Background:Axons,crucial for impulse transmission and cellular trafficking,are thought to be primary targets of neurodegeneration in Parkinson’s disease(Pd)and dementia with Lewy bodies(dLB).Axonal degeneration occurs early,preceeding and exceeding neuronal loss,and contributes to the spread of pathology,yet is poorly described outside the nigrostriatal *** insula,a cortical brain hub,was recently discovered to be highly vulnerable to pathology and plays a role in cognitive deficits in Pd and *** aim of this study was to evaluate morphological features as well as burden of proteinopathy and axonal degeneration in the anterior insular sub-regions in Pd,Pd with dementia(Pdd),and ***:α-Synuclein,phosphorylated(p-)tau,and amyloid-βpathology load were evaluated in the anterior insular(agranular and dysgranular)subregions of post-mortem human brains(n=27).Axonal loss was evaluated using modified Bielschowsky silver staining and quantified using *** damage was comprehensively studied using immunofluorescent multi-labelling and 3d confocal laser-scanning ***:Compared to Pd and Pdd,dLB showed significantly higherα-synuclein and p-tau pathology load,argyrophilic grains,and more severe axonal loss,particularly in the anterior agranular ***,the dysgranular insula showed a significantly higher load of amyloid-βpathology and its axonal density correlated with cognitive performance.p-Tau contributed most to axonal loss in the dLB group,was highest in the anterior agranular insula and significantly correlated with CdR global scores for *** and myelin showed degenerative changes including swellings,demyelination,and detachment of the axon-myelin ***:Our results highlight the selective vulnerability of the anterior insular sub-regions to various converging pathologies,leading to impaired axonal integrity in Pd,Pdd and dLB,disrupting their functional properties and potentially con

In digital soil mapping(dSM),a fundamental assumption is that the spatial variability of the target variable can be explained by the predictors or environmental *** to adequately sample the predictors have been well documented,with the conditioned Latin hypercube sampling(cLHS)algorithm receiving the most attention in the dSM *** advances in sampling design,a critical gap remains in determining the number of samples required for dSM *** propose a simple workflow and function coded in R language to determine the minimum sample size for the cLHS algorithm based on histograms of the predictor variables using the Freedman-diaconis rule for determining optimal bin *** preprocessing was included to correct for multimodal and non-normally distributed data,as these can affect sample size determination from the *** on a user-selected quantile range(QR)for the sample plan,the densities of the histogram bins at the upper and lower bounds of the QR were used as a scaling factor to determine minimum sample *** technique was applied to a field-scale set of environmental covariates for a well-sampled agricultural study site near Guelph,Ontario,Canada,and tested across a range of *** results showed increasing minimum sample size with an increase in the QR *** sample size increased from 44 to 83 when the QR increased from 50% to 95% and then increased exponentially to 194 for the 99%*** technique provides an estimate of minimum sample size that can be used as an input to the cLHS algorithm.

The sedimentary sequence of the upper 4 m deposits in the Huanghe Harbour Pit is as follows, 1, distributary mouth bar deposits formed before 1964; 2. lower delta plain (tidal flat) and subaqueous delta lateral deposits formed in the period of 1964-1972; 3. deposits of crevasse splay and distributary channel near mouth, formed in the period of 1972-1974; 4. tidal flat deposits formed in the period of 1975-1984. The sequence shows superimposition of sedimentary environments parallel to coastline.

Microfluidic systems enable automated and highly parallelized cell culture with low volumes and defined liquid *** achieve this,systems typically integrate all functions into a single,monolithic device as a“one size fits all”***,this approach limits the end users’(re)design flexibility and complicates the addition of new functions to the *** address this challenge,we propose and demonstrate a modular and standardized plug-and-play fluidic circuit board(FCB)for operating microfluidic building blocks(MFBBs),whereby both the FCB and the MFBBs contain integrated valves.A single FCB can parallelize up to three MFBBs of the same design or operate MFBBs with entirely different *** operation of the MFBBs through the FCB is fully automated and does not incur the cost of an extra external *** use this modular platform to control three microfluidic large-scale integration(mLSI)MFBBs,each of which features 64 microchambers suitable for cell culturing with high spatiotemporal *** show as a proof of principle that we can culture human umbilical vein endothelial cells(HUVECs)for multiple days in the chambers of this ***,we also use the same FCB to control an MFBB for liquid dosing with a high dynamic *** results demonstrate that MFBBs with different designs can be controlled and combined on a single *** novel modular approach to operating an automated microfluidic system for parallelized cell culture will enable greater experimental flexibility and facilitate the cooperation of different chips from different labs.

Background Asymptomatic carriers of leucine-rich repeat kinase 2(LRRK2)gene mutations constitute an ideal population for discovering prodromal biomarkers of Parkinson’s disease(Pd).In this study,we aim to identify CSF candidate risk biomarkers of Pd in individuals with LRRK2 mutation *** We measured the levels of CSF total-(t-),oligomeric(o-)and phosphorylated S129(pS129-)α-syn,total-tau(tTau),phosphorylated threonine 181 tau(pTau),amyloid-beta 40(Aβ-40),amyloid-beta-42(Aβ-42)and 40 inflammatory chemokines in symptomatic(n=23)and asymptomatic(n=51)LRRK2 mutation carriers,subjects with a clinical diagnosis of Pd(n=60)and age-matched healthy controls(n=34).General linear models corrected for age and gender were performed to assess differences in CSF biomarkers between the *** that varied significantly between the groups were then analyzed using backward-elimination logistic regression analysis to identify an ideal biomarkers panel of prodromal *** discriminant function analysis revealed low levels of CSF t-α-syn,high levels of CSF o-α-syn and TNF-αbest discriminated asymptomatic LRRK2 mutation carriers from both symptomatic Pd and healthy *** the discriminative power using receiver operating curve analysis,an area under the curve>0.80 was *** The current study suggests that CSF t-,o-α-syn and TNF-αare candidate risk biomarkers for the detection of Pd at the prodromal *** findings also highlight the dynamic interrelationships between CSF proteins and the importance of using a biomarkers’panel approach for an accurate and timely diagnosis of Pd.

AIM: To investigate relapse predictors in chronic hepatitis C (CHC) patients with end-of-treatment response (ETR), after pegylated interferon-α (PegIFN-α) and ribavirin treatment. METHOdS: In a retrospective study we evaluated a spectrum of predictors of relapse after PegIFN-α and ribavirin treatment in 86 CHC patients with ETR. Viral loads were determined with real-time reverse transcrip-tion polymerase chain reaction. Hepatitis C virus geno-typing was performed by sequencing analysis. Patients with genotype 1 were treated for 48 wk with 180 μg PegIFN-α2a or 1.5 μg/kg PegIFN-α2b once weekly plus ribavirin at a dosage of 1000 mg/d for those under 75 kg or 1200 mg/d for those over 75 kg. Patients with geno- types 2 and 3 were treated for 24 wk with 180 μgPegIFN-α2a or 1.5 μg/kg PegIFN-α2b once weekly plus ribavirin at a dosage of 800 mg/d. RESULTS: In all ETR patients, binary logistic regression analysis identif ied absence of complete early virological response (cEVR) (OR 27.07, 95% CI: 3.09-237.26, P d body mass index > 26 kg/m2 (OR: 8.27, 95% CI: 2.22-30.84, P dependent predictors of relapse. When cEVR patients were analyzed exclusively, ALP prior to therapy d the only predictor of relapse. CONCLUSION: Lower levels of ALP prior to, during and after therapy seem to be associated with a higher risk of relapse in CHC patients with ETR.

AIM To compare features of hepatocellular carcinoma(HCC) in Hispanics to those of African Americans and *** Patients treated for HCC at an urban tertiary medical center from 2005 to 2011 were identified from a tumor registry. data were collected retrospectively, including demographics, comorbidities, liver disease characteristics, tumor parameters, treatment, and survival(OS) outcomes. OS analyses were performed using Kaplan-Meier *** One hundred and ninety-five patients with HCC were identified: 80.5% were male, and 22% were age 65 or older. Mean age at HCC diagnosis was 59.7 ± 9.8 years. Sixty-one point five percent of patients had Medicare or Medicaid; 4.1% were uninsured. Compared to African American(31.2%) and White(46.2%) patients, Hispanic patients(22.6%) were more likely to have diabetes(P = 0.0019), hyperlipidemia(P = 0.0001), nonalcoholic steatohepatitis(NASH)(P = 0.0021), end stage renal disease(P = 0.0057), and less likely to have hepatitis C virus(P d to African Americans, Hispanics were more likely to meet criteria for metabolic syndrome(P = 0.0491), had higher median MELd scores(P = 0.0159), ascites(P = 0.008), and encephalopathy(P = 0.0087). Hispanic patients with HCC had shorter OS than the other racial groups(P = 0.020), despite similarities in HCC parameters and treatment. CONCLUSION In conclusion, Hispanic patients with HCC have higher incidence of modifiable metabolic risk factors including NASH, and shorter OS than African American and White patients.
