Disruption of a microvitellogenin gene impairs eggshell formation in Mythimna separata

Microvitellogenin(mVg) belongs to the lepidopteran-specific lipoprotein＿11 super family and is thought to be involved in insect reproduction. However, the function of mVg in reproduction has not been established. In this study, we identified and characterized MsmVg in Mythimna separata, a destructive insect pest of grain crops. MsmVg is comprised of a 1,310 bp open reading frame(ORF) encoding 437 amino acids. Structural analysis showed that MsmVg contains three conserved domains in the lipoprotein＿11 super family, including the all-α-N-terminal domain(NTD), a Pseudomonas putida homologous domain(PPD), and an all-β-c-terminal domain(cTD). MsmVg was highly expressed in mature female adults and enriched in the ovaries of 3-day-old female adults. Disruption of MsmVg by RNA interference did not change the expression of MsmVg, vitellogenin(MsVg) or the vitellogenin receptor(MsVgR) in fat bodies, but it inhibited their transcription in ovaries. Phenotype analysis showed that knockdown of MsmVg did not affect yolk deposition in ovaries, but impaired eggshell formation and led to a reduction in the reproductive capacity. Furthermore, knockdown of MsmVg significantly inhibited the expression of genes associated with eggshell formation(MschP1-3 and MsFcP3c). Taken together, these data suggest that MsmVg plays an important role in female reproduction by affecting eggshell formation in M. separata, which provides a potential target for managing this insect pest.

Knowledge-Driven Possibilistic clustering with Automatic cluster Elimination

Traditional Fuzzy c-Means (FcM) and Possibilistic c-Means (PcM) clustering algorithms are data-driven, and their objective function minimization process is based on the available numeric data. Recently, knowledge hints have been introduced to form knowledge-driven clustering algorithms, which reveal a data structure that considers not only the relationships between data but also the compatibility with knowledge hints. However, these algorithms cannot produce the optimal number of clusters by the clustering algorithm itself; they require the assistance of evaluation indices. Moreover, knowledge hints are usually used as part of the data structure (directly replacing some clustering centers), which severely limits the flexibility of the algorithm and can lead to knowledge misguidance. To solve this problem, this study designs a new knowledge-driven clustering algorithm called the PcM clustering with High-density Points (HP-PcM), in which domain knowledge is represented in the form of so-called high-density points. First, a new data density calculation function is proposed. The Density Knowledge Points Extraction (DKPE) method is established to filter out high-density points from the dataset to form knowledge hints. Then, these hints are incorporated into the PcM objective function so that the clustering algorithm is guided by high-density points to discover the natural data structure. Finally, the initial number of clusters is set to be greater than the true one based on the number of knowledge hints. Then, the HP-PcM algorithm automatically determines the final number of clusters during the clustering process by considering the cluster elimination mechanism. Through experimental studies, including some comparative analyses, the results highlight the effectiveness of the proposed algorithm, such as the increased success rate in clustering, the ability to determine the optimal cluster number, and the faster convergence speed. copyright © 2024 The Authors. Multiscale confinement nitridation in molybdenum carbide for efficient hydrogen production

The molybdenum carbide(Mo_(2)c)has been regarded as one of the most cost-efficient and stable electrocatalyst for the hydrogen evolution reaction(HER)by the virtue of its Pt-like electronic ***,the inherent limitation of high density of empty valence band significantly reduces its catalytic reactivity by reason of strong hydrogen desorption ***,we propose a multiscale confinement synthesis method to design the nitrogen-rich Mo_(2)c for modulating the band structure via decomposing the pre-coordination bonded polymer in a pressure-tight tube sealing ***-bonded c/N-Mo in the coordination precursor constructs a micro-confinement space,enabling the homogeneous nitrogenization in-situ happened during the formation of Mo_(2)***,the evolved gases from the precursor decomposition in tube sealing system establish a macro-confinement environment,preventing the lattice N escape and further endowing a continuous *** the multiscale confinement effects,the nitrogen-rich Mo2c displays as high as 25%N-Mo concentration in carbide lattice,leading to a satisfactory band ***,the constructed nitrogen-rich Mo_(2)c reveals an adorable catalytic activity for HER in both alkaline and acid *** is anticipated that the multiscale confinement synthesis strategy presents guideline for the rational design of electrocatalysts and beyond.

cellulose macrofibers (MFs) are gaining increasing interest as natural and biodegradable alternatives to fossil-derived polymers for both structural and functional applications. However, simultaneously achieving their exceptional mechanical performance and desired functionality is challenging and requires complex processing. Here, we reported a one-step approach using a tension-assisted twisting (TAT) technique for MF fabrication from bacterial cellulose (Bc). The TAT stretches and aligns Bc nanofibers pre-arranged in hydrogel tubes to form MFs with compactly assembled structures and enhanced hydrogen bonding among neighboring nanofibers. The as-prepared Bc MFs exhibited a very high tensile strength of 1 057 MPa and exceptional lifting capacity (over 340 000 when normalized by their own weight). Moreover, due to the volume expansion of Bc nanofibers upon water exposure, Bc MFs quickly harvested energy from environmental moisture to untwist the bundled networks, thus generating a torsional spinning with a peak rotation speed of 884 r/(min·m). The demonstrated rapid and intense actuation response makes the MFs ideal candidates for diverse humidity-response-based applications beyond advanced actuators, remote rain indicators, intelligent switches, and smart curtains.

应用材料模拟软件JMatPro模拟TTA曲线、ccT曲线和TTT曲线,可以获得c55E材料的相变温度。通过获得的相变温度,制定了c55E钢链板等温淬火工艺。结果表明,根据材料模拟软件JMatPro制定的等温淬火工艺参数为淬火温度850℃,等温温度310℃,等温时间25 min;等温淬火后链板硬度满足技术要求,其链条的强度、疲劳等性能接近采用同等工艺处理的50crV材料。

In order to verify the feasibility of applying high-Jc Nb_(3)Sn strand in fusion magnet,a full-size cable-in-conduit conductor(cIcc)with short twist pitch(STP)cable pattern was manufactured and tested in SULTAN facility at SPc,*** levels of cyclic electromagnetic(EM)load were applied on the sample stepwise,no visible decrease of current sharing temperature(TcsT was observed until the EM load increased to 80 kA×10.8 T,after that the Tcs decreased dramatically with the EM cycles,which suggested that irreversible deformation,causing a change in the strain state,or even damage has occurred in the superconducting *** investigating the reason which caused the conductor performance degradation,the tested conductor was dissected for metallographic *** segments which subjected to different EM loads were extracted from one of the legs,the geometric feature changes of the cable cross-sections were analyzed and compared.A good correlation was found between the decrease of the Tcs and deformation of the cable cross section.A mass of cracks were found on the sub-elements of strands in the segment which subjected to highest EM load,but the amount of crack is much lower in other *** the analyses,it is speculated that the critical EM load which causes irreversible degradation is between 850 kN/m and 870 kN/m for this *** results could be a reference in high-Jc Nb_(3)Sn cIcc design.

本文利用自洽聚类分析(Self-consistent clustering analysis,ScA)方法研究了2D c/Sic在单轴压缩载荷下的渐进损伤行为,ScA方法通过应变集中张量对网格单元进行聚类,在不显著降低计算精度的前提下,大幅度降低了模型的自由度,使模型的计算效率得以提高。整个方法由离线和在线两个阶段组成:离线阶段,利用k-means算法对高保真度的复合材料单胞进行分解、聚类并计算不同聚类间的相互作用张量,最终生成降阶模型;在线阶段,基于降阶模型求解离散的Lippmann-Schwinger方程组获取力学响应。将ScA方法应用于2D c/Sic压缩强度的预报,当聚类总数量为64时,与试验相比,压缩强度求解的计算精度与传统有限元相比降低了1%,但整体计算效率提升了34倍。当不考虑离线阶段花费的聚类时间,即事先已知材料的细观构型对其力学行为进行求解时,其一次在线计算的时间仅为6 s,计算效率比传统有限元提升了104倍,在结构性能快速设计、结构状态快速预报等领域,有着广阔的应用前景。

簸箕柳×三蕊柳种特异性KASP标记开发及种间杂交子代鉴别

【目的】开发簸箕柳和三蕊柳种特异性KASP引物,为开展簸箕柳和三蕊柳杂交选育速生、抗虫新种质的真实性鉴定提供参考。【方法】利用簸箕柳和三蕊柳重测序数据,开展全基因组SNP位点分析,筛选出种特异性的SNP位点并设计KASP引物,利用SeqHunter2检测设计引物在簸箕柳中的通用性,并利用簸箕柳和三蕊柳自然群体材料对合成的引物进行实验验证,筛选出簸箕柳和三蕊柳种特异性KASP引物,并对它们的杂交子代进行真实性鉴别。【结果】将三蕊柳和簸箕柳重测序的数据比对到三蕊柳基因组,共获得个6598144个SNPs。经过筛选,最终在簸箕柳中检测到674144个和三蕊柳相比为纯合突变的位点。从每条染色体上随机选取100个位点用于KASP引物设计,在1900个SNPs位点中,750个SNP位点可以成功设计出KASP引物。从每条染色体上选取2组,共选取38组引物,通过SeqHunter2检测得到11组引物在簸箕柳中是通用的。利用簸箕柳和三蕊柳自然群体DNA对合成的10组通用引物进行种内保守和种间差异性检测,获得4组在簸箕柳、三蕊柳和种间杂交子代中聚类明显的引物。引物Stri08_82809、Stri14_11602、Stri14_12274和Stri17_10731在簸箕柳自然群体中分别扩增出纯合G/G、T/T、A/A和T/T位点,在三蕊柳自然群体分别扩增出纯合A/A、c/c、G/G和c/c位点,而在真实杂交子代中分别扩增出G/A、T/c、A/G和T/c杂合位点。利用上述4组引物对簸箕柳×三蕊柳的杂交子代进行鉴定,发现在31个杂交子代中,29个个体扩增出杂合位点,说明同时遗传了2个亲本的特征位点,为真实的种间杂交子代。有2个个体扩增的位点为纯合且和母本相同,说明这2个子代为簸箕柳花粉污染的子代。【结论】本研究获得了簸箕柳和三蕊柳种间特异性KASP标记4组,可以准确快速的开展簸箕柳和三蕊柳种间杂交子代的真实性鉴别,为进一步簸箕柳和三蕊柳种间杂交及回交创制抗虫新品种提供了技术支持。

Plant anatomy is patterned early during leaf development which suggests studying the spatial–temporal transcriptomes of primordia will help identify critical regulative and functional *** successfully isolated the leaf primordia tissues from the c3grass rice and the c4grass foxtail millet by laser capture microdissection(LcM)and studied the gene expression throughout leaf developmental *** data analysis uncovered the conserved expression patterns of certain gene clusters both in rice and foxtail millet during leaf *** revealed genes and transcription factors involved in vein formation,stomatal development,and suberin *** identified 79 candidate genes associated with functional regulation of c4anatomy *** phenotype of the candidate genes revealed that knock-out of a putative polar auxin transport related gene NAL1 resulted significantly reduced veinal space in rice *** present work provides a foundation for future analyses of genes with novel functions in grasses and their role in leaf development,in particular the role in leaves with a contrasting c3vs.c4biosynthetic pathway.
