Objective To uncover the mechanisms underlying the development of colorectal cancer(CRC),we applied bioinformatic analyses to identify key genes and experimentally validated their possible roles in CRC onset and *** We performed Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis on differentially expressed genes(DEGs),constructed a protein-protein interaction(PPI)network to find the top 10 hub genes,and analyzed their expression in colon adenocarcinoma(COAD)and rectum adenocarcinoma(READ).We also studied the correlation between these genes and immune cell infiltration and prognosis and validated the expression of SLC9A2 in CRC tissues and cell lines using qRT-PCR and Western *** experiments were conducted in vitro to investigate the effects of SLC9A2 on tumor growth and *** We found 130 DEGs,with 45 up-regulated and 85 down-regulated in *** analysis indicated that these DEGs were primarily enriched in functions related to the regulation of cellular pH,zymogen granules,and transmembrane transporter *** pathway analysis revealed that the DEGs played pivotal roles in pancreatic secretion,rheumatoid arthritis,and the IL-17 signaling *** identified 10 hub genes:CXCL1,SLC26A3,CXCL2,MMP7,MMP1,SLC9A2,SLC4A4,CLCA1,CLCA4,and *** enrichment analysis showed that these hub genes were predominantly involved in the positive regulation of *** expression analysis revealed that CXCL1,CXCL2,MMP1,and MMP7 were highly expressed in CRC,whereas CLCA1,CLCA4,SLC4A4,SLC9A2,SLC26A3,and ZG16 were expressed at lower *** analysis revealed that 5 key genes were significantly associated with the prognosis of *** mRNA and protein expression levels of SLC9A2 were markedly reduced in CRC tissues and cell ***,SLC9A2 overexpression in SW480 cells led to a notable inhibition of cell proliferation,migration,and *** blotting analysis revealed tha

An underwater vest containing an antioxidant MXene hydrogel for sensitive recognition of fish locomotion

The perception of fish locomotion is important for understanding their adaptive behaviors and ethological ***,the main strategy used for extracting fish attitudes involves the use of a vision-based monitoring system,which is limited in its range of observation and cannot perform tracking for long ***,we report the use of a wearable tagging electronic device,referred to as an underwater vest,to capture the surrounding flow field disturbances triggered by swimming or momentary postural *** of these goals were achieved by integrating a pair of pseudocapacitive pressure-sensing units and a flexible circuit ***,additional conditions,such as variable hydraulic pressures and minimal changes in fish posture,require high stability and sensitivity of the sensing ***,hybrid hydrogel electrodes were developed through cross-linking MXene with holey-reduced graphene oxide nanosheets and further modification with 1-ethyl-3-methylimidazolium dicyanamide ionic liquids,which increased the interfacial capacitance and long-term interfacial activity of the ***,the sensing unit exhibited ultrahigh sensitivity(S_(max)~136,207 kPa^(−1))in an aquatic environment for 60 days and superior high-pressure resolution(10 Pa)within a wide working range of 1 ***,an underwater vest integrated with such sensing units clearly distinguished and recorded fish *** believe that the designed device may open avenues in flow field monitoring and ocean current detection and provide new insights into the development of sensitive underwater tagging.

Plastic offers a new niche for microorganisms,the *** everincreasing emission of plastic waste makes it critical to understand the microbial ecology of the plastisphere and associated ***,we present a global fingerprint of the plastisphere,analyzing samples collected from freshwater,seawater,and terrestrial *** plastisphere assembles a distinct microbial community that has a clearly higher heterogeneity and a more deterministically dominated assembly compared to natural *** coexistence patterns—loose and fragile networks with mostly specialist linkages among microorganisms that are rarely found in natural habitats—are seen in the *** microbiomes generally have a great potential to metabolize organic compounds,which could accelerate carbon *** involved in the nitrogen cycle are also altered in the plastisphere,especially in freshwater plastispheres,where a high abundance of denitrifiers may increase the release of nitrite(aquatic toxicant)and nitrous oxide(greenhouse gas).Enrichment of animal,plant,and human pathogens means that the plastisphere could become an increasingly mobile reservoir of harmful *** findings highlight that if the trajectory of plastic emissions is not reversed,the expanding plastisphere could pose critical planetary health challenges.

Exploring the structure-activity relationship between the performance of gas sensors and the structure of semiconductor metal oxide(SMO)nanomaterials is crucial for understanding and designing gas-sensing materials and overcoming the application limitations of SMO-based gas *** of a single SMO microstructure provides a promising solution to address this scientific problem due to its controllable *** this study,we control the grain boundary(GB)density of Fe_(2)O_(3)nanomaterials using a simple solvothermal *** have similar chemical compositions and crystal phases,providing an ideal platform for studying the influence of the GB density on the gas-sensing ***-sensing tests showed that the Fe_(2)O_(3)-1 sensor with medium GB density and the Fe_(2)O_(3)-2 sensor with high GB density had higher sensitivity and selectivity than the Fe_(2)O_(3)-0 sensors with low GB density before reaching the optimal operating ***,when the GB density increased,the response to acetone decreased slightly,whereas the optimal operating temperature *** work highlights the unique performance of the GB density in enhancing the gas sensitivity of a single SMO.

Superconducting SnTe-type topological crystalline insulators(TCIs)are predicted to host multiple Majorana zero modes(MZMs)which can coexist in a single *** level(FL)close to the Dirac points of topological surface states is helpful for detecting ***,the TCI SnTe is a heavily p-type semiconductor which is very difficult to modify to n-type via doping or *** this work,we fabricate the atomically flat Sn_(1-x)Pb_(x)Te/Pb heterostructure by molecular beam epitaxy,and make the p-type Sn_(1-x)Pb_(x)Te become n-type through changing the interface *** scanning tunnelling microscope,we find the Dirac points of Sn_(1-x)Pb_(x)Te/Pb heterostructure are always above the FL due to the Fermi level pinning(FLP)induced by topological surface states at atomically flat *** increasing the interface roughness,the FLP effect is suppressed and then the Dirac points of p-type Sn_(1-x)Pb_(x)Te can be tuned very close to or even below the *** work provides a new method for tuning the FL of SnTe-type TCI which has potential application in novel topological superconductor device.

探究荞麦壳黄酮对人肝癌HepG2细胞凋亡和迁移机制的影响。在用MTT比色法评价荞麦壳黄酮对人肺癌A549细胞、人宫颈癌Hela细胞、人肝癌HepG2细胞和人正常肝L-02细胞抑制效果的基础上,将HepG2细胞株作为试验对象,通过细胞形态学观察、Hoechst 33345/碘化丙啶(PI)双染、JC-1荧光探针检测、Annexin V-FITC/PI流式细胞术、伤口愈合试验和蛋白免疫印迹法,分析荞麦壳黄酮预处理对HepG2细胞凋亡和迁移的影响。结果表明:荞麦壳黄酮能够有效抑制A549细胞、Hela细胞和HepG2细胞的细胞活性,对HepG2细胞抑制效果最显著,50μg/mL干预HepG2细胞48 h后,细胞存活率为53.62%,且对L-02细胞活性无影响。与未处理组相比,荞麦壳黄酮以剂量依赖性的方式抑制HepG2细胞增殖,降低线粒体膜电位,下调凋亡蛋白Bcl-2、Bcl-xl表达,上调FADD、cleaved-Caspase-3蛋白表达,并降低迁移蛋白MMP 9表达,且效果均显著高于阳性对照芦丁。研究结果说明荞麦壳黄酮具有抗肿瘤能力,其抑制HepG2细胞增殖可能与诱导线粒体、死亡受体细胞凋亡和抑制细胞迁移有关。

Although tumor cell membranes with broad-spectrum antigens have been explored for cancer vaccines for decades,their relatively poor capacity to stimulate immune responses,especially cellular immune responses,has limited their ***,we presented a novel bacterial and cancerous cell membrane fusogenic liposome for co-delivering cell membrane-derived antigens and ***,a programmed death-ligand 1(PD-L1)inhibitor,JQ-1,was incorporated into the formulation to tackle the up-regulated PD-L1 expression of antigen-presenting cells(APCs)upon vaccination,thereby augmenting its anti-tumor *** fusogenic liposomes demonstrated significantly improved cellular uptake by APCs and effectively suppressed PD-L1 expression in bone marrow-derived dendritic cells(BMDCs)in *** subcutaneous vaccination,the nano-vaccines efficiently drained to the tumor-draining lymph nodes(TDLNs),and significantly inhibited PD-L1 expression of both dendritic cells(DCs)and macrophages within the TDLNs and *** a result,the liposomal vaccine induced robust innate and cellular immune responses and inhibited tumor growth in a colorectal carcinoma-burden mouse *** summary,the fabricated cell membrane-based fusogenic liposomes offer a safe,effective,and easily applicable strategy for tumor immunotherapy and hold potential for personalized cancer immunotherapy.

Inflammatory cytokine storms can trigger disease exacerbation and even death and have reached a consensus in the clinical treatment of acute organ ***,the existing strategies remain a great challenge to efficiently suppress inflammatory cytokine storms for promoting organ repair and ***,fully human umbilical cord(UC)-derived adhesive materials(UCAM)that integrate decellularized extracellular matrix(ECM)nanofiber hydrogel and homologous mesenchymal stem cells(MSCs)are demonstrated to greatly suppress inflammatory cytokine storms,demonstrating high efficacy in treating acute liver failure(ALF)in rats with 90%*** UC-derived adhesive materials have the capacity to secrete a significant quantity of cytokines by MSCs to recruit activated immune cells to migrate into their ECM nanofiber networks,segregating them away from the infection area and thereby greatly suppressing the inflammatory cytokine *** expected,the UC-derived adhesive materials can significantly promote hepatocyte proliferation to achieve functional recovery and regeneration of the liver,significantly improving the survival rate in *** fully human UC-derived adhesive materials provide a new avenue in suppressing inflammatory cytokine storms for promoting organ regeneration that would be really utility in clinical organ transplantation-related treatment.

Background and Aims:Increasing utilization of extended criteria donor leads to an increasing rate of early allograft failure after liver ***,consensus of definition of early allograft failure is ***:A retrospective,multicenter study was performed to validate the liver Graft Assessment Following Transplantation(L-GrAFT)risk model in a Chinese cohort of 942 adult patients undergoing primary liver transplantation at three Chinese centers.L-GrAFT(L-GrAFT7 and L-GrAFT10)was compared with existing models:the Early Allograft Failure Simplified Estimation(EASE)score,the model of early allograft function(MEAF),and the Early Allograft Dysfunction(EAD)*** and multivariate logistic regression were used to find risk factors of L-GrAFT high-risk ***:L-GrAFT7 had an area under the curve of 0.85 in predicting 90-day graft survival,significantly superior to MEAF[area under the curve(AUC=0.78,p=0.044)]and EAD(AUC=0.78,p=0.006),while there was no statistical significance between the predicting abilities of L-GrAFT7 and EASE(AUC=0.84,p>0.05).Furthermore,L-GrAFT7 maintains good predicting ability in the subgroup of high-donor risk index(DRI)cases(AUC=0.83 ***,p=0.007 ***,p=0.014)and recipients of donors after cardiac death(AUC=0.92 ***,plirubin level,units of packed red blood cells,and the DRI score were selected as independent risk factors of a L-GrAFT7 high-risk ***:The accuracy of L-GrAFT7 in predicting early allograft failure was validated in a Chinese multicenter cohort,indicating that it has the potential to become an accurate endpoint of clinical practice and transitional study of machine perfusion.
