Differences in leaf cuticular wax induced by whole-genome duplication in autotetraploid sour jujube

Drought-resistant plants exhibit strong water retention capability.In this regard,the autotetraploid sour jujube leaves exhibit better water retention than diploid leaves.Morphological comparisons and physiological comparisons of diploid and autotetraploid leaves showed that the autotetraploid leaves had thicker leaf cuticles and more leaf wax accumulation than the diploid leaves,which could reduce cuticle permeability and improve the drought tolerance of leaves.In this study,the cuticular wax crystalloids on the adaxial and abaxial sides of young and mature jujube leaves were observed in the two ploidy types,and unique cuticular wax crystalloids covering a large area of the cuticle on autotetraploid sour jujube leaves may provide an advantage in reducing leaf non-stomata transpiration and improving plant drought tolerance.Based on the transcriptome,115 differentially expressed genes between diploids and autotetraploids were further analyzed and found to be involved in the accumulation of cuticular wax components,including terpenoids,fatty acids,and lipids,as well as ABC transporter and wax biosynthetic process.Finally,14 genes differentially expressed between glossy autotetraploid leaves and nonglossy diploid leaves,such as LOC107414787,LOC107411574 and LOC107413721,were screened as candidate genes by qRT-PCR analysis.This findings provided insights into how polyploidization improved drought tolerance.

【目的】研究秋水仙素溶液质量浓度、处理时间和预培养时间对‘京枣39’离体多倍体诱导的影响,优化枣树良种‘京枣39’的增殖和生根培养基,建立‘京枣39’的离体多倍体诱导体系,获得多倍体植株。【方法】采用完全随机区组试验设计,探索不同秋水仙素溶液质量浓度和处理时间组合对‘京枣39’不定芽再生的影响,不同预培养时间对‘京枣39’离体多倍体诱导的影响,不同生长调节剂组合对‘京枣39’增殖培养的影响,不同基本培养基和生长调节剂组合对‘京枣39’组培苗生根培养的影响。【结果】(1)当秋水仙素溶液质量浓度为80 mg/L、处理48 h时‘京枣39’刻伤叶片存活率为(50.00±7.07)%,分化系数为(2.06±0.17)。(2)最佳预培养时间为6 d,并施以80 mg/L秋水仙素浓度处理48 h,获得了纯合四倍体‘京枣39’。(3)最佳增殖培养基为MS+0.8 mg/L 6-BA+0.4 mg/L IBA+30 g/L麦芽糖,pH=5.8,平均增殖系数为(4.22±0.22)。(4)最佳生根培养基为改良1/2MS+0.8 mg/L IBA+30 g/L麦芽糖,pH=5.8,平均根长为(5.22±0.19) cm;平均生根数为(3.20±0.22)。【结论】初步建立了‘京枣39’的离体多倍体诱导体系,获得了‘京枣39’的多倍体植株,并优化了其增殖培养及生根培养方法。本研究为优良枣树品种‘京枣39’的种质创新提供了切实可行的技术方法,为多倍体诱导在枣树以及其他果树中的应用奠定了试验基础。

Surficial modification enabling planar Al growth toward dendrite-free metal anodes for rechargeable aluminum batteries

Al metal possesses ultrahigh theoretical volumetric capacity of 8,040 m Ah cm^(-3),and gravimetric capacity of 2,980 m Ah g^(-1),and thus is highly attractive for electrochemical energy storage.However,it suffers from several issues,such as the dendrite formation,during Al stripping-deposition cycling,which has been verified to account for the short circuit and limited cyclic performance.Herein,we use a facile and applicable method to in-situ reconstruct the Al anode surface with F-Al-O chemical bonds,which could preferentially induce the planar growth of Al along the interface plane,thus leading to the dendrite-free morphology evolution during the cycling.Benefiting from F-Al-O chemical bonds on the surface of Al anodes,long lifespan of symmetric cells can be realized even under 1 m A cm^(-2)and 1 m Ah cm^(-2).Coupling the F-Al anode with graphite-based cathodes,high-voltage dual-ion Al metal batteries can be achieved with long-term cycle stability up to 1,200 cycles(at 0.5 m A cm^(-2)),surpassing the counterparts using pristine Al metal anode.Furthermore,the effectiveness of this surficial modification strategy is also elucidated with the aid of theoretical calculation.This work provides novel insights on low-cost and facile strategies against the Al dendrite growth in aluminum batteries.

Melittin,a classical antimicrobial peptide,is a highly potent antitumor agent.However,its significant toxicity seriously hampers its application in tumor therapy.In this study,we developed novel melittin analogs with pH-responsive,cell-penetrating and membranelytic activities by replacing arginine and lysine with histidine.After conjugation with camptothecin(CPT),CPT-AAM-1 and CPT-AAM-2 were capable of killing tumor cells by releasing CPT at low concentrations and disrupting cell membranes at high concentrations under acidic conditions.Notably,we found that the C-terminus of the melittin analogs was more suitable for drug conjugation than the N-terminus.CPT-AAM-1 significantly suppressed melanoma growth in vivo with relatively low toxicity.Collectively,the present study demonstrates that the development of antitumor drugs based on pH-responsive antimicrobial peptide-drug conjugates is a promising strategy.

The benefits of PCSK9 inhibitors in patients with acute coronary syndrome:a systematic review and meta-analysis

Background:Proprotein convertase subtilisin/kexin 9(PSCK9)inhibitors have been beneficial for many patients with hyperlipidemia.The objective of this study was to investigate the benefit of PSCK9 inhibitors in patients with acute coronary syndrome(ACS).Methods:We systematically searched PubMed,EMBASE,and Cochrane Clinical Trials(published before January 2023;no language restriction)to compare the treatment of patients with ACS using PCSK9 inhibitors and placebo.The primary end points were major adverse cardiovascular events,nonfatal myocardial infarction,cardiogenic death,stroke,hospitalization for recurrent ACS,and coronary revascularization.Fixed-or random-effects models were used to assess the aggregated data.Results:Of the 1686 identified studies,5 were eligible and included in our analysis(of a total of 38,005 participants,18,609 cases were placed in the PCSK9 inhibitor treatment group and 19,396 cases in the placebo group).Compared with the placebo group,PCSK9 inhibitors significantly reduced the major adverse cardiovascular events(odds ratio[OR]:0.83;95%confidence interval[CI]:0.77–0.88;P<0.00001)for patients following ACS.The incidence of nonfatal myocardial infarction(relative risk:0.80;95%CI:0.74–0.87;P<0.00001),cardiovascular death(OR:0.96;95%CI:0.83–1.10;P=0.56),stroke(OR:0.74;95%CI:0.63–0.88;P=0.0007),hospitalization for recurrent ACS(OR:0.57;95%CI:0.40–0.83;P=0.003),or coronary revascularization(OR:0.82;95%CI:0.76–0.88;P<0.00001)all demonstrated a significant decrease in the comparison between the 2 groups.Conclusion:This meta-analysis demonstrated that treatment with PCSK9 inhibitors in patients with ACS reduced the probability of multiple cardiovascular events and improved patient prognosis.

Understanding the driving mechanism of the divergence of hoarding strategies of animals(i.e.,scatter-hoarding vs.larder-hoarding)is crucial to understand the ecological and evolutionary implications of plant-animal interactions.Zhang et al.(2022)compiled a global-scale dataset from the published literature including 183 species of seed-hoarding rodents,and concluded that phylogenetic conservatism,functional traits,and environmental factors counted for the hoarding behavior divergence of rodents.A more thought-provoking question may be asked:Do these variables contribute equally or not to explain the divergence of hoarding strategies?Here,we re-analyzed Zhang et al's(2022)dataset using partial Ralk calculation(Ives 2019)to parse out the relative contri-lik butions of these variables.

Perenniality:From model plants to applications in agriculture

To compensate for their sessile nature,plants have evolved sophisticated mechanisms enabling them to adapt to ever-changing environments.One such prominent feature is the evolution of diverse life history strategies,particularly such that annuals reproduce once followed by seasonal death,while perennials live longer by cycling growth seasonally.This intrinsic phenology is primarily genetic and can be altered by environmental factors.Although evolutionary transitions between annual and perennial life history strategies are common,perennials account for most species in nature because they survive well under year-round stresses.This proportion,however,is reversed in agriculture.Hence,perennial crops promise to likewise protect and enhance the resilience of agricultural ecosystems in response to climate change.Despite significant endeavors that have been made to generate perennial crops,progress is slow because of barriers in studying perennials,and many developed species await further improvement.Recent findings in model species have illustrated that simply rewiring existing genetic networks can lead to lifestyle variation.This implies that engineering plant life history strategy can be achieved by manipulating only a few key genes.In this review,we summarize our current understanding of genetic basis of perenniality and discuss major questions and challenges that remain to be addressed.

Risk factors for lymph node metastasis in superficial esophageal squamous cell carcinoma

In this editorial,we comment on the article by Wang et al published in the recent issue of the World Journal of Gastroenterology in 2023.We focused on identifying risk factors for lymph node metastasis(LNM)in superficial esophageal squamous cell carcinoma(SESCC)patients and how to construct a simple and reliable clinical prediction model to assess the risk of LNM in SESCC patients,thereby helping to guide the selection of an appropriate treatment plan.The current standard treatment for SESCC is radical esophagectomy with lymph node dissection.However,esophagectomy is associated with considerable morbidity and mortality.Endoscopic resection(ER)offers a safer and less invasive alternative to surgical resection and can enable the patient's quality of life to be maintained while providing a satisfactory outcome.However,since ER is a localized treatment that does not allow for lymph node dissection,the risk of LNM in SESCC limits the effectiveness of ER.Understanding LNM status can aid in determining whether patients with SESCC can be cured by ER without the need for additional esophagectomy.Previous studies have shown that tumor size,macroscopic type of tumor,degree of differentiation,depth of tumor invasion,and lymphovascular invasion are factors associated with LNM in patients with SESCC.In addition,tumor budding is commonly associated with LNM,recurrence,and distant metastasis,but this topic has been less covered in previous studies.By comprehensively evaluating the above risk factors for LNM,useful evidence can be obtained for doctors to select appropriate treatments for SESCC patients.
