Reversal of tamoxifen resistance by artemisinin in ER+breast cancer:bioinformatics analysis and experimental validation

Breast cancer is the leading cause of cancer-related deaths in women worldwide,with Hormone Receptor(HR)+being the predominant subtype.Tamoxifen(TAM)serves as the primary treatment for HR+breast cancer.However,drug resistance often leads to recurrence,underscoring the need to develop new therapies to enhance patient quality of life and reduce recurrence rates.Artemisinin(ART)has demonstrated efficacy in inhibiting the growth of drug-resistant cells,positioning art as a viable option for counteracting endocrine resistance.This study explored the interaction between artemisinin and tamoxifen through a combined approach of bioinformatics analysis and experimental validation.Five characterized genes(ar,cdkn1a,erbb2,esr1,hsp90aa1)and seven drug-disease crossover genes(cyp2e1,rorc,mapk10,glp1r,egfr,pgr,mgll)were identified using WGCNA crossover analysis.Subsequent functional enrichment analyses were conducted.Our findings confirm a significant correlation between key cluster gene expression and immune cell infiltration in tamoxifen-resistant and-sensitized patients.scRNA-seq analysis revealed high expression of key cluster genes in epithelial cells,suggesting artemisinin’s specific impact on tumor cells in estrogen receptor(ER)-positive BC tissues.Molecular target docking and in vitro experiments with artemisinin on LCC9 cells demonstrated a reversal effect in reducing migratory and drug resistance of drug-resistant cells by modulating relevant drug resistance genes.These results indicate that artemisinin could potentially reverse tamoxifen resistance in ER-positive breast cancer.

Organophosphate esters in sediment from Taihu Lake,China:Bridging the gap between riverine sources and lake sinks

Surface sediment samples from Taihu Lake in China and its inflow rivers,along with two lake sediment core samples,were collected and analyzed for organophosphate esters(OPEs).The concentrations of total OPEs varied from 28.60 ng/g to 158.72 ng/g(median:54.25 ng/g)in river surface sediment and from 62.57 ng/g to 326.84 ng/g(median:86.37 ng/g)in lake sediment.Tributyl phosphate(TnBP)was the predominant compound in river surface sediment,and tris(2-butoxyethyl)phosphate was predominant in lake sediment.High contamination occurred in the north-west region,which was related to the high level of urbanization and high usage of OPEs.The sediment–water partition coefficients of OPEs(log_(Koc))were calculated,showing a significant correlation with logKow(p<0.05).The concentration and composition of OPEs in two sediment cores varied due to the different sampling locations,with more OPE species found in the northern region than in the southern one.Principal component analysis and positive matrix factorization indicated that sewage discharges,vehicle emissions,and atmospheric deposition were the possible sources of OPEs in Taihu Lake sediments.Tris(1-chloro-2-propyl)phosphate,tris(2-chloroethyl)phosphate,and TnBP were the main OPEs causing ecological risks.

Flexible multilayer MEMS coils and their application in energy harvesters

Electromagnetic vibration energy harvesters are promising for the power supply of wireless sensor nodes,small electronic devices,and wearable electronics.Conventional electromagnetic harvesters usually increase output by increasing the size of coils and magnets,limiting the improvement of energy conversion efficiency and power density.In this study,multilayer microelectromechanical system(MEMS)coils were prepared using flexible electronics,and their high integration performance in arbitrary space was utilized to greatly improve the utilization of the space magnetic field by the electromagnetic harvester.The core magnet of the generator was magnetically balanced to achieve levitation,which improved the sensitivity and reduced fatigue damage compared with traditional spring structures.The wound coils on the top and bottom of the magnet and the flexible coils on the sides worked together to improve the energy efficiency and output of the devices.The output performance of the device with different number distributions was simulated using mathematical models to obtain the optimal structural parameters.The results show that by introducing flexible multilayer MEMS coils on the side surface of the energy harvester,the open-circuit voltage of the energy generators increased from 7 to 10 V by more than 43%.Flexible multilayer MEMS coils can enhance energy conversion rates and possess compact dimensions,making them suitable for integration onto complex surfaces.After the vibration energy harvesting system testing,the maximum peak power of the harvester was 7.1 m W at an acceleration of1 g and a resonant frequency of 11 Hz with a resistor of 3.5 kΩinternal resistance.Moreover,a 470μF capacitor can be charged to 3.5 V within 10 s to drive a hygrothermograph to work for more than 80 s and can supply a light bulb continuously.This strategy shows the great potential of vibration-energy-driven electromagnetic generators for powering small electronics in limited spaces.

A high-performance electromagnetic energy harvester for scavenging ultra-low frequency vibration energy of human foot movement

In our daily lives,low-frequency kinetic energy primarily manifests as vibrations.However,effective harnessing of lowfrequency kinetic energy remains a formidable challenge.This paper proposes a rope-driven rotor that rotates around an axis and consists of an ultra-high-molecular-weight polyethylene(UHMWPE)wire wrapped around a metal shaft.The rotor can convert ultra-low frequency vibration/linear motion into rapid rotation by pressing the top at low frequencies and driving the rope for a quick release.The harvester can generate up to 36.25 m W power using a 0.1-mm-diameter UHMWPE wire as the rotor when periodically pressed down to 20 mm at a frequency of 1 Hz.A simple power generation floor is assembled,generating 28.58-m W power with a matching load at a frequency of 1.5 Hz.Moreover,the harvester can increase the charging voltage of a 0.47-F supercapacitor from 0 to 6.8 V within 10 min.In addition,the harvester can harvest energy through a light finger press motion,and the energy obtained can also support the continuous operation of multiple electronic devices concurrently.This study introduces an effective method for harvesting ultra-low frequency energy and has great prospects in the field of power generation floor and human movement energy harvesting.

Highly crystalline,highly stable n-type ultrathin crystalline films enabled by solution blending strategy toward organic single-crystal electronics

The development of n-type semiconductor is still far behind that of p-type semiconductor on account of the challenges in enhancing carrier mobility and environmental stability.Herein,by blending with the polymers,n-type ultrathin crystalline thin film was successfully prepared by the method of meniscus-guided coating.Remarkably,the n-type crystalline films exhibit ultrathin thickness as low as 5 nm and excellent mobility of 1.58 cm^(2) V^(-1) s^(-1),which is outstanding in currently reported organic n-type transistors.Moreover,the PS layer provides a high-quality interface with ultralow defect which has strong resistance to external interference with excellent long-term stability,paving the way for the application of n-type transistors in logic circuits.

Evaluation of Attending Medical Teams and Their Impact on the DRG Components of Cholecystectomy

Objective:This study assessed the role of the attending medical team in the cost control of the cholecystectomy DRG components.Methods:The association between team structure,workflow,and treatment outcomes was analyzed using a mixed-methods approach combining quantitative data and qualitative interviews from 628 patients.Results:Inter-professional teamwork significantly affected length of stay,treatment costs,and recurrence rates,with experienced teams performing better in terms of emergency response and collaborative efficiency.Patient satisfaction was generally high,indicating that good teamwork enhances treatment outcomes.Significance:The study highlights the importance of optimizing team configuration to improve the quality,efficiency,and cost control of healthcare.

Aiming for suppressing side-mode and spectrum broadening,a slit beam-shaping method and super-Gaussian apodization processing for femtosecond laser point-by-point(PbP)inscription technology of fiber Bragg gratings(FBGs)are reported here.High-quality FBGs,featuring narrow bandwidth of less than 0.3 nm,high reflectivity above 85%,low insertion loss(0.21 dB),and low cladding loss(0.82 dB),were obtained successfully.By a semi-automatic PbP inscription process,an array consisting of six FBGs,exhibiting almost no side-mode peaks with high suppression ability and narrow bandwidth,was fabricated along three independently developed single-mode fibers with an interval of 20 mm.

针对1/4车主动悬架系统,提出一种基于遗传算法(genetic algorithm,GA)的变论域模糊比例-积分-微分(proportional-integral-differential,PID)控制方法.在建立主动悬架系统模型的基础上,引入变论域思想设计模糊PID控制器.为进一步改善控制器减振效果,采用GA来优化变论域中伸缩因子描述函数的参数.结果表明:相比PID、模糊PID与未优化的变论域模糊PID等控制方法,基于GA的变论域模糊PID控制方法在降低车身垂向加速度、改善乘坐舒适性方面具有优越性;所提控制方法对簧载质量和车辆行驶速度不确定性具备较强的鲁棒性.
