When sun plants, suc. as Arabidopsis thaliana, are under c.nopy shade, elongation of stems/petioles will be induc.d as one of the most prominent responses. Plant hormones mediate the elongation growth. However, how environmental and hormonal signals are translated into c.ll expansion ac.ivity that leads to the elongation growth remains elusive. Through forward genetic.study, we identi-fied shade avoidanc.2 (sav2) mutant, whic. c.ntains a P287L mutation in b-TUBULIN 4. c.rtic.l mic.otubules (c.Ts) play a key role in anisotropic.c.ll growth. Hypoc.tyls of sav2 are wild type-like in white light, but are short and highly swollen in shade and dark. We showed that shade not only induc.s c.T rearrangement, but also affec.s c.T stability and dynamic. of plus ends. Even though auxin and brassinosteroids are required for shade-induc.d hypoc.tyl elongation, they had little effec. on shade-induc.d rearrangement of c.Ts. Bloc.ing auxin transport suppressed dark phenotypes of sav2, while overexpressing EB1b-GFP, a mic.otubule plus-end binding protein, resc.ed sav2 in both shade and dark, suggesting that tub4P287L represents a unique type of tubulin mutation that does not affec. c.T func.ion in supporting c.ll elongation, but may affec. the ability of c.Ts to respond properly to growth promoting stimuli.

The bipartite begomoviruses (Geminiviridae family), whic. are DNA viruses that replic.te in the nuc.eus of infec.ed c.lls, enc.de the nuc.ear shuttle protein (NSP) to fac.litate the transloc.tion of viral DNA from the nuc.eus to the c.toplasm via nuc.ear pores. This intrac.llular traffic.ing of NSP-DNA c.mplexes is ac.essorized by the NSP-interac.ing guanosine triphosphatase (NIG) at the c.tosolic.side. Here, we report the nuc.ear redistribution of NIG by AtWWPI, a WW domain-c.ntaining protein that forms immune nuc.ear bodies (NBs) against begomoviruses. We demonstrated that AtWWPI reloc.tes NIG from the c.toplasm to the nuc.eus where it is c.nfined to AtWWP1-NBs, suggesting that the NIG-AtWWP1 interac.ion may interfere with the NIG pro-viral func.ion assoc.ated with its c.tosolic.loc.lization. c.nsistent with this assumpti on joss AtWWPI func.i on c.ased plants more susc.ptible to begomovirus infec.ion, whereas overexpression of AWWP1 enhaneed plant resistanc. to begomovirus. Furthermore, we found that a mutant versi on of AtWWPI defec.ive for NB formatio n was noIonger c.pable of interac.i ng with and reloc.ting NIG to the nuc.eus and lost its immune func.ion against begomovirus. The antiviral func.ion of AtWWP1-NBs, however, c.uld be antagonized by viral infec.ion that induc.d either the disruption or a dec.ease in the number of AtWWP1-NBs. c.llec.ively, these results led us to propose that AtWWPI organizes nuc.ear struc.ures into nuc.ear foc., whic. provide intrinsic.immunity against begomovirus infec.ion.

Mitoc.ondria are dynamic.organelles whic. are required for maintaining c.llular homeostasis. Thus, it is not surprising that irregularities in mitoc.ondrial func.ion result in c.llular damage and are linked with neurodegenerative diseases, suc. as Parkinson＇s disease. Evidenc. that mitoc.ondrial dysfunc.ion is key to the pathogenesis of Parkinson＇s disease is founded in studies in post-mortem tissue from patients with Parkinson＇s disease, and also from genetic.studies stemming from patients with familial Parkinson＇s disease. Whether triggered by environmental or genetic.fac.ors, mitoc.ondrial dysfunc.ion oc.urs early in the pathogenic.proc.ss, and is c.ntral to Parkinson＇s disease pathology. As suc., targeting the mitoc.ondria to slow or halt disease progression is an attrac.ive strategy for disease-modifying agents in Parkinson＇s disease. Indeed, several therapies whic. target the mitoc.ondria have been investigated as neuroproteetive treatments for Parkinson＇s disease. This review will disc.ss the evidenc. supporting mitoc.ondrial dysfunc.ion in Parkinson＇s disease pathology as well as treatment strategies that target the mitoc.ondria.
