Anodal transcranial direct current stimulation(AtDCS)has been shown to alleviate cognitive impairment in an APP/PS1 model of Alzheimer’s disease in the preclinical stage.However,this enhancement was only observed immediately after AtDCS,and the long-term effect of AtDCS remains unknown.In this study,we treated 26-week-old mouse models of Alzheimer’s disease in the preclinical stage with 10 AtDCS sessions or sham stimulation.The Morris water maze,novel object recognition task,and novel object location test were implemented to evaluate spatial learning memory and recognition memory of mice.Western blotting was used to detect the relevant protein content.Morphological changes were observed using immunohistochemistry and immunofluorescence staining.Six weeks after treatment,the mice subjected to AtDCS sessions had a shorter escape latency,a shorter path length,more platform area crossings,and spent more time in the target quadrant than sham-stimulated mice.The mice subjected to AtDCS sessions also performed better in the novel object recognition and novel object location tests than sham-stimulated mice.Furthermore,AtDCS reduced the levels of amyloid-β42 and glial fibrillary acidic protein,a marker of astrocyte activation,and increased the level of neuronal marker NeuN in hippocampal tissue.These findings suggest that AtDCS can improve the spatial learning and memory abilities and pathological state of an APP/PS1 mouse model of Alzheimer’s disease in the preclinical stage,with improvements that last for at least 6 weeks.

BACKGROUND Wild rats have the potential to hold zoonotic infectious agents that can spread to humans and cause disease.AIM To better understand the composition of gut bacterial communities in rats is essential for preventing and treating such diseases.As a tropical island located in the south of China,Hainan province has abundant rat species.Here,we examined the gut bacterial composition in wild adult rats from Hainan province.METHODS Fresh fecal samples were collected from 162 wild adult rats,including three species(Rattus norvegicus,Leopoldamys edwardsi,and Rattus losea),from nine regions of Hainan province between 2017-2018.RESULTS We analyzed the composition of gut microbiota using the 16S rRNA gene amplicon sequencing.We identified 4903 bacterial operational taxonomic units(30 phyla,175 families,and 498 genera),which vary between samples of different rat species in various habitats at various times of the year.In general,Firmicutes were the most abundant phyla,followed by Bacteroidetes(15.55%),Proteobacteria(6.13%),and Actinobacteria(4.02%).The genus Lactobacillus(20.08%),unidentified_Clostridiales(5.16%),Romboutsia(4.33%),unidentified_Ruminococcaceae(3.83%),Bacteroides(3.66%),Helicobacter(2.40%)and Streptococcus(2.37%)were dominant.CONCLUSION The composition and abundance of the gut microbial communities varied between rat species and locations.This work provides fundamental information to identify microbial communities useful for disease control in Hainan province.

目的:鉴定参与梨火疫菌侵染的重要Sec依赖分泌蛋白酶。创新点:构建了梨火疫菌Sec依赖分泌蛋白酶编码基因在侵染寄主植物过程中的时序表达图谱。方法:利用生物信息学与大肠杆菌Pho A检测体系两者结合,在全基因组水平筛选并鉴定梨火疫菌的Sec依赖分泌蛋白酶;利用逆转录实时定量聚合酶链反应(RT-q PCR),分析Sec依赖分泌蛋白酶编码基因在梨火疫菌侵染寄主植物过程中转录表达的时序变化。结论:梨火疫菌含有完整的Sec分泌系统,可由此分泌至少15种蛋白酶,其中11种蛋白酶可能在病原菌侵染寄主植物过程中发挥功能。

As an important herbaceous plant, Scutellaria baicalensis Georgi(Chinese skullcap) is geographically widespread and commonly used throughout the world. In the Chinese medicine market, S. baicalensis has been divided into two primary types, "Ku Qin"(WXR) and "Tiao Qin"(TST). Moreover, TST is also divided into different grades according to the diameter of roots. To explore the distribution patterns of the contents of five biologically activate ingredients(FBAI), we used six-year-old cultivated S. baicalensis and analyzed its growth characteristics as well as the quality difference among different types and diameters in roots. Throughout the entire root, we discovered that contents of the FBAI all initially increased and subsequently decreased from the top to the bottom of the roots. The baicalin content of WXR was less than that of TST. On the contrary, the contents of baicalein, wogonin, and oroxylin A in WXR were up to about two times higher than that in TST. We also found that the 0 to 40 cm part of the S. baicalensis root possessed about 87% of the root biomass and about 92% of the contents of the active ingredients.

To determine the risk, malignant degree and clinical progression of prostate cancer （PCa） associated with mouse double-minute 2 protein （MDM2） T309G variants, a meta-analysis was performed on all eligible published studies. Odds ratios （ORs） with 95% confidence intervals （CIs） were estimated to assess these associations in seven studies that included 5151 cases and 1003 controls. In the overall analysis, the 309G allele was significantly associated with a decreased PCa risk （0R=0.85, 95% CI： 0.74-0.97）; this was also the case for the homozygous comparison （0R--0.72, 95% Ch 0.55-0.95） and the dominant genetic model （0R=0.79, 95% Ch 0.65-0.96）. The 309G allele was also found to be significantly associated with lower degrees of PCa malignancy （0R=0.85, 95% Ch 0.75-0.96） in the overall analysis, as well as in the heterozygous comparison （0R=0.79, 95% Ch 0.65-0.96）, homozygous comparison （0R=0.76, 95% Ch 0.58-0.98） and dominant genetic model （0R=0.81, 95% CI： 0.68-0.96）. Furthermore, grouping analysis showed that the 309G allele in Caucasians was significantly correlated with a decreased PCa risk （0R=0.77, 95% Ch 0.61-0.96）; this was also the case in the homozygous comparison （0R=0.51, 95% Ch 0.31-0.86）. The grouping analysis also showed that the 309G variant in Caucasians was significantly associated with a lower degree of PCa malignancy in all of the genetic models. In addition, we found that the 309G variant in Caucasians was significantly associated with a slower PCa clinical progression in all of the genetic models. In summary, our meta-analysis showed that the MDM2 309G variant was significantly associated with a decreased PCa risk, lower malignant degree and slower clinical progression in Caucasians, but there was no obvious association in the Asian population.

Role of multi-detector computed tomography for biliary complications after liver transplantation

AIM: To investigate the diagnostic performance of multi-detector computed tomography(MDCT) in detecting biliary complications after orthotopic liver transplantation(OLT). METHODS: Eighty-three consecutive OLT recipients, who presented with clinical or biochemical signs of biliary complications, underwent MDCT examination. Two experienced radiologists assessed MDCT images in consensus to determine biliary complications. Final confirmation was based on percutaneous transhepatic cholangiography or endoscopic retrograde cholangiography in 58 patients, surgery in four patients, liver biopsy in 10, and clinical and sonography follow-up in 11 patients. RESULTS: Biliary complications were eventually confirmed in 62 of 83 patients(74.7%), including anastomotic biliary strictures in 32, nonanastomotic biliarystrictures in 21, biliary stones in nine(5 with biliary strictures), anastomotic bile leak in five, and biloma in six(all with nonanastomotic strictures, and 2 with biligenic hepatic abscess). Twenty-one patients had no detection of biliary complications. The sensitivity, specificity, accuracy, positive predictive value and negative predictive value of MDCT for detecting biliary strictures were 90.6%, 86.7%, 89.2%, 92.3% and 83.9%, respectively. For detecting biliary stones, anastomotic bile leak and biloma, the sensitivity, specificity, accuracy, positive predictive value and negative predictive value of MDCT were all 100%.CONCLUSION: MDCT is a useful screening tool for detecting biliary complications after OLT.

马克思在《1857-1858年经济学手稿》中科学揭示了资本的生产界限、流通界限以及资本本身的界限。资本的生产界限指资本要不断实现自身的价值增殖,就必然要突破自身的生产界限,使其内含的必要劳动趋近于零。一旦必要劳动完全为零,即工人的劳动被完全排挤掉,资本就无法再榨取工人的剩余劳动,无法再实现自身的价值增殖。此时,资本就走向了自身的反面,其对价值增殖的无限追求反而成为否定自身价值增殖的力量。资本的流通界限则由两方面构成,一方面,资本的流通界限表现为社会对产品的"总需要"或"总消费量"对资本价值增殖的制约和限制;另一方面,资本的流通界限还表现为流通时间和流通费用对资本增殖本性的制约和限制。资本要想实现价值增殖,就要不断地提高劳动生产力,不断排挤工人的劳动,由此就形成了资本自身既要吸收活劳动又要排挤活劳动这一现实的矛盾,这既是资本的内在矛盾,同时也构成资本本身的界限。资本作为一种矛盾性的存在,在不断实现价值增殖,不断努力突破自身界限的过程中,必然会走向自身的反面,实现自我的历史性扬弃。由此,资本现实运动的必然指向和根本解决之道只能是走向"共同生产"。

BACKGROUND Interleukin(IL)-34 is a pro-inflammatory cytokine involved in tumor development.The role of IL-34 in the proliferation and epithelial-mesenchymal transition(EMT)of gastric cancer(GC)remains to be investigated.AIM To investigate whether and how IL-34 affects the proliferation of GC cells and EMT.METHODS Using immunohistochemical staining,the expression of IL-34 protein was detected in 60 paired GC and normal paracancerous tissues and the relationship between IL-34 and clinicopathological factors was analyzed.The expression of IL-34 mRNA and protein in normal gastric epithelial cell lines and GC was detected using quantitative real-time polymerase chain reaction(qRT-PCR)and western blotting,respectively.Stable IL-34 knockdown and overexpression in AGS cell lines were established by lentiviral infection and validated by qRT-PCR and western blotting.The cholecystokinin-8 assay,clone formation assay,cell scratch assay,and transwell system were used to detect GC cell proliferation,clone formation,migration,and invasion capacity,respectively.The effects of IL-34 on the growth of GC transplant tumors were assessed using a subcutaneous transplant tumor assay in nude mice.The effects of IL-34 on the expression level of EMT-associated proteins in AGS cells were examined by western blotting.RESULTS Expression of IL-34 protein and mRNA was higher in GC cell lines than in GES-1 cells.Compared to matched normal paraneoplastic tissues,the expression of IL-34 protein was higher in 60 GC tissues,which was correlated with tumor size,T-stage,N-stage,tumor,node and metastasis stage,and degree of differentiation.Knockdown of IL-34 expression inhibited the proliferation,clone formation,migration,and invasion of AGS cells,while overexpression of IL-34 promoted cell proliferation,clone formation,migration,and invasion.Furthermore,the reduction of IL-34 promoted the expression of E-cadherin in AGS cells but inhibited the expression of vimentin and N-cadherin.Overexpression of IL-34 inhibited E-cadherin expression but promoted e

AIM:To describe and compare pathologic findings in eyes enucleated after superselective ophthalmic arterial chemotherapy(SOAC)or SOAC with intravenous chemotherapy(IVC)for retinoblastoma.METHODS:Medical records between January 1st,2014 and June 30th,2017 were retrospectively analyzed,and pathologic findings were recorded.This study included 36 eyes from 22(61.1%)male and 14(38.9%)female patients.Nineteen of 36(52.8%)eyes received SOAC(mean=3,range=1-7)as primary treatment,and 17 of 36(47.2%)eyes received SOAC(mean=3.7,range=1-10)after IVC(mean=6.1,range=2-11).Tumor extension including choroidal invasion(n=9,25%),optic nerve invasion(n=5,13.9%)and anterior segment invasion(n=5,13.9%)were recorded.RESULTS:Histopathologic evidence of ischemic damage in the retina and choroid was found in 28(77.8%)eyes.Thrombosed blood vessels were identified in 9(25%)eyes,including orbital artery in the retrobulbar orbit(n=1),intrascleral vessels(n=4),and chorioretinal vessels(n=6).Fibrotic changes were found in extraocular muscles(n=5,13.9%)and optic nerve(n=5,13.9%).Varying degrees of scleral degeneration were found in all eyes.In statistical analysis,there was no significant difference in clinical and pathologic changes between SOAC group and SOAC with IVC group except for optic nerve invasion(P=0.047).CONCLUSION:SOAC for retinoblastoma can result in ocular toxicity,and SOAC with IVC do not increase the toxicity but reduced the incidence of optic nerve invasion.
