macrophages exist in various tissues,several body cavities,and around mucosal surfaces and are a vital part of the innate immune system for host defense against many pathogens and *** possess binary m1/m2 macrophage polarization settings,which perform a central role in an array of immune tasks via intrinsic signal cascades and,therefore,must be precisely *** crucial questions about macrophage signaling and immune modulation are yet to be *** addition,the clinical importance of tumor-associated macrophages is becoming more widely recognized as significant progress has been made in understanding their ***,they are an integral part of the tumor microenvironment,playing a part in the regulation of a wide variety of processes including angiogenesis,extracellular matrix transformation,cancer cell proliferation,metastasis,immunosuppression,and resistance to chemotherapeutic and checkpoint blockade ***,we discuss immune regulation in macrophage polarization and signaling,mechanical stresses and modulation,metabolic signaling pathways,mitochondrial and transcriptional,and epigenetic ***,we have broadly extended the understanding of macrophages in extracellular traps and the essential roles of autophagy and aging in regulating macrophage ***,we discussed recent advances in macrophages-mediated immune regulation of autoimmune diseases and ***,we discussed targeted macrophage therapy to portray prospective targets for therapeutic strategies in health and diseases.

＜正＞ The expression of COX - 1 increases in rat pheochromocytoma （PC12） cells following differentiation by nerve growth factor （NGF）. Therefore, the aim of this study was to determine if NGF affected the expression of prostaglandin E2（ PGF2） receptor subtypes in PC12 cells. We tested the effects of PGE2（EP1 - 4 agonist）, sulprostone （EP3/1 agonist）, ONO- Di - 004 （EP1 agonist）, ONO - AE1 -259 （EP2 agonist）, ONO - AE - 248 （EP3 agonist） and ONO - AE1 - 329 （EP4 agonist）. PC12 cells were culture for 32 h （ ± 50 ng/ml NGF） and [3h] cAmP and [3h]inositol phosphate （[3h]iP） production was assayed in response to 1 μm agonists （ ± 1 μm forskolin）. None of the agonists tested affected [3h] iP production. PGE2 and ONO - AE1 - 259 increased [3h]cAmP in nondifferentiatedcells and ONO - AE1 - 329 was active in NGF - treated cells （P ＜ 0.05）. PGF2, sulprostone and ONO - AE - 248 inhibited forskolin - stimulated [3h] cAmP in undifferentiated cells （P ＜ 0. 01） and this response was attenuated in NGF - treated cells （P ＜ 0.05 for PGE2 and ONO - AE - 248）. in conclusion, the predominant effect of NGF on PC12 cells is to down - regulate the Gi - coupled EP3 receptor subtype.

Formulation and characterization of extruded and spheronized pellets using pectin and crosslinking agents

The aim of this study was to prepare pellets of gliclazide by extrusion spheronization process by use of two methods in situ cross linking method and interfacial complexation method and to study the effect of different cross linking agents on the formed pellets. Extrusion/spheronization being an established technique for producing spherical pellets so this technique was used to prepare pellets. The gliclazide pellets prepared by extrusion-spheronization techniques and then the formed pellets were coated with pectin solution by interfacial complexation method. The effect of cross-linkers calcium chloride and aluminium chloride concentration, by in situ cross linking on properties of gliclazide pellets like swelling and drug release were studied. The formed pellets were subjected to swelling, analysis of morphology, in dissolution and vivo studies. most of the pellets were of acceptable shape. From the results calcium chloride when coated with pectin, retarded drug release. As the concentration of calcium chloride increased, it led to slow release of the drug. This may be due to the water solubility of calcium salt, which induced cross linking with pectin. Thus pellets with calcium chloride cross linked with pectin were beneficial, because it retarded the release of the drug. Though the release from interfacial coated pellets was retarded, it was less as compared to the in situ formed pellets. The in vivo studies on alloxan-induced diabetic rats indicated the significant hypoglycemic effect that was observed 24 h after oral administration of optimized pellets. Thus, the developed and optimized pellets were suitable for prolonged systemic absorption of gliclazide to maintain lower blood glucose level and improved patient compliance. The results suggested that the in situ cross linking pellets were able to retard the release of gliclazide greater than the interfacial complexation method. Therefore, this approach has been effectively achieved.

introduction: Burkina Faso experiences regular cases of streptococcus pneumoniae meningitis. As part of the strategy to reduce cases of meningitis, the 13-valent pneumococcal conjugate vaccine (PCV-13) has been introduced in the Expanded Programme on immunisation (EPi). Despite these efforts, there are some cases of pneumococcal meningitis including both vaccine and non-vaccine serotypes. The objective of this study was to describe the pneumococcal sequence types (sT) circulating in Burkina Faso before the introduction of the 13-valent pneumococcal conjugate vaccine (PCV-13). methods: it was a descriptive cross-sectional study that took place from 27th October 2013 to 7th January 2014. s. pneumoniae strains were collected in Burkina Faso and multi Locus sequence Typing (mLsT) was performed at the Pneumococcal Laboratory at the Centers for Disease Control and Prevention (CDC) in the UsA (United states of America). mLsT consists of 4 steps: amplification, purification, sequencing and interpretative reading of the results. The amplification used 7 primers consisting of sequences of aroe, gdh, gki, recP, spi, xpt, ddl genes. Results: Of 37 strains tested, 10 serotypes were identified. serotype 1 was prevalent in 48.7% (18/37) followed by serotype 25F in 10.8% (4/37). serotypes 5 and 12F/12A/12B/44/46 were 8.1% (3/37) each. serotype 1 contained 5 sTs including sT303 24.3% (9/37), sT217 8.1% (3/37) and sT618 8.1% (3/37);followed by serotype 25F with sT105 10.8% (4/37), serotype 5 with sT289 8.1% (3/37) and serogroup 12F/12A/12B/44/46 with sT 989 8.1% (3/37). Conclusion: Pneumococci are characterised by their great variability both in number of serotypes and in sT within the same serotype. Thus, 10 serotypes have been identified. Also, within serotype 1, 5 different sTs have been described. These data indicate the complexity of the pneumococcus which is strongly involved in purulent bacterial meningitis at national level. Nutritional screening in a University hospital:Comparison between Oncologic and Non-Oncologic Patients

Background & Aims: We compared the screening nutritional data of patients with malignant and non-malignant disease, and classified their nutritional risk according to the primary tumor’s site. methods: subjective Global Assessment was applied to 3008 patients within 48 h of admission to a public university. subjects were divided into Oncologic Group (n = 576) or Non-Oncologic Group (n = 2432) according to the presence of neoplasms or other diseases respectively. These data were compared by the t-student test and classic chi-square test, with statistical significance set at p sults: The Oncologic Group presented a higher prevalence of weight loss (60.3% vs. 49.3%), quantitative (40.7% vs. 28.5%) and qualitative (16.4% vs. 7.6%) alterations in the food intake pattern, gastrointestinal symptoms and muscle waste (23.2% vs. 13.2%) as compared to the Non-Oncologic Group. Also, there was a higher prevalence of nutritional disturbances in the Oncologic Group, with 49.5% of the subjects moderately (or suspected to be malnourished) and 12.7% severely mal-nourished. severe malnourishment was mainly observed in patients with head and neck (25%), upper digestive tract (21.9%) and soft tissue and bones (17.9%) tumors. Conclusion: When compared to other hospitalized patients, patients with neoplastic disease were in higher nutritional risk.

Predisposing factors’effects on mortality of oak (Quercus) and hickory (Carya) species in mature forests undergoing mesophication in Appalachian Ohio

Background:mature oak(Quercus spp.)and hickory(Carya spp.)trees are gradually being replaced by more shadetolerant tree species across the eastern U.s.,likely due to fire suppression and increased *** and hickories are highly valuable to wildlife;therefore,studying their mortality patterns can provide information on the longevity of habitat quality for many animal *** mortality has most often been studied following large oak decline events,but background mortality rates in forests with aging oak and hickory canopies warrant equal attention,especially in the context of widespread oak and hickory regeneration ***:We studied background mortality rates of five oak and one hickory species over a 23–25 year time period(1993–1995 to 2018),using 821/20th hectare permanent plots on the marietta Unit of the Wayne National Forest in southeastern *** calculated mortality rates based on remeasurement of individual trees for white oak(Quercus alba),chestnut oak(Quercus montana),northern red oak(Quercus rubra),black oak(Quercus velutina),scarlet oak(Quercus coccinea),and pignut hickory(Carya glabra).For each of these species other than scarlet oak,we also modeled the relationships of mortality probability with a priori topographic,soil,stand structural,and individual tree covariates,using a mixed-effects logistic regression ***:The species with the highest mortality rate was scarlet oak(61.3%),followed by northern red oak(41.4%),black oak(26.7%),pignut hickory(23.9%),white oak(23.4%),and chestnut oak(19.1%).in our models,northern red oak mortality was associated with more mesic slope positions,shallower solums,more acidic soils,and older stand *** hickory and chestnut oak mortality rates were associated with higher basal areas on the plot,while white oak mortality showed the opposite ***:Our data suggest that red oak subgenus trees in mature forests of our area will become increasingly uncommon relative t
