Comparison of Responses to Mn Deficiency Between the UK Wheat Genotypes Maris Butler, Paragon and the Australian Wheat Genotype C8MM

Wheat grown in Mn-deficient soil has been widely observed to produce much reduced yields. Breeding for Mn-efficient wheat genotypes adapted to Mn-deficient soils would represent a long-term solution for wheat agronomy. To characterize the physiological basis of Mn efficiency in wheat genotypes would facilitate the breeding programs for producing Mn-efficient wheat. Using a solution culture and a soil culture system in the present study, a Mn-efficient UK wheat genotype Maris Butler and a Mn-inefficient UK wheat genotype Paragon have been compared with a Mn-efficient Australian wheat genotype C8MM in the responses to Mn deficiency in order to characterize the Mn efficiency in these wheat genotypes. Results showed that in solution culture, Marls Butler grown under Mn deficiency had 77% relative dry matter yield of control plants that were grown under Mn sufficiency, whereas CSMM and Paragon had 60% and 58% relative dry matter yield of their respective controls. Results from the soil culture demonstrated that relative dry matter yield remained high for Maris Butler and C8MM （53% and 56%, respectively）, whereas the value for Paragon dropped to 33%. In terms of dry matter yield and photosynthetic efficiency, Maris Butler demonstrated Mn efficiency in both solution culture and soil culture, whereas C8MM showed Mn efficiency only in soil culture. Results also demonstrated that under Mn-depleted supply in soil, plants of C8MM had a significantly higher ability in Mn uptake, whereas plants of Marls Butler showed a higher internal Mn use efficiency in comparison with plants of Paragon. Results from the present study indicate that the ability of C8MM to accumulate higher amounts of Mn is the basis of the improved Mn efficiency of this genotype in comparison with Paragon, and in Marls Butler there is a higher internal use of Mn expressed as an improved photosynthetic efficiency in conferring its Mn efficiency. Based on the first type of Rayleigh Sommerfeld diffraction formula, an imaging model of circular aperture-array structure digital sun sensor optical system is developed. Then a 6×6 circular aperture-array structure optical system is designed. The results of numerical simulation show that the optical system is designed well and is conformed to the requirements of miniaturization and high accuracy of sun sensor.

Background Radiotherapy and adjuvant cisplatin chemotherapy were the mainstream in the treatment of nasopharyngeal carcinoma(NPC) and effectively improved the outcome and reduced tumor ***,radiotherapy and chemotherapy resistance in the course of treatment,which is an important cause of failure in NPC therapy,has become more common. Therefore,there is an urgent need to explore new therapeutic strategies or new adjuvant drugs. Aim However,little information about autophagy in nasopharyngeal carcinoma cells is available. In this study,we investigated the effects of autophagy inhibitor on ER stress-induced autophagy in nasopharyngeal carcinoma *** Two human nasopharyngeal-carcinoma-derived cell lines,CNE-1 and CNE-2,were used to investigate the relationship between induction by cisplatin (DDP),ionizing radiation(IR),2-deoxy-D-glucose(2-DG),tunicamycin(TM),3-methyladenine (3-MA,an autophagy inhibitor),or their combination treatment of ER stress and activation of *** cell proliferation was detected by MTT assay and colony formation assay. Meanwhile,Annexin V-FITC/PI double staining,JC-1 staining and DAPI staining were used to observe ***,ER stress(GRP78) and autophagy(beclinl,LC3) markers were assayed by Western blotting,while autophagy was also measured by immunocytochemistry utilizing beclinl *** Combining 3-MA with DDP,IR,2-DG or TM resulted in enhanced cell death compared with each agent along by MTT assay and colony formation assay. The Hexokinase inhibitor 2-Deoxy-D-glucose(2-DG) is a synthetic glucose analogue that inhibits glycolysis and glycosylation and blocks cell *** is also a classical induce of endoplasmic reticulum(ER) *** is undergoing clinical trials as an anticancer *** studies have shown that sensitivity of breast cancer cells to TNF-related apoptosis-inducing ligand (TRAIL) -induced apoptosis is largely correlated with the expression levels of TRAIL death receptors on the cell ***,fresh breast cancer isolates and breast cancer tissue sections express generally low levels of death receptors for *** clinical potential of TRAIL in the treatment of breast cancer may therefore be limited unless given with agents that increase the cell surface expression of TRAIL death *** this study we report that whether 2-DG and TRAIL enhanced sensitivity of the cells to apoptosis -induced by *** addition, which protein play the decisive role in this signaling *** result indicate that 2-DG did not kill breast cancer cells,but enhanced TRAIL-induced apoptosis in breast cancer *** 2-DG sensitizes breast cancer cells to TRAIL-induced apoptosis by down-regulation of RIP1 and up-regulation of *** suggested that 2-DG can become a promising agent to increase the therapeutic response to TRAIL in breast cancer.

Tumor metastasis is the main performance of cancer,also it is the crucial reason for mortality in *** cells through the natural barrier which consisting of the basement membrane(BM) and extra cellular matrix(ECM) is the most important step in tumor invasion and metastasis,so protecting this barrier is the key to reduce *** is a mammalian endo-beta-glucuronidase which is only found to promotes the cleavage of heparan sulfate(HS),it is of significance in tumor cell invasion and *** chemotherapeutic reagents can have a strong anti-tumor activity on tumor cells,meanwhile,they may lead to invasion and migration,this is appeared in the clinical *** report in this study that activation of the endoplasmic reticulum stress(ER stress) is involved in enhance invasion and migration by two chemotherapeutic reagents,Tunicamycin(TM) and Adriamycin(ADM).And we hypothesized that is associated with activation of *** support this,we identified heparanase inhibitor OGT-2115 and Low molecular weight heparin(LMWH) that can directly target heparanase to inhibit the expression and activity of heparanase in breast cancer *** found that invasion and migration can be suppressed by heparanase ***,our findings illustrate how heparanase inhibitors are linked to breast cancer cells through heparanase control,and they offer a strong rationale to develop heparanase-based therapeutics for metastasis under treatment with chemotherapeutic reagents.

Real-world evidence on the efficacy and safety of vonoprazanamoxicillin dual therapy for Helicobacter pylori treatment in elderly patients

BACKGROUND A dual therapy regimen containing amoxicillin is a common treatment option for the eradication of Helicobacter pylori(***).While substantial research supports the efficacy and safety of vonoprazan and amoxicillin(VA)dual therapy in the general population,there is still a lack of studies specifically focusing on its safety in elderly *** To evaluate efficacy and safety of VA dual therapy as first-line or rescue treatment for *** in elderly *** As a real-world retrospective study,data were collected from elderly patients aged 60 years and above who accepted VA dual therapy(vonoprazan 20 mg twice daily+amoxicillin 1000 mg thrice daily for 14 days)for *** eradication in the Department of Gastroenterology at Peking University First Hospital between June 2020 and January *** status was evaluated by^(13)C-urease breath test 6 weeks after *** adverse events(AEs)during treatment were *** In total,401 cases were ***-one cases were excluded due to loss to follow-up,lack of re-examination,or unwillingness to take *** total of 380 included cases comprised 250 who received VA dual therapy as first-line treatment and 130 who received VA dual therapy as rescue *** was successfully eradicated in 239 cases(95.6%)in the first-line treatment group and 116 cases(89.2%)in the rescue treatment *** overall incidence of AEs was 9.5%for both ***,9.2%of patients experienced an AE in the first-line treatment group and 10.0%in the rescue treatment *** patients discontinued treatment due to AE,with a discontinuation rate of 1.3%.No serious AE *** The VA dual therapy regimen as a first-line treatment and a rescue therapy was effective and safe for elderly patients aged 60 and older.

Aim Activation of the unfolded protein response(UPR) is involved in resistance of nasopharyngeal-carcinoma(NPC) cells,and this is associated with glucose-regulated protein 78 (GRP78).So we investigate that inhibiting GRP78 expression can reverse drug-resistance on NPC ***,cell proliferation can be reduced and cell apoptosis can be *** Two human nasopharyngeal-carcinoma-derived cell lines,HNEl and HNEl/DDP,are used to examine the relationship between endoplasmic reticulum stress(ER Stress) and drug resistance by treated with cisplatin(DDP).The cell proliferation is detected by MTT ***,PI staining,PI/Annexin V staining,JC-1 staining are used to observe ***,GRP78 and caspase-3 expression are assayed by Western *** After treated with DDP,cell proliferation of HNEl/DDP is more than HNEl by MTT assays and cell apoptosis of HNEl/DDP is less than HNEl by PI staining,PI/Annexin V staining and JC-1 *** of GRP78 expression can reverse drug-resistance on NPC cells by PI/Annexin V *** Overall,these results indicate that HNE1 is more sensitive than HNE1/DDP after treated with *** of GRP78 expression can reverse cisplatin-resistance on NPC *** of GRP78 gives us a new revelation of targeted therapy of NPC *** we want further study of this experiment in vivo and in clinical trails.
