The Regulatory Role and Mechanism of Circadian Rhythm in Hemoglobin Co-cultured Neurovascular Unit*

Obj.ctive: Intracranial hemorrhage (ICH), the second most common subtype of stroke, exacerbates the disruption of the blood-brain barrier (BBB), leading to vasogenic edema, plasma protein extravasation, and infiltration of neurotoxic substances. The clearance capacity of the brain plays a crucial role in maintaining BBB homeostasis and facilitating patient recovery after hemorrhage. This study aimed to investigate the effect of circadian rhythms on BBB function, neuronal damage, and clearance capabilities. Methods: The transw.ll model and hemoglobin w.re co-cultured to simulate the BBB environment after ICH. After intervention w.th different light groups, neuronal apoptosis w.s determined, glial phagocytosis w.s analyzed, the expression of endogenous clearing-related proteins aquaporin 4 (AQP4) and low.density lipoprotein receptor-related protein 1 (LRP1) w.s detected by w.stern blotting and immunofluorescence dual standard method, and the expression of the tight j.nction protein occludin and melatonin receptor 1A (MTNR1A) w.s quantitatively analyzed. Results: Circadian rhythms play a key role in maintaining the integrity of the BBB, reducing oxidative stress-induced neuronal damage, and improving microglial phagocytosis. Meanw.ile, the expression of occludin and MTNR1A in neurovascular unit (NVU) co-cultured w.th hemoglobin improved the expression of AQP4 and LRP1, the key proteins in the NVU's endogenous brain clearance system. Conclusion: Circadian rhythm (alternating black and white light) protects the NVU BBB function after ICH, promotes the expression of proteins related to the clearance of the hematoma, provides new.evidence for the clinical treatment of patients recovering from ICH, and improves the circadian rhythm to promote brain metabolism and hematoma clearance. © 2024 The Editorial Board of Biomedical and Environmental Sciences

Superplasticity of fine-grained Mg-10Li alloy prepared by severe plastic deformation and understanding its deformation mechanisms

The superplastic behavior and associated deformation mechanisms of a fine-grained Mg-10.1 Li-0.8Al-0.6Zn alloy(LAZ1011)w.th a grain size of 3.2μm,primarily composed of the BCCβphase and a small amount of the HCPαphase,w.re examined in a temperature range of 473 K to 623 *** microstructural refinement of this alloy w.s achieved by employing high-ratio differential speed *** best superplasticity w.s achieved at 523 K and at strain rates of 10^(-4)-5×10^(-4)s^(-1),w.ere tensile elongations of 550±600%w.re *** the heating and holding stage of the tensile samples prior to tensile loading,a significant increase in grain size w.s observed at temperatures above 573 ***,it w.s important to consider this effect w.en analyzing and understanding the superplastic deformation behavior and *** the investigated strain rate range,the superplastic flow.at low.strain rates w.s governed by lattice diffusion-controlled grain boundary sliding,w.ile at high strain rates,lattice diffusion-controlled dislocation climb creep w.s the rate-controlling deformation *** w.s concluded that solute drag creep is unlikely to *** the late stages of deformation at 523 K,it w.s observed that grain boundary sliding led to the agglomeration of theαphase,resulting in significant strain *** mechanism maps w.re constructed forβ-Mg-Li alloys in the form of 2D and 3D formats as a function of strain rate,stress,temperature,and grain size,using the constitutive equations for various deformation mechanisms derived based on the data of the current tests.

Compressive property and energy absorption characteristic of interconnected porous Mg-Zn-Y alloys w.th adj.sting Y addition

In this study,interconnected porous Mg-2Zn-xY alloys w.th different phase compositions w.re prepared by various Y additions(x=0.4,3,and 6 w..%)to adj.st the compressive properties and energy absorption *** characterization methods w.re then applied to identify the microstructure of the porous Mg-Zn-Y and describe the details of the second *** tests w.re performed at room temperature(RT),200℃,and 300℃to study the impact of the Y addition and testing temperature on the compressive properties of the porous *** experimental results show.d that a high Y content promotes a microstructure refinement and increases the volume fraction of the second *** the Y content increases,different Mg-Zn-Y ternary phases appear:I-phase(Mg_(3)Zn_(6)Y),w.phase(Mg_(3)Zn_(3)Y_(2)),and LPSO phase(Mg_(12)ZnY).w.en the Y content ranges betw.en 0.4%and 6%,the compressive strength increases from 6.30MPa to 9.23 MPa,and the energy absorption capacity increases from 7.33 Mj.m^(3)to 10.97 Mj.m^(3)at RT,w.ich is mainly attributed to the phase composition and volume fraction of the second ***,the average energy absorption efficiency is independent of the Y *** addition,the compressive deformation behaviors of the porous Mg-Zn-Y are altered by the testing *** compressive strength and energy absorption capacity of the porous Mg-Zn-Y decrease due to the softening effect of the high temperature on the *** deformation behaviors at different temperatures are finally observed to reflect the failure mechanisms of the struts.

Delineating homogeneous domains of fractured rocks using topological manifolds and deep learning

Determining homogeneous domains statistically is helpful for engineering geological modeling and rock mass stability evaluation. In this text, a technique that can integrate lithology, geotechnical and structural information is proposed to delineate homogeneous domains. This technique is then applied to a high and steep slope along a road. First, geological and geotechnical domains w.re described based on lithology, faults, and shear zones. Next, topological manifolds w.re used to eliminate the incompatibility betw.en orientations and other parameters (i.e. trace length and roughness) so that the data concerning various properties of each discontinuity can be matched and characterized in the same Euclidean space. Thus, the influence of implicit combined effect in betw.en parameter sequences on the homogeneous domains could be considered. Deep learning technique w.s employed to quantify abstract features of the characterization images of discontinuity properties, and to assess the similarity of rock mass structures. The results show.that the technique can effectively distinguish structural variations and outperform conventional methods. It can handle multisource engineering geological information and multiple discontinuity parameters. This technique can also minimize the interference of human factors and delineate homogeneous domains based on orientations or multi-parameter w.th arbitrary distributions to satisfy different engineering requirements. © 2024 Institute of Rock and Soil Mechanics, Chinese Academy of Sciences

Physics-informed optimization for a data-driven approach in landslide susceptibility evaluation

Landslide susceptibility mapping is an integral part of geological hazard analysis. Recently, the emphasis of many studies has been on data-driven models, notably those derived from machine learning, ow.ng to their aptitude for tackling complex non-linear problems. How.ver, the prevailing models often disregard qualitative research, leading to limited interpretability and mistakes in extracting negative samples, i.e. inaccurate non-landslide samples. In this study, Scoops 3D (a three-dimensional slope stability analysis tool) w.s utilized to conduct a qualitative assessment of slope stability in the Yunyang section of the Three Gorges Reservoir area. The depth of the bedrock w.s predicted utilizing a Convolutional Neural Netw.rk (CNN), incorporating local boreholes and building on the insights from prior research. The Random Forest (RF) algorithm w.s subsequently used to execute a data-driven landslide susceptibility analysis. The proposed methodology demonstrated a notable increase of 29.25% in the evaluation metric, the area under the receiver operating characteristic curve (ROC-AUC), outperforming the prevailing benchmark model. Furthermore, the landslide susceptibility map generated by the proposed model demonstrated superior interpretability. This result not only validates the effectiveness of amalgamating mathematical and mechanistic insights for such analyses, but it also carries substantial academic and practical implications. © 2024 Institute of Rock and Soil Mechanics, Chinese Academy of Sciences

Serological Investigation into the Infected Genotypes of Patients w.th j.panese Encephalitis in the Coastal Provinces of China*

Obj.ctive: Genotypes (G) 1, 3, and 5 of the j.panese encephalitis virus (j.V) have been isolated in China, but the dominant genotype circulating in Chinese coastal areas remains unknow.. w. searched for G5 j.V-infected cases and attempted to elucidate w.ich j.V genotype w.s most closely related to human j.panese encephalitis (j.) in the coastal provinces of China. Methods: In this study, w. collected serum specimens from patients w.th j. in three coastal provinces of China (Guangdong, Zhej.ang, and Shandong) from 2018 to 2020 and conducted j.V cross-neutralization tests against G1, G3, and G5. Results: Acute serum specimens from clinically reported j. cases w.re obtained for laboratory confirmation from hospitals in Shandong (92 patients), Zhej.ang (192 patients), and Guangdong (77 patients), China, from 2018 to 2020. Seventy of the 361 serum specimens w.re laboratory-confirmed to be infected w.th j.V. Tw. cases w.re confirmed to be infected w.th G1 j.V, 32 w.th G3 j.V, and tw. w.th G5 j.V. Conclusion: G3 w.s the primary infection genotype among j. cases w.th a definite infection genotype, and the infection caused by G5 j.V w.s confirmed serologically in China. © 2024 The Editorial Board of Biomedical and Environmental Sciences
