The regulation of spermatogonial proliferation and apoptosis is of great significance for maintaining *** single-cell RNA sequencing(scRNA-seq)analysis of the testis was performed to identify genes upregulated in *** scRNAseq analysis,we identified the spermatogonia upregulated gene origin recognition complex subunit 6(Orc6),which is involved in DNA replication and cell cycle regulation;its protein expression in the human and mouse testis was detected by western blot and *** explore the potential function of Orc6 in spermatogonia,the C18-4 cell line was transfected with control or Orc6 ***,5-ethynyl-2-deoxyuridine(EdU)and terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assays,flow cytometry,and western blot were used to evaluate its effects on proliferation and *** was revealed that ORC6 could promote proliferation and inhibit apoptosis of C18-4 *** RNA sequencing and bioinformatics analysis indicated that Orc6 was involved in the activation of wingless/integrated(Wnt)/β-catenin *** blot revealed that the expression ofβ-catenin protein and its phosphorylation(Ser675)were significantly decreased when silencing the expression of *** findings indicated that Orc6 was upregulated in spermatogonia,whereby it regulated proliferation and apoptosis by activating Wnt/β-catenin signaling.

Over the past few years,quantum computation based on superconducting circuits has achieved remarkable progress.A milestone occurred in 2019 when Google released Sycamore,a processor with 54 qubits,and claimed quantum supremacy by performing in just 100 s a specific computation which would take a classical supercomputer,as stated by Google’s team,10000 years to complete.[1]In 2021,a strong quantum advantage was demonstrated by Pan and his colleagues from the University of Science and Technology of China,using a quantum processor named Zuchongzhi,which has 66 functional qubits.[2]This year,the record of the number of quantum qubits has been lifted to 127 qubits.[3]Indeed,the number of qubits is limited to a few hundreds due to the finite space of dilution refrigerators,where the superconducting qubits must be placed to be isolated from thermal ***,this number is still several orders of magnitude away from the requirement of quantum error correction,which is essential for general-purpose quantum computers.[4–8].

Plastic characterization of metals by combining nanoindentation test and finite element simulation

Materials with the same elastic modulus E and representative stress and strain （σr,εr） present similar indentation-loading curves, whatever the value of strain hardening exponent n. Based on this definition, a good approach was proposed to extract the plastic properties or constitutive equations of metals from nanoindentation test combining finite element simulation. Firstly, without consideration of strain hardening, the representative stress was determined by varying assumed representative stress over a wide range until a good agreement was reached between the computed and experimental loading curves. Similarly, the corresponding representative strain was determined with different hypothetical values of strain hardening exponent in the range of 0-0.6. Through modulating assumed strain hardening exponent values to make the computed unloading curve coincide with that of the experiment, the real strain hardening exponent was acquired. Once the strain hardening exponent was determined, the initial yield stress ay of metals could be obtained by the power law constitution. The validity of the proposed methodology was verified by three real metals： AISI 304 steel, Fe andA1 alloy.

A novel TOA estimation method with effective NLOS error reduction

It is well known that non-line-of-sight （NLOS） error has been the major factor impeding the enhancement of accuracy for time of arrival （TOA） estimation and wireless positioning. This article proposes a novel method of TOA estimation effectively reducing the NLOS error by 60%, comparing with the traditional timing and synchronization method. By constructing the orthogonal training sequences, this method converts the traditional TOA estimation to the detection of the first arrival path （FAP） in the NLOS multipath environment, and then estimates the TOA by the round-trip transmission （RTT） technology. Both theoretical analysis and numerical simulations prove that the method proposed in this article achieves better performance than the traditional methods.

Hepatocyte growth factor combined with insulin like growth factor-1 improves expression of GATA-4 in mesenchymal stem cells cocultured with cardiomyocytes

Background In a suitable microenvironment, bone marrow mesenchymal stem cells （BMSCs） can transdifferentiate into myocardial cells whose special gene can be expressed as structural proteins. Growth factor （GF） plays an important role in the cell migration, survival and differentiation. However, the effect of GF on the cellular differentiation is not well understood. In this study, the hepatocyte growth factor （HGF） and insulin like growth factor-1 （IGF-1） were used in the mixed culture of BMSCs and myocardial cells and the effects of these growth factors on the GATA-4 expression of BMSCs were investigated.Methods BMSCs were isolated from the marrow of rabbit femurs and tibias and foetal rabbit ventricular myocytes were isolated with trypsin sequential digestion. These two kinds of cells were cocultured in a ratio of 1：1 for 6 weeks; cocultured cells with added HGF and IGF-1 were the experimental group. The differentiated BMSCs were collected using the laser capture, microdissection system and their RNA isolated. Immunocytochemical staining, transmission electron microscopy and reverse transcription-polymerase chain reaction were used to evaluate the transformation of the stem cells into cardiomyocytes like cells.Results When cultured separately, BMSCs did not express a-actin and the stem cells had many nucleoli. However, when cocultured with cardiomyocytes, BMSCs expressed a-actin and the cardiac transcription factor GATA-4 and showed cardiomyocyte like ultrastructure. In comparison with the control group, the experimental group exhibited the enhanced expression level of GATA-4. The GATA-4 expression of BMSCs increased gradually following the addition of HGF and IGF-1, reached the maximal level after two weeks and decreased slightly thereafter.Conclusions BMSCs can transdifferentiate into cardiomyocytes like cells and express the cardiac transcription factor GATA-4 after being cocultured with myocardial cells. The effect of chromophores to the glass transition temperature of polyimide ensemble has been investigated by means of molecular dynamics simulation in conjunction with barrier analysis. Simulated Tg results indicated a good agreement with experimental value. This study showed the MD simulation could estimate the effect of chromophores to the Tg of polyimide ensemble conveniently and an estimation approach method had a surprising deviation of Tg from experiment. At the same time, a polyimide structure with higher barrier energy was designed and validated by MD simulation.

A Convenient Fluorescent-labeled Assay for in vitro Measurement of DNA Mismatch Repair Activity

Objective The assay of DNA mismatch repair （MMR） activity can be used as a biomarker for environmental condition detection and human disease diagnosis. Radioactive 32p-endlabeled DNA containing mismatch is extensively used as the substrate for MMR activity analyses. The aim of the present study is to develop a simple non-radioactive, but equally specific and sensitive method for the MMR activity assay. Methods A fluorescent label was chosen to replace the radioactive isotope label. Sensitive evaluation of the fluorescent label was carried out for the first time, and then the fluorescent label was compared with the isotope label in the MMR activity and DNA binding assays. Result LOD （limit of detection） of the fluorescent label was about 0.1 fmol and the relative signal strength displayed a pretty good linear relationship. Moreover, the fluorescent label method has equivalent sensitivity and performance as compared with the classical radioactive method in experiments. Conclusion In light of the sensitivity, reproducibility, safety, rapidity and long lifespan of the fluorescent label, this improved method can be applied to evaluation of biologic and toxic effects of environmental pollutants on man and other forms of life.

The Permian Taiyuan and Shanxi formations exposed in Shandong Province,eastern North China,contain abundant spores and *** this study,a total of 42 genera and 146 species of spores and pollen from these Permian formations,native to northern China,are identified and related to the three epochs of the Permian Period(Cisuralian,Guadalupian,and Lopingian Epochs)as two assemblages:Assemblage I—the Laevigatosporites-Granulatisporites assemblage,inferred as the Cisuralian(~298.9-272.9 Ma);and,Assemblage II—the Gulisporites-Sinulatisporites assemblage,inferred as the Guadalupian(~272.9-259.1 Ma).Assemblage I represents growing ferns,whereas Assemblage II represents *** assemblage division and analysis indicated that the palaeoclimate of the study area during Early-Middle Permian time was dominated by warm and humid conditions,and later in the Middle Permian changed into moderately dry conditions.
