Treatment with a JAK1/2 inhibitor ameliorates murine autoimmune cholangitis induced by IFN overexpression

The interferon(IFN)signaling pathways are major immunological checkpoints with clinical significance in the pathogenesis of *** have generated a unique murine model named ARE-Del,with chronic overexpression of IFNγ,by altering IFNγ***,these mice develop an immunologic and clinical profile similar to patients with primary biliary cholangitis,including high titers of autoantibodies and portal *** hypothesized that the downregulation of IFN signaling pathways with a JAK1/2 inhibitor would inhibit the development and progression of *** study this hypothesis,ARE-Del^(+/−)mice were treated with the JAK1/2 inhibitor ruxolitinib and serially *** inhibition resulted in a significant reduction in portal inflammation and bile duct damage,associated with a significant reduction in splenic and hepatic CD4^(+)T cells and CD8^(+)T ***,ruxolitinib inhibited the secretion of the proinflammatory cytokines IFNγand TNF from splenic CD4^(+)T ***,ruxolitinib treatment also decreased the frequencies of germinal center B(GC B)cells and T follicular helper(Tfh)cells and led to lower serological AmA *** note,liver and peritoneal macrophages were sharply decreased and polarized from m1 to m2 with a higher level of IRF4 expression after ruxolitinib ***,ruxolitinib inhibited the secretion of IL-6,TNF and mCP1 and the expression of STAT1 but promoted the expression of STAT6 in macrophages in vitro,indicating that m1 macrophage polarization to m2 occurred through activation of the STAT6-IRF4 *** data highlight the significance,both immunologically and clinically,of the JAK/STAT signaling pathway in autoimmune cholangitis.

Liver-resident NK cells are distinct from conventional NK cells and play an important role in the maintenance of liver *** liver-resident NK cells participate in autoimmune cholangitis remains ***,we extensively investigated the impact of NK cells in the pathogenesis of autoimmune cholangitis utilizing the well-established dnTGFβRII cholangitis model,NK cell-deficient(Nfil3−/−)mice,adoptive transfer and in vivo antibody-mediated NK cell *** data demonstrated that disease progression was associated with a significantly reduced frequency of hepatic NK *** of NK cells resulted in exacerbated autoimmune cholangitis in dnTGFβRII *** further confirmed that the DX5−CD11c^(hi) liver-resident NK cell subset colocalized with CD4^(+) T cells and inhibited CD4^(+) T cell *** expression microarray analysis demonstrated that liver-resident NK cells had a distinct gene expression pattern consisting of the increased expression of genes involved in negative regulatory functions in the context of the inflammatory microenvironment.

Autoimmune cholangitis arises from abnormal innate and adaptive immune responses in the liver,and T cells are critical drivers in this ***,little is known about the regulation of their functional behavior during disease *** previously reported that mice with T cell-restricted expression of a dominant negative form of transforming growth factor beta receptor type Ⅱ(dnTGFβ RII)spontaneously develop an autoimmune cholangitis that resembles human primary biliary cholangitis(PBC).Adoptive transfer of CD8^(+) but not CD4^(+) T cells into Rag1^(−/−)mice reproduced the disease,demonstrating a critical role for CD8^(+) T cells in PBC ***,we used SOmAscan technology to perform proteomic analysis of serum samples from dnTGFβRII and B6 control mice at different *** addition,we analyzed CD8 protein profiles after adoptive transfer of splenic CD8^(+) cells into Rag1^(−/−)*** use of the unique SOmAscan aptamer technology revealed critical and distinct profiles of CD8 cells,which are key to biliary *** total,254 proteins were significantly increased while 216 proteins were significantly decreased in recipient hepatic CD8^(+) cells compared to donor splenic CD8^(+) *** contrast to donor splenic CD8^(+) cells,recipient hepatic CD8^(+) cells expressed distinct profiles for proteins involved in chemokine signaling,focal adhesion,T cell receptor and natural killer cell-mediated cytotoxicity pathways.

COmPETING REACTIONS OF EXISTING Ni SILICIDE AND Ni OR Si INDUCED BY THERmAL ANNEALING AND meV Si ION BEAm mIXING

The competing reactions between existing Ni silicides surrounded by Si and Ni were investigated by thermal annealing and meV Si ion beam mixing. With high energy irradiation, the energy deposition at both interfaces, Ni/Ni silicide and Ni silicide/Si, is equal. Two meV He^- RBS and TEm were used to obtain the reacted layer composition and epitaxial orientation, respectively. Also glancing angle Co K_a. X-ray diffraction was utilized to identify phase formation. The main results indicate that the existing silicides preferentially react with Ni layer, and that there are pronounced differences of Ni silicide phase transition between thermal annealing and meV Si ion beam mixing, even though the mixing was performed in radiation enhanced diffusion regime. The results can be explained in term of the heat of silicide formation and surface energy change.

The Aerosol-monsoon Climate System of Asia:A New Paradigm

This commentary is based on a series of recent lectures on aerosol-monsoon interactions I gave at the Beijing Normal University in August 2015. A main theme of the lectures is on a new paradigm of ＂An Aerosol-monsoon-Climate-System＂, which posits that aerosol, like rainfall, cloud, and wind, is an integral component of the monsoon climate system, influencing monsoon weather and climate on m1 timescales. Here, salient issues discussed in my lectures and my personal perspective regarding interactions between atmospheric dynamics and aerosols from both natural and anthropogenic sources are summarized. my hope is that under this new paradigm, we can break down traditional disciplinary barriers, advance a deeper understanding of weather and climate in monsoon regions, as well as entrain a new generation of geoscientists to strive for a sustainable future for one of the most complex and challenging human-natural climate sub-system of the earth.

Assessment of mercury(II) bioavailability using a bioluminescent bacterial biosensor: Practical and theoretical challenges

Critical methodological challenges in the microbial biosensor approach to assessing Hg（II） bioavailability were evaluated from the perspective of analytical chemists. The main challenge stems from the fact that the chemical speciation of Hg（II） in natural waters exerts a major control on its bioavailability, yet its natural complexation equilibria are extensively altered during conventional bioassays. New data, obtained using a bioluminescent Hg（II）-biosensor, that illustrate these challenges are presented and potential solutions proposed.

Predictive value of hypo-osmotic swelling test to identifyviable non-motile sperm

Aim: To determine the predictive value of the hypo-osmotic swelling (HOS) test to identify viable, non-motile sperm. methods: Semen samples from 20 men with severe asthenozoospermia underwent traditional seminal analysis, eosin-nigrosin (EN) staining and the HOS test. A further EN stain was then performed on a HOS pre-treated aliquot and a total of 2000 further sperm examined. Results: The median sperm density was 5.1 million/mL (IQR 4.3-13.1) and the median motility was 3.0 % (IQR 0-7). Seven samples showed complete asthenozoospermia. Initial EN staining showed 59 % viability (range 48-69) despite the poor standard parameters and 47 % (range 33-61) in the complete asthenozoospermia subgroup. The HOS test showed 49.9 % reacted overall (range 40-59) and 41.7 % (range 22-61) in the complete asthenozoospermia subgroup. The combined HOS/EN stain showed the positive predictive value of the HOS test to identify viable sperm was 84.2 % overall and 79.7 % in the complete asthenozoospermia subgroup. Conclusion: The HOS test can effectively predict sperm viability in patients with severe and complete asthenozoospermia.

Reproductive biology of an old endemic murid rodent of Australia, the Spinifex hopping mouse, Notomys alexis:adaptations for life in the arid zone

The Spinifex hopping mouse(Notomys alexis Thomas,1922)is an arid adapted Australo–Papuan old endemic rodent that undergoes boom and bust population *** this communication,we review our findings on the timing of reproduction and the potential reproductive rate of this *** investigate the first question,the reproductive condition of adult females,and occurrence of immatures,in a population on a cattle station in central Australia was determined and,for the second,data from a laboratory colony compared to those previously published on closely related species in other *** findings show that,at least in the population at the times that monitoring was performed,reproductive activity was only taking place in spring and early summer,whereas the laboratory study indicates that females have a similar gestation length and litter size to those of most close relatives occurring in other *** have extremely small testes and store relatively few *** findings suggest that Notomys alexis might show some seasonality of reproduction at least in this region of central Australia and that this species does not have a higher reproductive rate than that of related species in other,more predictable,environments.

Industry mix and Curvilinear Spillovers from FDI in China

This paper uses industry and province specific Chinese industrial data to demonstrate a potential causal link between two strands of the FDI literature. The first strand suggests that the impact of spillovers from inward FDI is less robust in middle-income economies than in either high-income or low-income economies. The second strand suggests diminishing returns of inward FDI on horizontal labor productivity in low-technology industries but not in high-technology industries. This paper suggests a link between these two phenomena. Specifically, if both FDI intensity and industry mix vary with the level of economic development, then an industry-dependent relationship between inward FDI and horizontal spillovers could cause middle-income economies to derive fewer benefits from inward FDI than either high- or low-income economies. This paper also verifies the curvilinear relationship between FDI in low-technology industries and horizontal labor productivity without relying on problematic FDI from Hong Kong, Taiwan and macao.
