As a precise and versatile tool for genome manipulation,the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)platform holds promise for modifying fish traits of *** the aim of reducing transgene introgression and controlling reproduction,upscaled disease resistance and reproductive intervention in catfish species have been studied to lower the potential environmental risks of the introgression of escapees as transgenic *** advantage of the CRISPR/Cas9-mediated system,we succeeded in integrating the cathelicidin gene(As-Cath)from an alligator(Alligator sinensis)into the target luteinizing hormone(lh)locus of channel catfish(Ictalurus punctatus)using two delivery systems assisted by double-stranded DNA(dsDNA)and single-stranded oligodeoxynucleotides(ssODNs),*** this study,high knock in(KI)efficiency(22.38%,64/286)but low ontarget events was achieved using the ssODN strategy,whereas adopting a dsDNA as the donor template led to an efficient on-target KI(10.80%,23/213).The on-target KI of As-Cath was instrumental in establishing the lh knockout(LH^(–)_As-Cath^(+))catfish line,which displayed heightened disease resistance and reduced fecundity compared with the wild-type(WT)sibling ***,administration of human chorionic gonadotropin(HCG)and luteinizing hormone-releasing hormone analogue(LHRHa)can restore the reproduction of the transgenic fish ***,we replaced the lh gene with an alligator cathelicidin transgene and then administered hormone therapy to move towards complete reproductive control of diseaseresistant transgenic catfish in an environmentally responsible *** strategy not only effectively improves consumer-valued traits but also guards against unwanted introgression,providing a breakthrough in aquaculture genetics to confine fish reproduction and prevent the establishment of transgenic or domestic genotypes in the natural environment.

Wide-field linear structured illumination microscopy(LSIM)extends resolution beyond the diffraction limit by moving unresolvable high-frequency information into the passband of the microscopy in the form of moiré***,due to the diffraction limit,the spatial frequency of the structured illumination pattern cannot be larger than the microscopy cutoff frequency,which results in a twofold resolution improvement over wide-field *** Letter presents a novel approach in point-scanning LSIM,aimed at achieving higher-resolution improvement by combining stimulated emission depletion(STED)with point-scanning structured illumination microscopy(ps SIM)(STED-ps SIM).The according structured illumination pattern whose frequency exceeds the microscopy cutoff frequency is produced by scanning the focus of the sinusoidally modulated excitation beam of STED *** experimental results showed a 1.58-fold resolution improvement over conventional STED microscopy with the same depletion laser power.

To fully exploit enormous data generated by intelligent devices in edge computing,edge federated learning(EFL)is envisioned as a promising *** distributed collaborative training in EFL deals with delay and privacy issues compared to traditional centralized model ***,the existence of straggling devices,responding slow to servers,degrades model *** consider data heterogeneity from two aspects:high dimensional data generated at edge devices where the number of features is greater than that of observations and the heterogeneity caused by partial device *** large number of features,computation overhead on the devices increases,causing edge devices to become *** incorporation of partial training results causes gradients to be diverged which further exaggerates when more training is performed to reach local *** this paper,we introduce elastic optimization methods for stragglers due to data heterogeneity in edge federated ***,we define the problem of stragglers in ***,we formulate an optimization problem to be solved at edge *** customize a benchmark algorithm,FedAvg,to obtain a new elastic optimization algorithm(FedEN)which is applied in local training of edge *** mitigates stragglers by having a balance between lasso and ridge penalization thereby generating sparse model updates and enforcing parameters as close as to local *** have evaluated the proposed model on MNIST and CIFAR-10 *** experiments demonstrate that our approach improves run time training performance by achieving average accuracy with less communication *** results confirm the improved performance of our approach over benchmark algorithms.

Biochar is a carbon-neutral or even carbon-negative material produced through thermal decomposition of plant-and animal-based biomass under oxygen-limited ***, there has been an increasing interest in the application of biochar as an adsorbent, soil ameliorant and climate mitigation approach in many types of ***-contaminated soil remediation using biochar has been intensively investigated in small-scale and pilot-scale trials with obtained beneficial results and multifaceted *** so far, the study and application of biochar in contaminated sediment management has been very limited, and this is also a worldwide problem. Nonetheless, there is reason to believe that the same multiple benefits can also be realized with these sediments due to similar mechanisms for stabilizing contaminants. This paper provides a review on current biochar properties and its use as a sorbent/amendment for metal-contaminated soil/sediment remediation and its effect on plant growth, fauna habits as well as microorganism communities. In addition, the use of biochar as a potential strategy for contaminated sediment management is also discussed, especially as regards in-situ planning. Finally, we highlight the possibility of biochar application as an effective amendment and propose further research directions to ensure the safe and sustainable use of biochar as an amendment for remediation of contaminated soil and sediment.

Health institutions in the People’s Republic of Chinaare under considerable challenge. Innovations in management systems may support the challenge to increase both efficiency and effectiveness. The balanced scorecard (BSC) is one such innova-tion. We have used a model to explore the factors that may impact the diffusion of the BSC in Chinese government hospitals. In particular, we concentrate on three factors — the outer context, the nature of the innovation and the communication and influence. The outer context and the nature of the innovation do provide an inducement to implement a BSC. The strength of the communica-tion and influence may affect the level of take-up. However, we recognise that they may choose the BSC not for any apparent technical excellence but for the legitimation that Western man-agement practices may bring to local managers.

A new vertically aligned nanocomposite(VAN)structure based on two-dimensional(2D)layered oxides has been designed and self-assembled on both LaAlO_(3)(001)and SrTiO3(001)*** new VAN structure consists of epitaxially grown Co_(3)O_(4) nanopillars embedded in the Bi_(2)WO_(6) matrix with a unique 2D layered structure,as evidenced by the microstructural *** property measurements show that the new Bi_(2)WO_(6)-Co_(3)O_(4) VAN structure exhibits strong ferromagnetic and piezoelectric response at room temperature as well as anisotropic permittivity *** work demonstrates a new approach in processing multifunctional VANs structure based on the layered oxide systems towards future nonlinear optics,ferromagnets,and multiferroics.
