Bac.ground： The tools of synthetic.biology have enabled researc.ers to explore multiple sc.entific.phenomena by direc.ly engineering signaling pathways within living c.lls and artific.al protoeells. Here, we explored the potential for engineered living c.lls themselves to assemble signafing pathways for non-living protoc.lls. This analysis serves as a preliminary investigation into a potential origin of proc.sses that may be utilized by c.mplex living systems. Spec.fic.lly, we suggest that if living c.lls c.n be engineered to direc. the assembly of genetic.signaling pathways from genetic.biomaterials in their environment, then insight c.n be gained into how naturally oc.urring living systems might behave similarly. Methods： To this end, we have modeled and simulated a system c.nsisting of engineered c.lls that c.ntrol the assembly of DNA monomers on mic.opartic.e sc.ffolds. These DNA monomers enc.de genetic.c.rc.its, and therefore, these mic.opartic.es c.n then be enc.psulated with minimal transc.iption and translation systems to direc. protoc.ll phenotype. The modeled system relies on multiple previously established synthetic.systems and then links these together to demonstrate system feasibility. Results： In this spec.fic.model, engineered c.lls are induc.d to synthesize biotin, whic. c.mpetes with biotinylated, c.rc.it-enc.ding DNA monomers for an avidinized-mic.opartic.e sc.ffold. We demonstrate that multiple synthetic.motifs c.n be c.ntrolled in this way and c.n be tuned by manipulating parameters suc. as induc.r and DNA c.nc.ntrations. c.nc.usions： We expec. that this system will provide insight into the origin of living systems as well as serve as a tool for engineering living c.lls that assemble c.mplex biomaterials in their environment.

New c.inic.l approac.es are imperative beyond the widely adopted National c.mprehensive c.nc.r Network (Nc.N) guidelines, utilized by prominent c.nc.r institutions. c.nc.r is the leading c.use of death among individuals younger than 85 years within the United States. Despite signific.nt tec.nologic.l advanc.s, inc.uding the expenditure of hundreds of billions, treatment outc.mes and overall survival have not notably improved for most types of advanc.d c.nc.r over the last several dec.des. Over the past 24 years, Envita Medic.l c.nters has pioneered a unique form of personalized treatment approac. for late-stage and refrac.ory c.nc.r patients, introduc.ng groundbreaking innovations in the field. Our integrated algorithm utilizes advanc.d genomic., transc.iptomic., and highly tailored immunotherapy, resulting in remarkable outc.me improvements. This study presents Envita’s innovative personalized treatment algorithms and examines the response outc.mes of 199 late-stage c.nc.r patients treated at Envita Medic.l c.nters over a two-year period. c.mpared to standard of c.re and palliative c.emotherapy, Envita’s treatment demonstrated a remarkable 35-fold improvement in overall response rates (Figure 1). Moreover, 88% of the patients, the majority presenting with Stage 3 or 4 c.nc.r, experienc.d a 43-fold improvement in quality of life with minimal side effec.s, as c.mpared to standard of c.re c.emotherapy and palliative c.re. This revolutionary suc.ess is attributed to Envita’s personalized therapeutic.algorithms, whic. inc.rporate c.stomized immunotherapy. Envita’s prec.sion c.re approac. has also ac.ieved a 100% better response rate c.mpared to over 65 global c.emotherapy c.inic.l trials with more than 2700 patients. The results from this study suggest that a wider utilization of Envita’s personalized approac. c.n signific.ntly benefit patients with late-stage and refrac.ory c.nc.r.

The normalization of quantitative real time RT-Pc. (qRT-Pc.) is important to obtain ac.urate gene expression data. The most c.mmon method for qRT-Pc. normalization is to use referenc., or house- keeping genes. However, there is emerging evidenc. that even referenc. genes c.n be regulated under different c.nditions, qRT-Pc. has only rec.ntly been used in terms of zebrafish gene expression studies and there is no validated set of referenc. genes. This study c.arac.erizes the expression of nine possible referenc. genes during zebrafish embryonic.development and in a zebrafish tissue panel. All nine referenc. genes exhibited variable expression. The β-ac.in, EF1α and Rp113α genes c.mprise a validated referenc. gene panel for zebrafish developmental time c.urse studies, and the EF1α, Rp113α and 18S rRNA genes are more suitable as a referenc. gene panel for zebrafish tissue analysis. Importantly, the zebrafish GAPDH gene appears unsuitable as referenc. gene for both types of studies.

Today, the oil and gas industry, and in partic.lar hydraulic.frac.uring operations, have c.me under inc.easing pressure from regulators and the public.to reduc. emissions. As the industry evolves, oil and gas produc.rs are in the position of evaluating alternative tec.nologies whic. will support their objec.ives of reduc.ng their overall emissions profile and c.rbon footprint. As a response, the deployment of tec.nology and solutions to reduc. emissions related to hydraulic.frac.uring applic.tions has rec.ntly ac.elerated, c.eating various options to address these industry c.allenges. BJ Energy Solutions and West Virginia University have been working on the applic.tion and emissions c.arac.erization of various hydraulic.frac.uring tec.nologies. A study was c.nduc.ed to evaluate the effic.enc. and resultant emissions from various tec.nologies, inc.uding natural gas rec.proc.ting engines, diesel-natural gas dual-fuel engines, large (>24 MW) gas turbines, and direc. drive turbines. The study involved the development of an emissions model with the purpose of estimating total emissions of c.rbon dioxide (c.2), nitrous oxide (N2O) and exhaust methane (c.4) slip, all Greenhouse Gases (GHGs), and c.nverted to tons of c.2 equivalent emissions per day of operation. The model inputs are the required Hydraulic.Horsepower (HHP) based on pumping rate and pressure for various shale play sc.narios. The model c.lc.lates emissions from the TITAN, whic. is a direc.-drive turbine model fielded by BJ, using data c.llec.ed following U.S. Environmental Protec.ion Agenc. (EPA) testing protoc.ls. The model also c.lc.lates and c.mpares other hydraulic.frac.uring tec.nologies utilizing published Original Equipment Manufac.urer (OEM) data. Mainstream silic.n photonic.integrated c.rc.its are based on c.mpac. and low-loss silic.n-on-insulator(SOI)waveguide platforms. However, monolithic.SOI-based photonic. provides only a limited number of func.ional devic. types. Here, to extend the on-c.ip c.pabilities, we propose a general heterogeneous integration approac. to embed highly nonlinear Ⅲ-Ⅴ(Al Ga As) photonic. into the SOI platform. We develop low-loss AlGaAs-on-SOI photonic.c.rc.its with integrated Si waveguides and showc.se sub-milliwatt-threshold(-25 mW) Kerr frequenc. c.mb generation in ultrahigh-QAlGaAs mic.orings(Q over 10^(6)) at the telec.m bands. Our demonstration c.mplements existing mature Si photonic. tec.nology with effic.ent nonlinear func.ionalities provided by Ⅲ-Ⅴ and propels c.nventional Si photonic. into emerging nonlinear photonic.applic.tions towards fully c.ip-based nonlinear engines.
