Ki-67 Change in Anthracyline-containing Neoadjuvant Chemotherapy Response in Breast Cancer

Objective Anthracycline-containing regimens are irreplaceable in neoadjuvant chemotherapy(NAC)for breast cancer(BC)at ***,30% of early breast cancer(EBC)patients are resistant to anthracycline-containing chemotherapy,leading to poor prognosis and higher ***-67 is associated with the prognosis and response to therapy,and it changes after *** A total of 105 BC patients who received anthracycline-containing NAC were ***,the optimal model of Ki-67 was selected,and its predictive efficacy was ***(IHC)was used to determine the estrogen receptor(ER),progesterone receptor(PR),and human epidermal growth factor receptor 2(HER-2)status and Ki-67 *** in situ hybridization(FISH)was used to verify the HER-2 when the IHC score was 2+.Results The post-NAC Ki67 level after treatment with anthracycline drugs was lower than pre-NAC Ki-67(19.6%±23.3%vs.45.6%±23.1%,P<0.001).Furthermore,patients with the Ki-67 decrease had a border line higher pathological complete response(pCR)rate(17.2%vs.0.0%,P=0.068),and a higher overall response rate(ORR)(73.6%vs.27.8%,P<0.001),when compared to patients without the Ki-67 *** ΔKi-67 and ΔKi-67%were valuable markers for the prediction of both the pCR rate and *** area under the curve(AUC)for ΔKi-67 on pCR and ORR was 0.809(0.698-0.921)and 0.755(0.655-0.855),respectively,while the AUC for ΔKi-67% on pCR and ORR was 0.857(0.742-0.972)and 0.720(0.618-0.822),*** logistic regression model 1 revealed thatΔKi-67 was an independent predictor for both pCR[odds ratio(OR)=61.030,95% confidence interval(CI)=4.709-790.965;P=0.002]and ORR(OR=10.001,95%CI:3.044-32.858;P<0.001).Multivariate logistic regression model 2 revealed thatΔKi-67%was also an independent predictor for both pCR(OR=408.922,95%CI=8.908-18771.224;P=0.002)and ORR(OR=5.419,95%CI=1.842-15.943;P=0.002).Conclusions The present study results suggest thatΔKi67 andΔKi67%are candidate predictor

Regulation of a vacuolar proton-pumping P-ATPase MdPH5 by MdMYB73 and its role in malate accumulation and vacuolar acidification

As the main organic acid in fruits,malate is produced in the cytoplasm and is then transported into the *** accumulates by vacuolar proton pumps,transporters,and channels,affecting the taste and flavor of *** the three types of proton pumps(V-ATPases,V-PPases,and P-ATPases),the P-ATPases play an important role in the transport of malate into *** this study,the transcriptome data,collected at different stages after blooming and during storage,were analyzed and the results demonstrated that the expression of MdPH5,a vacuolar proton-pumping P-ATPase,was associated with both pre-and post-harvest malate ***,MdPH5 is localized at the tonoplast and regulates malate accumulation and vacuolar *** addition,MdMYB73,an upstream MYB transcription factor of MdPH5,directly binds to its promoter,thereby transcriptionally activating its expression and enhancing its *** this way,MdMYB73 can also affect malate accumulation and vacuolar ***,this study clarifies how MdMYB73 and MdPH5 act to regulate vacuolar malate transport systems,thereby affecting malate accumulation and vacuolar pH.

Sugars are involved in plant growth,fruit quality,and signaling ***,understanding the mechanisms involved in soluble sugar accumulation is essential to understand fruit ***,we report that Md PFPβ,a pyrophosphatedependent phosphofructokinase gene,regulates soluble sugar accumulation by enhancing the photosynthetic performance and sugar-metabolizing enzyme activities in apple(Malus domestica Borkh.).Biochemical analysis revealed that a basic helix-loop-helix(b HLH)transcription factor,Mdb HLH3,binds to the Md PFPβpromoter and activates its expression,thus promoting soluble sugar accumulation in apple *** addition,Md PFPβoverexpression in tomato influenced photosynthesis and carbon metabolism in the ***,we determined that Mdb HLH3 increases photosynthetic rates and soluble sugar accumulation in apple by activating Md PFPβ*** results thus shed light on the mechanism of soluble sugar accumulation in apple leaves and fruit:Mdb HLH3 regulates soluble sugar accumulation by activating Md PFPβgene expression and coordinating carbohydrate allocation.

Scattering of Water Waves by Dual Symmetric Inclined Floating Porous Barriers Using the DBEM

Shear-thinning Catechol-modified Chitosan Hydrogel Loaded with Silver Nanoparticles for Endoscopic Submucosal Dissection

Shear-thinning hydrogels have emerged for endoscopic submucosal dissection, while wound intervention after surgery has rarely been mentioned. Herein, a catechol-modified chitosan hydrogel with shear-thinning property was developed for simultaneously facilitating endoscopic submucosal dissection and postoperative wound healing. Benefiting from the shear-thinning and self-healing characteristics, the asprepared hydrogel showed easily endoscopic injectability. It also performed very well as submucosal cushion, which could remain above 70% after injection for 120 min in ex vivo porcine large intestine model. In fact, the cushion height of normal saline dramatically decreased to 46% of the initial height at 30 min. Ag nanoparticles encapsulated into the network endowed the hydrogel with almost reached 100% antibacterial effect against E. coli and S. aureus. The hemolysis ratio of the hydrogel was calculated to be as low as 0.8%. Combined with good hemocompatibility and cytocompatibility, the as-prepared hydrogel displayed much higher in vivo wound closure and healing efficacy than normal saline. These results demonstrated the superiority of the shear-thinning chitosan hydrogel in facilitating clinical endoscopic submucosal dissection surgery.

A Study of 10 Rotating Radio Transients Using Parkes Radio Telescope

Rotating Radio Transients(RRATs)are a relatively new subclass of pulsars that emit detectable radio bursts *** analyzed 10 RRATs observed using the Parkes telescope,with eight of these observed via the ultra-wide-bandwidth low-frequency(UWL)*** measured the burst rate and produced integrated profiles spanning multiple frequency bands for three *** also conducted a spectral analysis on both integrated pulses and individual pulses of three *** of their integrated pulses follow a simple power law,consistent with the known range of pulsar spectral *** average spectral indices of single pulses are-0.9,-1.2,and-1.0 respectively,which are within the known range of pulsar spectral ***,we find that the spreads of single-pulse spectral indices for these RRATs(ranging from-3.5 to+0.5)are narrower compared to what has been observed in other ***,the average spectral index and scatter of single pulses are both relatively *** the remaining five RRATs observed at the UWL receiver,we also provide the upper limits on fluence and flux *** addition,we obtain the timing solution of PSR *** analysis shows that PSRs J1919+1745,J1709-43,and J1649-4653 are potentially nulling pulsars or weak pulsars with sparse strong pulses.

Identification of the ice channel is the basic technology for developing intelligent ships in ice-covered waters,which is important to ensure the safety and economy of *** the Arctic,merchant ships with low ice class often navigate in channels opened up by *** in the ice channel often depends on good maneuverability skills and abundant experience from the captain to a large *** ship may get stuck if steered into ice fields off the *** this circumstance,it is very important to study how to identify the boundary lines of ice channels with a reliable *** this paper,a two-staged ice channel identification method is developed based on image segmentation and corner point *** first stage employs the image segmentation method to extract channel *** the second stage,an intelligent corner regression network is proposed to extract the channel boundary lines from the channel region.A non-intelligent angle-based filtering and clustering method is proposed and compared with corner point regression *** training and evaluation of the segmentation method and corner regression network are carried out on the synthetic and real ice channel *** evaluation results show that the accuracy of the method using the corner point regression network in the second stage is achieved as high as 73.33%on the synthetic ice channel dataset and 70.66%on the real ice channel dataset,and the processing speed can reach up to 14.58frames per second.

Significantly Enhanced Melt Memory Effect of Metallocene-made Isotactic Polypropylene Containing Talc

The melt memory effect is a widely observed phenomenon in semi-crystalline polymers. In practical applications, various additives are usually introduced into polymers, which may affect their melt memory behavior. In this work, the effect of talc on the melt memory effect of metallocene-made isotactic polypropylene(M-PP) was investigated in detail by using the differential scanning calorimetry. The results indicated that the introduction of talc significantly strengthened the melt memory effect of M-PP. Specifically, the upper limit temperature of Domain II increased from 161 ℃ to 174 ℃, resulting in a substantial widening of the temperature range of Domain IIa from 1 ℃ to 14 ℃. Analysis of the crystal orientation of the M-PP containing talc cooled from various Ts suggested that the remarkably enhanced melt memory effect could be ascribed to the stabilization of oriented nuclei facilitated by talc. This stabilizing effect was likely attributable to the prefreezing effect or the sorption interaction between talc and the M-PP chains.

Grain boundaries,including low-angle,high-angle,and coinci-dence boundaries,are intrinsic interfaces in polycrystalline *** heat-treatment at sufficiently high temperatures,grain boundary migration occurs because the grain boundaries are ther-modynamically ***,grain growth occurs due to a decrease in grain boundary area after heat-treatment[1].In general,there are two types of grain growth:normal grain growth,where grains grow uniformly,and abnormal grain growth,where one or a few grains selectively grow and engulf surround-ing smaller grains,leading to the formation of large grains,some-times several millimeters or greater in size[2].The classical tech-nique to achieve abnormal grain growth is the strain-annealing method,which involves subjecting the material to a small strain followed by annealing[3,4].This method has been reported to ob-tain single-crystal wires or plates[1].Additionally,abnormal grain growth can be achieved by plastic straining at high temperatures,resulting in the fabrication of large grains or single crystals[5].However,the above methods are not applicable to components with complicated shapes due to the inevitable deformation ***,Omori et *** a simply method of abnor-mal grain growth to fabricate ultra-large grains,even single crys-tals,in Cu-Al-Mn[1],Fe-Mn-Al-Ni[2],and Fe-Mn-Al-Ni-Cr[6]*** method involves applying cyclic heat treatment between a high temperature BCC single-phase region and a low tempera-ture BCC+FCC two-phase region[1,2,6-12].Note that the cyclic heat treatment resulted in the formation of BCC sub-grain struc-tures responsible for abnormal grain growth[1,2,6-12].In addition,yang et *** that abnormal grain growth occurred at above 900℃,and large-grains,even single crystals,were obtained only through annealing cast Cu-based polycrystalline alloys contained coherent nano-particles[13,14].Notably,the cyclic heat treatment and annealing cast alloys are deform
