Oral everolimus inhibits intimal proliferation in injured carotid artery in rats

Background Everolimus, a derivative of sirolimus, is a potent immunosuppressant that has important anti-proliferative properties. In the present study, we demonstrated the inhibiting neointimal hyperplasia in injured carotid arteries in rats by using two different doses of everolimus administrated via the oral route for a long time. Methods A rat model of carotid artery injury was established by balloon inflation. Eighty rats were randomly divided into the sham-operated group （n=20）, injury group （n=20）, low dosage of everolimus group （n=20）, and high dosage of everolimus group （n=20）. The low close of everolimus （1.5 mg/kg） was given one day before injuring the carotid artery by balloon, followed by 0.75 mg/kg per day for 28 days via intragastric gavage. High dose everolimus （2.5 mg/kg） was given one day before injuring the carotid artery by balloon, followed by 1 mg/kg per day for 28 days. Expression of eukaryotic translation initiation factor 4E （elF-4E） and phosphorylation of ribosomal proteinS6 kinase 1 （P70S6K） were determined by reverse transcription-polymerase chain reaction and Western blotting analysis. Results In the injured carotid artery, neointimal hyperplasia was normally observed four weeks after injury. Everolimus inhibited neointimal hyperplasia after balloon injury in a dose dependent manner. At the same time, the study demonstrated that everolimus reduced the expression of P-P70S6K, elF-4E, transforming growth factor （TGF）-131 and of proliferating cell nuclear antigen （PCnA）. Conclusions Everolimus significantly inhibited neointimal hyperplasia of the injured carotid artery. The effect depended on dosaqe and was associated with the reduction of phosphorylation of P70S6K and the elF-4E expression level.

Introduction To improve compliance with voiding diaries in children with primary monosymptomatic nocturnal enuresis(PMnE),a new modified 3-day weekend frequency-volume chart(FVC)was designed,and the compliance and validity of this modified FVC was evaluated by comparing with the International Children's Continence Society(ICCS)recommended voiding *** A total of 1200 patients with PMnE were enrolled in the study from 13 centers in China and were randomly assigned to record this modified FVC or the ICCS-recommended voiding *** primary outcome measure was the compliance,assessed by comparing the completing index and the quality score of diaries between two *** secondary outcome measure was the validity,evaluated by comparing the constituent of subtypes,micturition parameters and response rate to *** Among the 1200 participants enrolled in the study,447 patients completed the ICCS-recommended voiding diary and 469 completed the modified *** diurnal completing index and the quality score of the modified FVC group were better than those of the ICCS *** addition,there was no significant difference between these two groups in the subtype classification,or in the response rate to *** The modified FVC could be applied to obtain the voiding characteristics of children with PMnE as the ICCS-recommended voiding diary does and offers a reasonable and better choice for children with PMnE from the unselected population in the future.

Background In China,lack of evidence remains a significant challenge for the national initiative to promote physical activity(PA).We aimed to quantify the beneficial effects of meeting or maintaining the recommended PA level[150 minutes per week(min/wk)of moderate PA or 75 min/wk of vigorous PA or an equivalent combination]on incident cardiovascular disease(CVD)among Chinese *** We included 100,560 participants without history of CVD from three cohorts in the Prediction for Atherosclerotic Cardiovascular Disease risk in China(China-PAR)*** proportional hazards models were used to estimate hazard ratios(HRs)and 95%confidence intervals(CIs)for CVD events and its subtypes,including stroke,coronary heart disease,heart failure,and CVD *** During a median follow-up of 7.3 years(range:6–15 years),777,163 person-years and 4693 incident CVD events were *** with participants who were inactive at baseline,the multivariable adjusted HR(95%CI)of developing CVD was 0.74(0.69–0.79)for those who met recommended moderate to vigorous physical activity(MVPA)level at ***,the risk of CVD incidence was reduced with increment of MVPA(Ptrend1),and the HR(95%CI)of highly-active versus inactive category was 0.62(0.56–0.68).Compared with individuals who were inactive both at the baseline and follow-up,those keeping active over the period of follow-up had a substantial lower risk of incident CVD with the HR(95%CI)of 0.57(0.43–0.77).Conclusions The findings demonstrated that meeting and maintaining the recommended MVPA level could reduce the cardiovascular *** adoption of the PA recommendations would have considerable health impacts to the Chinese population.

It's a challenge for detecting the therapeutic targets of a polypharmacological drug from variations in the responsed networks in thedfferentiated populations with complex diseases,as stable coronary heart ***,in an adaptive,31-center,randomized,double-blind trial invoving 920 patients with moderate symptomatic stable angina treated by 14-day Danhong injection(DHI),a kind of polyphamacological drug with high quality control,or placebo(0.9%saline),with 76-day following-up,we firstly confrmed that DHl couldincrease the proportion of patients with clinically significant changes on angina-frequency assessed by Seattle Angina Questionnaire(ASAQ-AF220)(12.78%at Day 30,95%confidence interval[C]5.86-19.71%,P=0.0003,13.82%at Day 6C0,95%CI 6.82-20.82%,P=0.0001and 8.95%at Day 90,95%CI 2.06-15.85%,P=0.01).We also found that there were no significant differences in new-onset major vascularevents(P=0.8502)and serious adverse events(P=0.9105)between DHl and *** performing the RnA sequencing in 62 selectedpatients,we developed a systemic modular approach tp identfy differentilly expressed modules(deMs)of DHI with the Z_(summay)valueless than 0 compared with the control group,calculated by weighted gene co-expression network analysis(WGCnA),and sketched out thebasic framework on a modular map with 25 functional modules targeted by ***,the effective therapeutic module(ETM),defined as the highest corelation value with the phenotype alteration(SAQ-AF,the change in SAQ-AF at Day 30 from baseline)calculatedby WGCnA,was identifed in the population with the best effect(ASAQ-AF240),which is related to anticoagulation and regulation ofcholesterol *** assessed the modular flexbility of this ETM using the global topological D value based on Euclidean distance,which is corelated with phenotype alteration(r^(2):0.8204,P=0.019)by linear *** study identified the ant-angina therapeuticmodule in the effective population treated by the multi-target

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SnR） ≥ 10. All data with SnR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

Intrahepatic cholangiocarcinoma(iCCA)can originate from the large bile duct group(segment bile ducts and area bile ducts),small bile duct group(septal bile ducts and interlobular bile ducts),and terminal bile duct group(bile ductules and canals of Hering)of the intrahepatic biliary tree,which can be histopathological corresponding to large duct type iCCA,small duct type iCCA and iCCA with ductal plate malformation pattern,and cholangiolocarcinoma,*** challenge in pathological diagnosis of above subtypes of iCCA falls in the distinction of cellular morphologies,tissue structures,growth patterns,invasive behaviors,immunophenotypes,molecular mutations,and surgical *** these reasons,this expert consensus provides nine recommendations as a reference for standardizing and refining the diagnosis of pathological subtypes of iCCA,mainly based on the 5th edition of the World Health Organization Classification of Tumours of the Digestive System.

目的了解2015-2021年中国不同地区临床分离葡萄球菌属细菌的分布及耐药变迁。方法按CHInET中国细菌耐药监测网统一的技术方案对临床分离的葡萄球菌采用纸片法和商品化药敏试验测试仪(机器法)进行药物敏感性试验;收集2015-2021年CHInET的葡萄球菌耐药监测数据,使用WHOnET 5.6软件统计并按2021年CLSI折点标准判读结果。结果2015-2021年共分离非重复葡萄球菌属细菌204771株,其中金黄色葡萄球菌136731株(66.8%),凝固酶阴性葡萄球菌68040株(33.2%),7年间两者检出率均无明显变化。金黄色葡萄球菌主要分离自呼吸道标本(38.9±5.1)%、伤口脓液分泌物(33.6±4.2)%、血液(11.9±1.5)%;凝固酶阴性葡萄球菌主要分离自血液(73.6±4.2)%、脑脊液(12.1±2.5)%、胸腹水(8.4±2.1)%。金黄色葡萄球菌的科室来源主要为ICU(17.0±7.3)%、门急诊(11.6±1.7)%、外科(11.2±0.9)%;凝固酶阴性葡萄球菌的科室来源主要为ICU(32.2±9.7)%、门急诊(12.8±4.7)%、内科(11.2±1.9)%。金黄色葡萄球菌、凝固酶阴性葡萄球菌中甲氧西林耐药株的总体检出率分别为32.9%、74.1%,7年间MRSA检出率从42.1%下降至29.2%,MRCnS检出率从82.1%下降至68.2%。除甲氧苄啶-磺胺甲噁唑外,MRSA对各类抗菌药物的耐药率均高于MSSA。MRSA对庆大霉素、利福平、左氧氟沙星耐药率有所下降;MRCnS对庆大霉素、红霉素、利福平、甲氧苄啶-磺胺甲噁唑的耐药率有所下降,对左氧氟沙星的耐药率有所上升。未发现对万古霉素耐药的葡萄球菌菌株。对利奈唑胺耐药的MRCnS有所增多,7年间耐药率从0.2%上升至2.3%。结论葡萄球菌属细菌仍是革兰阳性菌中最主要的细菌。MRSA和MRCnS亦仍然是革兰阳性菌中最主要的耐药细菌。至今未发现对万古霉素和利奈唑胺耐药的金黄色葡萄球菌,但已有利奈唑胺耐药的MRCnS菌株检出,应加强监测和关注。
