An experimental study of using Chai Lai Prescription to treat in vitro rabbit models of reflux esophagitis

Background Chai Lai Prescription is a Chinese herbal compound which is used to sooth the liver,strengthen the spleen and harmonize the stomach for descending adverse *** initiated the study to investigate its mechanism of treating in vitro rabbit reflux esophagitis *** Adult male Japanese white rabbits,weighing 1.8-2.2 kg,were divided into five groups of three each,which were：normal control group （Krebs buffer,pH7.4）,esophagitis model group （Krebs buffer,pH5.8）,esophagitis model proup＋low-dose Chinese herbal medicine protection group （0.6 mg·ml1·kg1）,esophagitis model group＋moderate-dose Chinese herbal medicine protection group （6 mg·ml1·kg1）,esophagitis model group＋high-dose Chinese herbal medicine protection group （60 mg·ml1·kg1）.The RT-PCR method was used to test the influence of Chai Lai Prescription on IL-1 and IL-6 in in vitro rabbit models of *** treated the in vitro models with different doses of Chinese herbal *** Esophageal mucosa were filled with various ***-6 and IL-1β mRNA expression was increased in rabbit esophageal mucosa stimulated with *** herbal medicine significantly reduced the levels of IL-6 and IL-1β mRNA expression in the in vitro cultured rabbit esophageal *** Chinese herbal medicine to treat in vitro models of RE,we found that the IL-6 and IL-1β mRNA expression levels went down,near to or lower than the normal control levels,compared with the group treated with acidified buffer *** Chai Lai Prescription lowered the IL-1β and IL-6 cytokine mRNA levels and protected the esophageal mucosa in the in vitro models of reflux esophagitis,suggesting that the traditional Chinese herbal compound may be able to treat reflux esophagitis by inhibiting the its inflammatory mediators.

Background Visfatin, a visceral fat-derived adipocytokine, plays a significant physiological function in lipid metabolism. However, the precise function of visfatin and its regulation by thyroid hormones are still unknown. This study observed the plasma visfatin concentrations in subjects with hyperthyroidism and hypothyroidism in vivo, and investigated the possible reaulation mechanism between visfatin and tri-iodothvronine （T3） in vitro as a further interpretation.

Reversal of multidrug resistance in renal cell carcinoma by short hairpin RNA targeting MDR1 gene

Background Over-expression of P-glycoprotein （P-gp）, encoded by the MDR1 gene, confers multidrug resistance （MDR） in renal cell carcinoma （RCC） and is a major reason for unsuccessful chemotherapy. This study aimed to determine the effct of RNA interference （RNAi） on the reversal of MDR in human RCC. Methods We designed and selected one short hairpin RNA （shRNA） targeting MDR1 gene, which is stably expressed from integrated plasmid and transfected by lentivirus fluid in human RCC A498 cell. Results The MDRl-targeted RNAi resulted in decreased MDR1 gene mRNA level （P 〈0.001）, almost abolished P-gp expression and reversed MDR to different chemotherapy drugs in the RCC A498 cell line. Conclusion MDR could be reversed by RNAi in human RCC A498 cell line, which may be used for clinical application

Developing new polymeric semiconductors with excellent device performance is essential for organic electronics. Herein, we synthesized two new thiazoloisoindigo(Tz II)-based polymers, namely, P(Tz II-d Th-d Th) and P(Tz II-d Th-d Tz), by copolymerizing thiophene-flanked Tz II with bithiophene and bithiazole, respectively. Owing to the more electron-deficient nature of bithiazole than bithiophene, P(Tz II-d Th-d Tz)possesses deeper LUMO/HOMO levels of-3.45/-5.47 e V than P(Tz II-d Th-d Th)(-3.34/-5.32 e V). The organic field-effect transistor(OFET) devices based on P(Tz II-d Th-d Th) exhibited p-type behaviors with an average hole mobility value as high as 1.43 cm^(2)·V^(-1)·s^(-1), while P(Tz II-d Th-d Tz)showed typical ambipolar characteristics with average hole and electron mobilities of 0.38 and 0.56 cm^(2)·V^(-1)·s^(-1). In addition, we compared the performances of both polymers with other Tz II-based polymers reported in our previous work, and showed that the charge carrier polarity can be manipulated by adjusting the number of the thiophene units between the acceptor unit. As the increase of the number of thiophene rings,charge carrier polarity shifts from electron-dominated ambipolar transport to hole-dominated ambipolar transport and then to unipolar hole transport in OFETs, which provides an effective molecular design strategy for further optimization of polymer OFET performance.

Transcatheter arterial chemoembolization combined with PD-1 inhibitors and Lenvatinib for hepatocellular carcinoma with portal vein tumor thrombus

BACKGROUND Hepatocellular carcinoma(HCC)patients complicated with portal vein tumor thrombus(PVTT)exhibit poor prognoses and treatment *** To investigate efficacies and safety of the combination of PD-1 inhibitor,transcatheter arterial chemoembolization(TACE)and Lenvatinib in HCC subjects comorbid with *** From January 2019 to December 2020,HCC patients with PVTT types Ⅰ-Ⅳ were retrospectively enrolled at Beijing Ditan *** were distributed to either the PTL or TACE/Lenvatinib(TL)*** median progression-free survival(mPFS)was set as the primary endpoint,while parameters like median overall survival,objective response rate,disease control rate(DCR),and toxicity level served as secondary *** Forty-one eligible patients were finally recruited for this study and divided into the PTL(n=18)and TL(n=23)*** a median follow-up of 21.8 months,the DCRs were 88.9%and 60.9%in the PTL and TL groups(P=0.046),***,mPFS indicated significant improvement(HR=0.25;P<0.001)in PTL-treated patients(5.4 months)compared to TL-treated(2.7 months)*** were no treatment-related deaths or differences in adverse events in either *** A triplet regimen of PTL was safe and well-tolerated as well as exhibited favorable efficacy over the TL regimen for advanced-stage HCC patients with PVTT types Ⅰ-Ⅳ.

Studying the system state of coexistence regions will peek into to reveal microscopic interactions between different phases of a thermodynamic ***,there is no effective method to study thermodynamic nature of the coexistence black hole regions for the failure of the equation of *** at these coexistence states,in this work,we develop a general approach by introducing two new ratio *** first one is the ratio of the horizon radii of the saturated coexistence small and large black holes,and the second one measures that of the small black hole molecule number to the total molecule *** demonstrate that the first parameter can serve as an order parameter to characterize the first-order phase *** study also shows that the black hole state in the coexistence region is uniquely determined by these two introduced parameters bounded between 0 and *** results are quite significant in the analytical study of phase transition and the microscopic nature of black hole in the coexistence regions.

Bismuth(Bi),as an alloy-based anode material,has attracted much atte ntion in the developme nt of sodiumion hybrid capacitors(SIHCs)due to its high theoretical ***,the volume expansion of the Bi-based anode during the sodiation/desodiation process results in limited rate *** the present work,a porous Bi-based composite was constructed by a one-step hydrothermal method,and Bi was encapsulated in ligninderived nitrogen-doped porous carbon(Bi@LNPC)after *** obtained Bi nanoparticles could effectively adapt to the strain and shorten the diffusion distance of Na^(+).In addition,porous carbon skeleton provides a rigid conductive network for electronic ***,the assembled sodium-ion half-cell with Bi@LNPC anode shows ultra-high-rate *** the current density was enhanced from 0.1 to 50 A·g^(-1),the specific capacity decreased slightly from 351.5 to 342.8 mAh·g^(-1).Even at an extremely high current density of 200 A·g^(-1),it retains 81.3%capacity retention when compared to a current density of 1 A·g^(-1).The SIHCs assembled by Bi@LNPC show a high energy density of 63 Wh·kg^(-1).This work provides an effective method for developing high-rate Bi anode materials for sodium-ion hybrid capacitors(SIHCs)and sodium-ion batteries(SIBs).
