Two kinds of cyclic aryl ester dimers have been synthesized by reaction of phthaloyl dichloride with bisphenols via interfacial polycondensation. The cyclic dimers readily undergo anionic ring-opening polymerization or copolymerization in the melt by using sodium benzoate as the initiator, producing linear, high molecular weight polyesters. The contents of cyclic dimers in the homopolymers P1, P2, and copolymer P12 are 13.7%, 10.2%, 2.9%, respectively, which indicates that ring-opening copolymerization of cyclic dimers may impel the conversion of cyclic dimers and decrease the content of cyclic dimers in the resulting copolymer. Moreover, the isothermal chemorheology of the ring-opening copolymeriza- tion of cyclic dimers indicates that the reactive molten mixture has low shear viscosity and the viscosity increases slowly in the initial stage of ring-opening polymerization.

Objective Pseudogenes are initially regarded as nonfunctional genomic sequences,but some pseudogenes regulate tumor initiation and progression by interacting with other genes to modulate their transcriptional *** receptor family 7 subfamily E member 47 pseudogene(OR7E47P)is expressed broadly in lung tissues and has been identified as a positive regulator in the tumor microenvironment(TME)of lung adenocarcinoma(LUAD).This study aimed to elucidate the correlation between OR7E47P and tumor immunity in lung squamous cell carcinoma(LUSC).Methods Clinical and molecular information from The Cancer Genome Atlas(TCGA)LUSC cohort was used to identify OR7E47P-related immune genes(ORIGs)by weighted gene correlation network analysis(WGCNA).Based on the ORIGs,2 OR7E47P clusters were identified using non-negative matrix factorization(NMF)clustering,and the stability of the clustering was tested by an extreme gradient boosting classifier(XGBoost).LASSO-Cox and stepwise regressions were applied to further select prognostic ORIGs and to construct a predictive model(ORPScore)for *** Botling cohorts and 8 immunotherapy cohorts(the Samstein,Braun,Jung,Gide,IMvigor210,Lauss,Van Allen,and Cho cohorts)were included as independent validation *** OR7E47P expression was positively correlated with immune cell infiltration and enrichment of immune-related pathways in LUSC.A total of 57 ORIGs were identified to classify the patients into 2 OR7E47P clusters(Cluster 1 and Cluster 2)with distinct immune,mutation,and stromal *** to Cluster 1,Cluster 2 had more infiltration by immune and stromal cells,lower mutation rates of driver genes,and higher expression of immune-related *** clustering performed well in the internal and 5 external validation *** on the 7 ORIGs(HOPX,STX2,WFS,DUSP22,SLFN13,GGCT,and CCSER2),the ORPScore was constructed to predict the prognosis and the treatment *** addition,the ORPScore was a bette

目的分析胎龄25%是双胎中仅一胎患BPD的危险因素(OR=3.370,95%CI:1.500~7.568,P<0.05),胎龄大是双胎均患BPD的保护因素(P<0.05)。对双胎中仅一胎为BPD组进行组内条件logistic回归分析,提示小于胎龄儿是双胎内个体发生BPD的危险因素(OR=5.017,95%CI:1.040~24.190,P<0.05)。结论双胎早产儿BPD的发生不仅与胎龄相关,也与双胎出生体重差异、小于胎龄儿密切相关。

Histone lysine methylation is known to be involved in the epigenetic regulation of gene expression in all eukaryotes including plants. Here we show that the rice SDG714 is primarily responsible for dimethylation but not trimethylaUon on histone H3K9 in vivo. Overexpression of YFP-SDG714 in Arabidopsis significantly inhibits plant growth and this inhibition is associated with an enhanced level of H3K9 dimethylation. Our microarray results show that many genes essential for the plant growth and development were downregulated in transgenic Arabidopsis plants overexpressing YFP-SDG714. By chromatin immunoprecipitation analysis, we show that YFP-SDG714 is targeted to specific chromatin regions and dimethylate the H3Kg, which is linked with heterochromatinization and the downregulation of genes. Most interestingly, when YFP-SDG714 production is stopped, the inhibited plants can partially restore their growth, suggesting that the perturbation of gene expression caused by YFP-SDG714 is revertible. Taken together, our results point to an important role of SDG714 in H3K9 dimethylation, suppression of gene expression and plant growth, and provide a potential method to regulate gene expression and plant development by an on-off switch of SDG714 expression.

BACKGROUND: Several studies have shown that KAI1 inhibits tumor metastasis, but its mechanism is not clear. The present study aimed to determine the role of KAI1 in lymphatic metastasis, specifically in pancreatic cancer. METHODS: The KAI1 gene was transfected into the pancreatic cancer cell line MIA PaCa-2 and PANC-1 by using liposomes and selected by G418, and the protein was measured by Western blotting. After successful infection, the cell growth curve was studied by MTT, vascular endothelial growth factor C(VEGF-C) secretion by pancreatic cancer cell were measured by ELISA. The KAI1 and pCMV transfected MIA PaCa-2 cells were renamed as MIA PaCa-2-K and MIA PaCa-2-p. These two kinds of cells were injected into the subcuticular layer of nude mice; both tumor growth and metastasis through the lymphatic nodes were assessed. Lymphangiogenesis in tumors was measured by immunohistochemistry. RESULTS: The VEGF-C secretion was significantly reduced in MIA PaCa-2 cells compared with PANC-1 cells after being transfected with the KAI1 gene. The growth rate of subcutaneous tumors was similar after the injection of MIA PaCa-2-K, MIA PaCa-2, and MIA PaCa-2-p. MIA PaCa-2-K tumors showed slower lymphangiogenesis and lymph node metastasis compared with MIA PaCa-2 and MIA PaCa-2-p tumors. CONCLUSION: The overexpression of KAI1 inhibits the lymphangiogenesis and lymph node metastasis of MIA PaCa-2 pancreatic tumors.

OBJECTIVE Cognitive deficit is a common comorbidity in temporal lobe epilepsy(TLE) and that is not well controlled by current therapeutics. Currently,how epileptic seizure affects cognitive performance remains largely unclear. The subiculum is the major output of the hippocampus, which projects to entorhinal cortex and other more distinct brain regions. Physiologically,the subiculum codes spatial working memory and navigation information including place, speed, and ***, prior studies have noted the importance of the subiculum in the beginning, spreading, and generalizing process of hippocampal seizure. How seizure-activated neurons in subiculum participate in cognitive impairment remains largely elusive. METHODS In this study, we sought to label the subicular seizure-activated c-fos+neurons with a special promoter with enhanced synaptic activity-responsive element E-SARE in the subiculum,combined with chemogenetics and designer receptors exclusively activated by designer drugs(DREADDs), Ca2+fiber photometry approaches, and behavioral tasks, to reveal the role of these neurons in cognitive impairment in epilepsy. RESULTS We found that chemogenetic inhibition of subicular seizure-tagged c-fos+neurons(mainly CaMK Ⅱ α+glutamatergic neurons) alleviates seizure generalization and improves cognitive performance in the hippocampal CA3 kindling TLE model. While inhibition of seizure-labeled c-fos+GABAergic interneuron shows no effect on seizure and cognition. As a comparison, chemogenetic inhibition of the whole subicular CaMK Ⅱ α+neuron impairs cognitive function in na?ve mice in basal condition. Notably, inhibition of subicular seizure-tagged c-fos+neurons enhances the recruitment of cognitionresponsive c-fos+neurons via increasing neural excitability during cognition tasks. CONCLUSION Our results demonstrate that subicular seizure-activated c-fos+neurons contribute to cognitive impairment in TLE, suggesting seizure-tagged c-fos+neurons as the potential

目的观察布瑞哌唑对成人急性期精神分裂症患者的疗效和安全性。方法将成人急性期精神分裂症患者随机分为试验组与对照组,试验组给予口服布瑞哌唑每日2~4 mg,对照组给予口服阿立哌唑每日10~20 mg,2组均治疗6周。比较2组治疗后有效率,比较2组阳性和阴性症状量表(PANSS)总评分、临床总体印象-严重程度(CGI-S)评分、个人和社会功能量表(PSP)评分、PANSS阳性症状分量表评分和PANSS阴性症状分量表评分治疗前后的变化,观察2组患者治疗相关不良事件的发生情况。结果试验组184例、对照组186例。治疗6周末,试验组和对照组的治疗有效率分别为79.50%(140例/184例)和82.40%(150例/186例),临床总体印象-改善量表(CGI-I)评分分别为(2.00±1.20)和(1.90±1.01)分,在统计学上差异均无统计学意义(均P>0.05)。治疗6周后试验组和对照组的PANSS总分较基线变化分别为(-30.70±16.96)和(-32.20±17.00)分,2组间差异无统计学意义(P>0.05)。治疗前后,试验组和对照组的CGI-S评分变化分别为(-2.00±1.27)和(-1.90±1.22)分,PSP评分变化分别为(18.80±14.77)和(19.20±14.55)分,PANSS阳性症状评分变化分别为(-10.30±5.93)和(-10.80±5.81)分,PANSS阴性症状评分变化分别为(-6.80±5.98)和(-7.30±5.15)分,在统计学上差异均无统计学意义(P>0.05)。试验组治疗相关不良事件发生率与对照组相比差异无统计学意义(69.00%vs.64.50%,P>0.05)。结论布瑞哌唑相比于阿立哌唑治疗精神分裂症的非劣效性成立,疗效及安全性相当。
