Identification and validation of stable quantitative trait loci for yield component traits in wheat

Grain weight and grain number are important yield component traits in wheat and identification of underlying genetic loci is helpful for improving ***,we identified eight stable quantitative trait loci(QTL)for yield component traits,including five loci for thousand grain weight(TGW)and three for grain number per spike(GNS)in a recombinant inbred line population derived from cross yangxiaomai/Zhongyou 9507 across four *** grain size is a major determinant of grain weight,we also mapped QTL for grain length(GL)and grain width(GW).***-2D,***-3B,***-5A and ***-7A.2 for TGW co-located with those for grain ***-2D also had a consistent genetic position with ***-2D,suggesting that the pleiotropic locus is a modulator of trade-off effect between TGW and *** and linkage mapping showed that TaGL3-5A and WAPO-A1 were candidate genes of ***-5A and ***-7A.2,*** developed Kompetitive allele specific PCR(KASP)markers linked with the stable QTL for yield component traits and validated their genetic effects in a diverse panel of wheat cultivars from the Huang-Huai River Valley ***-based genotyping analysis further revealed that the superior alleles of all stable QTL for TGW but not GNS were subject to positive selection,indicating that yield improvement in the region largely depends on increased *** analyses with previous studies showed that most of the QTL could be detected in different genetic backgrounds,and ***-7A.1 is likely a new *** findings provide not only valuable genetic information for yield improvement but also useful tools for marker-assisted selection.

Comprehensive evaluation of 20 pomegranate(Punica granatum L.) cultivars in China

Recent investigations on pomegranate products have significantly increased and successfully drawn consumers’ attention to nutritional and medicinal values, promoting the pomegranate industry’s development worldwide. However, little information on pomegranates grown in China is available. Morphological and chemical characterizations of fruits and arils from 20 pomegranate cultivars in six regions of China were investigated. Combined with overall scores by principal component analysis, ‘Yushiliu No. 1’, ‘Taishanhong No. 2’, ‘Tunisia’ and ‘Mollar’ were promising cultivars, and Chinese researchers bred the first two. It was surprising that ‘Mollar’ had bigger fruit size and more aril moisture grown in China than in Spain. Cultivars with higher anthocyanin content in arils were ‘Turkey’, ‘Moyu’ and ‘Red Angel’, which might be used as the source of natural red food colourants. While red husk ‘Hongruyi’ and ‘Hongshuangxi’ with higher vitamin C, aril moisture and lower titratable acid in arils, might also be promising cultivars for further various utilization. Furthermore, the comparison of ‘Tunisia’ fruits from four regions revealed that cultivation locations had more influence on fruit traits than genotypes. Maturity index classification was established for Chinese pomegranate cultivars. Therefore, the results would provide a valuable guide for agricultural cultivation, industrial utilization, and breeding.

Synergistic effects of AAGL and anti-PD-1 on hepatocellular carcinoma through lymphocyte recruitment to the liver

Objective:Therapy for hepatocellular carcinoma(HCC)is a major challenge,and targeted therapies provide only a modest benefit in terms of overall *** with antibodies to programmed cell death protein 1(PD-1)/PD-L1 can restore the functions of tumor-infiltrating T cells in HCC and has shown clinical efficacy in 20%of patients with advanced *** approaches are urgently needed to treat HCC and to augment the efficacy of ***:Tumor-bearing mice were treated with Agrocybe aegerita galectin(AAGL)alone or in combination with anti-PD-1,and the tumor sizes and lifespans of mice were *** analysis,cytokine analysis,flow cytometry analysis of the number and proportion of immune cell subsets in the liver and spleen,and molecular and cellular analyses of tumors were used to define the underlying ***:AAGL significantly inhibited the growth of liver tumors in a dose-dependent ***,AAGL increased the expression of multiple cytokines and chemokines in tumor-bearing mouse livers;this effect was associated with the activation and migration of T cells and macrophages,in agreement with the in vitro ***,the aggregation of T cells and macrophages induced by AAGL in tumor-bearing mouse livers clearly enhanced the response to PD-1 blockade ***:The results showed that AAGL induced the activation and migration of lymphocytes to the liver,and that the combination of AAGL and anti-PD-1 may be a promising strategy for HCC treatment.

Superior Toughened Biodegradable Poly(L-lactic acid)-based Blends with Enhanced Melt Strength and Excellent Low-temperature Toughness via In situ Reaction Compatibilization

The toughened poly(L-lactic acid)/poly(butylene succinate-butylene terephthalate)(PLLA/PBST) blend with enhanced melt strength and excellent low temperature toughness and strength was prepared by melt compounding through in situ compatibilization reaction in presence of multifunctional epoxy compound(ADR).The PLLA/PBST blend was an immiscible system,and the compatibility of the PLLA/PBST blend was improved after adding *** and GPC curves confirmed the formation of the PLLA-g-PBST copolymer,which improved the interfacial bonding of the blend and therefore the PLLA/PBST/ADR blend showed excellent melt strength and mechanical *** the PLLA/PBST/ADR blend with 70/30 PLLA/PBST content,the complex viscosity increased significantly with increasing ADR ***,the tensile strength,elongation at break and impact strength all increased obviously with increasing the ADR *** elongation at break of the blend reached the maximum value of 392.7%,which was 93.2 times that of neat *** the impact strength of the blend reached the maximum value of 74.7 k J/m~2,which was 21.3 times that of neat ***,the PLLA/PBST/ADR blend exhibited excellent lowtemperature toughness and *** –20 ℃,the elongation at break of the PLLA/PBST/ADR blend was as high as 93.2%,and the impact strength reached 18.8 k J/m~***,the tensile strength of the blend at low temperature was also high(64.7 MPa),which was beneficial to the application of PLA in the low temperature *** addition,the PLLA/PBST/ADR blend maintaind good biodegradability,which was of great significance to the wide application of PLLA.

Rhinovirus(RV)is a common pathogen that causes respiratory tract infection and can cause outbreaks in hos-pitals and welfare institutions.A cluster of respiratory diseases occurred in a primary school in Shenyang City,Liaoning Province,China,in *** this outbreak,a total of 31 students had symptoms similar to those of upper respiratory tract infection,mainly cough and sore *** them,27 throat swabs were collected and identified for respiratory pathogens by TaqMan low-density array(TLDA),quantitative real-time poly-merase chain reaction(PCR),reverse transcription-nested PCR and whole-genome *** of the 27 specimens,24 tested positive for RV,and 21 RV viral protein 1 sequences were obtained,of which 15(71.43%)were identified as RV-A49,while 2 RV-A20 and 4 sequences from 2 specimens were RV-A30 coin-fected with *** addition,one whole-genome sequence(WGS)of RV-A49 was obtained,and three unique amino acid mutations were found compared to 23 WGS of RV-A49 from *** conclusion,this outbreak of upper respiratory tract infection is caused by RV,mainly RV-A49.

Effect of water loss during curing on hydration reaction and hydrates conversion in calcium aluminate cement-bonded castables

Free water available in calcium aluminate cement(CAC)-bonded castables is crucial for the hydration of CAC and the conversion of hydration products in the curing and drying processes,as both the hydration and conversion reactions are dissolution–precipitation *** elucidate the effect of different levels of free water loss upon the hydration of CAC,the conversion of hydration products and the mechanical strength of the CAC-bonded castables,the CAC-bonded castables were subjected to sealed and unsealed curing conditions at 50℃ and drying at 110℃.The results demonstrate that the fast removal of free water during unsealed curing would hinder the conversion from 2CaO·Al_(2)O_(3)·8H_(2)O to 3CaO·Al_(2)O_(3)·6H_(2)O and consequently prevent the deterioration of *** a comparison,although sealed-cured samples have less water loss and high degree of hydration of CAC,they still show lower strength than the unsealed samples after *** following drying process further accelerates the hydration of residual calcium aluminate clinkers for both the sealed and unsealed samples,but still does not favor the conversion from 2CaO·Al_(2)O_(3)·8H_(2)O to 3CaO·Al_(2)O_(3)·6H_(2)O in the unsealed-cured samples.

Ulcerative colitis(UC)is a chronic inflammatory bowel disease characterized by persistent inflammation of the colon and disrupted intestinal *** mori(Sangzhi)alkaloids(SZ-A),derived from twigs of mulberry,were approved by the National Medical Products Administration in 2020 for treating type 2 diabetes *** evidence has confirmed that SZ-A also alleviates non-alcoholic fatty liver disease and ameliorates inflammation,indicating its potential to address inflammation in ***,the treatment of UC faces challenges due to low drug delivery efficiency and short retention *** overcome these challenges,an injectable and adherent in-situ thermo-sensitive hydrogel containing SZ-A was developed for rectal drug delivery,utilizing the thermo-sensitive polymers Poloxamer 407and *** thermo-sensitive hydrogel system was designed with a moderate gelation temperature of 32±0.5℃,a short gelation time of 64 s,a p H range of 7-10,high moisturizing capability exceeding 90%,and moderate mechanical strength of 4-5 *** a rat model with UC,the in situ thermo-sensitive hydrogel significantly extended the retention time at the colonic site and enabled sustained release after rectal *** of UC were markedly reduced following rectal administration of SZ-A thermosensitive ***,the release of inflammatory factors,such as interleukin-1β(IL-1β),IL-6,IL-18,tumor necrosis factor-α(TNF-α),and transforming growth factor-β1(TGF-β1),significantly decreased in the SZ-A thermo-sensitive hydrogel *** integrity of the colonic mucosal barrier was significantly enhanced following the application of SZ-A thermo-sensitive *** conclusion,rectal administration of SZ-A in situ thermo-sensitive hydrogel effectively alleviated UC symptoms,inhibited the secretion of inflammatory factors,and promoted the repair of the colonic mucosal *** approach holds promise as a potential treatment for UC.

Branch number is an important agronomic trait in peach(Prunus persica)trees because plant architecture affects fruit yield and *** breeders can select varieties with different tree architecture,the biological mechanisms underlying architecture remain largely *** this study,a pillar peach(‘Zhaoshouhong’)and a standard peach(‘Okubo’)were compared.‘Zhaoshouhong’was found to have significantly fewer secondary branches than‘Okubo’.Treatment with the synthetic strigolactone(SL)GR24 decreased branch *** analysis indicated that PpTCP18(a homologous gene of Arabidopsis thaliana BRC1)expression was negatively correlated with strigolactone synthesis gene expression,indicating that PpTCP18 may play an important role in peach *** one-hybrid,electrophoretic mobility shift,dual-luciferase assays and PpTCP18-knockdown in peach leaf buds indicated that PpTCP18 could increase expression of PpLBO1,PpMAX1,and ***,transgenic Arabidopsis plants overexpressing PpTCP18 clearly exhibited reduced primary rosette-leaf ***,lncRNA sequencing and transient expression analysis revealed that lncRNA5 targeted PpTCP18,significantly increasing PpTCP18 *** results provide insights into the mRNA and lncRNA network in the peach SL signaling pathway and indicate that PpTCP18,a transcription factor downstream of SL signaling,is involved in positive feedback regulation of SL *** role of PpTCP18 may represent a novel mechanism in peach branching *** study improves current understanding of the mechanisms underlying peach branching and provides theoretical support for genetic improvement of peach tree architecture.
