Head and neck squamous cell carcinoma(HNSCC)is a very aggressive disease with a poor prognosis for advanced-stage *** clinical,genomic,and cellular studies have revealed the highly heterogeneous and immunosuppressive nature of *** signifcant advances in multimodal therapeutic interventions,failure to cure and recurrence are common and account for most *** is becoming increasingly apparent that tumor microenvironment(TME)plays a critical role in HNSCC tumorigenesis,promotes the evolution of aggressive tumors and resistance to therapy,and thereby adversely affects the prognosis.A complete understanding of the TME factors,together with the highly complex tumor-stromal interactions,can lead to new therapeutic interventions in ***,different molecular and immune landscapes between HPV^(+ve) and HPV^(-ve)(human papillomavirus)HNSCC tumors offer new opportunities for developing individualized,targeted chemoimmunotherapy(CIT)*** review highlights the current understanding of the complexity between HPV^(+ve) and HPV^(-ve) HNSCC TME and various tumor-stromal cross-talk modulating processes,including epithelial-mesenchymal transition(EMT),anoikis resistance,angiogenesis,immune surveillance,metastatic niche,therapeutic resistance,and development of an aggressive tumor ***,we summarize the recent developments and the rationale behind CIT strategies and their clinical applications in HPV^(+ve) and HPV^(-ve) HNSCC.

Synthesis and Antimicrobial Activity of New 2,5-Disubstituted 1,3,4-Oxadiazoles and 1,2,4-Triazoles and Their Sugar Derivatives

A series of new 2,5-disubstituted-l,3,4-oxadiazole and 1,2,4-triazole derivatives were synthesized by hetero- cyclization of acid hydrazide I and thiosemicarbazide derivative 2. Furthermore, the acyclic C-nucleoside analogs were prepared by cyclization of their corresponding sugar hydrazones by reaction with acetic anhydride. The antimicrobial activity of the prepared compounds was evaluated and some of the synthesized compounds revealed good activities against fungi.

Antibody to El peptide of hepatitis C virus genotype 4 inhibits virus binding and entry to HepG2 cells in vitro

AIM： To analyze the neutralizing activity of antibodies against E1 region of hepatitis C virus （HCV）. Specific polydonal antibody was raised via immunization of New Zealand rabbits with a synthetic peptide that had been derived from the E1 region of HCV and was shown to be highly conserved among HCV published genotypes. METHODS： Hyper-immune HCV E1 antibodies were incubated over night at 4 ℃ with serum samples positive for HCV RNA, with viral loads ranging from 615 to 3.2 million IU/mL. Treated sera were incubated with HepG2 cells for 90 min. Blocking of viral binding and entry into cells by anti E1 antibody were tested by means of RTPCR and flow cytometry. RESULTS： Direct immunostaining using FITC conjugated E1 antibody followed by Flow cytometric analysis showed reduced mean fluorescence intensity in samples pre-incubated with E1 antibody compared with untreated samples. Furthermore, 13 out of 18 positive sera （72%） showed complete inhibition of infectivity as detected by RT-PCR. CONCLUSION： In house produced E1 antibody, blocks binding and entry of HCV virion infection to target cells suggesting the involvement of this epitope in virus binding and entry. Isolation of these antibodies that block virus attachment to human cells are useful as therapeutic reagents.

Comparison of urinary telomerase,CD44,and NMP22 assays for detection of bladder squamous cell carcinoma

Background:Squamous cell carcinoma(SCC)of the bladder is common in many regions around the *** is very poor,as most cases are diagnosed at an advanced stage due to a lack of affordable and valid screening markers for this type of *** diagnostic accuracy of urinary nuclear matrix protein-22(NMP22),telomerase activity,and CD44 were evaluated in urine samples of patients with bladder *** and methods:We conducted a case-control study comprised of 60 consecutive newly diagnosed bladder SCC patients diagnosed by cystoscopy and histopathological examination,and controls were 60 outpatients with benign urologic conditions and healthy clinic *** samples collected from each subject underwent testing for NMP22,telomerase activity,and *** and correlational statistical analysis of cases and controls were carried out and receiver operating characteristic curve analysis was used to determine optimal cut-off points for the three ***:Area under the curve was calculated at 0.96,0.93,and 0.62 for NMP22,telomerase,and CD44,*** levels of NMP22 and telomerase activity were significantly higher in the SCC group compared to controls(p<0.001).Urine CD44 levels were not significantly higher in the SCC group compared to controls(p=0.111).The overall sensitivity of NMP22,telomerase,and CD44 was 96.7%,87%,and 45%,respectively,while the specificity was 85%,88.6%,and 86.7%,***:Urinary telomerase activity,followed by NMP22 urine levels,showed high diagnostic yield and could hold potential promise as urinary biomarkers for the diagnosis of bladder SCC.

Barrett's esophagus:Prevalence and risk factors in patients with chronic GERD in Upper Egypt

AIM： To determine the prevalence and possible risk factors of Barrett＇s esophagus （BE） in patients with chronic gastroesophageal reflux disease （GERD） in ei Minya and Assuit, Upper Egypt. METHODS： One thousand consecutive patients with chronic GERD symptoms were included in the study over 2 years. They were subjected to history taking including a questionnaire for GERD symptoms, clinical examination and upper digestive tract endoscopy. Endoscopic signs suggestive of columnar-lined esophagus （CLE） were defined as mucosal tongues or an upward shift of the squamocolumnar junction. BF was diagnosed by pathological examination when specialized intestinal metaplasia was detected histologically in suspected CLE. pH was monitored in 40 patients. RESULTS： BE was present in 7.3% of patients with chronic GERD symptoms, with a mean age of 48.3 ± 8.2 years, which was significantly higher than patients with GERD without BE （37.4 ± 13.6 years）. Adenocarcinoma was detected in eight cases （0.8%）, six of them in BE patients. There was no significant difference between patients with BE and GERD regarding sex, smoking, alcohol consumption or symptoms of GERD. Patients with BE had significantly longer esophageal acid exposure time in the supine position, measured by pH monitoring. CONCLUSION： The prevalence of BE in patients with GERD who were referred for endoscopy was 7.3%. BE seems to be associated with older age and more in patients with nocturnal gastroesophageal reflux.
