Polymorphisms of the oxidant enzymes glutathione S-transferase and glutathione reductase and their association with resistance of Plasmodium falciparum isolates to antimalarial drugs

Objective:To investigate the association between amplification of the two regulatory genes controlling glutathione(GSH) levels,glutathione reductase(PfGR) and glutathione S-transferase (PfGST) genes and sensitivity of Plasmodium falciparum(***) isolates collected from different malaria endemic areas of Thailand to standard antimalarial ***:A total of 70 *** isolates were collected from endemic areas of multi-drug resistance (Tak,Chantaburi and Ranong Provinces) during the year *** in vitro assessment of antimalarial activity of *** clones(K1- and Dd2 chloroquine resistant and 3D7- chloroquine sensitive) and isolates to chloroquine,quinine,mefloquine and arteusnate was performed based on SYBR Green modified ***:68(97.14%),11(15.71%) and 28(40%) isolates respectively were classified as chloroquine-,quinine- and mefloquine-resistant isolates. With this limited number of *** isolates included in the analysis,no significant association between amplification of PfGST gene and sensitivity of the parasite to chloroquine, quinine,mefloquine and quinine was *** on PCR analysis,Dd2,Kl and 3D7 clones all contained only one copy of the PfGST *** isolates(70) also carried only one copy number of PfGST *** appears to be an association between amplification of PfGR gene and chloroquine *** 3D7 and Dd2 clones were found to carry only one PfGR gene copy, whereas the K1 clone carried two gene ***:Chloroquine resistance is likely to be a consequence of multi-factors and enzymes in the GSH system may be partly involved. Larger number of parasite isolates are required to increase power of the hypothesis testing in order to confirm the involvement of both genes as well as other genes implicated in glutathione metabolism in conferring chloroquine resistance.

Synergistic Effects of the ZnCl_2-SiCl_4 Modified TiCl_4/MgCl_2/THF Catalytic System on Ethylene/1-Hexene and Ethylene/1-Octene Copolymerizations

The copolymerizations of ethylene with 1-hexene or 1-0ctene by using TiC14/MgCI2/THF catalysts modified with different metal halide additives （ZnCI2, SIC14, and the combined ZnC12-SiCl4） were investigated based on catalytic activity and copolymer properties. It was found that the catalyst modified with mixed ZnC12-SiC14 revealed the highest activities for both ethylene/1-hexene and ethylene/1-0ctene copolymerization. The increase in activities was due to the formation of acidic sites by modifying the catalysts with Lewis acids. Based on the FTIR measurements, the characteristic C--O--C peaks of the catalysts modified with metal halide additives were slightly shifted to lower wavenumber when compared to the unmodified catalyst. This showed that the modified catalysts could generate more acid sites in the TiC14/MgC12/THF catalytic system leading to an increase in activities as well as comonomer insertion （as proven by IaC-NMR）. However, Lewis acid- modifications did not affect the microstructure of the copolymers obtained. By comparison on the properties of copolymers prepared with the unmodified catalyst, it was found that polymers with ZnC12 and/or SIC14 modification exhibited a slight decrease in melting temperature, crystallinity and density. It is suggested that these results were obtained based on the different amount of ct-olefins insertion, regardless of the types of Lewis acids and comonomer.

Ultrasound-assisted Low Density Solvent Based Dispersive Liquid-liquid Microextraction for Determination of Phthalate Esters in Bottled Water Samples

A technique of ultrasound-assisted low density solvent based dispersive liquid-liquid microextraction was developed for the determination of four phthalate esters, including dimethyl phthalate（DMP）, diethyl phthalate（DEP）, di-n-butyl phthalate（DnBP） and di（2-ethylhexyl） phthalate（DEHP） in bottled water samples. A low density solvent, toluene, was selected as extraction solvent. In the extraction process, a mixture of 15 μL of toluene（extraction solvent） and 100 μL of methanol（disperser solvent） was rapidly injected into 1.0 mL of water samples. A cloudy solution was formed after ultrasounded for 5 min, and then centrifuged at 5000 r/min for 5 min. The enriched analytes in the floa- ting phase were determined by means of gas chromatograph. Under the optimum conditions, the enrichment factors were found to be in a range of 29--67, and the recoveries were ranged from 81.2% to 103.9%. The limits of the detection were in a range of 3.8--5.6 μg/L. The proposed method was applied to the extraction and determination of phthalate esters in bottled water samples, and the concentrations of phthalate esters found in the water samples were below the allowable levels.

Antimalarial activity and toxicity of Garcinia mangostana Linn.

Seawater(SW)-acclimated Nile tilapia,Oreochromis niloticus,reared under a salinity 30 environment had lower growth and survival than the freshwater(FW)-acclimated *** was hypothesized that cells of the SW-acclimated fish had not been able to synthesize an adequate level of a compatible osmolyte,myo-inositol(MI),in adjusting to the salinity 30 *** this study,MI supplements,at 250,500,and 750 mg/kg pellets,were provided to the fish through *** the 30-day feeding trial,the following parameters were determined:final body weights;survival;biomass increase;feed conversion ratio(FCR);plasma osmolality and ions;and two transcripts in the gills mips250 and mipa1 encoding enzymes responsible for MI *** SW-acclimated *** receiving 500-mg MI supplement had significantly higher survival,biomass increase,and lower FCR than those of the SW-acclimated fish receiving no *** 500-mg MI supplemental level,the increasing values of plasma osmolality and Na+observed in SW-acclimated fish were significantly *** transcript mipa1,but not mips250,was markedly up-regulated in the SW-acclimated ***,compared with that of the FW-acclimated ***,MI at 500-mg supplement attenuated the up-regulation *** study suggests that MI supplement at the optimum level enhanced the performance of SW-acclimated ***,and through yet unknown mechanisms,attenuated some of their physiological responses to the osmotic stress.

Assessment of in vitro sensitivity of Plasmodium vivax fresh isolates

Objective:To compare the applicability of the SYBK Grcen-Ⅰ assay with the standard schizont maturalion assay,for determination of sensitivity of Plasmodium vivax(***) to chloroquine and a new antifolale WR ***:The study was conducted at Mae Tao Clinic for migrant workers,Tak Province during April 2009 to July 2010.A total of 64 blood samples(1 mL blood collected into sodium heparinized plastic tube) were collected from patients with monoinfection with *** malaria prior to treatment with standard regimen of a 3-day chloroquine. In vitro sensitivity of *** isolates was evaluated by schizont maturation inhibition and SYBR Green-Ⅰ ***:A total of 30 out of 64 blood samples collected from patients with *** malaria were successfully analyzed using both the microscopic schizont maturation inhibition and SYBR Green-I *** failure rates of the schizont maturation inhibition assay(50%) and the SYBR Green-I assay(54%) were similar(P=0.51).The median IC_(10)s,IC_(50)s and IC_(90)s of both chloroquine and WR99210 were not significantly different from the clinical isolates of *** *** on the cut-off of 100 nM,the prevalences of chloroquine resistance determined by schizont maturation inhibition and SYBR Green-I assays were 19 and 11 isolates,*** strength of agreement between the two methods was very poor for both chloroquine and ***:On the basis of this condition and its superior sensitivity,the microscopic method appears better than the SYUK Green-I Green assay for assessing in vitro sensitivity of fresh *** isolates to antimalarial drugs.

Determination of Trihalomethanes in Water Samples Using Headspace Solid-Phase Microextraction Gas Chromatography

The chlorination process is one of the water treatment method used for the disinfection of water. The disinfection by products are trihalomethanes such as chloroform, dichloromethane, dibromochloromethane and bromoform. A headspace solid-phase microextraction method has been developed for determination oftrihalomethanes in water samples. The experimental parameters such as the stirring rate, extraction time, extraction temperature and desorption time were investigated. The linearity, detection limits and percentage recovery were evaluated. The optimum conditions were stirring rate 800 rpm/min, extraction time 6 min, extraction temperature 20 ~C, desorption time 2.5 min and desorption temperature 220 ~C. The detection limits were 0.01 ~g/L and the recoveries were in the range of 86-110 %, The proposed method was successfully applied to determination of THM4 in tap water samples. The THM4 contents were varied depending on the sample sites and the season. The total THM4 contents in cool, summer and rainy season were in the range of 27.58-41.89, 32.06-60.73 and 46.26-69.87 p.g/L, respectively. Confirmation of the detected compounds in water samples were performed by gas chromatograph-mass spectrometer. The mass spectra of the target compounds in water samples is in good agreement with trihalomethanes standard spectra.

Proteomics analysis of antimalarial targets of Garcinia mangostana Linn.

Objective:To investigate possible protein targets for antimalarial activity of Garcina mangostana Linn.(***)(pericarp)in 3D7 Plasmodium falciparum clone using 2-dimensional electrophoresis and liquid chromatography mass-spectrometry(LC/MS/MS).Methods:3D7 Plasmodium falciparum was exposed to the crude ethanolic extract of *** Linn.(pericarp)at the concentrations of 12μg/mL(1C_(50)level:concentration that inhibits parasite growth by 50%)and 30μg/mL(1C_(90)level:concentration that inhibits parasite growth by 90%)for 12 *** proteins were separated by 2-dimensional electrophoresis and identified by LC/MS/***:At the IC_(50)concentration,about 82%of the expressed parasite proteins were matched with the control(non-exposed),while at the IC_(90)concentration,only 15%matched proteins were *** selected protein spots from parasite exposed to the plant extract at the concentration of 12μg/mL were identified as eneymes that play role in glycolysis pathway,i.e.,phosphoglyeerate mutase putative,L-lactate dehydrogenase/glyceraldehyde-3-phosphate dehydrogenase,and fruetose-bisphosphate aldolase/phosphoglyeerate *** proteosome was found in parasite exposed to 30μg/mL of the ***:Results suggest that proteins involved in the glycolysis pathway may be the targets for antimalarial activity of *** Linn.(pericarp).

Preliminary investigation on the prevalence of malaria and HIV co-infection in Mae Sot District, Tak Province of Thailand

Objective: To preliminarily investigate the prevalence of HIV co-infection in patients with malaria in Mae Sot District, Tak Province of ***: The study was a retrospective study on blood samples collected from a total of 256 patients with malaria(all species and severity) who attended Mae Tao clinic for migrant workers, Tak Province during 2005-2007(148 samples) and 2010-2012(108 samples). Malaria diagnosis was performed based on microscopic examination of patients' blood smears. Chemiluminescent microparticle immunoassay and gel particle passive agglutination were employed for the detection of HIV antigen in patients' plasma. Results: Plasmodium falciparum(P. falciparum) and Plasmodium vivax(P. vivax) are the two predominant malaria species with the ratio of about 1: 1 to 1.5:1. Most of the P. falciparum cases were presented with acute uncomplicated signs and symptoms with highest parasitemia of 1 045 000 asexual parasites/μL bloods. The prevalence of malaria and HIV co-infection during 2005-2007 was 1.35%(2/148 cases, 1 each for P. falciparum and P. vivax co-infection), but was increased to 2.78%(3/108 cases, 2 and 1 for P. falciparum and P. vivax co-infection, respectively) during ***: The increasing trend of prevalence of malaria and HIV co-infection in Mae Sot, Tak province was of a great concern on either pharmacodynamics or pharmacokinetics aspect. The study in a larger numbers of malaria patients in different endemic areas throughout the country with different time periods is underway.
