Prohibitin(PHB),an evoluti on arily con served mitochondrial inner membra ne protein,is highly expressed in cells that require strong mitoch on drial ***,we dem on strated that the deleti on of Phb in spermatocytes results in impaired mitochondrial *** addition,PHB expression in the mitochondrial sheath of human sperm has a significantly negative correlation with mitochondrial reactive oxygen species levels,but a positive one with mitochondrial membrane potential and sperm *** results suggest that mitochondrial PHB expression plays a role in sperm ***,the mechanism of PHB-mediated regulation of sperm motility remai ns unk ***,we dem on strate for the first time that PHB interacts with protei n kinase B(AKT)and exists in a complex with phospho-PHB(pT258)and phospho-AKT in the mitochondrial sheath of murine sperm,as determined using colocalization and coimmunoprecipitation *** blocking AKT activity using wortmannin(a phosphatidylinositol 3-kinase[PI3K]inhibitor),murine sperm have significantly(Pof phospho-PHB(pT258)and the total and progressive ***,significantly(Power levels of phospho-PI3K P85 subunit a+γ(pY199 and pY46)and phospho-AKT(pS473;pT308)are found in sperm from infertile asthenospermic and oligoasthenospermic men compared with no rmospermic subjects,which suggest a reduced activity of the PI3K/AKT pathway in these in fertile ***,these sperm from infertile subjects also have a significantly(Power level of phospho-PHB(pT258).Collectively,our findings suggest that the interaction of PHB with AKT in the mitochondrial sheath is critical for sperm motility,where PHB phosphorylation(pT258)level and PI3K/AKT activity are key regulatory factors.

Contents of four active components in different commercial crude drugs and preparations of Danshen (Salvia miltiorrhiza)

AIM: To detect the contents of four active components of Salvia miltiorrhiza in various commercially available danshen crude drugs and preparations. METHoDS: Commercially available danshen crude drugs from different sources, as well as danshen pills and intravenous injection preparations containing danshen alone or in combination with other herbs were collected. The composition of these danshen samples was analyzed using HPLC. Specifically, the amounts of magnesium tanshinoate B (MTB), danshensu, isotanshinone IIA, and ctyptotanshinone were determined. In some of these samples, the content of MTB was further confirmed by liquid chromatography-tandem mass spectrometer (LC-MS)/MS method. RESULTS: There were great variations in the amount of the four active ingredients in the commercially available danshen crude drugs and drug preparations in this study. Mining of plant-derived antimicrobials is the major focus at current to counter antibiotic resistance. This study was conducted to characterize the antimicrobial activity and mode of action of linalyl anthranilate(LNA) against carbapenemase-producing Klebsiella pneumoniae(KPC-KP). LNA alone exhibited bactericidal activity at 2.5%(V/V), and in combination with meropenem(MPM) at 1.25%(V/V). Comparative proteomic analysis showed a significant reduction in the number of cytoplasmic and membrane proteins,indicating membrane damage in LNA-treated KPC-KP cells. Up-regulation of oxidative stress regulator proteins and down-regulation of oxidative stress-sensitive proteins indicated oxidative stress. Zeta potential measurement and outer membrane permeability assay revealed that LNA increases both bacterial surface charge and membrane permeability. Ethidium bromide influx/efflux assay showed increased uptake of ethidium bromide in LNA-treated cells, inferring membrane damage. Furthermore, intracellular leakage of nucleic acid and proteins was detected upon LNA treatment. Scanning and transmission electron microscopies again revealed the breakage of bacterial membrane and loss of intracellular materials. LNA was found to induce oxidative stress by generating reactive oxygen species(RoS) that initiate lipid peroxidation and damage the bacterial membrane. In conclusion, LNA generates RoS, initiates lipid peroxidation, and damages the bacterial membrane, resulting in intracellular leakage and eventually killing the KPC-KP cells.

This study is a spectral analysis of the formant frequencies and formant movements of the diphthongs[ia,ai,ua,au,ie,ei,uo, ou,ye] that occur in open(C)V syllables in Peking dialect from both male and female *** main findings are as follows.(i)Regardless of the type of diphthongs,formant undershoot is larger for the high vowel elements than the low or mid vowel elements of the diphthongs.(ii) Formant undershoot is more pronounced for the high vowel elements of the diphthongs[ia,ai,ua,au] with larger formant movement than those of the diphthongs[ie,ei,uo,ou,ye]with smaller formant movement.(iii) Formant undershoot is larger for the diphthongs[ai,au,ei,ou] in Peking dialect than those in ***,(iv) the patterns of the formant undershoot for the diphthongs are similar in male and female speech.

The study^1 evaluates the quantal theory by analyzing the articulatory-acoustic relations in the four Cantonese palatal vowels[i:y:i y]through investigating the sensitivity of vowel formant frequencies to variation of linguo-palatal constriction location and that of constriction size along the vocal tract length during the vowels,using KMA AG5 0 *** articulatory-acoustic data obtained in this study show non-linear relationship between the up-down and front-back displacements in linguo-palatal constriction during the vowels and the acoustic consequences,which is similar to what the quantal theory *** displacements in constriction location and constriction size result in large spectral *** level of sensitivity of formant frequencies to variation of tongue displacement is lower for the unrounded[i:i]than the rounded counterparts[y:y],suggesting the quantal nature is more apparent in[i:i]than[y:y].In general,for the Cantonese palatal vowels the levels of sensitivity of F1 to the variation of constriction location and that of constriction size are similar,whereas F2 is more sensitive to the variation of constriction size than to the variation of constriction *** latter is similar to the articulatory-acoustic relations in the Knglish vowels reported in the previous studies.

Seropharmacological Study on osteogenic Effects of Post-Absorption Ingredients of An osteoprotective Herbal Formula

objective： To further investigate the in vitro effects of an osteoprotective herbal formula ＂ELP＂（Herba Epimedii, Fructus Ligustri Lucidi and Fructus Psora/eae） using seropharmacological approach. Methods： Rats were fed with ELP or its individual component herbs for 2 days. The serum containing the post-absorbed ingredients of the herbal items were collected for cell culture using UMR106 cell, RAW264.7 cell and mesenchymal stem cell （MSC） isolated from the bone marrow of the rats. The effects of the herbal-containing serum on cell toxicity were detected by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay; bromodeoxyuridine assay was conducted to measure the cell proliferation of UMR106 cell and MSC; cell activity was measured using colorimetric method, and mRNA expression of runt-related transcription factor 2 （Runx2）, alkaline phosphatase （ALP） and osteopontin （oPN） of UMR106 and MSC as well as matrix metalloproteinase 9 （MMP-9）, tartrate-resistant acid phosphatase （TRAP） and cathepsin K of RAW264.7 were analyzed using real-time reverse-transcription polymerase chain reaction. Results： ELP and its component serum exhibited no cytotoxic effects on the cells. The ELP-containing serum increased the proliferation of UMR106 cell and MSC by 25.7% and 14.4%, respectively and the alkaline phosphatase activity of MSC was increased by 42.6%. on the contrary, it inhibited the RAW264.7 cell differentiation by 29.2%. ELP serum upregulated the Runx2 expression of UMR and MSC by 1.18 fold and 1.27 fold, respectively. It also upregulated ALP and oPN expression in MSC by 1.69- and 2.12-fold, respectively. on the other hand, ELP serum down-regulated MMP-9 and cathepsin K expression of RAW264.7 cell by 0.46- and 0.36-fold, respectively. Conclusions： The serum of the animals fed with ELP contains active ingredients which are effective in promoting osteogenesis and inhibiting osteoclastogenesis.

Characterizing the Urban Temperature Trend Using Seasonal Unit Root Analysis:Hong Kong from 1970 to 2015

This paper explores urban temperature in Hong Kong using long-term time series. In particular, the characterization of the urban temperature trend was investigated using the seasonal unit root analysis of monthly mean air temperature data over the period January 1970 to December 2013. The seasonal unit root test makes it possible to determine the stochastic trend of monthly temperatures using an autoregressive model. The test results showed that mean air temperature has increased by 0.169~ C （10 yr） - 1 over the past four decades. The model of monthly temperature obtained from the seasonal unit root analysis was able to explain 95.9% of the variance in the measured monthly data -- much higher than the variance explained by the ordinary least-squares model using annual mean air temperature data and other studies alike. The model accurately predicted monthly mean air temperatures between January 2014 and December 2015 with a root-mean-square percentage error of 4.2%. The correlation between the predicted and the measured monthly mean air temperatures was 0.989. By analyzing the monthly air temperatures recorded at an urban site and a rural site, it was found that the urban heat island effect led to the urban site being on average 0.865~C warmer than the rural site over the past two decades. Besides, the results of correlation analysis showed that the increase in annual mean air temperature was significantly associated with the increase in population, gross domestic product, urban land use, and energy use, with the R2 values ranging from 0.37 to 0.43.
