ocean. Data from ice cores suggests that this single dinoflagellate genotype was adapted to extreme cold and clone library screening found that it was occasionally the only microbial eukaryotic genotype found in winter ice cores. The findings of this study could improve our understanding of the diverse dinoflagellate genotypes occurring in these perennially cold microbial ecosystems.

Silicon photonic integrated devices used for nonlinear optical signal processing play a key role in ultrafast switching,computing,and modern optical ***,current devices suffer from limited operation speeds and low conversion efficiencies due to the intrinsically low nonlinear index of *** this paper,we experimentally demonstrate enhanced optical nonlinearity in a silicon–organic hybrid slot waveguide consisting of an ultranarrow slot waveguide coated with a highly nonlinear organic *** fabricated slot area is as narrow as 45 nm,which is,to the best of our knowledge,the narrowest slot width that has been experimentally reported in silicon slot *** nonlinear coefficient of the proposed device with a length of 3 mm is measured to be up to 1.43×10^(6) W^(-1)km^(-1).Based on the nanostructure design,the conversion efficiencies of degenerate four-wave mixing showed enhancements of more than 12 dB and 5 dB compared to those measured for an identical device without the organic material and a silicon strip waveguide,*** a proof of concept,all-optical canonical logic units based on the prepared device with two inputs at 40 Gb/s are *** obtained logic results showed clear temporal waveforms and wide-open eye diagrams with error-free performance,illustrating that our device has great potential for use in high-speed all-optical signal processing and high-performance computing in the nodes and terminals of optical networks.

HL-2A is the first divertor tokamak in China. The vacuum system is one of the important parts of the HL-2A tokamak, which has to be work well during the physics campaign in 2004. As the tokamak machine is separated from the operation staff during discharges, to guarantee the safe and reliable operation of the vacuum system, it is necessary to watch the vacuum system outside the machine hall with an automatic monitoring system. The paper describes the design and manufacture of the monitoring system.

In an investigation of blood-sucking insects and arboviruses, a virus(YN12243) was isolated from Culicoides samples collected in the Sino-Burmese border region of Yunnan Province, China. The virus caused cytopathic effect(CPE) in C6/36 cells and passaged stably. Polyacrylamide gel analysis showed that the genome of YN12243 was composed of 12 segments of double-stranded RNA(dsRNA), with a distribution pattern of 6-6. The nucleotide and amino acid sequences of the coding region(1.12 segments) were17,803 bp and 5,925 amino acids in length, respectively. The phylogenetic analysis of VP1 protein(RdRp) revealed that YN12243 belonged to genus Seadornavirus of family Reoviridae, and further analysis indicated that YN12243 belongs to the Banna virus(BAV) genotype A2. Additionally, YN12243 was located in the same evolutionary cluster as BAV strains isolated from different mosquito species, suggesting that the BAV isolated from Culicoides does not have species barriers. These results indicate that Culicoides can also be a vector for BAV. In view of the hematophagous habits of Culicoides on cattle, horses, deer, and other large animals, as well as the possibility of spreading and causing a variety of animal arboviral diseases, it is important to improve infection detection and monitor the BAV in large livestock.
