Background:The association between 1,3-β-D-glucan(BDG)levels and in-fections caused by Pseudomonas aeruginosa or Streptococcus pneumoniae,and the stability of BDG under different storage conditions are unclear.Methods:Strains of Pseudomonas aeruginosa and S.pneumoniae were grown in medium and human serum.The BDG concentrations in culture superna-tants were measured.The specificity and stability of BDG were also evaluated.Results:P.aeruginosa produced high levels of BDG in Luria-Bertani medium(>4×10^(4)pg/mL)and human serum(527.0 pg/mL),whereas S.pneumoniae produced low levels of BDG in THY medium(175.6 pg/mL)and human serum(78.3 pg/mL).The BDG produced by these two bacteria was specifically degraded by 1,3-β-D-glucanase.BDG was degraded when stored at different temperatures,decreasing by 22.5%and 9.3%at−20℃and−70℃,respectively,for 63 days;by 30.7%at 4℃for 12 days;and by 12.6%and 22.0%at 37℃for 6 and 12 h.Conclusion:BDG false-positivity must be considered in patients with bacteremia caused by P.aeruginosa when diagnosing invasive fungal infection.Human serum samples for the BDG test in medical facilities should be tested as soon as possible or stored at low temperatures before testing.

Fruit wart is an important appearance trait influencing consumer preferences of bitter gourd(Momordica charantia L.).The molecular genetic mechanisms underlying fruit wart formation in bitter gourd are largely unknown.In this study,genetic analysis based on four generations showed that fruit wart formation in bitter gourd was controlled by a single dominant locus named as Fwa.The Fwa locus was initially mapped into a 4.82 Mb region on pseudochromosome 4 by BSA-seq analysis and subsequently narrowed down to a 286.30 kb region by linkage analysis.A large F2population consisting of 2360 individuals was used to screen recombinants,and the Fwa locus was finally fine mapped into a 22.70 kb region harboring four protein-coding genes through recombination analysis.MC04g1399,encoding an epidermal patterning factor 2-like protein,was proposed as the best candidate gene for Fwa via sequence variation and expression analysis.In addition,a 1-bp insertion and deletion(InDel)variation within MC04g1399 was converted to a cleaved amplified polymorphic sequence(CAPS)marker that could precisely distinguish between the warty and non-warty types with an accuracy rate of 100%among a wide panel of 126 bitter gourd germplasm resources.Our results not only provide a scientific basis for deciphering the molecular mechanisms underlying fruit wart formation but also provide a powerful tool for efficient genetic improvement of fruit wart via marker-assisted selection.

Interannual variations of the eddy kinetic energy(EKE)related to two types of winter circulation events(“O”and“U”)were investigated based on the outputs of the Ocean General Circulation Model(OGCM)for the Earth Simulator(OFES)and the corresponding energetic analyses.Results show that the EKE is strong and extends eastward to offshore the Vietnam coast about 2°,associated with the weaker South China Sea western boundary current(SCSwbc)in“O”type years,while the EKE is weak and high value that can be attained is narrowed along the coast,associated with the stronger SCSwbc in“U”type years.The energy budget shows that the wind stress and barotropic/baroclinic instability are important factors to regulate the EKE in“U”and“O”years.For“U”years,under a strong winter monsoon forcing,the SCSwbc strengthen,the directly wind work and barotropic conversion from the mean kinetic energy(MKE)to EKE are weak,thus the EKE decrease corresponding to the baroclinic conversion from the kinetic energy to potential energy.However,the situation is reversed in“O”years.Under the influence of El Niño events,wind stress forces can weaken SCSwbc and enhance EKE in pattern“O”,whereas La Niña events have relatively weaker influences.The barotropic conversion rate in“O”type is nearly eight times of the“U”type.The pressure work and advection term are the main sources to greatly suppress EKE in the SCSwbc region.

Genetic load and inbreeding are recognized as important factors to be considered in conservation programs.Elevated levels of both can increase the risk of population extinction by negatively impacting fitness-related characters in many species of plants and animals,including humans(inbreeding depression).Genomic tech-niques are increasingly used in measuring and understanding genetic load and inbreeding and their importance in evolution and conservation.We used whole genome resequencing data from two sibling grouse species in subarctic Eurasia to quantify both.We found a large range of inbreeding measured as FROH(fraction of runs of homozygosity)in individuals from different populations of Chinese Grouse(Tetrastes sewerzowi)and Hazel Grouse(T.bonasia).FROH estimated from genome-wide runs of homozygosity(ROH)ranged from 0.02 to 0.24 among Chinese Grouse populations and from 0.01 to 0.44 in Hazel Grouse.Individuals from a population of Chinese Grouse residing in the Qilian mountains and from the European populations of Hazel Grouse(including samples from Sweden,Germany and Northeast Poland)were the most inbred(FROH ranged from 0.10 to 0.23 and 0.11 to 0.44,respectively).These levels are comparable to other highly inbred populations of birds.Hazel Grouse from northern China and Chinese Grouse residing in the Qinghai-Tibetan Plateau showed relatively lower inbreeding levels.Comparisons of the ratio between deleterious missense mutations and synonymous mutations revealed higher levels in Chinese Grouse as compared to Hazel Grouse.These results are possibly explained by higher fixation rates,mutational melt down,in the range-restricted Chinese Grouse compared to the wide-ranging Hazel Grouse.However,when we compared the relatively more severe class of loss-of-function muta-tions,Hazel Grouse had slightly higher levels than Chinese Grouse,a result which may indicate that purifying selection(purging)has been more efficient in Chinese Grouse on this class of mutations.

Observation of continental shelf wave propagating along the eastern Taiwan Strait during Typhoon Meranti 2016

This study is the fi rst to depict typhoon-induced continental shelf wave(CSW)propagation in the eastern Taiwan Strait(TWS)during the passage of Typhoon Meranti in 2016 using tidal gauge data and along-track satellite altimeter data.The strong amplitude response of sea level oscillations(within the range of 0.30–0.54 m)as a free,barotropic CSW after Meranti,which impacted the TWS,was clearly detected in time and frequency(in bands of 64–81 h)using wavelet and cross-wavelet analyses.The measured group and phase speeds were consistent with the dispersion curves for CSW with the fi rst-mode derived from the cross-shelf sections of the eastern TWS,with the mean speeds reaching 3 and 5.6±0.7 m/s,respectively.Coincidentally,the typhoon-induced sea level anomaly(SLA)was also captured by the satellite altimeter before this CSW entered into the TWS.Using the theoretical cross-shore CSW modes to fi t the SLA data,the results indicated that the fi rst three wave modes can interpret this CSW event appeared in the southern TWS very well,with the fi rst mode being the dominant one.

针对碳毡装配过程中碳毡占位、边缘毛刺复杂,人工检测效率低、成本高、接触产生不良影响等问题,设计了一种基于机器视觉的碳毡装配检测算法系统。使用Python语言调用相机获取图像并进行预处理。随后,对图像应用改进的Sobel算子进行初步的边缘检测,获取边缘的粗略坐标信息。接着,采用改进的插值法亚像素边缘检测算法对这些粗略坐标进行细化,得到更精确的坐标信息。然后,利用最小二乘法对这些精确坐标进行直线拟合,得到图像中的四个角点。最终,通过对这些角点的位置关系进行判断和分析。实验结果表明,本系统可以稳定进行碳毡装配检测,具有一定的鲁棒性。

为了提高焊缝跟踪系统的精准度,提出了一种基于Mask R-CNN和结构光的焊缝特征参数检测系统。通过Mask R-CNN对焊缝结构光图像提取线结构光边界信息,再通过灰度重心法完成对结构光中心线的提取;对中心线图像进行斜率分析,来确定焊缝特征点坐标;最后将坐标信息传输至PLC中,通过PLC控制十字滑台带动焊枪进行焊缝的跟踪操作。避免了强光、灰尘和噪声等干扰,提高了焊缝特征信息检测的准确性。通过与Faster R-CNN和YOLOv5的预测实验对比,Mask R-CNN在焊缝检测中的准确度和稳定性更高。

The advances in understanding the South China Sea （SCS） western boundary current （SCSwbc） have been reviewed since the works of Dale （1956） and Wyrtki （1961） in the middle of the 20th century. The features of the pattern of SCSwbc and the oceanic phenomena associated with it are focused on. The current is driven mainly by monsoon over the SCS and partially by winds over the tropical Pacific governed by the island rule. The SCSwbc exhibits strong seasonal variation in its direction and patterns. In winter, the current is strong and flows southwestward along the South China shelf and slope from the east of Dongsha Islands to the northern central Vietnamese coast, then turns to the south along the central and southern Vietnamese coast, and finally partially exits the SCS through the Karimata Strait. In summer and early fall, the SCSwbc can be divided into three segments based on their characteristics. The southern segment is stable, flowing northward from the Karimata Strait up to about 11 N, where it separates from the coast forming an eastward offshore current. The separation of the current from Vietnamese coast induces some striking features, such as upwelling and cold sea-surface temperature. The middle segment off the central Vietnamese coast may have a bimodal behavior： northward coastal current and meandering current in early summer （June–July）, and cyclonic gyre in later summer and early fall （August–September）. The northern segment is featured by the summer SCS Warm Current on the South China shelf and a southwestward subsurface current along the continental slope.

11-year satellite altimeter sea surface height (SSH) anomaly data from January 1993 to December 2003 are used to present the dominant spatial patterns and temporal variations of the South China Sea (SCS) surface circulation through Empirical Orthogonal Function (EOF) analysis. The first three EOF modes show the obvious seasonal variations of SSH in the SCS. EOF mode one is generally characterized by a basin-wide circulation. Mode two describes the double-cell basin scale circulation structure. The two cells were located off west of the Luzon Island and southeast of Vietnam, respectively. EOF mode three presents the mesoscale eddy structure in the western SCS, which develops into a strong cyclonic eddy rapidly from July to September. EOF mode one and mode three are also embedded with interannual signals, indicating that the SCS surface circulation variation is influenced by El Ni o events prominently. The strong El Ni o of 1997/98 obviously changed the SCS circulation structure. This study also shows that there existed a series of mesoscale eddies in the western SCS, and their temporal variation indicates intra-seasonal and in-terannual signals.

Direct synthesis of high-quality graphene on dielectric substrates without a transfer process is of vital importance for a variety of applications.Current strategies for boosting high-quality graphene growth,such as remote metal catalyzation,are limited by poor performance with respect to the release of metal catalysts and hence suffer from a problem with metal residues.Herein,we report an effective approach that utilizes a metal-containing species,copper acetate,to continuously supply copper clusters in a gaseous form to aid transfer-free growth of graphene over a wafer scale.The thus-derived graphene films were found to show reduced multilayer density and improved electrical performance and exhibited a carrier mobility of8500 cm~2 V-1s-1.Furthermore,droplet-based hydrovoltaic electricity generator devices based on directly grown graphene were found to exhibit robust voltage output and long cyclic stability,in stark contrast to their counterparts based on transferred graphene,demonstrating the potential for emerging energy harvesting applications.The work presented here offers a promising solution to organize the metal catalytic booster toward transfer-free synthesis of high-quality graphene and enable smart energy generation.
