Functional analysis of UG7207D3 associated with abamectin resistance in Tetranychus cinnabarinus(Boisduval)

Uridine diphosphate(UDP)-glycosyltransferases(UGTS)are widely distributed within living organisms and share roles in biotransformation of various lipophilic endo-and xenobiotics with activated UDP sugars.In this study,it was found that the activity of UGTs in abamectin-resistant(AbR)strain was significantly higher(2.3-fod)than that in susceptible strain(SS)of Tetramychus cinnabarinus.Further analysis showed that 5-nitrouracil,the inhibitor ofUGTs,could enhance the lethal effect of abamectin on mites.From the previous microarray results,we found an UGT gene(UGT201D3)overexpressed in AbR strain.Quantitative PCR analysis showed that UGT20ID3 was highly expressed and more inducible with abamectin exposure in the AbR strain.After silencing the tran-scription of UGT201D3,the activity of UGTs was decreased and the susceptibility to abamectin was increased in AbR strain whereas it was not in sS.Furthermore,UGT201D3 gene was then succefully expressed in Escherichia coli.The recombinant UGT20ID3 exhibited a-naphthol activity(2.810.43 nmol mg protein/min),and the enzyme activity could be inhibited by abamectin(inhibitory concentration at 50%:57.50±3.54 μmol/L).High-performance liquid chromatography analysis demonstrated that the recombinant UGT201D3 could efctively deplete abamectin(15.77%±3.72%)incubating with 150 ug protein for 6 h.These results provided direct evidence that UGT201D3 was involved in abamectin resistance in T cinnabarinus.

目的评价我国健康教育机构服务效果，找出存在的问题，并给出切实可行的解决方案。方法采用熵权优劣解距离法（technique for order preference by similarity to an ideal solution, TOPSIS）与秩和比法（rank-sum ratio, RSR）模糊联合的方法对我国31个省（自治区、直辖市）的健康教育机构服务效果进行综合评价。结果研究选取14项健康教育机构服务效果评价指标。其中指标权重排名前两位为手机短信覆盖人次数、音像制品数量，后两名为主办网站数、与电视台合办栏目数。经过熵权TOPSIS法评价显示，各地区Ci值在0.022～0.665之间；RSR法评价结果显示，健康教育机构服务效果处在较差、一般、良好、优秀的地区数分别为2、13、13和3个。两者结果与模糊联合模型结果总体趋势基本一致。结论熵权TOPSIS法和RSR法模糊联合可以较好的评价我国健康教育机构服务效果。我国健康教育机构服务存在内容和地区不平衡的现象，应增大健康教育宣传和人力资源的总投入，针对各地区的差异提出因地制宜的措施，以提升健康教育机构服务效果，增强居民幸福感和满足感。

Ubiquitin-specific protease 21 promotes tumorigenicity and stemness of colorectal cancer by deubiquitinating and stabilizing ZEB1

BACKGROUND Colorectal cancer(CRC)is one very usual tumor together with higher death rate.Ubiquitin-specific protease 21(USP21)has been confirmed to take part into the regulation of CRC progression through serving as a facilitator.Interestingly,the promotive function of USP21 has also discovered in the progression of CRC.ZEB1 has illustrated to be modulated by USP7,USP22 and USP51 in cancers.However,the regulatory functions of USP21 on ZEB1 in CRC progression need more invest-igations.AIM To investigate the relationship between USP21 and ZEB1 in CRC progression.METHODS The mRNA and protein expressions were assessed through RT-qPCR,western blot and IHC assay.The interaction between USP21 and ZEB1 was evaluated through Co-IP and GST pull down assays.The cell proliferation was detected through colony formation assay.The cell migration and invasion abilities were determined through Transwell assay.The stemness was tested through sphere formation assay.The tumor growth was evaluated through in vivo mice assay.RESULTS In this work,USP21 and ZEB1 exhibited higher expression in CRC,and resulted into poor prognosis.Moreover,the interaction between USP21 and ZEB1 was further investigated.It was demonstrated that USP21 contributed to the stability of ZEB1 through modulating ubiquitination level.In addition,USP21 streng-thened cell proliferation,migration and stemness through regulating ZEB1.At last,through in vivo assays,it was illustrated that USP21/ZEB1 axis aggravated tumor growth.CONCluSION For the first time,these above findings manifested that USP21 promoted tumorigenicity and stemness of CRC by deubiquitinating and stabilizing ZEB1.This discovery suggested that USP21/ZEB1 axis may provide novel sights for the treatment of CRC.

Intestinal failure-associated liver disease(IFALD)is a common complication of long-term parenteral nutrition that is associated with significant morbidity and mortality.It is mainly characterized by cholestasis in children and steatohepatitis in adults.Unfortunately,there is no effective approach to prevent or reverse the disease.Regulated cell death(RCD)represents a fundamental biological paradigm that determines the outcome of a variety of liver diseases.Nowadays cell death is reclassified into several types,based on the mechanisms and morphological phenotypes.Emerging evidence has linked different modes of RCD,such as apoptosis,necroptosis,ferroptosis,and pyroptosis to the pathogenesis of liver diseases.Recent studies have shown that different modes of RCD are present in animal models and patients with IFALD.Understanding the pathogenic roles of cell death may help uncover the underlying mechanisms and develop novel therapeutic strategies in IFALD.In this review,we discuss the current knowledge on how RCD may link to the pathogenesis of IFALD.We highlight examples of cell death-targeted interventions aiming to attenuate the disease,and provide perspectives for future basic and translational research in the field.

Carbon nanofibers embedded with Fe-Co alloy nanoparticles via electrospinning as lightweight high-performance electromagnetic wave absorbers

As a lot of electromagnetic pollution and interference issues have emerged,to overcome electromagnetic interference,prevent electromagnetic hazards,and develop new high-performance electromagnetic wave(EMW) absorbers have become a significant task in the field of materials science.In this paper,a three-dimensional(3D) carbon nanofibers network with core-shell structure,embedded with varied molar ratios of iron and cobalt(4:0,3:1,2:2,1:3,0:4),was effectively synthesized(Fe/Co@C-CNFs) via electrospinning.The phase,microstructure,magnetic and EMW absorption properties were studied.It is discovered that Fe/Co@C-CNFs doped with iron:cobalt=1:1 have excellent EMW absorption capacity.When the matching thickness is 1.08 mm,the minimum reflection loss(RL) value is-18.66 dB,while the maximum effective absorption bandwidth(EAB) reaches 4.2 GHz(13.9-18 GHz) at a thickness of 1.22 mm.This is owing to the absorbers' superior impedance matching and multiple reflections as well as the conductivity,dielectric,and magnetic losses of carbon nanofibers embedded with Fe-Co alloy particles.In addition,the radar cross section(RCS) of the absorbers has been calculated by CST Studio Suite,showing that the absorbing coating can effectively reduce the RCS at various angles,especially for Fe/Co@C-CNFs doped with iron:cobalt=1:1.These findings not only provide new insights for the preparation of light-weight and high-performance electromagnetic wave absorbers,but also contribute to energy storage and conversion.

Hydrodynamics and OCM reaction performance in a bubbling fluidized bed reactor and a riser reactor

The reasonable reactor design is of great importance for increasing the C_(2) yield(C2H4 and C2H6)of the oxidative coupling of methane(OCM),and the OCM reactor should remove the heat released in reactions quickly and efficiently and minimize the consecutive reaction of ethylene to carbon oxides.The fluidized bed reactor is characterized by excellent heat transfer,superior mass transport,and large handling capacity,while fewer studies focused on large-scale fluidized bed reactors for the OCM reaction.Therefore,large cold-model experiments and computational fluid dynamics simulations were conducted to investigate hydrodynamics and the OCM reaction performance in a large-scale bubbling fluidized bed(BFB)and a large-scale riser.In the BFB reactor,consecutive reactions of ethylene are acute because of the strong gas back-mixing,high solids holdup,and non-uniform solids distribution.While the consecutive reactions of ethylene are negligible due to the plug flow structure and low solids holdup in the riser reactor.Further,both reactors can achieve isothermal operation for the OCM process.The C_(2) selectivity of 45.4% and C_(2) yield of 21.1% are obtained in the riser reactor,increasing by 20.3% and 5.8% individually than that in the BFB reactor.This study provides useful information and reference to the OCM reactor designandcommercialization.

Glucagon-like-peptide-1 receptor agonists and the management of type 2 diabetes-backwards and forwards

This editorial is stimulated by the article by Alqifari et al published in the World Journal of Diabetes(2024).Alqifari et al focus on practical advice for the clinical use of glucagon-like-peptide-1(GLP-1)receptor agonists(GLP-1RAs)in the management of type 2 diabetes and this editorial provides complementary information.We initially give a brief historical perspective of the development of GLP-1RAs stimulated by recognition of the‘incretin effect’,the substantially greater insulin increase to enteral when compared to euglycaemic intravenous glucose,and the identification of the incretin hormones,GIP and GLP-1.In addition to stimulating insulin,GLP-1 reduces postprandial glucose levels by slowing gastric emptying.GLP-1RAs were developed because native GLP-1 has a very short plasma half-life.The majority of current GLP-1RAs are administered by subcutaneous injection once a week.They are potent in glucose lowering without leading to hypoglycaemia,stimulate weight loss in obese individuals and lead to cardiovascular and renal protection.The landscape in relation to GLP-1RAs is broadening rapidly,with different formulations and their combination with other peptides to facilitate both glucose lowering and weight loss.There is a need for more information relating to the effects of GLP-1RAs to induce gastrointestinal symptoms and slow gastric emptying which is likely to allow their use to become more effective and personalised.

Facile intercalation of alkali ions in WO_(3)for modulated electronic and optical properties:Implications for artificial synapses and chromogenic application

Tungsten oxides(WO_(3))are widely recognized as multifunctional systems owing to the existence of rich polymorphs.These diverse phases exhibit distinct octahedra-tilting patterns,generating substantial tunnels that are ideally suited for iontronics.However,a quantitative comprehension regarding the impact of distinct phases on the kinetics of intercalated conducting ions remains lacking.Herein,we employ first-principles calculations to explore the spatial and orientational correlations of ion transport inγ-and h-WO_(3),shedding light on the relationship between diffusion barriers and the size of the conducting ions.Our findings reveal that different types and concentrations of alkali-metals induce distinct and continuous lattice distortions in WO_(3)polymorphs.Specifically,γ-WO_(3)is more appropriate to accommodate Li+ions,exhibiting a diffusion barrier and coefficient of 0.25 eV and 9.31×10^(-8)cm^(2)s^(-1),respectively.Conversely,h-WO_(3)features unidirectional and sizeable tunnels that facilitate the transport of K+ions with an even lower barrier and a high coefficient of 0.11 e V and 2.12×10^(-5)cm^(2)s^(-1),respectively.Furthermore,the introduction of alkali-metal into WO_(3)tunnels tends to introduce n-type conductivity by contributing s-electrons to the unoccupied W 5d states,resulting in enhanced conductivity and tunable electronic structures.These alkali metals in WO_(3)tunnels are prone to charge transfer,forming small polaronic states and modulating the light absorption in the visible and nearinfrared regions.These tunable electronic and optical properties,combined with the high diffusion coefficient,underscore the potential of WO_(3)in applications such as artificial synapses and chromogenic devices.

Evaluation of G3BP1 in the prognosis of acute and acute-on-chronic liver failure after the treatment of artificial liver support system

BACKGROUND The increased expression of G3BP1 was positively correlated with the prognosis of liver failure.AIM To investigate the effect of G3BP1 on the prognosis of acute liver failure(ALF)and acute-on-chronic liver failure(ACLF)after the treatment of artificial liver support system(ALSS).METHODS A total of 244 patients with ALF and ACLF were enrolled in this study.The levels of G3BP1 on admission and at discharge were detected.The validation set of 514 patients was collected to verify the predicted effect of G3BP1 and the viability of prognosis.RESULTS This study was shown that lactate dehydrogenase(LDH),alpha-fetoprotein(AFP)and prothrombin time were closely related to the prognosis of patients.After the ALSS treatment,the patient’amount of decreased G3BP1 index in difference of G3BP1 between the value of discharge and admission(difG3BP1)luSION This study indicated that G3BP1 could predict the prognosis of ALF or ACLF patients treated with ALSS.The combination of G3BP1,AFP and LDH could accurately evaluate the disease condition and predict the clinical endpoint of patients.

Dear Editor,We present the first time,a case of a patient developed cerebrospinal fluid(CSF)leak and pneumocranium following optic canal decompression(OCD).INTRODUCTION Indirect traumatic optic neuropathy(ITON)impairs visual functions and quality of life.Endoscopic transnasal optic canal decompression(ETOCD)is one of the standard treatment strategies for the ITON.During the ETOCD,the optic nerve sheath are usually incised for sufficient decompression of optic nerve after removal of optic canal,which is associated with complications like CSF leakage,ophthalmic artery injury,and optic nerve injury[1].Generally,the mild CSF leak is common and can heal spontaneously using conventional treatment,the severe CSF leak requires surgical repair[2].
