Efficacy of substances containing 3 types of active ingredients-saponins, flavones, and alkaloids in regulation of cytokines in autoimmune diseases a systematic review and Meta-analysis based on animal studies

OBJECTIVE: To investigate the efficacy of substances containing 3 types of active ingredients-saponins,flavones, and alkaloids on experimental animals with autoimmune diseases(AIDs).METHODS: The protocol for this systematic review and Meta-analysis was prospectively registered with PROSPERO(CRD42023395741). Searches were conducted in the China National Knowledge Infrastructure, Wanfang, Chinese Science and Technology Journals, China Biomedical, Pub Med,Cochrane Library, and Embase databases to screen for animal studies investigating the therapeutic effects of saponins, flavones, or alkaloids on autoimmune diseases;consequently, corresponding data extraction tables were prepared. Systematic Review Centre for Laboratory Animal Experimentation was used to assess the risk of methodological bias in the included literature. RevMan 5.4 was used for the Meta-analysis on the 8 serum cytokines.RESULTS: A total of 31 studies were included, all of which were randomized controlled studies. Meta-analysis indicated that substances rich in saponins, flavones, and alkaloids reduced serum levels of interleukin(IL)-1β[standardized mean difference(SMD) =-1.94, 95%confidence interval(CI)(-2.99,-0.90), P = 0.0003], IL-6 [SMD =-1.65, 95% CI(-2.33,-0.97,) P < 0.000 01],IL-17 [SMD =-2.41, 95% CI(-3.61,-1.20), P <0.0001], tumor necrosis factor(TNF)-α [SMD =-1.84, 95%CI(-2.61,-1.06), P < 0.0001], and interferon(IFN)-γ[SMD =-1.54, 95% CI(-2.43,-0.65), P = 0.0007], but increased serum levels of IL-4 [SMD = 1.30, 95% CI(0.15,2.44), P = 0.03) and IL-10 [SMD = 2.05, 95% CI(1.39,2.70), P < 0.000 01) in animal models. However, no significant regulatory effect of these three active components was observed on serum levels of IL-2 [SMD =-0.63, 95% CI(-1.82, 0.57), P = 0.30].CONCLUTIONS: Substances containing saponins,flavones, and alkaloids regulated the changes of immunerelated cytokines, it may be a novel dietary substance to relieve and control autoimmune diseases in the future.

Sound insulation measurement of building components combining diffuse acoustic field excitation and near-field acoustic holography reconstruction

A method for measuring the airborne sound insulation of building components combining diffuse acoustic field(DAF) excitation and near-field acoustic holography(NAH)radiation intensity reconstruction is proposed. In this method, the DAF is first used to excite the vibration of the component and obtain the incident sound power, then the normal sound intensity distribution with high spatial resolution on the surface of the component is reconstructed from the radiation sound pressure field using the NAH, and finally, according to the sound intensity distribution, the radiated sound power is calculated and the radiation heat areas are located, so as to realize the measurement of sound insulation and sound insulation defects of components. The experimental research in a sound insulation room shows that under the condition that the test distance and sampling interval are both0.04 m, the error of the sound insulation measured by this method compared to the sound pressure method is less than 3.3 d B in the 100–5000 Hz frequency band and less than1.3 d B in the 250–3150 Hz frequency band, and the positioning accuracy of round holes(diameter: 8 mm) and rectangular slots(length: 80 mm, width: 3 mm) is up to the centimeter level. At the same time, the method has strong stability under the influence of certain amount of reverberation and background noise. When reverberation time in the receiving room increases from 1.0 s to 3.4 s(step size: 0.6 s), and the signal-to-noise ratio decreases from 10 d B to 0 d B(step size: 5 d B), the sound volume measurement errors are within 0.8 d B and 0.3 d B, and the defect location errors are within 0.037 m and 0.035 m.The proposed method helps to improve the measurement capability of sound insulation characteristics of building components in the laboratory, and it is robust to the receiving room test environment.

The early life immune dynamics and cellular drivers at single-cell resolution in lamb forestomachs and abomasum

Background Four-chambered stomach including the forestomachs(rumen,reticulum,and omasum)and abomasum allows ruminants convert plant fiber into high-quality animal products.The early development of this four-chambered stomach is crucial for the health and well-being of young ruminants,especially the immune development.However,the dynamics of immune development are poorly understood.Results We investigated the early gene expression patterns across the four-chambered stomach in Hu sheep,at 5,10,15,and 25 days of age.We found that forestomachs share similar gene expression patterns,all four stomachs underwent widespread activation of both innate and adaptive immune responses from d 5 to 25,whereas the metabolic function were significantly downregulated with age.We constructed a cell landscape of the four-chambered stomach using single-cell sequencing.Integrating transcriptomic and single-cell transcriptomic analyses revealed that the immune-associated module hub genes were highly expressed in T cells,monocytes and macrophages,as well as the defense-associated module hub genes were highly expressed in endothelial cells in the four-stomach tissues.Moreover,the non-immune cells such as epithelial cells play key roles in immune maturation.Cell communication analysis predicted that in addition to immune cells,non-immune cells recruit immune cells through macrophage migration inhibitory factor signaling in the forestomachs.Conclusions Our results demonstrate that the immune and defense responses of four stomachs are quickly developing with age in lamb's early life.We also identified the gene expression patterns and functional cells associated with immune development.Additionally,we identified some key receptors and signaling involved in immune regulation.These results help to understand the early life immune development at single-cell resolution,which has implications to develop nutritional manipulation and health management strategies based on specific targets including key receptors and signaling pathways.

Tau Accumulation in the Spinal Cord Contributes to Chronic Inflammatory Pain by Upregulation of IL-1βand BDNF

Microtubule-associated protein Tau is responsible for the stabilization of neuronal microtubules under normal physiological conditions.Much attention has been focused on Tau’s contribution to cognition,but little research has explored its role in emotions such as pain,anxiety,and depression.In the current study,we found a significant increase in the levels of p-Tau(Thr231),total Tau,IL-1β,and brain-derived neurotrophic factor(BDNF)on day 7 after complete Freund's adjuvant(CFA)injection;they were present in the vast majority of neurons in the spinal dorsal horn.Microinjection of Mapt-shRNA recombinant adeno-associated virus into the spinal dorsal cord alleviated CFA-induced inflammatory pain and inhibited CFA-induced IL-1βand BDNF upregulation.Importantly,Tau overexpression was sufficient to induce hyperalgesia by increasing the expression of IL-1βand BDNF.Furthermore,the activation of glycogen synthase kinase 3 beta partly contributed to Tau accumulation.These findings suggest that Tau in the dorsal horn could be a promising target for chronic inflammatory pain therapy.

Identification of fractional order Hammerstein models based on mixed signals

An algorithm based on mixed signals is proposed,to solve the issues of low accuracy of identification algorithm,immeasurable intermediate variables of fractional order Hammerstein model,and how to determine the magnitude of fractional order.In this paper,a special mixed input signal is designed to separate the nonlinear and linear parts of the fractional order Hammerstein model so that each part can be identified independently.The nonlinear part is fitted by the neural fuzzy network model,which avoids the limitation of polynomial fitting and broadens the application range of nonlinear models.In addition,the multi-innovation Levenberg-Marquardt(MILM)algorithm and auxiliary recursive least square algorithm are innovatively integrated into the parameter identification algorithm of the fractional order Hammerstein model to obtain more accurate identification results.A simulation example is given to verify the accuracy and effectiveness of the proposed method.

Comprehensive Exergoeconomic Analysis and Optimization of a Novel Zero-Carbon-Emission Multi-Generation System based on Carbon Dioxide Cycle

This paper aims to conduct a comprehensive exergoeconomic analysis of a novel zero-carbon-emission multi-generation system and propose a fast optimization method combined with machine learning.The detailed exergoeconomic analysis of a novel combined power,freshwater and cooling multi-generation system is performed in this study.The exergoeconomic analysis model is established by exergy flow theory.A comprehensive exergy,exergoeconomic and environmental analysis is carried out.Five critical decision variables are researched to bring out effects on the multi-generation system exergoeconomic performance.A novel fast optimization method combining genetic algorithm and Bagging neural network is proposed.The advanced nature comparison is made between the proposed system and four similar cases.Results display that increasing the turbine inlet temperature can improve exergy efficiency and decrease the total product unit cost.The multi-generation system exergy destruction directly determines exergy efficiency and total exergy destruction cost rate.The total product unit cost in the cost optimal design case is reduced by 7.7%and 25%,respectively,compared with exergy efficiency optimal design case and basic design case.Compared with four similar cases,the proposed multi-generation system has great advantages in thermodynamic performance and exergoeconomic performance.This paper can provide research methods and ideas for performance analysis and fast optimization of multi-generation system.

Spatiotemporal mismatch of land use functions and land use efficiencies and their influencing factors:A case study in the Middle Reaches of the Yangtze River,China

Identification of the spatial mismatch between land use functions(LUFs)and land use efficiencies(LUEs)is essential to regional land use policies.However,previous studies about LUF-LUE mismatch and its driving factors have been insufficient.In this study,we explored the spatiotemporal mismatch of LUFs and LUEs and their influencing factors from 2000 to 2018 in the Middle Reaches of the Yangtze River(MRYR).Specifically,we used Spearman correlation analysis to reveal the trade-off relationship between LUFs and LUEs and determine the direction of the influencing factors on the LUF-LUE mismatch,adopted spatial mismatch analysis to measure the imbalance between LUFs and LUEs,and used the geographical detector model to analyze the factors influencing this spatial mismatch.The results showed that production function(PDF),living function(LVF),ecological function(ELF),agricultural production efficiency(APE),urban construction efficiency(UCE),and ecological services efficiency(ESE)all displayed significant spatial heterogeneity.The high trade-off areas were widely distributed and long-lasting in agricultural space and urban space,while gradually decreasing in ecological space.Wuhan and Changsha showed high spatial mismatch coefficients in urban space,but low spatial mismatch coefficients in agricultural space.Hunan generally presented high spatial mismatch coefficients in ecological space.Furthermore,the interaction of the proportion of cultivated area and transportation accessibility exacerbated the mismatch in agricultural space.The interaction effects of capital investment and technology innovation with other factors have the most intense impact on the mismatch in urban space.The internal factor for cultivated area interacts with other external factors to drastically affect ecological spatial mismatch.

Performance Design of High-Temperature Chloride Salts as Thermal Energy Storage Material

The chloride salts have great potential used as high-temperature thermal energy storage(TES) medium for the concentrated solar power system.In this study,LiCl,KCl and CaCl_(2) were selected as energy storage materials in order to further broaden the working temperature of ternary chloride salt and improve its energy storage density.The new high-temperature energy storage ternary chloride composed of LiCl,KCl,and CaCl_(2) was developed based on the phase diagram generated by FactSage.Three components of LiCl-KCl-CaCl_(2) with the mass ratio of 37.85%-53.38%-8.77%,30.90%-13.82%-55.28% and 1.78%-18.61%-79.61% were developed,of which the corresponding melting temperature(T_(m)) 340.93,433.57 and 626.85℃,respectively,was obtained either.Considering that T_(m) of the third group of salts was too high,only the first two groups of salts were tested.DSC test showed that the actual melting point was only 0.46% and 1.64% different from the melting point predicted by Factsage.The thermal properties of the two ternary chloride salts were also compared.The solid and liquid-specific heat of ternary salts was determined by DSC using sapphire as the standard reference.The vapor pressure and decomposition temperature of ternary chloride salts were investigated.The results showed that the vapor pressure of salt 1 was almost constant below 650℃ by FactSage.Meanwhile,the TG results showed that the upper working temperature of salt 1 was 650℃ under the air atmosphere.In addition,the ternary chloride salts after short-term cycling still exhibited excellent thermal properties,which revealed that these good thermal properties make them have broad application prospects in high-temperature thermal energy storage systems.

Pore-pressure and stress-coupled creep behavior in deep coal:Insights from real-time NMR analysis

Understanding the variations in microscopic pore-fracture structures(MPFS) during coal creep under pore pressure and stress coupling is crucial for coal mining and effective gas treatment. In this manuscript, a triaxial creep test on deep coal at various pore pressures using a test system that combines in-situ mechanical loading with real-time nuclear magnetic resonance(NMR) detection was conducted.Full-scale quantitative characterization, online real-time detection, and visualization of MPFS during coal creep influenced by pore pressure and stress coupling were performed using NMR and NMR imaging(NMRI) techniques. The results revealed that seepage pores and microfractures(SPM) undergo the most significant changes during coal creep, with creep failure gradually expanding from dense primary pore fractures. Pore pressure presence promotes MPFS development primarily by inhibiting SPM compression and encouraging adsorption pores(AP) to evolve into SPM. Coal enters the accelerated creep stage earlier at lower stress levels, resulting in more pronounced creep deformation. The connection between the micro and macro values was established, demonstrating that increased porosity at different pore pressures leads to a negative exponential decay of the viscosity coefficient. The Newton dashpot in the ideal viscoplastic body and the Burgers model was improved using NMR experimental results, and a creep model that considers pore pressure and stress coupling using variable-order fractional operators was developed. The model’s reasonableness was confirmed using creep experimental data. The damagestate adjustment factors ω and β were identified through a parameter sensitivity analysis to characterize the effect of pore pressure and stress coupling on the creep damage characteristics(size and degree of difficulty) of coal.

Defect Detection in c-Si Photovoltaic Modules via Transient Thermography and Deconvolution Optimization

Defects may occur in photovoltaic(PV)modules during production and long-term use,thereby threatening the safe operation of PV power stations.Transient thermography is a promising defect detection technology;however,its detection is limited by transverse thermal diffusion.This phenomenon is particularly noteworthy in the panel glasses of PV modules.A dynamic thermography testing method via transient thermography and Wiener filtering deconvolution optimization is proposed.Based on the time-varying characteristics of the point spread function,the selection rules of the first-order difference image for deconvolution are given.Samples with a broken grid and artificial cracks were tested to validate the performance of the optimization method.Compared with the feature images generated by traditional methods,the proposed method significantly improved the visual quality.Quantitative defect size detection can be realized by combining the deconvolution optimization method with adaptive threshold segmentation.For the same batch of PV products,the detection error could be controlled to within 10%.
