Cold plasma promotes Sertoli cell proliferation via AMPK–mTOR signaling pathway

This study investigated cold plasmas for multiple biological applications. Our previous work has found dielectric barrier discharge plasma improves chicken sperm quality. The number of Sertoli cells(SCs) decides spermatogenesis. However, whether cold plasma can regulate SC proliferation remains unclear. This study explored the effects of cold plasma on immature chicken SC proliferation and the regulation mechanism. Results showed that cold plasma exposure at 2.4 W for 30 s twice with an interval of 6 h produced(P<0.05) the maximum SC viability, cell growth, and cell cycle progression. SC proliferation-promoting effect of cold plasma treatment was regulated by increasing(P<0.05) the adenosine triphosphate production and the respiratory enzyme activity in the mitochondria. This process was potentially mediated by the adenosine monophosphate-activated protein kinase(AMPK)–mammalian target of rapamycin(m TOR) signaling pathway, which was regulated by the micro RNA(mi RNA) targeting regulation directly and by the intracellular reactive oxygen species homeostasis indirectly. The cold plasma treatment increased(P<0.01) the mi R-7450-5 p expression and led to a decreased(P<0.01) AMPKα1 level. On the other hand, mi R-100-5 p expression was reduced(P<0.05) and led to an increased(P<0.05) m TOR level in SCs. A single-stranded synthetic mi R-7450-5 p antagomir and a double-stranded synthetic mi R-100-5 p agomir reduced(P<0.05) the SC proliferation. However, this could be ameliorated(P<0.05) by the cold plasma treatment. Our findings suggest that appropriate cold plasma treatment provides a safe strategy to improve SC proliferation, which is beneficial to elevating male chicken reproductive capacity.

17β-Estradiol Regulates SKP2 Expression in Cultured Immature Boar Sertoli Cells Mainly via Estrogen Receptor β,cAMP-PKA and ERK1/2

Estrogen plays an important role in regulating testicular Sertoli cell number. Furthermore, S-phase kinase-associated protein 2 （SKP2） plays a central role in mammalian cell cycle progression. The objective of this study was to determine whether 17β-estradiol can regulate the expression of SKP2, and the Sertoli cell cycle, via estrogen receptor β （ERβ）, the cyclic adenosine monophosphate （cAMP）-protein kinase A （PKA） and extracellular signal-regulated kinase （ERK1/2） pathway. When cultured immature boar Sertoli cells were treated with 17β-estradiol, a time-dependent increase in SKP2 mRNA and protein level was observed by real-time PCR and Western blot, and 17β-estradiol activity peaked at 30 min. Treatment with ICI182780 and ERβ antagonist reduced 17β-estradiol-induced expression of SKP2 and proliferating cell nuclear antigen （PCNA）, while increasing the protein concentration of p27kip1. However, the effect of ERa antagonist on these parameters was lower than that of ICI 182780 and ERβ. Forskolin had a similar effect as 17β-estradiol on the expression of SKP2, PCNA and p27kip1, Rp-cAMP, H-89 and U0126 treatment reduced 17β-estradiol-induced changes, while H-89 also inhibited ERK1/2 activation. Therefore, 17β-estradiol mainly regulates SKP2 mRNA and protein expression via ERβ-cAMP-PKA and ERK1/2 activation. SKP2 and PCNA expression were positively correlated, while increased SKP2 expression likely resulted in p27kip1 degradation.

Fate of Fertilizer Nitrogen Applied to Crops Grown on Fluvo-Aquic Soil in Huang-Huai-Hai Plain

The fate of urea-and ammonium bicarbonate(ABC)-nitrogen (N) applied by prevailing traditional techniques to winter wheat (Triticum aestivum L.)or maize (Zea mays L.)grown in the fields of Fluvo-aquic soil was investigated using ^15N tracer-micro-plot technique.Results show that:(1) at maturity of wheat,N recovery in plants and N losses of urea and ABC applied at seeding in autumn were 31-39%,and 34-46%,respextively,while the corresponding figures for side-banding at 10 cm depth in early spring were 51-57%,and 5-12%;surface-broadcast of urea followed by irrigation at early spring was as efficient as the side-banding in improving N recovery in plants and reducing N loss,however,such techuique was found less satisfactory with ABC.(2)At the maturity of maize,N recovery in the plants and N loss of urea and ABC sidebanded at seedling stage or prior to tasseling ranged from 23% to 57%,and 9% to 26%,respectively.(3) Either in Wheat or in maize experiment,the majority of residual fertilizer N in soil profile (0-60cm) was in the form of biologically immobilized organic N,however,the contribution of ammonium fixation by clay minerals increased markedly nwith depth in soil profile.(4) Though the proportion of residual fertilizer N was generally highest in the top 20 cm soil layer,considerable reaidual N (nostly 6-11% of the N applied)was found in 60-100 cm soil layers.

The development of efficient perovskite light-emitting diodes(PeLEDs)relies strongly on the fabrication of perovskite films with rationally designed structures(grain size,composition,surface,etc.).Therefore,an understanding of structure-performance relationships is of vital importance for developing high-performance perovskite devices,particularly for devices with in-situ fabricated perovskite nanocrystal films.In this study,we reveal the vertical structure of an in-situ fabricated quasi-two-dimensional perovskite film.By combining time-of-flight secondary ion mass spectrometry,energy dispersive spectroscopy,grazing incidence wide-angle X-ray scattering(GIWAXS),and low-temperature photoluminescence spectra,we illustrate that the resulting in-situ fabricated DPPA_(2)Cs_(n-1)Pb_(n)(Br_(0.3)I_(0.7))_(3n+1)(DPPA^(+):3,3-diphenylpropylammonium)film has a gradient structure with a very thin layer of ligands on the surface,predominantly small-n domains at the top,and predominantly large-n domains at the bottom owing to the solubility difference of the precursors.In addition,GIWAXS measurements show that the domain of n=2 on the top layer has an ordered in-plane alignment.Based on the understanding of the film structure,we developed an in-situ fabrication process with ligand exchange to achieve efficient pure red PeLEDs at 638 nm with an average external quantum efficiency(EQE)of 7.4%.The optimized device had a maximum luminance of 623 cd/m^(2) with a peak EQE of 9.7%.

Metal–organic framework-derived Co Se2@N-doped carbon nanocubes for high-performance lithium-ion capacitors

Cobalt selenide （CoSe2） has garnered considerable attention as a prospective anode candidate for advanced lithium-ion storage,prompting comprehensive investigations.However,CoSe2-based anodes usually suffer from significant volume variation upon lithiation leading to unsatisfactory cycling stability.Herein,a versatile synthesis route is proposed for the in-situ fabrication of CoSe2nanoparticles embedded in N-dope carbon skeleton （CoSe2@NC） through annealing treatment and selenization of a metal–organic framework-derived （MOF）precursor.The N-doped carbon derived from the MOF serves not only as an excellent conductive substrate but also as a confined reactor,effectively inhibiting the structural instability and alleviating the inevitable volume change of CoSe2.Owing to their unique nanostructure,the as-prepared CoSe2@NC exhibits a high capacity of 745.9mAh·g-1at 0.1 A·g-1,while maintaining excellent rate capability and an impressive lifespan.Furthermore,the assembled lithium-ion capacitor （LIC） based on CoSe2@NC demonstrates an energy density of 130Wh·kg-1,a power density of 24.6 kW·kg-1,and remarkable capacity retention of 90.8%after 8000 cycles.These results highlight the great potential of CoSe2@NC for practical applications.

Reaction mechanism between Al_(2)O_(3)–MgO refractory materials and rare earth high-carbon heavy rail steel

Submerged entry nozzle(SEN)clogging is a major problem affecting the production quality of rare earth steel,and finding a suitable refractory outlet can significantly reduce production costs.To explore the relationship between refractory composition and interface interaction,unprotected coated Al_(2)O_(3)–MgO refractories and SiO2-coated Al_(2)O_(3)–MgO refractories were added to rare earth high-carbon heavy rail steel under laboratory conditions,and the Al_(2)O_(3)–MgO refractory was found to be more suitable.The results show that,from the epoxy resin side to the refractory side,the contour of the refractory interface reaction layer can be divided into two main layers:an iron-rich reaction layer and an iron-poor reaction layer.Calculations based on the spherical model suggest that the adhesion force is proportional to the size of the refractory particles and inclusions,and the same result applies to the surface tension.Controlling the inclusions at a smaller size has a specific effect on alleviating the erosion of refractories.Combined with the erosion mechanism of Al_(2)O_(3)–MgO refractories,the interface reaction mechanism between Al_(2)O_(3)–MgO refractories and molten steel was proposed,which provides ideas for solving SEN clogging.

Track correlation algorithm based on CNN-LSTM for swarm targets

The rapid development of unmanned aerial vehicle(UAV) swarm, a new type of aerial threat target, has brought great pressure to the air defense early warning system. At present, most of the track correlation algorithms only use part of the target location, speed, and other information for correlation.In this paper, the artificial neural network method is used to establish the corresponding intelligent track correlation model and method according to the characteristics of swarm targets.Precisely, a route correlation method based on convolutional neural networks (CNN) and long short-term memory (LSTM)Neural network is designed. In this model, the CNN is used to extract the formation characteristics of UAV swarm and the spatial position characteristics of single UAV track in the formation,while the LSTM is used to extract the time characteristics of UAV swarm. Experimental results show that compared with the traditional algorithms, the algorithm based on CNN-LSTM neural network can make full use of multiple feature information of the target, and has better robustness and accuracy for swarm targets.

Tuning Surface Molecular Design of Porous Carbon for Blue Energy Harvesting

Capacitive mixing is a promising blue energy technology due to its membrane-free electricity generation and long electrode life cycle.However,because of limited performance,existing systems do not lend themselves to practical implementation.Although it is a crucial factor directly influencing electrode behavior,surface chemistry has largely been overlooked in capacitive mixing.Here,we show that manipulating surface functionalization alone can tune the responses of electrodes to produce a high voltage rise without altering the pore structure of the electrodes.Our findings reveal that the spontaneous electrode potential of a surface-modified carbon electrode shifts negatively proportional to the surface charge due to the surface groups,which explains why and how manipulating the surface chemistry can improve the power generation capacity.

The role of acute normovolemic hemodilution in reducing allogeneic blood transfusion in glioblastoma surgery:a case-control study

Background Acute normovolemic hemodilution(ANH)was first introduced in glioblastoma surgery,and its role in reducing allogeneic blood transfusion was investigated in this study.Methods This study enrolled supratentorial glioblastoma patients who received total resection.In the ANH group,the patients were required to draw blood before the operation,and the blood will be transfused back to the patient during the operation.The association between ANH and clinical features was investigated.Results Sixty supratentorial glioblastoma patients were enrolled in this study,25 patients were allocated in the ANH group,and another 35 patients were included in the control group.ANH dramatically reduced the need for allogeneic blood transfusion(3[12%]vs 12[34.3%],P=0.049),and the blood transfusion per total of patients was dramatically decreased by the application of ANH(0.40±1.15 units vs 1.06±1.59 units,P=0.069).Furthermore,ANH also markedly reduced the requirement of fresh frozen plasma(FFP)transfusion(2[8%]vs 11[31.4%],P=0.030)and the volume of FFP transfusion per total of patients(32.00±114.46 mL vs 115.71±181.00 mL,P=0.033).The complication rate was similar between the two groups.Conclusions ANH was a safe and effective blood conservation technique in glioblastoma surgery.
