考虑环保风险的建筑供应链低碳策略研究

在全球共同呼吁生态全球化的今天,建筑业的绿色性和环保性是社会的重要关注点。文章着重于建筑业供应链低碳策略的研究,在阅读大量文献以及调研众多建筑企业之后,构建了建筑供应链绿色度评价体系,从多维度分析出供应链的影响因素。再者,通过Fermatean模糊集和SWARA方法确定了各因素评价的权重,采用灰色关联改进的TOPSIS方法对湖北省典型建筑项目的供应链绿色度进行了评估,最终根据研究结果,在考虑环保风险的基础上,为建筑业供应链管理者提出针对性低碳策略。

“双碳”目标是应对气候变化问题的重大战略。汽车行业大力推广电动汽车的使用,使得车辆周期的碳排放越来越值得关注。如何确定“双碳”背景下电动汽车供应链脆弱性影响因素并分析是电动汽车行业需要解决的问题。考虑专家意见的模糊性,采用毕达哥拉斯模糊DEMATEL确定影响因素间的相互关系,结合ANP确定各影响因素的优先级,提出了模糊DEMATEL-ANP模型。研究结果表明:行业生态不完善、市场需求不确定性、缺乏重构设计的能力、低碳减排指标的约束、原材料供应不稳定性、政策以及补贴调整是“双碳”背景下电动汽车供应链脆弱性重要影响因素。

生菜菌核病是由核盘菌Sclerotinia sclerotiorum引起的一种植物病害,主要危害生菜的茎基部。本研究以前期分离获得的贝莱斯芽胞杆菌Bacillus velezensis S297为研究对象,研究其对生菜菌核病的生防作用。结果显示,贝莱斯芽胞杆菌S297能抑制核盘菌菌丝的生长、菌核的生成和萌发。以5×10^(7)cfu/mL浓度S297发酵液处理核盘菌侵染离体生菜叶片36h后的防效达84.65%;贝莱斯芽胞杆菌S297与木霉菌协同使用,防效提高至97.52%,与50%啶酰菌胺1mg/mL处理的防效无显著差异。田间试验中,贝莱斯芽胞杆菌S297发酵液(5×10^(7)cfu/mL)防效为81.08%,与木霉菌协同使用防效为85.14%,与50%啶酰菌胺1 mg/mL处理无显著差异。贝莱斯芽胞杆菌S297对核盘菌的作用结果表明,菌株S297可使核盘菌菌丝变形,抑制核盘菌果胶甲基半乳糖醛酸酶(PMG)、多聚半乳糖醛酸酶(PG)和纤维素酶(CL)的酶活性,亦抑制核盘菌在侵染生菜过程中草酸的产生。

工业一体式阀门阀体加工存在多机床、多次装夹定位、多工序加工和多次转道等问题,使得阀体生产质量稳定性较差,生产效率较难满足自动化生产的需要。为此,在采用多种机床、配置复合式夹具及多工位转轴的基础上,应用自动平行料仓和自动控制技术,研究开发了可实现阀体两次装夹的多工序自动化加工产线。该项技术成果可以应用在阀门阀体及中小型机械零件自动化加工产线设计建设中,具有一定的参考借鉴价值。

目的研究白细胞介素(IL)⁃15对脂多糖(LPS)诱导的脓毒症心肌细胞凋亡的影响,并探讨其相关的作用机制。方法H9C2细胞分为对照组(常规培养的H9C2细胞)、凋亡组(建立LPS诱导的心肌细胞损伤模型,未进行其他干预)和干预组(采用10 ng/ml重组大鼠IL⁃15干预心肌H9C2细胞损伤模型6 h)。甲基噻唑基四唑(MTT)法和流式细胞仪检测IL⁃15干预对H9C2细胞增殖和凋亡的影响,四甲基罗丹明乙酯(TMRE)染色测定线粒体膜电位。10 mg/kg LPS腹腔注射建立小鼠脓毒症模型,采用腹腔注射含100μg/ml IL⁃15的生理盐水100μl进行干预。通过HE染色评估小鼠心肌病理损伤。ELISA法检测小鼠心肌组织氧化应激指标。实时荧光定量PCR和蛋白质印迹法检测小鼠心肌组织中血红素氧合酶(HO)⁃1、核转录相关因子2(Nrf2)mRNA和蛋白的表达。结果凋亡组H9C2细胞凋亡率[(47.9±5.1)%]明显高于对照组[(2.1±0.3)%],差异有统计学意义(P<0.05),而干预组H9C2细胞凋亡率[(20.7±2.7)%]则明显低于凋亡组(P<0.05)。LPS诱导可以显著抑制心肌细胞的增殖活性,IL⁃15能够抑制LPS诱导的心肌细胞活力降低(P<0.05)。凋亡组H9C2细胞线粒体膜电位相对值(2.6±4.6)明显低于对照组H9C2细胞线粒体膜电位相对值(7.4±5.2),差异有统计学意义(P<0.05);干预组H9C2细胞线粒体膜电位相对值(4.2±4.9)明显高于凋亡组(P<0.05)。LPS作用后显著增强了H9C2细胞线粒体膜电位的丧失,IL⁃15干预会减少LPS诱导的H9C2细胞线粒体膜电位的丧失。HE染色切片镜下观察显示小鼠脓毒症模型心肌组织出现明显损伤及炎症细胞浸润;IL⁃15干预可改善LPS诱导的小鼠心肌损伤。脓毒症模型小鼠心肌组织中超氧化物歧化酶(SOD)和总抗氧化能力(T⁃AOC)水平较正常小鼠明显降低,而丙二醛(MDA)水平则明显升高(P<0.05)。IL⁃15干预小鼠心肌组织SOD、T⁃AOC水平均升高,MDA水平均降低(P<0.05)。正常小鼠心肌组织中HO⁃1和Nrf2的mRNA和蛋白表达水平均较低;脓毒症模型小鼠心肌组织中HO⁃1、Nrf2的mRNA和蛋白表达水平较正常小鼠明显升高(P<0.05);IL⁃15干预小鼠心肌组织中HO⁃1、Nrf2的mRNA和蛋白表达水平进一步升高(P<0.05)。结论IL⁃15可减轻脓毒症诱导的心肌细胞凋亡,改善脓毒症小鼠心肌损伤程度。其机制可能与IL⁃15激活HO⁃1/Nrf2信号通路,进而抑制脓毒症心肌组织氧化应激有关。

Tumor necrosis factor alpha(TNF-a) is a cytokine that can potently stimulate the synthesis of a range of proinflammatory mediators in macrophages. The underlying epigenetic mechanism, however, is underexplored. Here we report that the transcriptional modulator megakaryocytic leukemia 1(MKL1) is associated with a histone H3 K4 methyltransferase activity. Re-ChIP assay suggests that MKL1 interacts with and recruits WDR5, a component of the COMPASS complex responsible for H3 K4 methylation, to the promoter regions of pro-inflammatory genes in macrophages treated with TNF-α. WDR5 enhances the ability of MKL1 to stimulate the promoter activities of proinflammatory genes. In contrast, silencing of WDR5 attenuates TNF-a induced production of pro-inflammatory mediators and erases the H3 K4 methylation from the gene promoters. Of interest, the chromatin remodeling protein BRG1 also plays an essential role in maintaining H3 K4 methylation on MKL1 target promoters by interacting with WDR5. MKL1 knockdown disrupts the interaction between BRG1 and WDR5. Together, our data illustrate a role for MKL1 in moderating the crosstalk between BRG1 and WDR5 to activate TNF-a induced pro-inflammatory transcription in macrophages.

China is among the top nitrous oxide（N2O）-emitting countries, but existing national inventories do not provide full-scale emissions including both natural and anthropogenic sources. We conducted a four-decade（1980–2020） of comprehensive quantification of Chinese N2O inventory using empirical emission factor method for anthropogenic sources and two up-to-date process-based models for natural sources. Total N2O emissions peaked at 2287.4（1774.8–2799.9） Gg N2O yr-1in 2018, and agriculture-developed regions, like the East, Northeast, and Central, were the top N2O-emitting regions. Agricultural N2O emissions have started to decrease after 2016 due to the decline of nitrogen fertilization applications, while, industrial and energetic sources have been dramatically increasing after 2005. N2O emissions from agriculture, industry,energy, and waste represented 49.3%, 26.4%, 17.5%, and 6.7% of the anthropogenic emissions in 2020,respectively, which revealed that it is imperative to prioritize N2O emission mitigation in agriculture,industry, and energy. Natural N2O sources, dominated by forests, have been steadily growing from 317.3（290.3–344.1） Gg N2O yr-1in 1980 to 376.2（335.5–407.2） Gg N2O yr-1in 2020. Our study produces a Full-scale Annual N2O dataset in China（FAN2020）, providing emergent counting to refine the current national N2O inventories.

Vaccines have been the primary remedy in the global fight against coronavirus disease 2019（COVID-19）.The receptor-binding domain（RBD） of the spike protein, a critical viral immunogen, is affected by the heterogeneity of its glycan structures and relatively low immunogenicity. Here, we describe a scalable synthetic platform that enables the precise synthesis of homogeneously glycosylated RBD, facilitating the elucidation of carbohydrate structure–function relationships. Five homogeneously glycosylated RBDs bearing biantennary glycans were prepared, three of which were conjugated to T-helper epitope(Tpep) from tetanus toxoid to improve their weak immune response. Relative to natural HEK293-derived RBD, synthetic RBDs with biantennary N-glycan elicited a higher level of neutralising antibodies against SARS-CoV-2 in mice. Furthermore, RBDs containing Tpepelicited significant immune responses in transgenic mice expressing human angiotensin-converting enzyme 2. Our collective data suggest that trimming the N-glycans and Tpepconjugation of RBD could potentially serve as an effective strategy for developing subunit vaccines providing efficient protection.
