A transient method with rectangular pulse heating has been developed to measure the thermal conductivity of highly porous materials such as activated carbon, zeolite and silica gel. By this method the thermal conductivity can be measured quickly and accu-rately. In this paper, a set of automatically controlled testing equiptnent is *** measuring method is analysed. The thermal conductivities of some samples, such as activated carbon and zeolite, are measured by the equipment. A group of useful data has been obtained.

Aim:To develop a complex high-throughput screening （HTS） assay based onscintillation proximity assay （SPA） technology for identification of novel peroxi-some proliferator-activated receptor gamma （PPARγ） ***:Full-length PPARγ and retinoid X receptor alpha （RXRα）,biotinylated PPAR responseelement （PPRE）,[3H]BRL49653 and streptavidin-coated FlashPlate or microbeadwere used to develop an HTS assay based on SPA ***‘ABCDE’method was validated against conventional hydroxyapatite （HA） assay and ap-plied to large-scale screening of 16 000 synthetic compounds and natural ***:（1） IC50values of positive control compounds （BRL49653 andtroglitazone） obtained from the‘ABCDE’method and HA assay were compa-rable and consistent with those reported elsewhere;（2） Approximately 178compounds,showing more than 70% competitive inhibition on BRL49653 bind-ing to PPARγ,were identified initially by the‘ABCDE’method （microbead）;（3）Secondary screening using FlashPlate and cross-reactivity studies with RARα,β,γ and RXRα,β,γ confirmed that 12 compounds possessed specific PPARγ bind-ing properties including 2 with IC50values less than 0.5 μmol/L and novel chemi-cal ***:The‘ABCDE’method using either FlashPlate ormicrobead,is a highly efficient,automatable,and robust tool to screen potentialPPARγ modulators in HTS *** application may be expanded to othernuclear receptors that form heterodimers upon activation.

本文描述了我国疟疾媒介嗜人按蚊(Anopheles anthropophagus)的唾腺染色体图。此蚊的唾腺染色体由五个臂组成。第1号染色体为性染色体,又称X-染色体,是近端着丝粒,只有一个臂,它是各臂中最短的,此臂分为5个区;第2号染色体是中心着丝粒,左、右臂约等长,两臂共分为16个区;第3号染色体为亚中心着丝粒,右臂是各臂中最长的,左臂则是常染色体中最短的,两臂共分为18个区。

16,17α-环氧-5α-和5β-孕甾-3β-醇-20-酮（1,2）用诺卡氏菌脱氢分别得到4,5,6, 7和8,5,6,7四个化合物,其中16,17α-环氧-⊿4-孕甾烯-3,20-双酮（5）是主要产物.1和2分别用节杆菌脱氢时则均得到两个20α-羟基的不饱和化合物15和16,其中⊿1,4-双酮16是主要产物.16,17α-环氧-16β-甲基-5α-⊿9（11）-孕甾烯-3β-醇-20-酮（3）用诺卡氏菌脱氢可得到12,13,14三个化合物,其中16,17α-环氧-16β-甲基-⊿4,9（11）-孕甾二烯-3,20-双酮（13）是主要产物.综上所述,5α和5β甾族化合物用诺卡氏菌脱氢主要脱去C4,5两个氢原子形成⊿4-烯-3-酮化合物,而采用节杆菌可使Cl,2和C4,5位同时脱氢形成⊿1,4-双烯-3-酮化合物.

缺氧对鸡成肌细胞肌纤维类型转化作用的机制探究

旨在利用氯化钴(CoCl_(2))模拟肌细胞缺氧,并结合转录组测序分析缺氧对肌纤维类型及相关基因的影响。本试验以鸡原代成肌细胞为研究对象,使用不同浓度氯化钴(0、50、100、200和400μmol·L^(-1))处理细胞,每组设3个重复孔。通过检测缺氧标志基因表达和观察细胞形态筛选氯化钴浓度,确定最适宜浓度用于建模。随后,将收集的细胞通过Illumina测序检测缺氧组和对照组鸡成肌细胞的转录表达谱,并分析差异基因和富集到的信号通路。结果表明,200μmol·L^(-1)氯化钴最适合构建鸡胚成肌细胞缺氧模型,通过转录组测序发现,在24和48 h时,缺氧组和对照组分别筛选出1 474个和2 028个差异表达基因,其中有959个差异基因在两个时间点共有。GO和KEGG分析发现,在缺氧环境下,鸡成肌细胞的信号受体活性调节、质膜组成成分、神经活性配体受体作用、PI3K-AKT、肌肉收缩等信号通路关键因子发生明显表达变化。测序和qRT-PCR结果一致显示,缺氧处理后慢肌纤维特异性基因如MYH7B、CSRP2显著上调,而快肌纤维特异性基因如MYH1F、MYH1D、TNNI2、SOX6显著下调,表明缺氧能够诱导鸡成肌细胞快肌纤维向慢肌纤维转换。本研究初步揭示了缺氧对鸡成肌细胞肌纤维类型转换的机制,并提供相关基因表达数据库,为未来家禽肉质和哺乳动物缺氧研究提供了重要参考。
