Background:Adults with obesity may display disturbed cardiac chronotropic responses during cardiopulmonary exercise testing,which relates to poor cardiometabolic health and an increased risk for adverse cardiovascular *** is unknown whether cardiac chronotropic incompetence(CI)during maximal exercise is already present in obese adolescents and,if so,how that relates to cardiometabolic ***:Sixty-nine obese adolescents(body mass index standard deviation score=2.23±0.32,age=14.1±1.2 years;mean±SD)and 29lean adolescents(body mass index standard deviation score=-0.16±0.84,age=14.0±1.5 years)performed a maximal cardiopulmonary exercise testing from which indicators for peak performance were *** resting heart rate and peak heart rate were used to calculate the maximal chronotropic response ***(lipid profile,glycemic control,inflammation,and leptin)was studied in fasted blood samples and during an oral glucose tolerance test within obese *** analyses were applied to examine associations between the presence of CI and blood or exercise capacity parameters,respectively,within obese ***:CI was prevalent in 32 out of 69 obese adolescents(46%)and 3 out of 29 lean adolescents(10%).C-reactive protein was significantly higher in obese adolescents with CI compared to obese adolescents without CI(p=0.012).Furthermore,peak oxygen uptake and peak cycling power output were significantly reduced(pmore common in the current cohort of obese adolescents,and is related to systemic inflammation and exercise intolerance.

metabolic cost of level,uphill,and downhill running in highly cushioned shoes with carbon-fiber plates

Background:Compared to conventional racing shoes,Nike Vaporfly 4% running shoes reduce the metabolic cost of level treadmill running by4%.The reduction is attributed to their lightweight,highly compliant,and resilient midsole foam and a midsole-embedded curved carbon-fiber *** investigated whether these shoes also could reduce the metabolic cost of moderate uphill(+3°) and downhill(-3°) *** tested the null hypothesis that,compared to conventional racing shoes,highly cushioned shoes with carbon-fiber plates would impart the same ~4%metabolic power(W/kg) savings during uphill and downhill running as they do during level ***:After familiarization,16 competitive male runners performed six 5-min trials(2 shoes × 3 grades) in 2 Nike marathon racing-shoe models(Streak 6 and Vaporfly 4%) on a level,uphill(+3°),and downhill(-3°) treadmill at 13 km/h(3.61 m/s).We measured submaximal oxygen uptake and carbon dioxide production during minutes 4-5 and calculated metabolic power(W/kg) for each shoe model and grade ***:Compared to the conventional shoes(Streak 6),the metabolic power in the Vaporfly 4% shoes was 3.83%(level),2.82%(uphill),and2.70%(downhill)less(all p metabolic power for uphill running was less compared to level running(p=0.04;effect size(ES)=0.561) but was not statistically different between downhill and level running(p=0.17;ES=0.356).Conclusion:On a running course with uphill and downhill sections,the metabolic savings and hence performance enhancement provided by Vaporfly 4% shoes would likely be slightly less overall,compared to the savings on a perfectly level race course.

Legume rhizobium symbiosis is initiated upon perception of bacterial secreted lipo-chitooligosaccharides （LCOs）. Perception of these signals by the plant initiates a signaling cascade that leads to nodule formation. Several studies have implicated a function for cytokinin in this process. However, whether cytokinin accu- mulation and subsequent signaling are an integral part of rhizobium LCO signaling remains elusive. Here, we show that cytokinin signaling is required for the majority of transcriptional changes induced by rhizo- bium LCOs. In addition, we demonstrate that several cytokinins accumulate in the root susceptible zone 3 h after rhizobium LCO application, including the biologically most active cytokinins, trans-zeatin and iso- pentenyl adenine. These responses are dependent on calcium- and calmodulin-dependent protein kinase （CCamK）, a key protein in rhizobial LCO-induced signaling. Analysis of the ethylene-insensitive mtein21 mtsickle mutant showed that LCO-induced cytokinin accumulation is negatively regulated by ethylene. Together with transcriptional induction of ethylene biosynthesis genes, it suggests a feedback loop negatively regulating LCO signaling and subsequent cytokinin accumulation. We argue that cytokinin accumulation is a key step in the pathway leading to nodule organogenesis and that this is tightly controlled by feedback loops.

Pseudogenes are frequently encountered noncoding sequences with a high sequence similarity to their protein-coding *** this reason,their presence is often considered troublesome in molecular *** pseudoxanthoma elasticum(PXE),a disease predominantly caused by mutations in ATPbinding cassette family C member 6(ABCC6),the presence of two pseudogenes complicates the analysis of sequence *** whole-exome sequencing(WES)becoming the standard of care in molecular diagnostics,we wanted to evaluate whether this technique is as reliable as gene-specific targeted enrichment analysis for the analysis of *** established a PCR-based targeted enrichment and next-generation sequencing testing approach and demonstrated that the ABCC6-specific enrichment combined with the applied mapping algorithm overcomes the complication of ABCC6 pseudogene aspecificities,contrary to *** propose a time-and cost-efficient diagnostic strategy for comprehensive and accurate molecular genetic testing of PXE,which is highly automatable.

Pitch size varies in official soccer matches and differently sized pitches are adopted for tactical purposes in small-sided training *** interactive team behaviour emerges under constraints,the authors evaluate the effect of pitch size(task) manipulations on interactive team behaviour in small-sided soccer *** 4-a-side(plus goalkeepers) small-sided games were played:a reference game(30×20 m),length manipulation(24×20 m),width manipulation(30×16 m),and a combination(24×16 m).Using position data(100Hz),three measures quantifying the teams' interaction were calculated:longitudinal inter-team distance,lateral inter-team distance,and surface area *** and standard deviations,correlations and coupling values were *** correlations were calculated over a 3-s window to evaluate interaction *** expected,a shorter pitch results in smaller longitudinal inter-team distance,lateral inter-team distance decreased for narrow pitches,and smaller total playing area resulted in decreased surface ***,a crossover effect was present;length and width manipulations also triggered changes in lateral and longitudinal direction ***-team distances and surface area difference differed significantly across *** patterns differed across conditions for all ***,highly tactically relevant,soccer teams seem to adapt their interactive behaviour according to pitch size in small-sided games.

Physical and chemical changes of rapeseed meal proteins during toasting and their effects on in vitro digestibility

Background： Toasting during the production of rapeseed meal（RSm） decreases ileal crude protein（CP） and amino acid（AA） digestibility. The mechanisms that determine the decrease in digestibility have not been fully elucidated. A high protein quality, low-denatured, RSm was produced and toasted up to 120 min, with samples taken every 20 min. The aim of this study was to characterize secondary structure and chemical changes of proteins and glucosinolates occurring during toasting of RSm and the effects on its in vitro CP ***： The decrease in protein solubility and the increase of intermolecular β-sheets with increasing toasting time were indications of protein aggregation. The contents of NDF and ADIN increased with increasing toasting *** of arginine, lysine and O-methylisourea reactive lysine（OmIU-RL） linearly decreased with increasing toasting time, with a larger decrease of OmIU-RL than lysine. First-order reactions calculated from the measured parameters show that glucosinolates were degraded faster than lysine, OmIU-RL and arginine and that physical changes to proteins seem to occur before chemical changes during toasting. Despite the drastic physical and chemical changes noticed on the proteins, the coefficient of in vitro CP digestibility ranged from 0.776 to 0.750 and there were no effects on the extent of protein hydrolysis after 120 min. In contrast, the rate of protein hydrolysis linearly decreased with increasing toasting time, which was largely correlated to the decrease in protein solubility, lysine and OmIU-RL observed. Rate of protein hydrolysis was more than 2-fold higher for the untoasted RSm compared to the 120 min toasted ***： Increasing the toasting time for the production of RSm causes physical and chemical changes to the proteins that decrease the rate of protein hydrolysis. The observed decrease in the rate of protein hydrolysis could impact protein digestion and utilization.

The effect of particle size and amount of inoculum on fungal treatment of wheat straw and wood chips

Background： The aim of this study was to optimize the fungal treatment of lignocellulosic biomass by stimulating the colonization. Wheat straw and wood chips were treated with Ceriporiopsis subvermispora and Lentinula edodes with various amounts of colonized millet grains（0.5, 1.5 or 3.0 % per g of wet weight of substrate） added to the substrates. Also, wheat straw and wood chips were chopped to either 0.5 or 2 *** of the fungal treatment after 0, 2, 4, 6, or 8 wk of incubation was determined by changes in chemical composition, in vitro gas production（IVGP） as a measure for rumen degradability, and ergosterol content as a measure of fungal ***： Incomplete colonization was observed for C. subvermispora treated wheat straw and L. edodes treated wood chips. The different particle sizes and amounts of inoculum tested, had no significant effects on the chemical composition and the IVGP of C. subvermispora treated wood chips. Particle size did influence *** treatment of wheat straw. The L. edodes treatment of 2 cm wheat straw resulted in a more selective delignification and a higher IVGP than the smaller particles. Addition of 1.5 % or 3 % L. edodes inoculum to wheat straw resulted in more selective delignification and a higher IVGP than addition of 0.5 % ***： Particle size and amount of inoculum did not have an effect on C. subvermispora treatment of wood chips. At least 1.5 % L. edodes colonized millet grains should be added to 2 cm wheat straw to result in an increased IVGP and acid detergent lignin（ADL） degradation.

原子力显微镜(Atomic force microscopy,AFm)作为一种纳米级高分辨率的探针扫描显微镜,在病毒形态学研究等领域有广泛的应用。为更好地利用AFm在接近病毒生理环境(液相)中观测单个病毒颗粒,选择合适的基底吸附病毒样品是重要的前提保障。本文介绍了一种病毒样品制备及AFm液相成像方法。首先制备疏水化处理的玻璃盖玻片作为吸附基底,再将观察样品腺病毒重组载体颗粒(Adenovirus type 5F35,Ad5F35)吸附于基底上,建立在液相下单个病毒颗粒的原子力显微镜成像方法。通过该方法获取了单个腺病毒颗粒在液相下的高分辨率纳米级高度图(height image),且病毒颗粒保持了约90nm的正确测量高度。实验结果表明疏水化处理的玻璃基底具有良好的吸附力,有助于病毒颗粒牢固地吸附在基底上,且不会引起较大的形变。本研究为AFm观测病毒等生物样品提供了一种较为合适的吸附基底和在液相中的AFm成像方法,对后期开展病毒形态结构、物理属性及其与宿主细胞受体相互作用的研究打下基础。
