Ticks are external parasitic arthropods that can transmit a variety of pathogens by sucking blood.Low-temperature tolerance is essential for ticks to survive during the cold winter.Exploring the protein regulation mechanism of low-temperature tolerance of Haemaphysalis longicornis could help to explain how ticks survive in winter.In this study,the quantitative proteomics of several tissues of H.longicornis exposed to low temperature were studied by data independent acquisition technology.Totals of 3699,3422,and 1958 proteins were identified in the salivary gland,midgut,and ovary,respectively.The proteins involved in energy metabolism,cell signal transduction,protein synthesis and repair,and cytoskeleton synthesis changed under low-temperature stress.The comprehensive analysis of the protein regulation of multiple tissues of female ticks exposed to low temperature showed that maintaining cell homeostasis,maintaining cell viability,and enhancing cell tolerance were the most important means for ticks to maintain vital signs under low temperature.The expression of proteins involved in and regulating the above cell activities was the key to the survival of ticks under low temperatures.Through the analysis of a large amount of data,we found that the expression levels of arylamine N-acetyltransferase,inositol polyphosphate multikinase,and dual-specificity phosphatase were up-regulated under low temperature.We speculated that they might have important significance in low-temperature tolerance.Then,we performed RNA interference on the mRNA of these 3 proteins,and the results showed that the ability of female ticks to tolerate low temperatures decreased significantly.

污水处理行业快速发展,各种治理技术层出不穷,在很大程度上解决了环境污染问题,但也不可避免地会造成环境影响。在“双碳”背景下,为选择更高效的污水处理技术,各种评价方法相继在污水处理行业中应用。本综述总结了城镇污水处理过程中应用较为广泛的评价研究现状,主要关注基于生命周期评价的环境影响评价,其中重点关注环境富营养化、全球变暖潜力等方面的影响;经济评价中通过对成本分析可发现在处理过程中能源成本很重要,由利润分析可发现回收利用沼气可以提高利润;此外,碳足迹和有机物毒性计算也逐渐成为研究重点。基于以上内容提出了“5E”评价系统,可切实解决城镇污水处理过程多角度综合评价问题,为行业可持续发展、“双碳”目标的实现提供支撑。要点:(1)简述了污水处理厂发展前景和优势。(2)综述了环境影响评价、经济评价、碳足迹分析、有机物毒性分析和技术分析在城镇污水处理中的应用。(3)提出了“5E”评价系统,对多角度综合评价的发展前景进行了展望。

近年来,全国范围内手足口病(hand, foot, and mouth disease,HFMD)的感染率逐渐上升,其中柯萨奇病毒A组10型(Coxsackievirus A10,CV-A10)是主要病原体之一,利巴韦林是临床上治疗儿童手足口病的常用药物之一。为了评价利巴韦林在体外对CVA10的抑制效果,本研究中,我们随机选取了19条2009-2021年的不同地区和不同年代的有代表性的中国内地优势流行基因型C基因型的CVA10毒株,通过细胞毒性实验(Cell counting kit-8,CCK8)来检测利巴韦林在RD细胞上对CVA10的抑制率。将抑制率大于85%的CVA10定义为对利巴韦林敏感组,抑制率小于15%的CVA10定义为对利巴韦林耐药组。结果显示利巴韦林在0.78mmol/L浓度下可以对CVA10产生较好的抑制作用并且对细胞的毒性较低。19株CVA10中有4株表现出对利巴韦林耐药性,2株表现出对利巴韦林敏感性,剩余13株的敏感性在85%~15%之间。通过7日龄ICR乳鼠感染实验比较对利巴韦林敏感组和耐药组毒株的致病力差异,通过观察乳鼠临床评分、生存率、体重及组织取材的滴度测定结果均表明对利巴韦林敏感组CVA10在小鼠上的致病力比耐药组强。进一步使用MEGA 7.0软件对影响CVA10复制和翻译的2C区氨基酸序列进行差异性分析,以分析可能的耐药位点。结果显示对利巴韦林耐药组和敏感组CVA10的2C区的327位和328位氨基酸位点存在差异。本研究发现当前国内流行的CVA10存在广泛对利巴韦林耐药现象,其中部分CVA10对利巴韦林完全耐药。因此,有必要加强对CVA10耐药株的监测,以便更好地指导临床医生正确地选择抗病毒药物,为个体化的治疗提供理论依据。

The parahippocampal gyrus-orbitofrontal cortex(PHG-OFC)circuit in humans is homologous to the postrhinal cortex(POR)-ventral lateral orbitofrontal cortex(vlOFC)circuit in rodents.Both are associated with visuospatial malfunctions in Alzheimer’s disease(AD).However,the underlying mechanisms remain to be elucidated.In this study,we explored the relationship between an impaired POR-vlOFC circuit and visuospatial memory deficits through retrograde tracing and in vivo local field potential recordings in 5XFAD mice,and investigated alterations of the PHG-OFC circuit by multi-domain magnetic resonance imaging(MRI)in patients on the AD spectrum.We demonstrated that an impaired glutamatergic POR-vlOFC circuit resulted in deficient visuospatial memory in 5XFAD mice.Moreover,MRI measurements of the PHG-OFC circuit had an accuracy of 77.33%for the classification of amnestic mild cognitive impairment converters versus non-converters.Thus,the PHG-OFC circuit explains the neuroanatomical basis of visuospatial memory deficits in AD,thereby providing a potential predictor for AD progression and a promising interventional approach for AD.

Microbes serve as the most important resource for drug discovery.During our screening for bioactive compounds from our natural products library,a pathogenic fungus,Microdochium majus strain 99049,from wheat was selected for further investigation.A new alkaloid named brocaeloid D(1),together with six previously characterized compounds(2–7)were identified.Compound 1 belongs to 4-oxoquinoline with C-2 reversed prenylation and a succinimide substructure.All the structures of these newly isolated compounds were determined by different means in spectroscopic experiments.The absolute configurations of 1 was further deduced from comparison of its CD spectrum with that of known compound 2.The bioactivities of these identified compounds were evaluated against several pathogenic microorganisms and cancer cell lines.Compounds 1–5 showed activity against HUH-7 human hepatoma cells with IC50 values of 80μg/mL.Compound 6 showed mild activity against HeLa cells(IC50=51.9μg/mL),weak anti-MTB activity(MIC=80μg/mL),and moderate anti-MRSA activity(MIC=25μg/mL),and compound 7 showed weak anti-MRSA activity(MIC=100μg/mL).

文章基于渗出砂岩型铀矿成矿机理和模式,认为渗出砂岩型铀成矿作用不同于渗入砂岩型铀矿成矿作用,其成矿预测和找矿标志也不同;提出了渗出砂岩型铀矿成矿作用两大基本关键条件识别标志:红杂色含矿建造原生成因和其中控矿灰色砂体的后生成因识别;在提出的红杂色沉积建造中渗出砂岩型铀矿“上红下黑、上下连通、红中找灰、灰中找矿”总体找矿新思路基础上,阐明渗出砂岩型铀成矿区域预测评价条件和标志:深部富铀富有机质沉积岩建造、区域构造、区域建造、放射性异常信息和综合预测标志等,提出并阐述“小凹陷成大矿”条件;系统建立渗出砂岩型铀矿床预测定位标志体系,特别是野外可识别的宏观标志,包括控矿构造、沉积建造、蚀变改造、铀矿化砂岩颜色、外来有机质特征等,并对比了渗入和渗出砂岩型铀成矿预测标志。提出的渗出砂岩型铀矿识别标志体系不仅对区分“渗入”和“渗出”两种矿化成因,而且对厘清控矿要素、指导成矿预测和找矿工程部署具有重要意义和价值。

Perovskite solar cells(PSCs)emerging as a promising photovoltaic technology with high efficiency and low manufacturing cost have attracted the attention from all over the world.Both the efficiency and stability of PSCs have increased steadily in recent years,and the research on reducing lead leakage and developing eco-friendly lead-free perovskites pushes forward the commercialization of PSCs step by step.This review summarizes the main progress of PSCs in 2020 and 2021 from the aspects of efficiency,stability,perovskite-based tandem devices,and lead-free PSCs.Moreover,a brief discussion on the development of PSC modules and its challenges toward practical application is provided.

手术治疗是口腔颌面部肿瘤的主要治疗手段,吞咽障碍是患者术后的常见并发症。吞咽障碍不但可导致误吸、营养不良、吸入性肺炎等严重后果,还可能引发心理问题和社会交往障碍,影响患者的生活质量。目前国内针对口腔颌面部肿瘤术后合并吞咽障碍的问题尚缺乏一个系统的评估和康复治疗管理方案。在前期临床实践的基础上,结合口腔颌面肿瘤患者术后吞咽障碍的特点及总结颌面肿瘤外科和康复领域专家的临床经验,回顾和总结国内外相关文献,组织相关专家经共同讨论,从吞咽障碍的筛查、评估和康复治疗管理的实施方案等核心内容方面达成共识,以期为临床相关工作提供参考。

Low temperature is a major environmental factor that limits plant growth and productivity.Although transient elevation of cytoplasmic calcium has long been recognized as a critical signal for plant cold tolerance,the calcium channels responsible for this process have remained largely elusive.Here we report that OsCNGC9,a cyclic nucleotide-gated channel,positively regulates chilling tolerance by mediating cytoplasmic calcium elevation in rice(Oryza sativa).We showed that the loss-of-function mutant of OsCNGC9 is defective in cold-induced calcium influx and more sensitive to prolonged cold treatment,whereas OsCNGC9 overexpression confers enhanced cold tolerance.Mechanistically,we demonstrated that in response to chilling stress,OsSAPK8,a homolog of Arabidopsis thaliana OST1,phosphorylates and activates OsCNGC9 to trigger Ca2+influx.Moreover,we found that the transcription of OsCNGC9 is activated by a rice dehydration-responsive element-binding transcription factor,OsDREB1A.Taken together,our results suggest that OsCNGC9 enhances chilling tolerance in rice through regulating cold-induced calcium influx and cytoplasmic calcium elevation.
