BACKGROUND Endoscopic drainage remains the treatment of choice for unresectable or inoperable malignant distal biliary obstruction(mDBO).AIm To compare the safety and efficacy of plastic stent(PS)vs self-expanding metal stent(SEmS)placement for treatment of *** This meta-analysis was developed according to Preferred Reporting Items for Systematic Reviews and meta-Analysis guidelines.A comprehensive search was performed in mEDLINE,Cochrane,Embase,Latin American and Caribbean Health Sciences Literature,and grey literature to identify randomized clinical trials(RCTs)comparing clinical success,adverse events,stent dysfunction rate,reintervention rate,duration of stent patency,and mean *** difference(RD)and mean difference(mD)were calculated and heterogeneity was assessed with I2 *** analyses were performed by SEmS *** Twelve RCTs were included in this study,totaling 1005 *** was no difference in clinical success(RD=-0.03,95%confidence interval[CI]:-0.01,0.07;I 2=0%),rate of adverse events(RD=-0.03,95%CI:-0.10,0.03;I2=57%),and mean patient survival(mD=-0.63,95%CI:-18.07,19.33;I2=54%)between SEmS vs PS ***,SEmS placement was associated with a lower rate of reintervention(RD=-0.34,95%CI:-0.46,-0.22;I2=57%)and longer duration of stent patency(mD=125.77 d,95%CI:77.5,174.01).Subgroup analyses revealed both covered and uncovered SEmS improved stent patency compared to PS(RD=152.25,95%CI:37.42,267.07;I2=98%and RD=101.5,95%CI:38.91,164.09;I2=98%;respectively).Stent dysfunction was higher in the covered SEmS group(RD=-0.21,95%CI:-0.32,-0.1;I²=205%),with no difference in the uncovered SEmS group(RD=-0.08,95%CI:-0.56,0.39;I²=87%).CONCLUSION While both stent types possessed a similar clinical success rate,complication rate,and patient-associated mean survival for treatment of mDBO,SEmS were associated with a longer duration of stent patency compared to PS.

N-Acetyltransferase 2 genetic polymorphisms and risk of colorectal cancer

AIm:To investigate the possible association between meat intake,cigarette smoking and N-acetyltransferase 2 (NAT2) genetic polymorphisms on colorectal cancer (CRC) ***:Patients with CRC were matched for gender and age to healthy *** intake and cigarette smoking were assessed using a specific frequency *** was extracted from peripheral blood and the genotypes of the polymorphism were assessed by polymerase chain reaction-restriction fragment length *** NAT2 alleles were studied (WT,m1,m2,m3 and m4) using specific digestion ***:A total of 147 patients with colorectal cancer (76 women and 90 men with colon cancer) and 212 controls were *** mean age of the two groups was 62 *** than half the subjects (59.8% in the case group and 51.9% in the control group) were NAT2 slow *** odds ratio for colorectal cancer was 1.38 (95% CI:0.90-2.12) in slow *** the number of women was small (n=76 in the case group),the cancer risk was found to be lower in intermediate (W/mx) acetylators [odds ratio (OR):0.55,95% confidence interval (95% CI):0.29-1.02].This difference was not observed in men (OR:0.56,95% CI:0.16-2.00).Among NAT2 fast acetylators (W/W or W/mx),meat consumption more than 3 times a week increased the risk of colorectal cancer (OR:2.05,95% CI:1.01-4.16).In contrast,cigarette smoking increased the risk of CRC among slow acetylators (OR:1.97,95% CI:1.02-3.79).CONCLUSION:The risk of CRC was higher among fast acetylators who reported a higher meat *** NAT2 acetylation was associated with an increased risk of CRC.

Dissolution method for delapril and manidipine combination tablets based on an absorption profile of manidipine

The present study describes the development and validation of a dissolution method for delapril （DEL） and manidipine （mAN） combination tablets, using a simulated absorption profile based on in vivo data for mAN. The suitable in vitro dissolution profile for this formulation was obtained using 900 mL of citrate buffer pH 3.2 at 37 ℃ ± 0.5 ℃ as dissolution medium and USP apparatus 2 （paddle） at 75 rpm. All samples were analyzed by a liquid chromatography （LC） method. Under these conditions, a significant linear relationship between the absorbed （calculated by deconvolution approach） and dissolved fractions of mAN was obtained （R=0.997） and an in vivo-in vitro （IVIV） correlation for this particular formulation containing mAN can be established. Validation parameters for dissolution methodology such as the specificity, linearity, accuracy and precision were also evaluated according to the international guidelines, giving results within the acceptable range. Therefore, the proposed dissolution conditions can be applied for the simultaneous release analysis of DEL and mAN from the solid dosage form, contributing to the improvement of the quality control of pharmaceutics and minimizing the number of bioavailability studies.

Formation of DNA Strand Breaks in Peripheral Lymphocytes of Rats After Exposure to Natural Sunlight

Objective This paper aims to evaluate the genotoxicity in peripheral blood lymphocytes of rats after exposure to sunlight at different time points of day in a tropical region of Brazil（5oS,42oW）.materials and methods Thirty Wistar-Hannover rats,three months old,were randomly divided into three groups of 10 animals each：Group I [control,without exposure to ultraviolet（UV） radiation],Group II（exposed to sunlight during 08：00 *** 10：00 a.m.）,and Group III（exposed to sunlight during 10：00 *** 12：00 a.m.）.After a week of exposure,peripheral blood samples were taken from the tail of these animals to prepare smears on two slides per *** 24 h after exposure to sunlight in Group III,a new collection was obtained to observe the repair *** alkaline comet assay was used in this study to evaluate the genotoxic activity of sunlight（P0.05）.Results There was no statistical difference between Group I and II（P=0.672）.On the other hand,the exposure to sunlight in Group III showed genotoxic action in comparison to the other groups（P0.0001）.Also,there was no significant repair in Group III R（P=0.407）.Conclusion This study has shown a genotoxic potential of sunlight（UVA-B） in lymphocytes of mammals from 10：00 *** 12：00 a.m.,due to a higher intensity of UV in this tropical region.

The existence of multigenic families in the mevalonate pathway suggests divergent functional roles for pathway components involved in the biosynthesis of plant sterols. Squalene epoxidases （SQEs） are key components of this pathway, and Squalene Epoxidase 1 （SQE1） has been identified as a fundamental enzyme in this biosynthetic step. In the present work, we extended the characterization of the remaining SQE family members, phylogenetically resolving between true SQEs and a subfamily of SQE-like proteins that is exclusive to Brassicaceae. Functional characterization of true SQE family members, Squalene Epox- idase 2 （SQE2） and Squalene Epoxidase 3 （SQE3）, indicates that SQE3, but not SQE2, contributes to the bulk SQE activity in Arabidopsis, with sqe3-1 mutants accumulating squalene and displaying sensitivity to ter- binafine. We genetically demonstrated that SQE3 seems to play a particularly significant role in embryo development. Also, SQE1 and SQE3 both localize in the endoplasmic reticulum, and SQE3 can functionally complement SQEI. Thus, SQE1 and SQE3 seem to be two functionally unequal redundant genes in the pro- motion of plant SQE activity in Arabidopsis.

BACKGROUND metabolic dysfunction-associated fatty liver disease corresponds to a clinical entity that affects liver function triggered by the accumulation of fat in the liver and is linked with metabolic *** To evaluate the effects of the intragastric balloon(IGB)in patients with metabolic dysfunction-associated fatty liver disease through the assessment of liver enzymes,imaging and several metabolic *** A comprehensive search was done of multiple electronic databases(mEDLINE,EmBASE,LILACS,Cochrane and Google Scholar)and grey literature from their inception until February *** criteria involved patients with a body mass index>25 kg/m2 with evidence or previous diagnosis of hepatic *** analyzed before and after 6 mo of IGB removal were alanine aminotransferase(IU/L),gamma-glutamyltransferase(IU/L),glycated hemoglobin(%),triglycerides(mg/dL),systolic blood pressure(mmHg),homeostatic model assessment,abdominal circumference(cm),body mass index(kg/m2)and liver volume(cm3).RESULTS Ten retrospective cohort studies evaluating a total of 508 patients were *** 6 mo of IGB placement,this significantly reduced alanine aminotransferase[mean difference(mD):10.2,95%confidence interval(CI):8.12-12.3],gammaglutamyltransferase(mD:9.41,95%CI:6.94-11.88),glycated hemoglobin(mD:0.17%,95%CI:0.03-0.31),triglycerides(mD:38.58,95%CI:26.65-50.51),systolic pressure(mD:7.27,95%CI:4.79-9.76),homeostatic model assessment(mD:2.23%,95%CI:1.41-3.04),abdominal circumference(mD:12.12,95%CI:9.82-14.41)and body mass index(mD:5.07,95%CI:4.21-5.94).CONCLUSION IGB placement showed significant efficacy in improving alanine aminotransferase and gamma-glutamyltransferase levels in patients with metabolic dysfunctionassociated fatty liver disease as well as improving metabolic markers related to disease progression.

Simultaneous Determination of Ten Endocrine Hormone Disrupters in Water Using SPE/LC-(ESI)mS-mS

The aim of this study was developed and validated an analytical method based on liquid chromatography and tandem mass spectrometry after solid phase extraction to monitorizing ten endocrine hormone disrupters in Lisbon drinking water system. Natural and synthetic hormones (17-β-estradiol, ethinylestradiol, estriol, estrone, progesterone, mestranol and diethylstilbestrol) and some industrial products (4-n-nonylphenol, 4-tert-octylphenol and bisphenol A) were studied. mass spectrometer detection parameters were optimized, such as the best conditions for the precursor ion formation, namely cone voltage, when applying negative and positive electrospray ionization, and also collision energy for mRm1 and mRm2 transitions. The best conditions of the solid phase extraction (SPE) using Waters Oasis HLB (6 mL, 200 mg) and Isolute C18 (EC) (6 ml, 1000 mg) were also optimized. The method was validated through the application of several statistical tests and the uncertainty estimation of the analytical assay. This method showed a very good linear range for all the studied analytes with determination coefficients (r2) between 0.9962 and 0.9999 and coefficients of variation lower than 4%. There were no significant differences between recoveries obtained with the studied matrices, like groundwater, surface water and water for human consumption. In these matrices, the recovery values varied between 32 and 95%. The limits of method detection were between 0.28 and 22 ng/L. The validated method was applied for the analysis of water samples from the EPAL (Empresa Portuguesa das águas Livres, S.A.) water supply system including tap water, spring water, groundwater, and river water. Some target compounds (bisphenol A, progesterone, 4-tert-octylphenol, and 4-n-nonylphenol) were found in trace amounts in analysed waters.

Influence of Agitation and Aeration on Single Cell Oil Production by Rhodotorula glutinis from Glycerol

In the present study, the yeast Rhodotorula glutinis has been assessed with the aim of producing microbial lipids from glycerol under different aeration conditions. For such a purpose, experiments were carried out in shake flasks, under different conditions of agitation (150 to 250 rpm) and aeration (2.5 to 5.0 of flask volume-to-medium volume ratio). Furthermore, their influence on fermentative parameters (lipid and cell concentration, biomass yield;lipid yield;and lipid volumetric productivity) has been investigated using a 22 full factorial design. The statistical analysis has revealed a strong influence of both variables on substrate consumption, lipid accumulation, cell growth and lipid productivity. As a whole, results suggest that higher aeration levels provide greater cell and lipid concentrations, and lipid volumetric productivity. The best results (4.5 g/L of lipids and QP = 0.95 g/L⋅day) were achieved at the highest aeration (5.0 flask volume-to-medium volume ratio) and agitation (250 rpm) levels. Their fatty acid profile showed that oleic acid was produced in greater quantity (53.5%), followed by linoleic acid (18.7%), palmitic acid (6.8%) and stearic acid (9.9%). The microbial oil presented viscosity of 39.3 cP at 50°C and free fatty acid content of 1.93% ± 0.08%. These are significant results and contribute to establishing operational conditions that maximize single-cell oil production from glycerol by Rhodotorula glutinis, i.e. an alternative source as renewable raw material for lipid-based biorefineries.
