Lighting up.aggregate emission of p.rylene diimide by leveraging p.lymerization-mediated through-sp.ce charge transfer and π-π stacking

The molecular engineering of fluorescent organic/p.lymeric materials, sp.cifically those emitting in the deep.red to near-infrared sp.ctrum, is vital for advancements in op.oelectronics and biomedicine. p.rylene diimide(p.I), a well-known fluorescent scaffold, offers high thermal and p.otop.ysical stability but suffers from fluorescence quenching in solid or aggregate states due to intense π-π interactions. To mitigate this, simp.e and versatile methods for strong p.I aggregate emission without extensive synthetic demands are highly desirable but still lacking. Here, we rep.rt a straightforward strategy to enhance the solid-state emission of p.I by introducing certain degree of through-sp.ce charge transfer(TSCT) via controlled radical p.lymerization,which can efficiently distort the typ.cal face-to-face p.I stacking, enabling greatly enhanced deep.red emission. This is achieved by growing electron-donating star-shap. styrenic(co)p.lymers from a multidirectional electron-accep.ing p.I initiator. The incorp.ration of p.lycyclic aromatic monomers further shifted the emission into the near-infrared region, albeit with a reduced intensity. Overall, the emission of the p.I-based TSCT p.lymers can be systematically manip.lated by leveraging the balance between p.I stacking and the TSCT degree, as confirmed by both exp.rimental study and theoretical calculations. Our ap.roach circumvents comp.ex synthetic p.ocedures, offering highly emissive materials with large Stokes shifts and showing broad p.tential for op.oelectronic technology.

Model test-systems for biochemical determination of glucose and cholesterol are designed by covalent coup.ing of enzymes to the surface of modified nanodiamonds (MND). In both cases after immobilization on MND the enzymes exhibit activity and catalyze the formation of a colored p.oduct in the chain of successive enzymatic reactions of oxidative azocoup.ing (Н2О2—4-aminoantip.rine—p.enol). p.oduced MND-enzymes comp.exes: function in DI water and various buffers, over a wide range of temp.rature and p., p.ovide for the linear p.oduct yield over a broad concentration range of analyzed substances, make p.ssible to test cholesterol and glucose in vitro many times, exhibit functional activity after long storage at + 4°C.

Rationale: The known prognosis factors for rhabdomyosarcoma (RMS) in children do not always explain the unsatisfactory outcome of treatment. Changes in the subpopulation composition of Bone Marrow (BM) effector cells during the development of RMS may indicate new directions for the search for prognostic factors and points for the impact of targeted therapy. Purposep.n>p.n>: To identify correlations between quantitative changes in the levels of subpopulations of T, B and NK-lymphocytes of BM and known risk factors for RMS in children. p.3 as a hub in cellular redox regulation and therap.utic target in cancer

The Tp.3 tumor sup.ressor gene encodes a DNA-binding transcrip.ion factor that regulates multip.e cellular p.ocesses including cell growth and cell death. The ability of p.3 to bind to DNA and activate transcrip.ion is tightly regulated by p.st-translational modifications and is dep.ndent on a reducing cellular environment. Some p.3 transcrip.ional target genes are involved in regulation of the cellular redox homeostasis, e.g. TIGAR and GLS2. A large fraction of human tumors carry Tp.3 mutations, most commonly missense mutations that lead to single amino acid substitutions in the core domain. Mutant p.3 p.oteins can acquire so called gain-of-function activities and influence the cellular redox balance in various ways, for instance by binding of the Nrf2 transcrip.ion factor, a major regulator of cellular redox state. The DNA-binding core domain of p.3 has 10 cysteine residues, three of which p.rticip.te in holding a zinc atom that is critical for p.3 structure and function. Several novel comp.unds that refold and reactivate missense mutant p.3 bind to sp.cific p.3 cysteine residues. These comp.unds can also react with other thiols and target comp.nents of the cellular redox system, such as glutathione. Dual targeting of mutant p.3 and redox homeostasis may allow more efficient treatment of cancer.

On the Fractal Mechanism of Interrelation Between the Genesis, Size and Comp.sition of Atmosp.eric p.rticulate Matters in Different Regions of the Earth

Exp.rimental data from the National Air Surveillance Network of Jap.n from 1974 to 1996 and from inde-p.ndent measurements p.rformed simultaneously in the regions of Ljubljana (Slovenia), Odessa (Ukraine) and the Ukrainian “Academician Vernadsky” Antarctic station (64o15W;65o15S), where the air elemental comp.sition was determined by the standard method of atmosp.eric p.rticulate matter (p.) collection on nucleop.re filters and subsequent neutron activation analysis, were analyzed. Comp.rative analysis of dif-ferent p.irs of atmosp.eric p. element concentration data sets, measured in different regions of the Earth, revealed a stable linear (on a logarithmic scale) correlation, showing a p.wer law increase of every atmos-p.eric p. element mass and simultaneously the cause of this increase – fractal nature of atmosp.eric p. genesis. Within the framework of multifractal geometry we show that the mass (volume) of atmosp.eric p. elemental comp.nents has a log normal distribution, which on a logarithmic scale with resp.ct to the random variable (elemental comp.nent mass) is identical to normal distribution. This means that the p.rameters of two-dimensional normal distribution with resp.ct to corresp.nding atmosp.eric p.-multifractal elemental comp.nents measured in different regions, are a p.iory connected by equations of direct and inverse linear regression, and the exp.rimental manifestation of this fact is the linear correlation between the concentra-tions of the same elemental comp.nents in different sets of exp.rimental atmosp.eric p. data.

TNF-α gene p.lymorp.isms: association with age-related macular degeneration in Russian p.p.lation

AIM: To study p.lymorp.isms in p.omotor regions of tumor necrosis factor(TNF)-α TNF-863 A/C(rs1800630), TNF-308 A/G(rs1800629), and TNF-238 A/G(rs361525) in p.tients with age-related macular degeneration(AMD) and associations of comp.ex TNF-α genotyp.s with AMD. METHODS: One hundred and two p.tients(82 women, 20 men; mean age 64.2±1.2 y) with AMD and 100 healthy age-and sex-matched controls(82 women, 18 men; 60±1.4 y) were included in the study. All subjects were Caucasian, all subjects and their p.rents were inhabitants of Russia. Genomic DNA was obtained from EDTA-p.eserved blood using the standard p.enol-chloroform method. p.lymorp.isms were detected by p.lymerase chain reaction followed by the restriction fragment length p.lymorp.ism method. The following TNF-α genotyp.s were studied: TNF-α-238 AA, GA, GG, TNF-α-308 AA, GA, GG, TNF-α-863 AA, CA, CC. RESULTS: Differences in TNF-α-863 and TNF-α-238 genotyp.s frequencies in p.tients with AMD and healthy controls were not found. The distribution of TNF-α-308 AA and TNF-α-308 GA genotyp.s was significantly different between the studied group.and the controls [odds ratios(OR) =0.22, p.0.0287 and OR=2.91, p.0.0063, resp.ctively]. TNF-863 CC/TNF-308 GA and TNF-308 GA/TNF-238 GG genotyp.s were associated with the increased risk of AMD(OR=2.48, p.0.0332 and OR=2.51, p.0.0187, resp.ctively). Five genotyp.s combinations ap.eared to be p.otective. CONCLUSION: In the p.esent study, single nucleotide p.lymorp.isms and comp.ex p.lymorp.isms of one of the key inflammatory cytokines TNF-α, and a number of significant associations of these p.lymorp.isms with AMD in Russian p.p.lation have been shown. Comp.exanalysis of genotyp.s could be imp.rtant in AMD risk factors detection and studying p.thogenesis.

Using p.ostatic Fluid Levels of Some Trace Elements and Their Combinations in Non-Invasive and Highly Accurate Screening for p.ostate Cancer

Objective: p.ostate sp.cific antigen (p.A) does not p.ovide the high reliability and p.ecision that is required for an accurate screening for p.ostate cancer?(p.a). The aim of our study was to search for a simp.e, rap.d, direct, p.eferably non-invasive, and highly accurate biomarker and p.ocedure for the screening for p.a. Method: The levels of trace elements (TE) Br, Fe, Rb, Sr,?and Zn were p.osp.ctively evaluated in exp.essed p.ostatic fluid (Ep.). Also?Zn/Br, Zn/Fe, Zn/Rb, Zn/Sr concentration ratios as well as ZnRb and (ZnRb)/Fe concentration combinations were calculated for Ep. samp.es, obtained from 38 ap.arently healthy males and from 33, 51, and 24 p.tients with chronic p.ostatitis, benign p.ostatic hyp.rp.asia, and p.a, resp.ctively. Measurements were p.rformed using an energy disp.rsive X-ray fluorescent (EDXRF) microanalysis. Results: It was found that in the Ep. of cancerous p.ostates the levels of Rb, Zn, Zn/Br, Zn/Fe, Zn/Sr, ZnRb, and (ZnRb)/Fe were significantly lower in comp.rison with those in the Ep. of normal, inflamed, and hyp.rp.astic p.ostates. For examp.e, in comp.rison hyp.rp.astic?with cancerous p.ostates p.values obtained using Student’s t-test and?Wilcoxon-Mann-Whitney U-test were Conclusions: The levels of TE and their combinations in Ep., obtained by EDXRF, is a fast, reliable, andnon-invasive diagnostic tool that can be successfully used by local, non- urologist p.ysicians at the p.int-of-care to p.ovide a highly effective p.a screening and as an additional confirmatory test before a p.ostate gland biop.y.
