Given the extremely high inter-patient heterogeneity of acute myeloid leukemia(AML),the identification of biomarkers for prognostic assessment and therapeutic guidance is *** surface markers(CSMs)have been shown to play an important role in AML leukemogenesis and *** the current study,we evaluated the prognostic potential of all human CSMs in 130 AML patients from The Cancer Genome Atlas(TCGA)based on differential gene expression analysis and univariable Cox proportional hazards regression *** using multi-model analysis,including Adaptive LASSO regression,LASSO regression,and Elastic Net,we constructed a 9-CSMs prognostic model for risk stratification of the AML *** predictive value of the 9-CSMs risk score was further validated at the transcriptome and proteome *** Cox regression analysis showed that the risk score was an independent prognostic factor for the AML *** AML patients with high 9-CSMs risk scores had a shorter overall and event-free survival time than those with low ***,single-cell RNA-sequencing analysis indicated that patients with high 9-CSMs risk scores exhibited chemotherapy ***,PI3K inhibitors were identified as potential treatments for these high-risk *** conclusion,we constructed a 9-CSMs prognostic model that served as an independent prognostic factor for the survival of AML patients and held the potential for guiding drug therapy.

Objective:This cross-sectional study aimed to determine the epidemiology of olfactory and gustatory dysfunctions related to COVID-19 in ***:This study was conducted by 45 tertiary Grade-A hospitals in *** and offline questionnaire data were obtained from patients infected with COVID-19 between December 28,2022,and February 21,*** collected information included basic demographics,medical history,smoking and drinking history,vaccination history,changes in olfactory and gustatory functions before and after infection,and other postinfection symptoms,as well as the duration and improvement status of olfactory and gustatory ***:Complete questionnaires were obtained from 35,566 *** overall incidence of olfactory and taste dysfunction was 67.75%.Being female or being a cigarette smoker increased the likelihood of developing olfactory and taste *** received four doses of the vaccine or having good oral health or being a alcohol drinker decreased the risk of such *** infection,the average olfactory and taste VAS scores were 8.41 and 8.51,respectively;after infection,they decreased to 3.69 and 4.29 and recovered to 5.83 and 6.55 by the time of the *** median duration of dysosmia and dysgeusia was 15 and 12 days,respectively,with 0.5%of patients having symptoms lasting for more than 28 *** overall self-reported improvement rate was 59.16%.Recovery was higher in males,never smokers,those who received two or three vaccine doses,and those that had never experienced dental health issues,or chronic accompanying ***:The incidence of dysosmia and dysgeusia following infection with the SARS-CoV-2 virus is high in *** and prognosis are influenced by several factors,including sex,SARS-CoV-2 vaccination,history of head-facial trauma,nasal and oral health status,smoking and drinking history,and the persistence of accompanying symptoms.

To the Editor:Growth hormone deficiency(GHD)impairs growth and development,affecting approximately 1 in 4000 children worldwide.[1]Recombinant human growth hormone(rhGH)has been standard practice since 1985.[2]However,compliance has been challenged by the need for frequent *** glycol rhGH(PEG-rhGH)was developed,improving protein stability and extending *** studies suggested that a weekly dosage of 0.20 mg/kg of PEG-rhGH offers improved growth rates and increased insulin-like growth factor-1 standard deviation score(IGF-1 SDS)without novel toxicities.[3]Here,we present a pooled analysis of two phase IV trials evaluating the efficacy and safety of various PEG-rhGH dosages in GHD.

We assessed the global incidence, mortality, and disability-adjusted life years(DALYs) associated with various liver diseases, including alcohol-related liver disease(ALD), hepatitis B/C virus infections(HBV or HCV), liver cancer, metabolic dysfunction-associated steatotic liver disease(MASLD), and other chronic liver diseases, from the 2019 Global Burden of Disease study. Additionally, we analyzed the global trends in hepatology research and drug development. From 2000 to 2019, prevalence rates increased for ALD, MASLD and other liver diseases, while they decreased for HBV, HCV, and liver cancer. Countries with a high socio-demographic index(SDI) exhibited the lowest mortality rates and DALYs. The burden of liver diseases varied due to factors like sex and region. In nine representative countries, MASLD,along with hepatobiliary cancer, showed highest increase in funding in hepatology research. Globally, the major research categories in hepatology papers from 2000 to 2019 were cancer, pathobiology, and MASLD. The United States(U.S.) was at the forefront of hepatology research, with China gradually increasing its influence over time. Hepatologists worldwide are increasingly focusing on studying the communication between the liver and other organs, while underestimating the research on ALD. Cancer, HCV, and MASLD were the primary diseases targeted for therapeutic development in clinical trials. However, the proportion of new drugs approved for the treatment of liver diseases was relatively low among all newly approved drugs in the U.S., China, Japan, and the European Union. Notably, there were no approved drug for the treatment of ALD in the world.

目的建立过度复制诱导原代小鼠软骨细胞衰老模型及小鼠自然衰老模型,研究KAT7在软骨细胞及组织衰老中的作用。方法二型胶原酶消化法获取小鼠膝关节软骨细胞,使用甲苯胺蓝染色及ColⅡ染色鉴定小鼠软骨细胞;Western blot和细胞免疫荧光法检测不同代次小鼠软骨细胞衰老信号相关蛋白及KAT7表达水平,SA-β-Gal染色确定细胞衰老情况。取22月龄老龄小鼠与2月龄年轻小鼠关节HE染色、番红O-固绿染色比较小鼠关节病理情况;免疫组化观察小鼠关节组织中KAT7和p53的表达情况。结果与对照组相比,衰老的小鼠软骨细胞KAT7蛋白及p21、p53表达水平显著升高,KAT7抑制剂WM-3835可抑制衰老软骨细胞KAT7和p21的蛋白表达。SA-β-Gal染色可见,第八代(P8)较第一代(P1)软骨细胞阳染明显增加。与年轻小鼠软骨组织相比,老龄小鼠软骨组织呈现近骨性分布,软骨表面完整性降低,肥大软骨细胞数量显著增多,KAT7和p53细胞阳染增加。结论 KAT7在软骨细胞衰老模型及老龄小鼠软骨组织中表达升高,提示KAT7可能与软骨细胞衰老相关。

阴阳辨证在八纲辨证中具有统率其他六纲的作用,是对病情进行更高层面或总的归纳,在中医临证过程中阴阳辨证贯穿疾病诊治过程的始末。阴阳辨证对于帕金森病而言尤为重要,不同症状表现、病情发展过程中病机的转化和中西医治疗都体现了阴阳对立制约、互根互用、消长转化的特点。该文从阴阳辨证的源流及应用、阴阳辨证是帕金森病辨证基础、阴阳辨证在帕金森病临证中的地位和作用3个方面论述了阴阳辨证产生的背景、应用的基础和价值,以“阴阳为纲”的辨证方法来指导帕金森病的诊治具有重要意义。

＜正＞ To explore the effects of Astragalosides （AST） and Astragals Saponin I （AS - I） on intracellular calcium concentration （[Ca2 + ]i） in senescent rats treated by hydrocortisone. The [Ca2 + ]i was measured by using double wavelength fluorescence spectrophotometer in thymocytes and hippocampus neurons intrasynaptosomes from mice. The results showed that [Ca2 + ]i in senescent thymus and hippocampus sitrasynaptosomes was significantly higher than [ Ca2 + ] i of adult thymus and hippocamal intrasynaptosomes. The high level [Ca2+ ]i of hippocampus caused memory impairment in senescent mice. When treated with AST or AS - I. the [ Ca2+ ] i in two kind cells decreased. Dexamamethasone （ DEX）. BayK8644,KCl and Glutamete （Glu） all elevated [Ca2+ ]i of thymocytes of neonate （7 days rats） and hippocampal neurons of fatal rats in vitro. When treated with AST or AS - I. the [ Ca2+ ]i in cultured thymocytes and hippocampus neurons stimulated by DEX,BayK8644,KC1 and Glu decreased.

Background:Panax notoginseng(PNE)is a prominent traditional Chinese medicine with extensive beneficial effects on the immune ***,the precise mechanism of PNE in treating inflammatory bowel disease(IBD)remains ***:We first used an extensive metabolomics approach utilizing UPLC-ESI-Q TRAP-MS/MS to identify the metabolite components of PNE aqueous ***,the mechanism of PNE in treating IBD was investigated through in silico analysis including RNA-seq analysis,Network pharmacology and Molecular *** a Drosophila toxin-induced intestinal inflammation model was employed to investigate ***:A total of 1,543 metabolites of PNE aqueous extract were characterized using UPLC-ESI-Q TRAP-MS/*** silico analyses showed that 97 IBD hub targets were targeted by 21 PNE *** Encyclopedia of Genes and Genomes results indicated that PNE may play an anti-IBD role through the Mitogen-activated protein kinase(MAPK)signaling pathway and other immune-related signaling ***,11 top hits compounds of PNE show a good affinity binding to IBD *** experimental results demonstrated that PNE can effectively improve the survival rate of adult Drosophila while also inhibit the excessive proliferation and differentiation of intestinal stem cells induced by sodium dodecyl ***,PNE notably lower the epithelial cell mortality,the accumulation of reactive oxygen species and the activation of oxidative stress-associated jun-Nterminal kinase(JNK)***:Our data suggests that PNE aqueous extract has a significant protective impact on the intestinal homeostasis of *** findings establish a basis for utilizing PNE in clinical investigations and managing IBD.
