NF-κB and ERK-signaling pathways contribute to the gene expression induced by cag PAI-positive-Helicobacter pylori infection

AIM： To elucidate the sequential gene expression profile in AGS cells co-cultured with wild-type Helicobacter pylori （H pylon） as a model of Hpylori-infected gastric epithelium, and to further examine the contribution of cag-pathogenicity islands （cagPAI）-coding type IV secretion system and the two pathways, nuclear factor kappa B （NF-κB） and extracellular signal-regulated kinases （ERK） on wild-type Hpylori-induced gene expression. METHODS： Gene expression profiles induced by Hpylori were evaluated in AGS gastric epithelial cells using cDNA microarray, which were present in the 4600 independent clones picked up from the human gastric tissue. We also analyzed the contribution of NF-κB and ERK signaling on H pylori-induced gene expression by using inhibitors of specific signal pathways. The isogenic mutant with disrupted cagE （△cagE） was used to elucidate the role of cagPAI-encoding type IV secretion system in the gene expression profile. RESULTS： According to the expression profile, the genes were classified into four clusters. Among them, the clusters characterized by continuous upregulation were most conspicuous, and it contained many signal transducer activity-associated genes. The role of cagPAI on cultured cells was also investigated using isogenic mutant cagE, which carries non-functional cagPAI. Then the upregulation of more than 80% of the induced genes （476/566） was found to depend on cagPAI. Signal transducer pathway through NF-κB or ERK are the major pathways which are known to be activated by cagPAI-positive H pylori. The role of these pathways in the whole signal activation by cagPAI-positive H pyloriwas analyzed. The specific inhibitors against NF-κB or ERK pathway blocked the activation of gene expression in 65% （367/566） or 76% （429/566） of the genes whose activation appealed to depend on cagPAI. This paper describes an evaluation and advice system for improving the manner of speaking in lectures. Our developed system evaluates the manner of speaking using decision trees. These decision trees are trained using features of filled pauses that are included in lecture speeches. Recently, it has been learned that filled pauses in speech influence the understanding of listeners and change the overall impression of the speech. Our decision-tree-based evaluation and advice system detects filled pauses using automatic speech recognition. The system then uses the features of the filled pauses in decision trees. Finally, the system evaluates the manner of speaking at three evaluation levels and provides advice to the speaker. In the evaluation experiment, our system worked well and advice given improved the manner of speaking in four out of five instances.

Objective: This study aim to assess the efficacy and safety of sunitinib in Japanese patients with metastatic renal cell carcinoma (mRCC) in general clinical practice. Patients and Methods: Non-selected fifty eight Japanese patients with mRCC were treated with sunitinib. Overall survival (OS) and time to treatment failure (TTF) were estimated. Response rate and safety profiles were also assessed. Results: Partial response, stable disease, and progressive disease were observed in 13 (22.4%), 26 (44.8%), and 19 (32.8%) patients, respectively. The median TTF was 5.4 months, and the median OS was 11.2 months. In the prior nephrectomy group, the median TTF was 9.0 months, and the median OS was 16.4 months. In the non-nephrectomy group, the median TTF was 1.1 months, and the median OS was 2.8 months. The most frequently occurring Grade 3/4 adverse events (AEs) were anorexia (17.2%), fatigue (12.1%), thrombocytopenia (13.8%), and anemia (12.1%). Conclusions: Sunitinib has a favorable risk/benefit profile in Japanese mRCC patients with a history of nephrectomy.

Diabetes mellitus is a powerful risk factor of coronary artery disease(CAD), leading to death and disability. In recent years, given the accumulating evidence that prediabetes is also related to increasing risk of CAD including cardiovascular events, a new guideline has been proposed for the treatment of blood cholesterolfor primary prevention of cardiovascular events. This guideline recommends aggressive lipid-lowering statin therapy for primary prevention in diabetes and other patients. The ultimate goal of patient management is to inhibit progression of systemic atherosclerosis and prevent fatal cardiovascular events such as acute coronary syndrome(ACS). Because disruption of atherosclerotic coronary plaques is a trigger of ACS, the high-risk atheroma is called a vulnerable plaque. Several types of novel diagnostic imaging technologies have been developed for identifying the characteristics of coronary atherosclerosis before the onset of ACS, especially vulnerable plaques. According to coronary angioscopic evaluation, atherosclerosis severity and plaque vulnerability were more advanced in prediabetic than in nondiabetic patients and comparable to that in diabetic patients. In addition, pharmacological intervention by statin therapy changed plaque color and complexity, and the dynamic changes in plaque features are considered plaque stabilization. In this article, we review the findings of atherosclerosis in prediabetes, detected by intravascular imaging modalities, and the therapeutic implications.

Provenance Variability during Damuda Sedimentation in the Talchir Gondwana Basin, India – A Statistical Assessment

The Talchir Gondwana basin houses the Talchir Formation at the base, succeeded by the Damuda Group and the Kamthi Formation in upward progression. The present study is an attempt to determine the provenance of the Damuda Group and its variability in terms of location, climate and tectonism through time from the composition of sandstone grains and detritus of the constituent Karharbari, Barakar and Barren Measures formations. The Damuda sandstones are composed of variable amounts of monocrystalline undulatory, nonundulatory and polycrystalline quartz grains, potash and plagioclase feldspars as well as metamorphic and sedimentary rock fragments in addition to heavy accessories. Palaeocurrent studies suggest that the Eastern Ghats Supergroup lying to the south of the basin served as the source area of the Damuda sediments. Plots of sandstone composition in tectonic setting discrimination diagrams suggest derivation of the detritus from craton interior, continental block and recycled orogen provinces. Statistical analyses indicate significant differences in the detrital modes of the sandstones of the Karharbari, Barakar and Barren Measures formations, which may be attributed to temporal and spatial variation of the provenance coupled with climate change in commensurate with Damuda sedimentation.

The succession of lithofacies of a part of the Barakar Formation of the Singrauli coalfield has been studied by statistical techniques. The lithologies have been grouped under five facies states viz. coarse-, medium-, and fine-grained sandstones, shale and coal for statistical analyses. Markov chain analysis indicates the arrangement of Barakar lithofacies in the form of fining-upward cycles. A complete cycle consists of conglomerate or coarse-grained sandstone at the base sequentially succeeded by medium- and fine-grained sandstones, shales and coal seam at the top. The entropy analysis puts the Barakar cycles in A-4 type of cyclicity, which consists of different proportions of lower-, side- and upper-truncated cycles of lithologic states. Regression analysis indicates a sympathetic relationship between total thickness of strata (net subsidence) and number of coal cycles and an antipathic relationship between average thickness and number of coal cycles. The cyclic sedimentation of the Barakar Formation was controlled by autocyclic process, which occurred due to the lateral migration of streams triggered by intrabasinal differential subsidence. In many instances, the clastic sediments issued from the laterally migrating rivers interrupted the sedimentation resulting in thinner cycles in areas where the numbers of cycles are more. Principal component and multivariate regression analyses suggest that the net subsidence of the basin is mostly controlled by number and thickness of sandstone beds and coal seams.

Rehabilitation devices help to recover the physical abilities of patients. This study aims to develop a portable rehabilitation device that is safe to use when?patients are holding it by hands. In a previous study, to realize a home rehabilitation device, a flexible spherical actuator that can provide motion to patients was developed. In this study, to measure the relative position between both handling stages, a 3D coordinate measuring device using three wire-type linear potentiometers and an embedded controller was proposed and tested. In this paper, the spherical actuator with built-in 3D coordinate measuring device is described. The measuring method and experimental results obtained are also presented. The tracking position control of the actuator using the measuring device was carried out. As a result, the position of the handling stages can be successfully controlled using the feedback signal from the tested?measuring device.

Despite recent advances in surgical techniques and perioperative management, the prognosis of pancreatic cancer(PCa) remains extremely poor. To provide optimal treatment for each patient with Pca, superior biomarkers are urgently needed in all phases of management from early detection to staging, treatment monitoring, and prognosis. In the blood of patients with cancer, circulating tumor cells(CTCs) and cell-free nucleic acids(cf NAs), such as DNA, m RNA, and noncoding RNA have been recognized. In the recent years, their presence in the blood has encouraged researchers to investigate their potential use as novel blood biomarkers, and numerous studies have demonstrated their potential clinical utility as a biomarker for certain types of cancer. This concept, called "liquid biopsy" has been focused on as a less invasive, alternative approach to cancer tissue biopsy for obtaining genetic and epigenetic aberrations that contribute to oncogenesis and cancer progression. In this article, we review the available literature on CTCs and cfN As in patients with cancer, particularly focusing on PCa, and discuss future perspectives in this field.
