Role of MMP-2 and MMP-9 and their natural inhibitors in liver fibrosis, chronic pancreatitis and non-specific inflammatory bowel diseases

BACKGROUND： There is a growing evidence that matrix metalloproteinase （MMP）-2 and MMP-9 （gelatinases） play an important role in the pathogenesis of numerous disorders, especially with inflammatory etiology and extracellular matrix （ECM） remodeling. Despite the fact that gelatinases involve in liver cirrhosis is provided in the literature, their role in the pathogenesis of chronic pancreatitis and non-specific inflammatory bowel diseases is still under investigation. DATA SOURCES： We carried out a PubMed search of Englishlanguage articles relevant to the involvement of gelatinases in the pathogenesis of liver fibrosis, pancreatitis, and non-specific inflammatory bowel diseases. RESULTS： The decreased activity of gelatinases, especially MMP-2, is related to the development of liver fibrosis, probably due to the decrease of capability for ECM remodeling. Similar situation can be found in chronic pancreatitis; however, reports on this matter are rare. The presence of non-specific inflammatory bowel diseases results in MMP-9 activity elevation. CONCLUSION： The fluctuation of gelatinases activity during liver fibrosis, chronic pancreatitis and non-specific inflammatory bowel diseases is observed, but the exact role of these enzymes demands further studies.

Molecular mimicry in Helicobacter pylori infections

Gram-negative bacteria Helicobacter pylori(H. pylori) colonize gastric mucosa in humans and increase the risk of serious diseases such as gastric and duodenal ulcers, stomach cancers and mucosa associated lymphoid tissue lymphoma. The role of H. pylori infection in the pathogenesis of several extragastric diseases has been suggested including immune thrombocytopenic purpura, iron deficiency anemia, vitamin D deficiency, cardiovascular diseases,diabetes mellitus and dermatological disorders. Also neurological diseases and even lung cancer have attracted researchers concern. The relation between H. pylori infection and a growth retardation in children has also been suggested. Many mechanisms of molecular mimicry between H. pylori and the host have been proposed as a pathogen strategy to manipulate the immune system of the host in order to remain unrecognized and avoid eradication. A lot of effort has been put into the demonstration of homologous sequences between H. pylori and host compounds. However, knowledge about how often autoantibodies or autoreactive T lymphocytes induced during H. pylori infections cause pathological disorders is insufficient. This review provides data on H. pylori antigenic mimicry and possible deleterious effects due to the induction of immune response to the components common to these bacteria and the host.

CO2 degassing and melting of metasomatized mantle lithosphere during rifting-Numerical study

Soil application of Beauveria bassiana GHA against apple sawfly, Hoplocampa testudinea （Hymenoptera： Tenthredinidae）： Field mortality and fungal persistence

Low impact alternatives to synthetic insecticides for the control of apple sawfly （Hoplocampa testudinea Klug） are scarce encumbering pest management in organic apple orchards. We investigated the soil persistence and field efficacy of the entomopathogenic fungus Beauveria bassiana （Balsamo） Vuillemin （BotaniGard） against apple sawfly under common organic orchard practices. We also assessed the efficacy of B. bassiana GHA and Metarhiziurn brunneum Petch （indigenous strain） against sawfly in the laboratory. Larvae treated with either fungus in the laboratory died faster than control larvae and displayed 49.4%-68.4% mycosis. In the field, B. bassiana density remained high in the week after application, during larval descent to the soil. Fungal density decreased to 25% at 49 d after application and to 0.4% after 55 weeks. Molecular markers revealed that the majority of fungal isolates recovered comprised the applied B. bassiana strain GHA. Larvae pupating in soil cages in the orchard for 49 d displayed 17% mycosis. The high efficacy under laboratory conditions was not seen in the field. B. bassiana application resulted in densities above the upper natural background level during the growing season, but reversion to background levels occurred within a year. It remains to be investigated whether this has a detrimental effect on nontarget organisms. Additional work is needed to bridge the knowledge gap between laboratory and field efficacy in orchards.

Ultra-hot Mesoproterozoic evolution of intracontinental central Australia

The Musgrave Province developed at the nexus of the North, West and South Australian cratons and its Mesoproterozoic evolution incorporates a 100 Ma period of ultra-high temperature （UHT） meta- morphism from ca. 1220 to ca. 1120 Ma. This was accompanied by high-temperature A-type granitic magmatism over an 80 Ma period, sourced in part from mantle-derived components and emplaced as a series of pulsed events that also coincide with peaks in UHT metamorphism. The tectonic setting for this thermal event （the Musgrave Orogeny） is thought to have been intracontinental and the lithospheric architecture of the region is suggested to have had a major influence on the thermal evolution. We use a series of two dimensional, fully coupled thermo-mechanical-petrological numerical models to investi- gate the plausibility of initiating and prolonging UHT conditions under model setup conditions appro- priate to the inferred tectonic setting and lithospheric architecture of the Musgrave Province. The results support the inferred tectonic framework for the Musgrave Orogeny, predicting periods of UHT meta- morphism of up to 70 Ma, accompanied by thin crust and extensive magmatism derived from both crustal and mantle sources. The results also appear to be critically dependent upon the specific location of the Mus^rave Province. constrained between thicker cratonic masses.

This opinion review considers the prevailing question of whether to screen or notto screen for adolescent idiopathic scoliosis. New and improved standards ofpeople-oriented care and person-centredness, as well as improved principles ofpreventive screening and guideline development, have been postulated andimplemented in health care systems and cultures. Recommendations addressingscreening for scoliosis differ substantially, in terms of their content, standards ofdevelopment and screening principles. Some countries have discontinued issuingrecommendations. In the last decade, a number of updated and newrecommendations and statements have been released. Systematically developedguidelines and recommendations are confronted by consensus and opinion-basedstatements. The dilemmas and discrepancies prevail. The arguments concentrateon the issues of the need for early detection through screening in terms of theeffectiveness of early treatment, on costs and cost-effectiveness issues, scientificand epidemiologic value of screenings, and the credibility of the sources ofevidence. The problem matter is of global scale and applies to millions of *** regards clinical and methodological dilemmas, but also the matter of vulnerableand fragile time of adolescence and, more generally, children’s rights. Thedecisions need to integrate people’s values and preferences – screening tests needto be acceptable to the population, and treatments need to be acceptable forpatients. Therefore we present one more crucial, but underrepresented in thediscussion, issue of understanding and implementation of the contemporaryprinciples of person-centred care, standards of preventive screening, andguideline development, in the context of screening for scoliosis.

Host pathogen interactions in Helicobacter pylori related gastric cancer

Helicobacter pylori (H. pylori), discovered in 1982, is a microaerophilic, spiral-shaped gram-negative bacterium that is able to colonize the human stomach. Nearly half of the world’s population is infected by this pathogen. Its ability to induce gastritis, peptic ulcers, gastric cancer and mucosa-associated lymphoid tissue lymphoma has been confirmed. The susceptibility of an individual to these clinical outcomes is multifactorial and depends on H. pylori virulence, environmental factors, the genetic susceptibility of the host and the reactivity of the host immune system. Despite the host immune response, H. pylori infection can be difficult to eradicate. H. pylori is categorized as a group I carcinogen since this bacterium is responsible for the highest rate of cancer-related deaths worldwide. Early detection of cancer can be lifesaving. The 5-year survival rate for gastric cancer patients diagnosed in the early stages is nearly 90%. Gastric cancer is asymptomatic in the early stages but always progresses over time and begins to cause symptoms when untreated. In 97% of stomach cancer cases, cancer cells metastasize to other organs. H. pylori infection is responsible for nearly 60% of the intestinal-type gastric cancer cases but also influences the development of diffuse gastric cancer. The host genetic susceptibility depends on polymorphisms of genes involved in H. pylori-related inflammation and the cytokine response of gastric epithelial and immune cells. H. pylori strains differ in their ability to induce a deleterious inflammatory response. H. pylori-driven cytokines accelerate the inflammatory response and promote malignancy. Chronic H. pylori infection induces genetic instability in gastric epithelial cells and affects the DNA damage repair systems. Therefore, H. pylori infection should always be considered a pro-cancerous factor.

Lipidomic insights into the response of Arabidopsis sepals to mild heat

Arabidopsis sepals coordinate flower opening in the morning as ambient temperature rises;however,the underlying molecular mechanisms are poorly understood. Mutation of one heat shock proteinencoding gene, HSP70-16, impaired sepal heat stress responses (HSR), disrupting lipid metabolism,especially sepal cuticular lipids, leading to abnormal flower opening. To further explore, to what extent,lipids play roles in this process, in this study, we compared lipidomic changes in sepals of hsp70-16 andvdac3 (mutant of a voltage-dependent anion channel, VDAC3, an HSP70-16 interactor) grown underboth normal (22 C) and mild heat stress (27 C, mild HS) temperatures. Under normal temperature,neither hsp70-16 nor vdac3 sepals showed significant changes in total lipids;however, vdac3 but nothsp70-16 sepals exhibited significant reductions in the ratios of all detected 11 lipid classes, except themonogalactosyldiacylglycerols (MGDGs). Under mild HS temperature, hsp70-16 but not vdac3 sepalsshowed dramatic reduction in total lipids. In addition, vdac3 sepals exhibited a significant accumulationof plastidic lipids, especially sulfoquinovosyldiacylglycerols (SQDGs) and phosphatidylglycerols (PGs),whereas hsp70-16 sepals had a significant accumulation of triacylglycerols (TAGs) and simultaneousdramatic reductions in SQDGs and phospholipids (PLs), such as phosphatidylcholines (PCs), phosphatidylethanolamines (PEs), and phosphatidylserines (PSs). These findings revealed that the impact ofmild HS on sepal lipidome is influenced by genetic factors, and further, that HSP70-16 and VDAC3differently affect sepal lipidomic responses to mild HS. Our studies provide a lipidomic insight intofunctions of HSP and VDAC proteins in the plant’s HSR, in the context of floral development.

Health hazards related to conidia of Cladosporium—biological air pollutants in Poland, central Europe

The spores of Cladosporium Link. are often present in the air in high quantities and produce many allergenic proteins, which may lead to asthma. An aerobiological spore monitoring program can inform patients about the current spore concentration in air and help their physicians determine the spore dose that is harmful for a given individual. This makes it possible to develop optimized responses and propose personalized therapy for a particular sensitive patient. The aim of this study was to assess the extent of the human health hazard posed by the fungal genus Cladosporium. For the first time, we have determined the number of days on which air samples in Poland exceeded the concentrations linked to allergic responses of sensitive patients, according to thresholds established by three different groups（2800/3000/4000 spores per 1 m^3 of the air）. The survey was conducted over three consecutive growing seasons（April–September, 2010–2012） in three cities located in different climate zones of Poland（Poznan, Lublin and Rzeszow）. The average number of days exceeding 2800 spores per cubic meter（the lowest threshold） ranged from 61（2010） through 76（2011） to 93（2012）, though there was significant variation between cities. In each year the highest concentration of spores in the air was detected in either Poznan or Lublin, both located on large plains with intensive agriculture. We have proposed that an effective, science-based software platform to support policy-making on air quality should incorporate biological air pollutant data,such as allergenic fungal spores and pollen grains.
