Identification of QTL and underlying genes for root system architecture associated with nitrate nutrition in hexaploid wheat

The root system architecture(RSA) of a crop has a profound effect on the uptake of nutrients and consequently the potential yield. However, little is known about the genetic basis of RSA and resource adaptive responses in wheat(Triticum aestivum L.). Here, a high-throughput germination paper-based plant phenotyping system was used to identify seedling traits in a wheat doubled haploid mapping population, Savannah×Rialto. Significant genotypic and nitrate-N treatment variation was found across the population for seedling traits with distinct trait grouping for root size-related traits and root distribution-related traits. Quantitative trait locus(QTL) analysis identified a total of 59 seedling trait QTLs. Across two nitrate treatments, 27 root QTLs were specific to the nitrate treatment. Transcriptomic analyses for one of the QTLs on chromosome 2 D, which was found under low nitrate conditions, revealed gene enrichment in N-related biological processes and 28 differentially expressed genes with possible involvement in a root angle response. Together, these findings provide genetic insight into root system architecture and plant adaptive responses to nitrate, as well as targets that could help improve N capture in wheat.

Background: modelling aboveground biomass(AGB) in forest and woodland ecosystems is critical for accurate estimation of carbon stocks. However, scarcity of allometric models for predicting AGB remains an issue that has not been adequately addressed in Africa. In particular, locally developed models for estimating AGB in the tropical woodlands of Ghana have received little attention. In the absence of locally developed allometric models, Ghana will continue to use Tier 1 biomass data through the application of pantropic models. Without local allometric models it is not certain how Ghana would achieve Tier 2 and 3 levels under the United Nations programme for reducing emissions from deforestation and forest degradation. The objective of this study is to develop a mixedspecies allometric model for use in estimating AGB for the tropical woodlands in Ghana. Destructive sampling was carried out on 745 trees(as part of charcoal production) for the development of allometric equations. Diameter at breast height(dbh, i.e. 1.3 m above ground level), total tree height(H) and wood density(ρ) were used as predictors for the models. Seven models were compared and the best model selected based on model efficiency,bias(%) and corrected Akaike Information Criterion. The best model was validated by comparing its results with those of the pantropic model developed by Chave et al.(Glob Chang Biol 20:3177–3190, 2014) using equivalence test and conventional paired ***: The results revealed that the best model for estimating AGB in the tropical woodlands is AGB =0.0580ρ((dbh)2 H)0.999. The equivalence test showed that this model and the pantropic model developed by Chave et al.(Glob Chang Biol 20:3177–3190, 2014) were equivalent within ±10% of their mean predictions(p-values mean(181.44 ± 18.25 kg) of the model predictions of the best model of this s

Frequency response analysis (FRA) is regarded as the most effective technique to detect mechanical faults of transformers. Over the years, FRA measurement data have been collected by utilities into transformer asset databases. The characteristic of FRA data is fundamentally determined by the transformer's equivalent electrical circuit, which consists of inductance and capacitance parameters that are windings' design and structure dependent. Different winding types tend to have different FRA characteristics, and a transformer's design information such as winding type, dimension etc. is often not known to the utility but critically important for asset management. This study reviews the state-of-the-art transformer FRA databases and application of machine learning techniques in this field, and proposes to apply a support vector machine (SVm) model onto the FRA data to identify the winding type. The SVm model is first trained by FRA traces of transformers with known winding types, and after testing, the SVm model is then applied to FRA traces with unknown winding information. A set of data from the UK's National Grid FRA database, was used to demonstrate and verify the SVm model. All transformers used in this study are 400/275/13 kV transmission transformers, which were designed using four different winding types, namely multiple layer, plain disc, interleaved disc and single helical windings. The proposed method can successfully identify the correct winding type.

Background and Aims: Accurate endoscopic detection of premalignant lesions and earlycancers in the colon is essential for cure, since prognosis is closely related to lesion size andstage. Although it has great clinical potential, autofluorescence endoscopy has limited tumorto-normal tissue image contrast for detecting small preneoplastic lesions. We have developed amolecularly specific, near-infrared fluorescent monoclonal antibody (CC49) bioconjugate whichtargets tumor-associated glycoprotein 72 (TAG72), as a contrast agent to improve fluorescencebased endoscopy of colon cancer. methods: The fluorescent anti-TAG72 conjugate was evaluated in vitro and in vivo in athymic nude mice bearing human colon adenocarcinoma (LS174T)subcutaneous tumors. Autofluorescence, a fluorescent but irrelevant antibody and the free fluorescent dye served as controls. Fluorescent agents were injected intravenously, and in vivowhole body fluorescence imaging was performed at various time points to determine pharmacokinetics, followed by ex vivo tissue analysis by confocal fluorescence microscopy and histology Results: Fluorescence microscopy and histology confirmed specific LS174T cell membrane targeting of labeled CC49 in vitro and ex vivo. In vivo fluorescence imaging demonstrated significant tumor-to-normal tissue contrast enhancement with labeled-CC49 at three hours postinjection, with maximum contrast after 48 h. Accumulation of tumor fluorescence demonstratedthat modification of CC49 antibodies did not alter their specific tumor-localizing properties, andwas antibody-dependent since controls did not produce detectable tumor fluorescence. Conclusions: These results show proof-of-principle that our near-infrared fluorescent-antibody probetargeting a tumor-associated mucin detects colonic tumors at the molecular level in real time,and offer a basis for future improvement of image contrast during clinical fluorescence endoscopy.

Background:Higher levels of cardiorespiratory fitness are associated with reduced asthma severity and increased quality of life in those with ***,the purpose of this study was to evaluate the effectiveness of a 6-month high-intensity interval training(HIIT)intervention in adolescents with and without ***:A total of 616 adolescents(334 boys;13.0±1.1 years,1.57±0.10 m,52.6±12.9 kg,mean±SD),including 155 with asthma(78 boys),were recruited as part of a randomized controlled trial from 5 schools(4 control and 1 intervention).The 221 intervention participants(116 boys;47 asthma)completed 6 months of school-based HIIT(30 min,3 times per week,10-30 s bouts at>90%age-predicted maximum heart rate with equal rest).At baseline,mid-intervention,post-intervention,and 3-month follow-up,measurements for 20-m shuttle run,body mass index(BmI),lung function,Pediatric Quality of Life Inventory,Paediatric Asthma Quality of Life Questionnaire,and Asthma Control Questionnaire were ***,69 adolescents(39 boys(of the 36 with asthma there were 21 boys))also completed an incremental ramp *** analysis,each group’s data(intervention and control)were divided into those with and without ***:Participants with asthma did not differ from their peers in any parameter of aerobic fitness,at any time-point,but were characterized by a greater *** intervention elicited a significant improvement in maximal aerobic fitness but no change in sub-maximal parameters of aerobic fitness,lung function,or quality of life irrespective of asthma *** in the intervention group maintained their BmI,whereas BmI significantly increased in the control group throughout the 6-month ***:HIIT represents an effective tool for improving aerobic fitness and maintaining BmI in adolescents,irrespective of asthma *** was well-tolerated by those with asthma,who evidenced a similar aerobic fitness to their healthy peers and responded equally

The phosphate adsorption and surface charge characteristics of the tropical and subtropical soils derived from different parent materials in China were determined, and their relations to soil mineralogy were *** results showed that all soil phosphate adsorption curves were well fitted by Freundlich equation and Langmuir equation. The maximum buffering capacity of P ranged from 66 to 9880 mg kg-1, with an increasing order of purple soil, skeletal soil, red soil, lateritic red soil, yellow soil and latosol; and the highest value was 149 times the lowest value, which indicated great differences among these soils in phosphate adsorption and supplying characteristics. The pHo (zero point of charge) values obtained by salt titrationpotential titration varied from 3.03 to 5.49, and the highest value was found in the latosol derived from basalt whereas the lowest value was found in the purple soil. The correlation analysis indicated that the main minerals responsible for phosphate adsorption in the soils were gibbsite, amorphous iron oxide and kaolinite; and the pHo was mainly controlled by kaolinite, gibbsite and oxides.
