Chemical composition, antiparasitic and cytotoxic activities of aqueous extracts of Ziziphus joazeiro Mart

Objective: To compare the in vitro antiparasitic activity of aqueous extracts from Ziziphus joazeiro leaves and stem bark against Trypanosoma cruzi, Leishmania braziliensis, and Leishmania infantum, as well as to evaluate its cytotoxicity in mammalian cells, in addition to identifying the chemical composition of the extracts. Methods: Ziziphus joazeiro leaf and stem bark aqueous extracts were prepared by cold extraction maceration and subjected to ultra-efficient liquid chromatography coupled to a quadrupole/time of flight system. The susceptibility assays used Trypanosoma cruzi CLB5 strains and promastigote forms of Leishmania braziliensis and Leishmania infantum for antiparasitic activity of the extracts. Moreover, mammalian fibroblasts NCTC clone 929 were used for cytotoxicity analysis. Results: Terpenoid compounds, flavonoids and phenolic acid were identified in extracts. The stem bark aqueous extracts presented more significant results in terms of antiparasitic activity compared with the leaf aqueous extracts, especially against Leishmania braziliensis and Leishmania infantum promastigote forms with an IC50 d moderate toxicity of the stem bark aqueous extracts, which is relevant information for the rational use of this plant part since it is widely used by the population. Conclusions: These preliminary results may contribute to the formulation of new therapeutic agents against this group of neglected diseases, so further investigations are required to delineate the mechanisms of action mainly of the aqueous extract of stem bark of Ziziphus joazeiro.

Electromagnetic Stimulation Combined with Aloe vera Increases Collagen Reorganization in Burn Repair

Objectives： Burns are shown as a clinical problem for their severity and multiple complications due to the time required to heal. Therapies that improve their healing are of great importance, especially for being minimally invasive, of low cost and best performance, all related to the speed and quality of healing. This study investigated the effects of the magnetic electro stimulator Haihuá Cd9 isolated or in association with Aloe vera in rats skin burns. Methods： Experimental groups （n = 30/group） were：（C） Carbopol gel; （F） A. vera/Carbopol gel; （H） Haihuá＋Carbopol gel; （H＋F） Haihuá＋A. vera/Carbopol gel. Samples were collected on the 7th, 14th, and 21st experimental days for structural and morphometric analysis, hydroxyproline and glycosaminoglycans quantification, zymography for MMP-2 and MMP-9 and Western Blotting for TGF-β1, VEGF, Collagen I and III. Key findings： The expression of TGF-β1 in H＋F was increased on the 7th day and of MMP-9 on the 7th and 14th days. The expression of VEGF increased in the first experimental periods and decreased in the last for the treated groups. There was an increase in the fibroblasts and birefringent collagen fibers in groups treated with Haihuá isolated or in association with A. vera in all periods. The quantification of collagen I increased, while collagen III decreased in H＋F. The higher amount of GAGs and MMP-2 active isoform was detected in H and H＋F during all periods. Conclusions： Considering the results of the present study, electromagnetic stimulation in association with the A. vera extract promoted an increase in the number of fibroblasts, GAGs content, MMP-2 activity, the deposition and organization of collagen fibers, favoring the repair of injuries to second degree burns, and may also present therapeutic potential in this injury type.

Water Use in Sugar and Ethanol Industry in the State of Sao Paulo(Southeast Brazil)

Brazil is the largest producer of ethanol from sugarcane in the world. While the ethanol industry is economically important to Brazil for several reasons, it also has a significant impact on the environment. Here we analyze the water consumptive use in the transformation of the feedstock (sugarcane) into ethanol and the impact of industrial byproduct effluents on water resources of the state of Sao Paulo, Brazil. Our estimates indicated that in the 2007-2008 harvest, 700 million m3 was withdrawn mainly from rivers and streams by 140 mills, and of this total 440 million m3 was consumed which yielded a water use of approximately 1.53 m3 ·water·tonˉ1 sugarcane or approximately 18 L·water·L-ˉ1 ethanol. At the same time, a total of 120 million m3 of vinasse by-product was produced in the state, equivalent to an organic load of approximately 3 billion kg·BOd during the harvest season or approximately 8 million kg·BOd·dˉ1. Although the water used by sugarcane mills has decreased in recent decades, it is still possible to further decrease the amount of water used by ethanol production. This would decrease the pressure on 1st order streams of the state from which most water is withdrawn. In addition, the enormous volume of vinasse production must be reduced because it exerts constant pressure on aquatic ecosystems, soil and groundwater due to the constant increase in the potassium (K) concentration in areas where it is used as a fertilizer.

Background: dengue is a Neglected tropical disease (NTds) with high incidence in Brazil. This disease is caused by dengue virus and is transmitted by Aedes aegypti mosquito. The search for new approaches for controlling of this disease is the subject of numerous studies. The aaNAT is a key enzyme in the metabolism of A. aegypti and is crucial in the sclerotization process, as well as regulation of circadian rhythm and inactivation of neurotransmitters. Computational techniques applied to studies of biological systems become an effective weapon in the mapping and management of 3d data structures, giving direction and guidance of potential ligands that can form stable complexes with targets of interest, using a Molecular docking approach. The present study was conducted by a virtual screening, followed by docking calculations, in order to find molecules that could inhibit aaNAT. In this study, we used available compounds in SAM database (Bioinformatics and Medicinal Chemistry Laboratory—Southwest Bahia State University, Jequié-Bahia, Brazil), PubChem and ZINC. Results: The result of dockings with selected ligands showed good energy affinities, presenting potential inhibitory interactions with the enzyme active site. Conclusions: The Coa-S-acetyl-tryptamine and 3-indoleacriloil-coenzyme-A showed the same binding energies -8.9 Kcal/Mol and were described as possible inhibitors of aaNAT.

The compositions of essential oils from Chenopodium ambrosioides L. and Philodendron bipinnatifidum Schott were determined, and the their potential effects on the nutrition and mortality of diabrotica speciosa were studied. The extraction of the oils was performed by hydrodistillation (2 h) using a modified Clevenger apparatus and the oils were subsequently subjected to analysis by gas chromatography/flame ionization detector (CG/FId) and gas chromatography/mass spectrometry (GC-MS). A completely randomized design with five treatments and four replications was adopted. The bean plants were sprayed with solutions of the oils dissolved in aqueous Tween 80 solutions at concentrations of 0 (water + Tween 80), 0.5%, 1.0%, 1.5% and 2.0% and then furnished to the insects with no choice available. Seven days after the application, the percentage of leaves with injury, degree of defoliation, the preference index for consumption and the percent of mortality of insects were evaluated. Neither of the essential oils caused a reduction in foliar injury, but antifeeding activity was observed, causing reduced feeding and increasing the mortality of adult d. speciosa insects.

The aim of this paper is to present the linear generator （Stirling solar dish） in the context of a HSSB （hybrid system solar biomass）, project P ＆ d （research and development P ＆ d 0041, in cooperation and partnership with CPFL--LIght and Force Paulista Company-Campinas, Piratininga, S.P. Brazil）. The other components of the system will be the solar ORC （organic Rankine cycle）, the rotary Stifling and the biomass gasifier. The integration of the complete system will be described in the paper, and is projected to be hydraulic one.

Prevalence of Low Levels of Vitamin d in Type 2 diabetes at the City of Mangueirinha, Paraná, Southern Brazil

Background: diabetes mellitus is a worldwide epidemic. In 2002 there were 173 million diabetic adults worldwide, and these numbers are expected to reach up to 300 million people by 2030. Meanwhile, vitamin d deficiency has its worldwide prevalence directly influenced by factors as solar radiation, skin color, latitude and seasons, cultural habits of populations such as clothing and food, and these factors are important to explain the different prevalences of vitamin d deficiency in the world. Methods: A prospective cross-sectional cohort study was conducted with patients in the outpatient clinic of the Health Unit of the city of Mangueirinha, Paraná, Southern Brazil. Fifty-four type 2 diabetic patients were evaluated (38 women and 16 men), aged 55.8 ± 12.6 years. The following variables were evaluated: age, ethnicity, presence of type 2 diabetes (dM2), hypertension, dyslipidemia, weight, BMI, WC, blood pressure, blood glucose, glycated hemoglobin, ionized calcium, PTH, 25-OH Vit, total cholesterol, HdL, LdL, urea, creatinine, uric acid and red cell/hematocrit. Results: mean BMI was 30.2 ± 4.4 kg/m2, indicating class 1 obesity in this population. Fasting glucose levels were approximately 169.8 ± 74.5 mg/dL. The 25-OH vitamin d values for this population were 23.4 ± 8.3 ng/mL, and 13% of them showed 25-OH vitamin d levels above 30 mg/dL. Fifty percent of those patients had vitamin d levels lower than 30 mg/dL, and 37% had less than 20 mg/dL. Conclusions: this study suggests that vitamin d is associated with low levels of vitamin d in type 2 diabetic patients. Supplementation of vitamin d should be considered in diabetic patients, when levels under 30 mg/mL are found.

背景：在美国降脂药物已获广泛应用。但是对各种降脂药物发生肌溶解症的危险性目前尚缺乏可靠的估计。目的：对门诊情况下，不同他汀及贝特类药物（单用或联用）治疗病人肌溶解症的发生率进行估计。设计、地点及病人：根据全美11个卫生保健计划申报数据建立不同他汀和贝特类用药起始队列。人选本队列的病例为1998年1月1H至2001年6月30日人组保健计划前至少用药180天的病人。人-时间按照单药治疗或他汀-贝特类联合治疗分类。主要结局指标：每治疗10000人一年肌溶解症的发生率、所需治疗例数以及发生肌溶解症的相对危险性。结果：在252460例采用降脂药物治疗的病人中，24例于治疗期间发生住院的肌溶解症。采用阿托伐他汀、普伐他汀或辛伐他汀单药治疗平均每10000人一年肌溶解症的发生率为0．44（95％可信区间[confidence interval，CI]，0．20～0．84）；西立伐汀为5．34（95％CI，1．46～13．68）；贝特类为2．82（95％CI，0．58—8．24；P=0．056）。未用药者（unexposed person—time）肌溶解症的发生率为0（95％CI，0—0．48，P=0．56）。阿托伐他汀、普伐他汀与一种贝特联川肌溶解症的发生率增至5．98（95％CI，0．72～216．0），西立伐他汀与叭特类联用增至1035（95％CI，389—2117）。每治疗1年，发现1例肌溶解症的所需治疗例数他汀单药治疗为2272例，采用他汀和贝特类联合治疗的年龄较大的糖尿病人为484例，采用西立伐他汀加贝特类治疗的病人为9．7到12．7例。结论：采用阿托伐他汀、普伐他汀及辛伐他汀单药治疗发生肌溶解症的危险相似而且很低；联合使用他汀及贝特类者危险增加，特别是在老年糖尿病病人。西立伐他汀联合贝特类每年治疗10例即可能有1例发生肌溶解症。

The essential oils from Mentha viridis L. and Mentha pulegium L. were characterized by gas chromatography coupled to mass spectrometry (GC-MS). These oils were obtained by hydrodistillation and presented linalool (40.70%), carvone (13.52%) and α-terpinene (8.56%) as the principal constituents in the essential oil from Mentha viridis L. Pulegone (50.01%), menthol (31.90%) and menthone (16.56%) were the principal constituents in the essential oil from Mentha pulegium L. These essential oils (in concentrations ranging from 3.91 to 500 μL·mL-1) showed satisfactory activities against Escherichia coli, Listeria monocytogenes, Salmonella choleraesuis and Staphylococcus aureus. The antioxidant activities with 2-deoxyribose and phosphomolybdenum and the reducing power (in concentrations ranging from 0.78 to 1000 μL·mL-1) were determined. The antioxidant activity was observed for the two oils evaluated by the phosphomolybdenum and 2-deoxyribose methods, whereas the essential oil from M. viridis presented low antioxidant activity in the reducing power assay.

Risk factors for osteoporosis in inflammatory bowel disease patients

Inflammatory bowel disease(IBd) patients exhibit higher risk for bone loss than the general population. The chronic inflammation causes a reduction in bone mineral density(BMd), which leads to osteopenia and osteoporosis. This article reviewed each risk factor for osteoporosis in IBd patients. Inflammation is one of the factors that contribute to osteoporosis in IBd patients, and the main system that is involved in bone loss is likely RANK/RANKL/osteoprotegerin. Smoking is a risk factor for bone loss and fractures, and many mechanisms have been proposed to explain this loss. Body composition also interferes in bone metabolism and increasing muscle mass may positively affect BMd. IBd patients frequently use corticosteroids, which stimulates osteoclastogenesis. IBd patients are also associated with vitamin d deficiency, which contributes to bone loss. However, infliximab therapy is associated with improvements in bone metabolism, but it is not clear whether the effects are because of inflammation improvement or infliximab use. Ulcerative colitis patients with proctocolectomy and ileal pouches and Crohn's disease patients with ostomy are also at risk for bone loss, and these patients should be closely monitored.
