The ultimate goal in Parkinson’s disease (PD) research remains the identification of treatments that are capable of slowing or even halting the progression of the disease. The failure of numerous past disease-modification trials in PD has been attributed to a variety of factors related not only to choosing wrong interventions, but also to using inadequate trial designs and target populations. In patients with clinically established PD, neuronal pathology may already have advanced too far to be modified by any intervention. Based on such reasoning, individuals in yet prediagnostic or prodromal disease stages, may provide a window of opportunity to test disease-modifying strategies. There is now sufficient evidence from prospective studies to define diagnostic criteria for prodromal PD and several approaches have been studied in observational cohorts. These include the use of PD-risk algorithms derived from multiple established risk factors for disease as well as follow-up of cohorts with single defined prodromal markers like hyposmia, rapid eye movement sleep behavior disorders, or PD gene carriers. In this review, we discuss recruitment strategies for disease-modification trials in various prodromal PD cohorts, as well as potential trial designs, required trial durations, and estimated sample sizes. We offer a concluding outlook on how the goal of implementing disease-modification trials in prodromal cohorts might be achieved in the future.

Antimicrobial resistance(AMR)poses a critical threat to global health and development,with environmental factors—particularly in urban areas—contributing significantly to the spread of antibiotic resistance genes(ARGs).However,most research to date has been conducted at a local level,leaving significant gaps in our understanding of the global status of antibiotic resistance in urban *** address this issue,we thoroughly analyzed a total of 86,213 ARGs detected within 4,728 metagenome samples,which were collected by the Meta SUB International Consortium involving diverse urban environments in 60 cities of 27 countries,utilizing a deep-learning based *** findings demonstrated the strong geographical specificity of urban environmental resistome,and their correlation with various local socioeconomic and medical *** also identified distinctive evolutionary patterns of ARG-related biosynthetic gene clusters(BGCs)across different countries,and discovered that the urban environment represents a rich source of novel *** study provides a comprehensive overview of the global urban environmental resistome,and fills a significant gap in our knowledge of large-scale urban antibiotic resistome analysis.

Functionally Graded Concrete （FGC） is fabricated at the Institute for Lightweight Structures and Conceptual Design （ILEK） by using a layer-by-layer technique with two different technological procedures： casting and dry spraying. Functional gradations are developed from two reference mixtures with diametrically opposed characteristics in terms of density, porosity, compression strength and elasticity modulus. In this study the first mixture consists of Normal Density Concrete （NDC）, with density about 2160 kg·m^-3 while the second mixture helps to obtain a very lightweight concrete, with density about 830 kg·m^-3. The FGC specimens have layers with different alternating porosities and provide superior deformability capacity under bulk compression compared to NDC specimens. In addition, the FGC specimens experienced a graceful failure behaviour, absorbing high amounts of energy during extended compression paths. The porosity variation inside the layout of tested specimens is inspired by the internal structure of sea urchin spines of heterocentrotus mammillatus, a promising role model for energy absorption in biomimetic engineering.

Rice is the staple food of more than half of the world's population,and major efforts are being made to improve rice *** yield is mainly determined by three components:number of panicles per plant,number of grains per panicle,and grain weight,which is positively associated with grain ***,grain size is an important agricultural trait for yield improvement in rice,and it is controlled by multiple genetic factors and environmental cues(Li et al.,2018).The final size of rice grains is largely determined by cell proliferation and cell expansion in the spikelet hull(husk),which sets the storage capacity of the grain and limits grain filling(Li et al.,2018).A number of quantitative trait loci for grain size have been identified and their underlying molecular functions *** include,for example,protein ubiquitination,chromatin modifications,transcription factors,RNA-mediated regulatory networks,and multiple plant hormones(Li and Li,2016).Among the plant hormones,brassinosteroids and cytokinin play pivotal roles in grain size ***,Liu et al.(2021)reported the discovery of an unexpected novel link between the brassinosteroid and the cytokinin signaling pathways with a major impact on rice grain size.

Computational Fluid Dynamics coupled with Discrete Element Method (CFD-DEM) is a commonly used numerical method to model gas-solid flow in fluidised beds and other multiphase systems. A significant limitation of CFD-DEM is the feasibility of the realistic simulation of large numbers of particles. Coarse-graining (CG) approaches, through which groups of multiple individual particles are represented by single, larger particles, can substantially reduce the total number of particles while maintaining similar system dynamics. As these three CG models have not previously been compared, there remains some debate, however, about the best practice in the application of CG in CFD-DEM simulations. In this paper, we evaluate the performance of three typical CG methods based on simulations of a bubbling fluidised bed. This is achieved through the use of a numerical validation framework, which makes full use of the high-resolution 3D positron emission particle tracking (PEPT) measurements to rigorously validate the outputs of CFD-DEM simulations conducted using various different coarse-graining models, and various different degrees of coarse-graining. The particle flow behaviours in terms of the particle occupancy field, velocity field, circulation time, and bubble size and velocity, are comprehensively analysed. It is shown that the CG simulation starts to fail when the size ratio between the bed chamber and the particles decreases to approximately 20. It is also observed, somewhat surprisingly, that the specific CG approach applied to interparticle contact parameters does not have a substantial effect on the simulation results for the bubbling bed simulations across a wide range of CG factors.

The tumor microenvironment is an ecosystem composed of multiple types of cells, such as tumor cells, immune cells, and cancerassociated fibroblasts. Cancer cells grow faster than non-cancerous cells and consume larger amounts of nutrients. The rapid growth characteristic of cancer cells fundamentally alters nutrient availability in the tumor microenvironment and results in reprogramming of immune cell metabolic pathways. Accumulating evidence suggests that cellular metabolism of nutrients, such as lipids and amino acids, beyond being essential to meet the bioenergetic and biosynthetic demands of immune cells, also regulates a broad spectrum of cellular signal transduction, and influences immune cell survival, differentiation, and anti-tumor effector function. The cancer immunometabolism research field is rapidly evolving, and exciting new discoveries are reported in high-profile journals nearly weekly. Therefore, all new findings in this field cannot be summarized within this short review. Instead, this review is intended to provide a brief introduction to this rapidly developing research field, with a focus on the metabolism of two classes of important nutrients-lipids and amino acids-in immune cells. We highlight recent research on the roles of lipids and amino acids in regulating the metabolic fitness and immunological functions of T cells, macrophages, and natural killer cells in the tumor microenvironment. Furthermore, we discuss the possibility of “editing” metabolic pathways in immune cells to act synergistically with currently available immunotherapies in enhancing anti-tumor immune responses.

Reversible switching of Kondo resonance in a single-molecule junction

The control of the Kondo effect is of great interest in single-molecule junction due to its potential applications in spin based ***,we demonstrate that the Kondo effect is reversibly switched on and off in an iron phthalocyanine(FePc)single-molecule junction by using a superconducting Nb *** a scanning tunneling microscope-based Nb-insulator-FePc-Au junction,we achieve a reversible switching between the Kondo dip and inelastic electronic tunneling spectra by simply adjusting the tip-sample distance to tune the tunnel coupling at low *** approaching the tip leads to the picking up of the molecule to the tip apex,which transfers the geometry of the single-molecule junction into a Nb-FePc-insulator-Au *** the molecule forms an effective magnetic impurity embedded into the superconducting ground states of the Nb tip,the out-gap Kondo dip switched to an in-gap Yu-Shiba-Rusinov *** results open up a new route for manipulating the Kondo effect within a single-molecule junction.

奥地利维也纳音乐与表演艺术大学(Universitat fur Musik und darstellende Kunst Wien,mdw)音乐声学系一维也纳声音风格研究所(Institut fur musikalische Akustik-Wiener Klangstil,WK)是致力于音乐声学、表演科学、心理声学与音乐心理学的跨学科研究所,近年来完成了许多具有国际影响力的科研项目。其中,在2021年9月开始的奥地利科学基金项目“维也纳的标志性声音:量化分析维也纳爱乐乐团新年音乐会”中,IWK团队采用可发现、可获取、可互操作以及可重复使用(Findable,Accessible,Interoperable,and Reusable,FAIR)的数据管理模式来探索这一著名文化名片的全方位特征,扩展了语义音乐信息的工作流程,有效助力了数字音乐学与表演科学的突破性发展,同时也向更广泛的数字音乐研究群体提供了能重复使用的数据集,并为跨领域出版实践的深入对话奠定了基础。
