Mechanical behavior and deformation mechanism of shape memory bulk metallic glass composites synthesized by powder metallurgy

The synthesis of martensitic or shape-memory bulk metallic glass composites(BMGCs)via solidification of the glass-forming melts requires the meticulous selection of the chemical composition and the proper choice of the processing parameters in order to ensure that the glassy matrix coexists with the desired amount of austenitic phase.Unfortunately,a relatively limited number of such systems,where austenite and glassy matrix coexist over a wide range of compositions,is available.Here,we study the effective-ness of powder metallurgy as an alternative to solidification for the synthesis of shape memory BMGCs.Zr_(48)Cu_(36)Al_(8)Ag_(8)matrix composites with different volume fractions of Ni_(50.6)Ti_(49.4)are fabricated using hot pressing and their microstructure,mechanical properties and deformation mechanism are investigated employing experiments and simulations.The results demonstrate that shape-memory BMGCs with tun-able microstructures and properties can be synthesized by hot pressing.The phase stability of the glass and austenitic components across a wide range of compositions allows us to examine fundamental as-pects in the field of shape memory BMGCs,including the effect of the confining stress on the martensitic transformation exerted by the glassy matrix,the contribution of each phase to the plasticity and the mechanism responsible for shear band formation.The present method gives a virtually infinite choice among the possible combinations of glassy matrices and shape memory phases,expanding the range of accessible shape memory BMGCs to systems where the glassy and austenitic phases do not form simul-taneously using the solidification route.

The demand for industrial enzymes is continually rising,fueled by the growing need to shift towards more sustainable industrial processes.However,making efficient enzyme production strains and identifying optimal enzyme expression conditions remains a challenge.Moreover,the production of the enzymes themselves comes with unavoidable impacts,e.g.,the need to utilize secondary feedstocks.Here,we take a more holistic view of bioprocess development and report an integrative approach that allows us to rapidly identify improved enzyme expression and secretion conditions and make use of cyanobacterial waste biomass as feed for supporting Pichia pastoris fermentation.We demonstrate these capabilities by producing a phytase secreted by P.pastoris that is grown on cyanobacterium hydrolysate and buffered glycerol-complex(BMGY)medium,with genetic expression conditions identified by high-throughput screening of a randomized se-cretion library.When our best-performing strain is grown in a fed-batch fermentation on BMGY,we reach over 7000 U/mL in three days.

Vision is one of our most precious senses,and its impairment has a high socio-economic impact.In the industrialized world,degenerative diseases of the retina lead to vision loss,particularly among the elderly.These degenerations include,for instance,retinitis pigmentosa,age-related macular degeneration,and diabetic retinopathy.Although treatments are evolving to manage late-stage symptoms of retinal degenerations,no effective therapies to recover vision loss exist.Retinal degeneration often involves loss or damage to specialized neural cells,such as photoreceptors,and their death stimulates the activation and proliferation of Müller cells(Salman et al.,2021).

Primarily driven by an increasingly sedentary lifestyle and hypercaloric nutritionally imbalanced diets,non-alcoholic fatty liver disease(NAFLD)has emerged as the most prevalent liver disease in the US and Europe.NAFLD constitutes an umbrella term that includes a spectrum of disease from non-alcoholic fatty liver(NAFL)to non-alcoholic steatohepatitis(NASH),NASH fibrosis and NASH cirrhosis that are all characterized by≥5%of all hepatocytes being steatotic in patients with little alcohol intake and no apparent alternative causes.

Long-term assist device patients admitted to ICU: Tips and pitfalls

Left ventricular assist device(LVAD)therapy is well-established in the treatment of end-stage cardiac failure.Indications are bridge to transplant(BTT),bridge to candidacy(BTC),bridge to recovery(BTR),and destination therapy(DT).The durability and adverse event(AE)rate of LVADs have improved over the years.However,due to donor shortage,the duration of support in the BTT population has increased tremendously;similarly,DT patients are on the device for a long time.Consequently,the number of readmissions of long-term LVAD patients has increased.In cases of severe AEs,intensive care unit(ICU)treatment can be necessary.Infectious complications are the most common AE.Furthermore,embolic or hemorrhagic strokes can occur due to foreign surfaces,acquired von Willebrand syndrome,and anticoagulation treatment.Another consequence of the coagulative status,in combination with the continuous flow,are gastrointestinal bleeding events.Moreover,in most patients,an isolated LVAD is implanted,and this involves the risk of late right heart failure.Adjustment of pump speed and optimization of the volume status can help solve this issue.Malignant arrhythmias,pre-existing or de novo after LVAD implantation,can be a life-threatening AE.Antiarrhythmic medical therapy or ablation are potential treatment options.As for specific LVADs,the Medtronic HeartWare TM ventricular assist device(HVAD)is not manufactured and distributed currently;however,4000 patients are still on the device.Pump thrombosis can occur,wherein thrombolytic therapy is the first-line treatment option.Additionally,the HVAD can fail to restart after controller exchange due to technical issues,and precautions must be taken.The Momentum 3 trial showed superior survival without pump exchange or disabling stroke in patients treated with the HeartMate 3■(HM3;Abbott,Abbott Park,IL,USA)device in comparison to the HeartMate II(HMII).However,in a few cases,a twisted graft or bio debris formation between the outflow graft and bend relief could be observed,causing outflow graft obstr

Corynebacterium glutamicum is a microbial production host established in the industry 60 years ago.It is mainly used for production of feed and food amino acids.As C.glutamicum strain development has been cutting edge since its discovery,it has been engineered for production of a plethora of valuable products.This review will focus on recent developments of C.glutamicum strain engineering for biotransformation and fermentation processes towards flavor and fragrance molecules as well as pigments and sweeteners.

Controlling fluid flow in capillaric circuits is a key requirement to increase their uptake for assay applications.Capillary action off-valves provide such functionality by pushing an occluding bubble into the channel using a difference in capillary pressure.Previously,we utilized the binary switching mode of this structure to develop a powerful set of fundamental fluidic valving operations.In this work,we study the transistor-like qualities of the off-valve and provide evidence that these structures are in fact functionally complementary to electronic junction field effect transistors.In view of this,we propose the new term capillaric field effect transistor to describe these types of valves.To support this conclusion,we present a theoretical description,experimental characterization,and practical application of analog flow resistance control.In addition,we demonstrate that the valves can also be reopened.We show modulation of the flow resistance from fully open to pinch-off,determine the flow rate-trigger channel volume relationship and demonstrate that the latter can be modeled using Shockley's equation for electronic transistors.Finally,we provide a first example of how the valves can be opened and closed repeatedly.

Hepatitis B virus morphogenesis

The hepatitis B virus (HBV) particle consists of an envelope containing three related surface proteins and probably lipid and an icosahedral nucleocapsid of approximately 30 nm diameter enclosing the viral DNA genome and DNA polymerase. The capsid is formed in the cytosol of the infected cell during packaging of an RNA pregenome replication complex by multiple copies of a 21-kDa C protein. The capsid gains the ability to bud during synthesis of the viral DNA genome by reverse transcription of the pregenome in the lumen of the particle. The three envelope proteins S, M, and L shape a complex transmembrane fold at the endoplasmic reticulum, and form disulfide-linked homo- and heterodimers. The transmembrane topology of a fraction of the large envelope protein L changes post- translationally, therefore, the N terminal domain of L (preS) finally appears on both sides of the membrane. During budding at an intracellular membrane, a short linear domain in the cytosolic preS region interacts with binding sites on the capsid surface. The virions are subsequently secreted into the blood. In addition, the surface proteins can bud in the absence of capsids and form subviral lipoprotein particles of 20 nm diameter which are also secreted.

Facile synthesis of ( β -chlorodifluoroethyl)phosphonates via chlorination reaction of difluoroalkyl diazo derivatives with HCl

An efficient chlorination reaction of in situ generated(β-diazo-α,α-difluoroethyl)phosphonates has been achieved with hydrochloric acid as a chlorine source under mild and operationally convenient conditions.The reaction does not need any catalyst and tolerates a wide scope of substrates,which affords the(β-chlorodifluoroethyl)phosphonate products in good to excellent yields.This reaction represents the first example of the halogenation of difluoroalkyl diazo compounds,and also provides an easy way for the synthesis of difluoromethylenephosphonate-containing compounds.

Direct molecular-level near-field plasmon and temperature assessment in a single plasmonic hotspot

Tip-enhanced Raman spectroscopy(TERS)is currently widely recognized as an essential but still emergent technique for exploring the nanoscale.However,our lack of comprehension of crucial parameters still limits its potential as a user-friendly analytical tool.The tip’s surface plasmon resonance,heating due to near-field temperature rise,and spatial resolution are undoubtedly three challenging experimental parameters to unravel.However,they are also the most fundamentally relevant parameters to explore,because they ultimately influence the state of the investigated molecule and consequently the probed signal.Here we propose a straightforward and purely experimental method to access quantitative information of the plasmon resonance and near-field temperature experienced exclusively by the molecules directly contributing to the TERS signal.The detailed near-field optical response,both at the molecular level and as a function of time,is evaluated using standard TERS experimental equipment by simultaneously probing the Stokes and anti-Stokes spectral intensities.Self-assembled 16-mercaptohexadodecanoic acid monolayers covalently bond to an ultra-flat gold surface were used as a demonstrator.Observation of blinking lines in the spectra also provides crucial information on the lateral resolution and indication of atomic-scale thermally induced morphological changes of the tip during the experiment.This study provides access to unprecedented molecular-level information on physical parameters that crucially affect experiments under TERS conditions.The study thereby improves the usability of TERS in day-to-day operation.The obtained information is of central importance for any experimental plasmonic investigation and for the application of TERS in the field of nanoscale thermometry.
