Aspirin suppresses hepatocellular carcinoma progression by inhibiting platelet activity

BACKGROUND Hepatocellular carcinoma(HCC)is the most common malignant liver disease in the ***(PLTs)are known to play a key role in the maintenance of liver homeostasis and the pathophysiological processes of a variety of liver *** is the most classic antiplatelet ***,the molecular mechanism of platelet action and whether aspirin can affect HCC progression by inhibiting platelet activity need further *** To explore the impact of the antiplatelet effect of aspirin on the development of *** Platelet-rich plasma,platelet plasma,pure platelet,and platelet lysate were prepared,and a coculture model of PLTs and HCC cells was ***-8 analysis,apoptosis analysis,Transwell analysis,and real-time polymerase chain reaction(RT-PCR)were used to analyze the effects of PLTs on the growth,metastasis,and inflammatory microenvironment of ***-PCR and Western blot were used to detect the effects of platelet activation on tumor-related signaling *** was used to block the activation and aggregation of PLTs both in vitro and in vivo,and the effect of PLTs on the progression of HCC RESULTS PLTs significantly promoted the growth,invasion,epithelial-mesenchymal transition,and formation of an inflammatory microenvironment in HCC *** PLTs promoted HCC progression by activating the mitogenactivated protein kinase/protein kinase B/signal transducer and activator of transcription three(MAPK/AKT/STAT3)signaling ***,aspirin inhibited HCC progression in vitro and in vivo by inhibiting platelet *** PLTs play an important role in the pathogenesis of HCC,and aspirin can affect HCC progression by inhibiting platelet *** results suggest that antiplatelet therapy has promising application prospects in the treatment and combined treatment of HCC.

A method to avoid the cycle-skip phenomenon in time-of-flight determination for ultrasonic flow measurement

The double-threshold method has been widely used in ultrasonic flow measurement to determine time-of-flight(TOF)due to its low cost and ease of *** of this method is negatively affected by the cycle-skip phenomenon which occurs frequently under inconstant working conditions,especially varied fluid *** paper proposes a method to suppress the phenomenon to facilitate reliable determination of TOF in ultrasonic flow ***,the double-threshold method is used to generate a feature point to segment the ***,based on the correlation coefficient and signal power,judgement factors of individual signal periods are calculated to determine signal ***,a valid zero crossing which has a constant lag from the onset is selected to determine the ***,the cycle-skip phenomenon is *** additional modifications are proposed to eliminate the influence of varied signal frequency and low sampling *** proposed method was validated by an experiment based on an ultrasonic water flow *** showed that the frequently appearing cycle-skip phenomenon can be successfully suppressed by the proposed method.

利用数据挖掘和网络药理学方法分析中药专利复方治疗肝纤维化的用药规律及机制。检索国家知识产权局中国专利公布公告网中治疗肝纤维化的中药专利复方,筛选整理后建立数据库,运用Microsoft Excel软件和中医传承计算平台(V3.0)对中药复方用药规律进行数据挖掘与分析。运用网络药理学方法探讨核心高频药对治疗肝纤维化的机制。最终筛选出专利复方147个,中药431味。高频药物包括黄芪、丹参、鳖甲和当归等,功效以补虚、活血化瘀和清热为主,核心高频药对黄芪-丹参治疗肝纤维化的主要活性成分为槲皮素、木犀草素、山奈酚和丹参酮IIA等,主要靶点为骨髓细胞瘤病毒癌基因(myelocytomatosis viral oncogene,MYC)、转录激活因子3(signal transducer and activator of transcription 3,STAT3)、丝氨酸/苏氨酸激酶1(serine/threonine kinase 1,AKT1)、雌激素受体1(estrogen receptor 1,ESR1)、FOS原癌基因(FOS proto-oncogene,FOS)、缺氧诱导因子1亚基α(hypoxia-inducible factor 1 alpha,HIF-1a)、肿瘤蛋白p53(tumor protein p53,TP53)、丝裂原活化蛋白激酶14(mitogen-activated protein kinase14,MAPK14)等,主要涉及磷脂酰肌醇3-激酶(phosphatidylinositol-3-kinase,PI3K)-蛋白激酶B(protein kinase B,Akt)、糖尿病并发症中晚期糖基化终产物及其受体(advanced glycation end products-receptor for advanced glycation end products,AGE-RAGE)、肿瘤坏死因子(tumor necrosis factor,TNF)、白细胞介素-17(interleukin-17,IL-17)、低氧诱导因子-1(hypoxia-inducible factor-1,HIF-1)、辅助性T细胞17(helper T cells 17,Th17)细胞分化信号等通路。结果表明:治疗肝纤维化的中药专利复方以补虚药、活血化瘀药和清热药为主,核心高频药对黄芪-丹参通过多成分、多靶点调控PI3K-Akt、AGE-RAGE、TNF和IL-17等通路来治疗肝纤维化。

Making use of the transformation relation among usual, normal, and antinormal ordering for the multimode boson exponential quadratic polynomial operators （BEQPO＇s）I we present the analytic expression of arbitrary matrix elements for BEQPO＇s. As a preliminary application, we obtain the exact expressions of partition function about the boson quadratic polynomial system, matrix elements in particle-number, coordinate, and momentum representation, and P representation for the BEQPO＇s.

专利中药复方是实用性较强的验方,但其治疗酒精性肝病(alcoholic liver disease,ALD)的用药规律和作用机制尚不明确。采用数据挖掘方法探讨中药专利复方治疗ALD的用药规律,运用生物信息学探索核心药对调节铁死亡治疗ALD的潜在作用机制。通过中医传承计算平台(V3.0)对治疗ALD的专利中药复方进行数据挖掘,提取核心药对。从GSE28619数据集、FerrDb和BATMAN-TCM数据库中获取“核心药对-ALD-铁死亡”共同分子靶点的差异表达基因(differentially expressed genes,DEGs)并分析其相关性和富集分析。构建机器学习模型筛选关键DEGs,构建列线图模型预测ALD的患病风险并验证。共获得229个专利中药复方,包含609味中药。高频药物有丹参、甘草等23味,以补虚药、利水渗湿药、清热药和活血化瘀药为主,丹参-山楂是核心药对。共有10个共同分子靶点DEGs,它们具有相互调控、介导促性腺激素释放激素、糖尿病并发症中晚期糖基化终产物及其受体等信号通路的作用。RF机器学习模型的预测效果最佳,磷酸丝氨酸转氨酶1(phosphoserine aminotransferase 1,PSAT1)、早期生长反应因子1(early growth response factor 1,EGR1)、醌氧化还原酶1(NADPH quinone oxidoreductase 1,NQO1)、醛酮还原酶1C3(aldo-keto reductase 1C3,AKR1C3)、JUN原癌基因(Jun proto-oncogene,JUN)是关键的DEGs。列线图模型能够准确地预测ALD的患病风险。结果表明:中药专利复方治疗ALD以扶正祛邪为基本原则,用药以补虚药为主,兼以利水渗湿、清热、活血化瘀等药物。核心药对丹参-山楂调节铁死亡治疗ALD可能与PSAT1、EGR1、NQO1等DEGs及其介导的促性腺激素释放激素、糖尿病并发症中晚期糖基化终产物及其受体等信号通路有关。

Design and construction of size-controlled CoO/CS catalysts for Fischer–Tropsch synthesis

Discourse relation classification is a fundamental task for discourse analysis,which is essential for understanding the structure and connection of *** discourse relation classification aims to determine the relationship between adjacent sentences and is very challenging because it lacks explicit discourse connectives as linguistic cues and sufficient annotated training *** this paper,we propose a discriminative instance selection method to construct synthetic implicit discourse relation data from easy-to-collect explicit discourse *** expanded instance consists of an argument pair and its sense *** introduce the argument pair type classification task,which aims to distinguish between implicit and explicit argument pairs and select the explicit argument pairs that are most similar to natural implicit argument pairs for data *** also propose a simple label-smoothing technique to assign robust sense labels for the selected argument *** evaluate our method on PDTB 2.0 and PDTB *** results show that our method can consistently improve the performance of the baseline model,and achieve competitive results with the state-of-the-art models.

This study explores the implementation of computed tomography(CT)reconstruction and simulation techniques for patient-specific valves,aiming to dissect the mechanical attributes of calcified valves within transcatheter heart valve replacement(TAVR)*** order to facilitate this exploration,it derives pertinent formulas for 3D multi-material isogeometric hyperelastic analysis based on Hounsfield unit(HU)values,thereby unlocking foundational capabilities for isogeometric analysis in calcified aortic valves.A series of uniaxial and biaxial tensile tests is executed to obtain an accurate constitutive model for calcified active *** mitigate discretization errors,methodologies for reconstructing volumetric parametric models,integrating both geometric and material attributes,are *** these analytical formulas,constitutive models,and precise analytical models to isogeometric analyses of calcified valves,the research ascertains their close alignment with experimental results through the close fit in displacement-stress curves,compellingly validating the accuracy and reliability of the *** study presents a step-by-step approach to analyzing themechanical characteristics of patient-specific valves obtained fromCT images,holding significant clinical implications and assisting in the selection of treatment strategies and surgical intervention approaches in TAVR procedures.

Improving the highly selective and sensitive binding of chemosensor to target guest is always very *** order to solve this issue,herein,the enrichment effect was introduced into the design of chemosensor molecule.A novel bi-fused-macrocyclic host molecule BPN1 was synthesized by bridging a pillar[5]arene and a naphthalene diimide(NDI)group through hydrogen-bond-rich *** the BPN1,the naphthalimide side ring is outside the cavity of the pillar[5]*** addition,Cr(VI)greatly threat human health and the environment due to its severe toxicity,and it is very important to develop effective chemosensor for sensitive and selective detection of Cr_(2)O_(7)^(2-)or its ion *** this paper,the novel bi-fused-macrocyclic host molecule BPN1 can recognize Cr_(2)O_(7)^(2-)with high selectivity and *** mechanism of BPN1 recognition of Cr_(2)O_(7)^(2-)was studied through experiments and density functional theory(DFT),the results show that BPN1 could supply enrichment effect to bind Cr_(2)O_(7)^(2-)through multiple weak interactions such as hydrogen bonds and anion-π,and achieve highly sensitive and selective detection of Cr_(2)O_(7)^(2-).It is a significant and feasible strategy for improve high selectivity and sensitivity of host to specific objects by using the enrichment effect of fused bi-macrocyclic.

Predictability of the upper ocean heat content in a Community Earth System Model ensemble prediction system

Upper ocean heat content(OHC)has been widely recognized as a crucial precursor to high-impact climate variability,especially for that being indispensable to the long-term memory of the *** the predictability of OHC using state-of-the-art climate models is invaluable for improving and advancing climate *** developed retrospective forecast experiments,based on a Community Earth System Model ensemble prediction system,offer a great opportunity to comprehensively explore OHC *** results indicate that the skill of actual OHC predictions varies across different oceans and diminishes as the lead time of prediction *** spatial distribution of the actual prediction skill closely resembles the corresponding persistence skill,indicating that the persistence of OHC serves as the primary predictive signal for its *** decline in actual prediction skill is more pronounced in the Indian and Atlantic oceans than in the Pacific Ocean,particularly within tropical ***,notable seasonal variations in the actual prediction skills across different oceans align well with the phase-locking features of OHC *** potential predictability of OHC generally surpasses the actual prediction skill at all lead times,highlighting significant room for improvement in current OHC predictions,especially for the North Indian Ocean and the Atlantic *** such improvements necessitates a collaborative effort to enhance the quality of ocean observations,develop effective data assimilation methods,and reduce model bias.
